THE ECOLOGY, PHYTOGEOGRAPHY AND ETHNOBOTANY OF GINSENG

  • Ginseng is the English common name for the species in the genus Panax. This article gives a broad botanical review including the morphological characteristics, ecological amplitude, and the ethnobotanical aspect of the genus Panax. The species of Panax are adapted for life in rich loose soil of partially shaded forest floor with the deciduous trees such as linden, oak, maple, ash, alder, birch, beech, hickory, etc. forming the canopy. Like their associated trees, all ginsengs are deciduous. They require annual climatic changes, plenty of water in summer, and a period of dormancy in winter. The plant body of ginseng consists of an underground rhizome and an aerial shoot. The rhizome has a terminal bud, prominent leafscars and a fleshy root in some species. It is perennial. The aerial shoot is herbaceous and annual. It consists of a single slender stem with a whorl of digitately compound leaves and a terminal umbel bearing fleshy red fruits after flowering. The yearly cycle of death and renascence of the aerial shoot is a natural phenomenon in ginseng. The species of Panax occur in eastern North America and eastern Asia, including the eastern portion of the Himalayan region. Such a bicentric generic distributional pattern indicates a close floristic relationship of the eastern sides of two great continental masses in the northern hemisphere. It is well documented that genera with this type of disjunct distribution are of great antiquity. Many of them have fossil remains in Tertiary deposits. In this respect, the species of Panax may be regarded as living fossils. The distribution of the species, and the center of morphological diversification are explained with maps and other illustrations. Chemical constituents confirm the conclusion derived from morphological characters that eastern Asia is the center of species concentration of Panax. In eastern North America two species occur between longitude $70^{\circ}-97^{\circ}$ Wand latitude $34^{\circ}-47^{\circ}$ N. In eastern Asia the range of the genus extends from longitude $85^{\circ}$ E in Nepal to $140^{\circ}$ E in Japan, and from latitude $22^{\circ}$ N in the hills of Tonkin of North Vietnam to $48^{\circ}$ N in eastern Siberia. The species in eastern North America all have fleshy roots, and many of the species in eastern Asia have creeping stolons with enlarged nodes or stout horizontal rhizomes as storage organs in place of fleshy roots. People living in close harmony with nature in the homeland of various species of Panax have used the stout rhizomes or the fleshy roots of different wild forms of ginseng for medicine since time immemorial. Those who live in the center morphological diversity are specific both in the application of names for the identification of species in their communication and in the use of different roots as remedies to relieve pain, to cure diseases, or to correct physiological disorders. Now, natural resources of wild plants with medicinal virtue are extremely limited. In order to meet the market demand, three species have been intensively cultivated in limited areas. These species are American ginseng (P. quinquefolius) in northeastern United States, ginseng (P. ginseng) in northeastern Asia, particularly in Korea, and Sanchi (P. wangianus) in southwestern China, especially in Yunnan. At present hybridization and selection for better quality, higher yield, and more effective chemical contents have not received due attention in ginseng culture. Proper steps in this direction should be taken immediately, so that our generation may create a richer legacy to hand down to the future. Meanwhile, all wild plants of all species in all lands should be declared as endangered taxa, and they should be protected from further uprooting so that a. fuller gene pool may be conserved for the. genus Panax.

한국산 도롱뇽의 포식압과 서식지에 따른 polyphenism (The different Polyphenism by the Level of Predation Risk and Habitat in Larval Salamander, Hynobius ieechii)

  • 본 연구는 서식지와 포식압에 따라 머리 크기가 서로 다르게 나타나는 polyphenism에 대하여 한국산 도롱뇽 유생을 대상으로 이루어졌다. 인왕산과 수락산, 경기도 광주에서 채집해 온 도롱뇽의 알(난괴)을 서식지 특성과 포식압에 따라 4가지 그룹으로 나누었다. 서식지는 계곡 형과 웅덩이 형 두 그룹으로 나누었고, 포식압은 높음과 포식압 없음, 두 그룹으로 나누었다. 포식압 수준은 하루에 포식자의 cue에 노출된 빈도로 나누었으며 높은 수준은 하루 세 번으로 하였다. 포식 cue는 버들치의 냄새를 추출하여 사용하였고, 알 채집일 다음날부터 부화 후 일주일까지 cue에 노출시켰다. Chemical cue 노출 처리가 끝나는 부화 후 일주일이 되는 날, 각 도롱뇽 유생의 머리 중 가장 넓은 부분과 눈이 있는 부분의 길이, 그리고 snout-vent length를 측정하였다. 눈이 있는 부분의 길이는 머리 중 가장 넓은 부분으로 나눈 수치를 이용하여 통계 처리를 하였으며 결과는 다음과 같다. 도롱뇽 유생의 머리 크기 비율은 포식압의 수준이 높음에 따라 의미 있게 증가하였다. 또한 계곡형 서식지에 사는 도롱뇽 유생의 머리는 버들치의 cue에 반응하여 포식압이 높을 때 머리 크기 비율이 의미 있게 증가하지만, 웅덩이형 서식지에 사는 도롱뇽 유생은 무의미한 결과가 나타났다. 그리고 snout-vent length의 길이는 서식지와 상관없이 높은 포식압에서 증가하는 경향이 나타났다.

35% Hydrogen Peroxide Gel의 미백효과 및 법랑질의 변화 (35% Hydrogen Peroxide Gel in the Whitening Effect and Enamel Changes)

  • 본 연구는 전문가 미백제로 많이 사용되고 있는 35% hydrogen peroxide를 사용하여 인공적으로 변색시킨 법랑질에 노출시킨 후 법랑질의 색, 경도 및 표면변화에 미치는 영향을 평가하여 다음과 같은 결론을 얻었다. 1. 35% HP 미백처리군의 색조변화량(${\Delta}E^*$)은 시간의 경과에 따라 색조 변화를 보였으며 통계적으로도 유의한 차이를 나타냈으며 NBS unit로 환산해본 결과 30분 노출군에서는 appreciable에 해당이 되며 1시간, 2시간 노출군에서 각각 much, very much에 해당이 되어 임상적인 의미를 지니는 것으로 나타났다(p < 0.05). 2. 법랑질 표면의 경도를 분석한 결과 대조군에서는 3주동안 거의 변화를 보이지 않은 반면 미백제 노출군에서는 법랑질 표면경도가 매우 낮게 나타났다. 특히 2시간 노출군에서 각 주마다 큰 변화를 보였으며 통계적으로도 유의한 차이를 나타냈다(P < 0.05). 3. 미백처리 전 후의 법랑질 표면에 대한 미세구조를 분석하기 위해 주사전자현미경을 이용하여 관찰한 결과 미백 후 노출시간이 길수록 법랑질 표면의 불규칙한 표면양상을 관찰할 수 있었으며 EDX를 사용하여 법랑질 표면의 성분 분석을 시행해본 결과 모든 군에서 Ca, P의 전반적인 감소를 볼 수 있었으나 통계적으로는 유의한 차이를 보이지 않았다(P > 0.05). 이상의 결과를 종합해 볼 때 35% HP 치아 미백은 미백노출 시간이 길어질수록 법랑질 표면에 영향을 미쳐 술후 치아의 시림이나 거칠어짐과 같은 부작용을 야기 할수 있으므로 짧은 기간 동안 정확한 방법으로 전문가에 의하여 시행되어야 할 것으로 사료된다.

Isolation of human mesenchymal stem cells from the skin and their neurogenic differentiation in vitro

  • Objectives: This aim of this study was to effectively isolate mesenchymal stem cells (hSMSCs) from human submandibular skin tissues (termed hSMSCs) and evaluate their characteristics. These hSMSCs were then chemically induced to the neuronal lineage and analyzed for their neurogenic characteristics in vitro. Materials and Methods: Submandibular skin tissues were harvested from four adult patients and cultured in stem cell media. Isolated hSMSCs were evaluated for their multipotency and other stem cell characteristics. These cells were differentiated into neuronal cells with a chemical induction protocol. During the neuronal induction of hSMSCs, morphological changes and the expression of neuron-specific proteins (by fluorescence-activated cell sorting [FACS]) were evaluated. Results: The hSMSCs showed plate-adherence, fibroblast-like growth, expression of the stem-cell transcription factors Oct 4 and Nanog, and positive staining for mesenchymal stem cell (MSC) marker proteins (CD29, CD44, CD90, CD105, and vimentin) and a neural precursor marker (nestin). Moreover, the hSMSCs in this study were successfully differentiated into multiple mesenchymal lineages, including osteocytes, adipocytes, and chondrocytes. Neuron-like cell morphology and various neural markers were highly visible six hours after the neuronal induction of hSMSCs, but their neuron-like characteristics disappeared over time (24-48 hrs). Interestingly, when the chemical induction medium was changed to Dulbecco's Modified Eagle Medium (DMEM) supplemented with fetal bovine serum (FBS), the differentiated cells returned to their hSMSC morphology, and their cell number increased. These results indicate that chemically induced neuron-like cells should not be considered true nerve cells. Conclusion: Isolated hSMSCs have MSC characteristics and express a neural precursor marker, suggesting that human skin is a source of stem cells. However, the in vitro chemical neuronal induction of hSMSC does not produce long-lasting nerve cells and more studies are required before their use in nerve-tissue transplants.

폴리케톤 섬유의 산소 플라즈마 처리에 따른 천연고무와의 계면접착 특성 (Interfacial Adhesion Properties of Oxygen Plasma Treated Polyketone Fiber with Natural Rubber)

  • 폴리케톤(polyketone, PK) 섬유는 고무의 강화재로 사용하기 위해서 최근 개발되고 있는 소재이다. 섬유의 고무와의 접착성을 향상시키기 위하여 플라즈마로 표면을 개질하였다. 산소 플라즈마 처리에 의한 섬유표면의 물리적 형태변화를 관찰하기 위하여 주사전자현미경과 원자현미경을 이용하여 관찰하였다. 섬유표면의 화학적 조성변화를 XPS (X-ray photoelectron spectroscopy)를 이용하여 알아보았다. 최종적으로 이러한 변화가 PK 섬유와 고무와의 계면접착력에 어떠한 영향을 미치는지를 microdroplet debonding 시험을 통해 분석하였다. 플라즈마 처리에 의하여 섬유표면에 산소함유기들이 증가하는 결과를 보였으며, 처리시간과 처리전력이 증가함에 따라 에칭에 의한 표면조도(RMS roughness)가 증가하였다. 그러나 장시간의 플라즈마 처리조건에서는 표면에 degradation이 발생하여 오히려 표면조도가 감소하는 결과를 보였다. PK 섬유와 고무와의 계면전단강도는 처리시간 60 s의 80 W, 처리전력 60 W의 180 s에서 처리한 경우에 최대 계면전단강도를 나타내었다. 그러나 그 이상으로 증가하면 degradation이 발생하면서 계면전단강도가 감소하였다.

조팝나무(Spiraea prunifolia var. simpliciflora)로부터 분리한 페놀 화합물의 신경세포 보호효과 (Neuroprotective effects of phenolic compounds isolated from Spiraea prunifolia var. simpliciflora)

  • 조팝나무(Spiraea prunifolia) 잎을 80% MeOH 수용액으로 추출한 뒤, 감압 농축한 추출물을 EtOAc, n-BuOH과 $H_2O$층으로 계통 분획을 실시하였다. n-BuOH분획에 대하여 $SiO_2$, ODS column chromatograph 및 중압분취 장비를 반복실시하여 2종의 phenolic glucoside화합물을 분리 및 정제하였다. NMR 및 Mass데이터를 해석하여, isosalicin (1)와 crenatin (2)로 구조 동정 하였다. 분리한 2종의 화합물에 대하여 UPLC-MS/MS 질량 분석기를 기반으로 다중반응검지 (MRM mode) 분석법을 이용하여 정량분석 하였다. 그 결과, crenatin은 9.53 mg/g 및 isosalicin은 0.65 mg/g이 조팝나무 잎 추출물에 함유된 것을 확인하였다. 조팝나무 잎에서 분리된 화합물이 $H_2O_2$에 의해 유발된 산화스트레스로부터 신경세포 보호효과를 검증한 결과, 신경모세포종인 SK-N-MC 세포에 대해서 화합물 1 및 2 (100 mM) 모두 세포 독성은 보이지 않았으며, 특히 화합물 2 (crenatin)는 $H_2O_2$처리에 따른 신경세포의 외형적 변화, 세포 손상, 세포 사멸을 억제하여 산화스트레스로부터 신경세포를 보호하는 효과가 있음을 확인하였다.

Safety and Effects on Lipid Parameters of Rubus coreanus and Atractylodes japonica in Ovariectomized Rats

  • Defatted methanol extracts of the medicinal plants, Rubus coreanus Miq. (RC) and Atractylodes japonica Koidzumi (AJ) were added at the levels of 0.1, 0.5, or $2\%$ (w/w) to high cholesterol diets and fed to ovariectomized Sprague-Dawley female rats, weighing 212.6 $\pm$ 1.8 g for four weeks. Weight gains were lower in RC and AJ groups than the control group, but there were no changes in uterus weights. Serum levels of triglyceride decreased by 20-$27\%$ in the experimental groups fed $0.1\%$ of each extract (O.1RC and O.1AJ), compared with that of control (Ovx). Serum cholesterol levels were not changed in the RC groups but increased in the group fed $2\%$ of the AJ extract. Liver levels of cholesterol and triglyceride were reduced in both the RC and AJ groups. Microscopic observation revealed that there were no morphological alterations in liver, lung, heart, spleen and kidney tissues of the experimental groups. Plasma levels of albumin, BUN, creatinine, sodium, potassium and phosphate in the IRC and AJ groups were in normal ranges. Serum GOT and GPT activities were, however, higher in the 2.0AJ than Ovx group. These results suggest that the extracts of the Rubus coreanus Miq. and Atractylodes japonica Koidzumi at dietary levels as low as $0.1\%$ may be utilized as hypotriglyceridemic ingredients for functional foods.

Unsaturated Polyester/Polyvinylacetate Semi-IPN의 경화반응속도에 따른 상분리현상 연구 (The Study on Phase Separation Development by Curing Reaction Rate for Unsaturated Polyester/Polyvinylacetate Semi-IPN)

  • 불포화폴리에스터/폴리비닐아세테이트 semi-IPN의 경화과정 중 경화속도와 상분리 확산속도의 상호관계 속에서 형성되는 상분리 모폴로지를 광산란 및 열분석 장치를 이용하여 연구하였다. 열경화성 고분자의 경화과정 중 열가소성 고분자의 확산에 기인한 물성의 변화를 측정하였고, 상분리를 수반하는 경화과정에서의 활성화에너지의 변화를 Flynn-Wall method를 이용하여 구하였다. 반응에 의하여 나타나는 상분리현상은 경화과정 중 다양한 상분리 거동을 나타내게 되는데, 폴리비닐아세테이트가 10 wt%일 경우에는 반응도중에 상분리가 일어나지 않고 냉각 후 상온에서 nucleation & growth 거동과 유사한 형태로 상분리가 발생하였고, 11.65 wt% 이상에서는 반응도중 spinodal decomposition으로 사료되는 상분리가 발생하였다. 또한 상분리현상이 경화속도에 영향을 미친 것을 활성화에너지의 변화거동으로부터 확인하였으며, 온도변화에 따라 총산란량을 측정하였고 이로부터 확산에 의한 상분리 속도 R(${\beta}_m$)를 비교하였다.

Paraquat Induces Apoptosis through a Mitochondria-Dependent Pathway in RAW264.7 Cells

  • Paraquat dichloride (N,N-dimethyl-4-4'-bipiridinium, PQ) is an extremely toxic chemical that is widely used in herbicides. PQ generates reactive oxygen species (ROS) and causes multiple organ failure. In particular, PQ has been reported to be an immunotoxic agrochemical compound. PQ was shown to decrease the number of macrophages in rats and suppress monocyte phagocytic activity in mice. However, the effect of PQ on macrophage cell viability remains unclear. In this study, we evaluated the cytotoxic effect of PQ on the mouse macrophage cell line, RAW264.7 and its possible mechanism of action. RAW264.7 cells were treated with PQ (0, 75, and $150{\mu}M$), and cellular apoptosis, mitochondrial membrane potential (MMP), and intracellular ROS levels were determined. Morphological changes to the cell nucleus and cellular apoptosis were also evaluated by DAPI and Annexin V staining, respectively. In this study, PQ induced apoptotic cell death by dose-dependently decreasing MMP. Additionally, PQ increased the cleaved form of caspase-3, an apoptotic marker. In conclusion, PQ induces apoptosis in RAW264.7 cells through a ROS-mediated mitochondrial pathway. Thus, our study improves our knowledge of PQ-induced toxicity, and may give us a greater understanding of how PQ affects the immune system.

Synthesis and Characterization of Cu(In,Ga)Se2 Nanostructures by Top-down and Bottom-up Approach

  • Nanomaterials have emerged as new building blocks to construct light energy harvesting assemblies. Size dependent properties provide the basis for developing new and effective systems with semiconductor nanoparticles, quantized charging effects in metal nanoparticle or their combinations in 2 and 3 dimensions for expanding the possibility of developing new strategies for photovoltaic system. As top-down approach, we developed a simple and effective method for the large scale formation of self-assembled Cu(In,Ga)$Se_2$ (CIGS) nanostructures by ion beam irradiation. The compositional changes and morphological evolution were observed as a function of the irradiation time. As the ion irradiation time increased, the nano-dots were transformed into a nano-ridge structure due to the difference in the sputtering yields and diffusion rates of each element and the competition between sputtering and diffusion processes during irradiation. As bottom-up approach, we developed the growth of CIGS nanowires using thermal-chemical vapor deposition (CVD) method. Vapor-phase synthesis is probably the most extensively explored approach to the formation of 1D nanostructures such as whiskers, nanorods, and nanowires. However, unlike binary or ternary chalcogenides, the synthesis of quaternary CIGS nanostructures is challenging because of the difficulty in controlling the stoichiometry and phase structure. We introduced a method for synthesis of the single crystalline CIGS nanowires in the form of chalcopyrite using thermal-CVD without catalyst. It was confirmed that the CIGS nanowires are epitaxially grown on a sapphire substrate, having a length ranged from 3 to 100 micrometers and a diameter from 30 to 500 nm.
