Proposals on Basic Data Based on Comparison of Changes in Clinical Laboratory Technologists' National Examination and Job Definition: Focused on Korea, Japan, and Taiwan (임상병리사국가시험 및 직무의 변천 비교를 중심으로 한 기초자료 제안: 한국, 일본, 대만을 중심으로)

  • This study examined the transition process of clinical laboratory technologists' national examination and job definition in Korea and compared the differences in the national examination between Korea and neighboring countries, such as Japan and Taiwan. In Korea, the number of questions made for it was 200 (1965), 200 (1977), 300 (1982), 250 (1992), 330 (2006), and 280 (2015). The practice of clinical physiology is important for real-time monitoring, given the characteristics of physiological testing. On the other hand, there are conflicts between other occupations in the working area. Clinical molecular biology needs to be established as a new major subject considering the diagnostic importance of molecular biological tests and the speed of science and technology development. Clinical laboratory operations provide policy and guidance recommendations to technologist staff. The proposed clinical laboratory technologists' national examination comprises major subjects: clinical biochemistry, clinical hematology, clinical transfusionology, clinical immunology, clinical microbiology, clinical molecular biology, clinical histology, clinical cytology, clinical physiology, and clinical laboratory operations. In addition, this study proposes the job definition of clinical laboratory technologists, revising various chemical or physiological testing to biomedical or physiological testing required for medical practice.

Intratesticular Injection of Hypertonic Saline: Non-Invasive Alternative Method for Animal Castration Model

  • Previous studies, including our own, have demonstrated that the intratesticular injection of hypertonic saline (20%) decreased serum testosterone level which was similar to the surgical castration in the rat, showing the state of chemical castration. In the present study, we further verify the efficacy of this less invasive method as an alternative of surgical orchidectomy in the andrological field. Sterilized 20% saline was directly injected into the adult male rats (750 ${\mu}l$ per testis). The tested rats were divided into 3 groups including intact group (intact), orchidectomy group (ORX) and saline injection group (SAL) after bilateral orchidectomy was performed at the same day of injection. All rats were sacrificed at 4 weeks after injection. The reproductive organs (testes, epididymis, seminal vesicles and prostates) were collected and used for DNA and protein pattern analyses. Also, patho-histological studies on the testes were performed. In contrast to the intact group, similar DNA damages of testis and seminal vesicle were appeared in ORX group and SAL group. The DNA degradations seemed to be the results of necrosis rather than apoptosis. In the protein pattern analysis, all the testing tissues exerted similar patterns in the ORX group and the SAL group compared to the those of intact group. Patho-histological studies revealed that severe degenerative changes in testicular seminiferous tubules and massive infiltration of immune cells in SAL group. The present study confirmed that direct injection of hypertonic saline into the testis caused the equivalent biochemical changes in the accessory sex organs as shown in the orchidectomized animals. These results suggest that hypertonic saline injection model could be a useful castration model which can substitute for surgical castration when its safety is secured through further study in the future.

Study on the Change of Physical Properties in Polyurethane Foam by NCO index at the Aging Condition (NCO index에 따른 폴리우레탄 폼의 노화 물성변화 연구)

  • Polyurethane foams were synthesized with different contents and kinds of catalysts to know change of properties under various NCO index. UTM(universal testing machine), DSC(differential scanning calorimetry), SEM(scanning electron microscope) and FT-IR(Fourier transform spectroscopy) were used for studying the PUF's physical properties change. Compressive strength of PUF increased with increasing contents of catalyst. Glass transition temperature(Tg) and compressive strength of PUF using PC-8 and 33LV catalyst, increased with increasing NCO index at the aging. According to the results of Infrared spectral analysis, reduction of NCO peak was found in gelling catalyst, because unreacted NCO reacted with polyurethane. Although Tg and compressive strength of PUF using TMR-2, unchanged with increasing NCO index at the aging, because trimerization of isocyanate.

Development and Validation of Primary Method for the Determination of Glucose in Human Serum by Isotope Dilution Liquid Chromatography Tandem Mass Spectrometry and Comparison with Field Methods

  • Glucose is a common medical analyte measuring in human serum or blood samples. The development of a primary method is necessary for the establishment of traceability in measurements. We have developed an isotope dilution liquid chromatography tandem mass spectrometry as a primary method for the measurement of glucose in human serum. Glucose and glucose-$^{13}C_6$ in sample were ionized in ESI negative mode and monitored at mass transfers of m/z 179/89 and 185/92 in MRM, respectively. Glucose was separated on $NH_2P$-50 2D column, and the mobile phase was 20 mM $NH_4OAc$ in 30% acetonitrile/70% water. Verification of this method was performed by the comparison with NIST SRMs. Our results agreed well with the SRM values. We have developed two levels of glucose serum certified reference material using this method and distributed them to the clinical laboratories in Korea as samples for proficiency testings. The expended uncertainty was about 1.2% on 95% confidence level. In proficiency testings, the results obtained from the clinical laboratories showed about 3.6% and 3.9% RSD to the certified values. Primary method can provide the traceability to the field laboratories through proficiency testings or certified reference materials.

  • Four component $Ag_2S$-PbS-$PbHAsO_4-Cu_2S$ and three component $Ag_2S$-PbS-$PbHAsO_4$ electrodes have been prepared and evaluated for the direct measurement of monohydrogen arsenate. The 3.0 : 0.5 : 1.0 : 0.25 (mole ratio, $Ag_2S$:PbS:$PbHAsO_4:Cu_2S$) composition is superior in terms of potentiometric response, stability, rapidity of response and reproducibility. Testing was done over the concentration range $10^{-1}$~$10^{-4}M\;HA_SO_4^{2-}$in 0.1F NH4Ac-NH4OH buffer solution at pH 8.50 with constant ionic strength. Interfering ions were $CN^-,\;I^-,\;S^{-2}$ and $Cl^-$.

  • A number of leukocytes increases when infected by a germ or virus. Detection of leukocyte levels can indicate of such medical informations as urogenital tract infection or other dysfunction. In this study, pentyl-3-thiophene-carboxlyate (PTC), pentyl-8-quinolinecarboxylate (PQC), and 2-Phenyl-4(N-tosyl-alanyloxyl)-thiazole (PTT) were synthesized, and the test strips were prepared with these substrates for quantifying leukocytes in urine. Among these substrates, the PTT test strip prepared in 0.5% borate buffer pH 8.0, 0.03% PTT, 0.1-0.8% PVP, and 1% decanol showed not only better color reaction but also an excellent application possibility to be used in automatic urine analyzer.

  • Poly(o-hydroxyamides)(PHAs) copolymers having oligo(oxy ethylene) pendant in the main chain were synthesized by solution polycondensation reaction at low temperature. Copolymer precursors were studied by fourier transform infrared(FT-IR), differential scanning calorimeter(DSC), thermogravimetric analyzer(TGA), universal testing machine(UTM) and limited oxygen index(LOI). The inherent viscosities of the PHAs measured at $35^{\circ}C$ in DMAc or DMAc/LiCl solution were in the range of 0.74~1.42 dL/g. Solubility of the precursors with higher oligo(oxy ethylene) unit was increased, but the PBOs were nearly insoluble in a variety of solvents. The degradation temperature of the copolymer precursors was recorded in the ranges of $408{\sim}664^{\circ}C$ in nitrogen and char yields showed 13~59% values at $900^{\circ}C$. The mechanical properties and flame retardancy of copolymer precursors decreased with higher oligo(oxy ethylene) unit.

A Study on the Ultrasonic Nondestructive Evaluation of Carbon/Carbon Composite Disks

  • It is desirable to perform nondestructive evaluation (NDE) to assess material properties and part homogeneity because the manufacturing of carbon/carbon brake disks requires complicated and costly processes. In this work several ultrasonic techniques were applied to carbon/carbon brake disks (322mm ad, 135mm id) for the evaluation of spatial variations in material properties that are attributable to the manufacturing process. In a large carbon/carbon disk manufactured by chemical vapor infiltration (CYI) method, the spatial variation of ultrasonic velocity was measured and found to be consistent with the densification behavior in CYI process. Low frequency (e.g., 1-5MHz) through-transmission scans based on both amplitude and time-of-flight of the ultrasonic pulse were used for mapping out the material property inhomogeneity. Images based on both the amplitude and the time-of-flight of the transmitted ultrasonic pulse showed significant variation in the radial direction. The radial variations in ultrasonic velocity and attenuation were attributed to a density variation caused by the more efficient densification of pitch impregnation near the id and od and by the less efficient densification away from the exposed edged of the disk. Ultrasonic velocities in the edges of the disk. Ultrasonic velocities in the thickness direction were also measured as a function of location using dry-coupling transducers ; the results were consistent with the densification behavior. However, velocities in the in-plane directions (circumferential and radial) seemed to be affected more by the relative contents of fabric and chopped fiber, and less by the void content.

Possible Use of NIR Spectroscopy for Soil Testing (토양검정에서 근적외 분광분석기의 이용 가능성)

  • Traditional methods of chemical analysis for the soil properties take time and produce harmful waste. The purpose of this research was to evaluate an NIR technique for measuring some soil properties that are rapid and accurate in soil fertility assessments. The NIR instrument (InfraAlyzer 500, Bran & Luebbe Co.) was used for obtaining spectral data from 140 finely ground soil for calibrations and validation estimating pH, CEC, extractable Ca, Mg, K, $SiO_2$, humic acid and EC. Partial least square regression analysis was used to develop a calibration of NIR spectroscopy method. The results indicated that NIR spectroscopy could be used as a routine nondestructive method quantitatively determining soil chemical properties quickly. However the NIR technique may require sample preparation to obtain even diffuse reflection spectra from the soil and data manipulations to obtain optimal predictions.

Catalytic Effects and Characteristics of Ni-based Catalysts Supported on TiO2-SiO2 Xerogel

  • The catalytic activities of nickel-based catalysts were estimated for oxidizing acetaldehyde of VOCs exhausted from industrial facilities. The catalysts were prepared by sol-gel methods of SiO2 and SiO2-TiO2 as a xerogel followed by impregnating Al2O3 powder with the nickel nitrate precursor. The crystalline structure and catalytic properties for the catalysts were investigated by use of BET surface area, X-ray diffraction (XRD), Xray photoelectron spectroscopy (XPS) and temperature programmed reduction (TPR) techniques. These results show that nickel oxide is transformed to NiAl2O4 spinel structure at the calcination temperature of 400 °C in response to the steps with after- and co-impregnation of Al2O3 powder in sol-gel process. The NiAl2O4 could suppress the oxidation reaction of acetaldehyde by catalysts. The NiO is better dispersed on SiO2-TiO2/Al2O3 support than SiO2/Al2O3 and SiO2-TiO2-Al2O3 supports. From the testing results of catalytic activities for oxidation of acetaldehyde, Catalysts showed a big difference in conversion efficiencies with the way of the preparation of catalysts and the loading weight of nickel. The catalyst of 8 wt.% Ni/TiO2-SiO2/Al2O3 showed the best conversion efficiency on acetaldehyde oxidation with 100% conversion efficiency at 350 °C.
