카카오(Theobroma cacao L.) 용매 분획물의 항산화 및 항염증 효과 (Anti-Oxidative and Anti-Inflammatory Effects of the Solvent Fraction from Theobroma cacao L. Extract)

  • 본 연구는 카카오를 화장품 소재로 활용하기 위하여 항산화 및 항염증 효과를 검증하여 기능성 화장품 소재로서의 가능성을 검토하였다. 아세톤 추출물을 이용하여 클로로포름층, 에틸아세테이트층, 부탄올층, 물층의 극성별로 분획을 실시하였다. 카카오 분획물의 전자공여능은 100 ${\mu}g/ml$에서 에틸아세테이트 분획물이 76.2%, 부탄올 분획물이 53.9%의 소거능을 나타내었다. 또한, superoxide anion radical 소거능 측정 결과, 에틸아세테이트 분획물과 부탄올 분획물이 50 ${\mu}g/ml$에서 각각 76.09%, 51.4%의 소거능이 나타났고, $Fe^{2+}$, $Cu^{2+}$의 첨가에 따른 지방산패 억제능에서 $Fe^{2+}$를 첨가한 군에서는 에틸아세테이트 분획물이 50 ${\mu}g/ml$에서 64%로 $Cu^{2+}$를 첨가한 군보다 저해능이 높았다. 항염증 효과 측정으로 hyaluronidase 저해 활성을 측정한 결과, 부탄올 분획물의 경우 100 ${\mu}g/ml$에서 53.04%의 저해활성을 나타내었으며, lipoxygenase 저해 활성 측정결과 10 ${\mu}g/ml$에서 51.32%의 저해활성을 나타내었다. 대식세포를 이용한 nitric oxide (NO) 저해활성을 측정한 결과 카카오 에틸아세테이트 분획물에서 가장 높은 NO 저해활성을 나타냄을 확인할 수 있었다. 또한 대식세포 내에서 에틸아세테이트 분획물 100 ${\mu}g/ml$ 농도에서 iNOS, COX-2 단백질 발현을 저해하였다.

추출방법을 달리한 미역 및 다시마 과립차의 품질 및 항산화 특성 (Quality and Antioxidant Characteristics of Granule Tea Prepared with Sea Tangle (Laminaria japonica) and Sea Mustard (Undaria pinnatifida) Powder as Affected by Extraction Method)

  • 본 연구에서는 해조류의 고부가가치 창출을 위하여 기호성과 상품성을 향상시킨 해조 과립차의 품질특성 및 항산화 특성을 비교 분석하였다. 미역, 다시마 미세분쇄분말을 열수 및 고압 추출한 후 동결건조하여 얻은 분말의 수율은 미역, 다시마 모두 열수 추출물에서 높았으며 알긴산 함량은 고압 추출물에서 높았다. 추출방법에 따른 미역, 다시마 과립의 흡습특성 및 알긴산 함량은 고압 추출물에서 높은 함량을 나타내었다. 총 폴리페놀 및 플라보노이드 함량은 유의적인 차이는 있으나 큰 차이는 없었다. 미역 과립차의 전자공여능을 비교분석한 결과 고압추출에서 높은 활성을 나타내는 반면 다시마의 과립차에서는 열수추출에서 다소 높은 활성을 보였다. 미역 과립차의 $ABTS^+$ radical 소거능은 열수추출에서 높은 활성을 보였고, 다시마 과립차의 경우 고압추출에서 높은 활성을 나타내었다. $Fe^{2+}$ chelating효과는 열수추출에서 높은 활성을 보였으며, TBARS에서는 고압추출에서 더 높은 값을 나타내었다. 관능평가에서는 미역 다시마 모두 고압추출에서 높은 기호도를 나타내었다.

  • 추출방법을 달리하여 달맞이꽃 에탄올 추출물의 주요성분과 항산화 및 항균 활성을 측정하였다. 상온교반 추출(SE, stirrer extraction), 초음파 추출(USE, ultrasonification extraction), 환류 추출(RE, reflux extraction), 고온가압추출(AE, autoclave extraction), 저온고압추출(LE, low temperature high pressure extraction)법을 사용한 결과 RE법이 추출수율(46.33%), 총 폴리페놀(463.05 mg GAE/g)과 플라보노이드(71.71 mg RHE/g) 함량이 다른 추출방법 보다 높았다. 달맞이꽃 추출물은 B. cereus에 대한 항균활성이 나타났으며, 추출방법에 따른 차이는 나타나지 않았다. DPPH와 ABTS radical 소거능, 환원력, 철이온 흡착능 시험에서도 RE법을 이용한 추출물이 각각 74.40%, 65.29%, 1.370 ($OD_{700}$), 90.14%를 나타내어 다른 추출방법에 비해 높았다. 달맞이꽃 추출물은 항산화 활성이 우수하여 유용한 천연물 소재로 사용이 가능하며, B. cereus가 주 원인이 되는 식중독의 예방이나 오염된 식품의 보존성 증진을 위한 천연 소재로 할용이 가능할 것으로 판단된다.

아미드옥심기와 인산기가 함유된 이온 교환 섬유의 합성 및 우라늄 흡착 특성 (Synthesis of ion Exchange Fiber Containing Amidoxime and Phosphoric Acid Groups and Its Uranium Adsorption Properties)

  • 전자선 전조사법을 이용하여 아크릴로니트릴 (AN)과 스티렌 (Sty)을 PP섬유에 그라프트반응시컨 PP-g-(AN/Sty) 공중합체를 합성하고, 이어서 아미드옥심화 및 인산화 반응을 수행하였다. 공단량체 중 AN의 양이 증가할수록 공중합체 내의 AN의 몰분율은 선형적으로 증가하였으며 공중합체 내에 그라프트 되어진 AN의 양은 공단량체 중 AN의 조성이 40 vol%에서 최대 45%를 나타내었다. 용매인 메탄올의 양이 증가함에 따라 공중합체 내에 도입되어진 AN의 몰분율은 감소하였다 반응온도에 따른 공중합체의 그라프트율은 반응온도 5$0^{\circ}C$까지 선형적으로 증가하였으며 이후 평형에 도달하였다. 이온 교환 섬유에 도입되어진 아미드옥심기는 하이드록실아민의 양이 증가함에 따라 증가하였으며 하이드록실아민 농도 9 wt%에서 최대 5.8 mmol/g을 나타내었다. 공중합체에 도입되어진 인산기는 인산의 농도가 0.5 N까지 증가하는 경향을 나타내었으며 이후 감소하였다. 우라늄 흡착 실험 결과, 우라늄 흡착량은 이관능성 이온 교환 섬유가 아미드옥심화 이온 교환체 및 인산화 이온 교환체보다 우수한 흡착량을 나타내었다.

Antioxidant activities of Erythrina stricta Roxb.using various in vitro and ex vivo models

  • Erythrina stricta, a deciduous tree widely used traditionally in indigenous system of medicine for various ailments such as rheumatism, fever, leprosy, epilepsy etc. The leaves of Erythrina stricta was extracted with ethanol (70%) and used for the evaluation of various in vitro antioxidant assays which includes H - donor activity, nitric oxide scavenging, superoxide anion scavenging, reducing ability, hydroxyl radical, hydrogen peroxide scavenging, total phenolic content, total flavonoid content, total antioxidant activity by thiocyanate and phosphomolybdenum method, metal chelating, $\beta$-carotene bleaching, total peroxy radical assays. The pro-oxidant activity was measured using bleomycin-dependent DNA damage. Ex vivo models like lipid peroxidation and erythrocyte haemolysis were also used to study the antioxidant property of the extract. The various antioxidant activities were compared with suitable standard antioxidants such as ascorbic acid, butylated hydroxyl toluene, $\alpha$-tocopherol, curcumin, quercetin and Trolox. The generation of free radicals viz. $O_2^{{\cdot}-}$, $OH^{\cdot}$, $H_2O_2$, $NO^{\cdot}$ and peroxyl radicals were effectively scavenged by the ethanolic extract of Erythrina stricta. In all the methods, the extract offered strong antioxidant activity in a concentration dependent manner. The total phenolic content, flavonoid content and total antioxidant activity in Erythrina stricta were determined as microgram (g) pyrocatechol, quercetin and $\alpha$-tocopherol equivalent/mg respectively. The extract did not exhibit any prooxidant activity when compared with ascorbic acid. The results obtained in the present study clearly indicates that Erythrina stricta scavenges free radicals and reduces lipid peroxidation, ameliorating the damage imposed by oxidative stress in different disease conditions and serve as a potential source of natural antioxidant.

Evaluation of Antioxidative Activity of Agrimonia pilosa-Ledeb Leaves on Non-lipid Oxidative Damage

  • Present study was conducted to evaluate the anti oxidative activity of the Agrimonia pilosa-Ledeb leaves on non-lipid oxidative damage. The antioxidative activity of methanolic (MeOH) extract of the Agrimonia pilosa-Ledeb leaves on non-lipid oxidation, including liposome oxidation, deoxyribose oxidation, protein oxidation, chelating activity against metal ions, scavenging activity against hydrogen peroxide, scavenging activity against hydroxyl radical and 2'-deoxyguanosine (2'-dG) oxidation were investigated. The MeOH extract of the Agrimonia pilosa-Ledeb leaves exhibited high anti oxidative activity in the liposome model system. Deoxyribose peroxidation was inhibited by the MeOH extract of the Agrimonia pilosa-Ledeb leaves and MeOH extract of the Agrimonia pilosa-Ledeb leaves provided remarkable protection against damage to deoxyribose. Protective effect of MeOH extracts of the Agrimonia pilosa-Ledeb leaves on protein damage was observed at $600{\mu}g$ level (82.05%). The MeOH extracts of the Agrimonia pilosa-Ledeb leaves at $300{\mu}g$ revealed metal binding ability (32.64%) for hydrogen peroxide. Furthermore, the oxidation of 2'-deoxyguanosine (2'-dG) to 8-hydroxy-2-deoxyguanosine (8-OH-2'dG) was inhibited by MeOH extracts of the Agrimonia pilosa-Ledeb leaves and scavenging activity for hydroxyl radical exhibited a remarkable effect. From the results in the present study on biological model systems, we concluded that MeOH extract of the Agrimonia pilosa-Ledeb leaves was effective in the protection of non-lipids against various oxidative model systems.

단삼, 도인, 당귀미 및 솔잎의 열수추출물이 지방산화에 미치는 영향 (Effect of Hot Water Extracts of Salvia miltiorrhiza Bge., Prunus persica Stokes, Angelica gigas Nakai and Pinus strobus on Lipid Oxidation)

  • This study was carried out to investigate the effects of herb extracts on lipid oxidation and free radical reaction in iron sources reacted with active oxygen species. The catalytic effects of active oxygen on lipid oxidation in oil emulsion tended to show more active in the order of OH, H2O2 and KO2. Herb extracts tended to show a little catalytic effect and active oxygen scavenging ability of herb extracts didn't show. But herb extracts played role as a strong chelating agents to bind iron if Fe2+ ion exist in oil emulsion. The contents of Fe2+ ion and total iron in Salvia miltiorrhiza Bge. and Angelica gigas Nakai were higher than those of Prunus persica Stockes and pinus strobus. The content of asocrbic acid in Pinus strobus showed the highest (26.97ppm) among several herb extracts. Electron donating abilities of Pinus strobus and Salvia miltiorrhiza Bge. were 79.54% and 77.11%, respectively, which were higher contents than those of Prunus persical Stokes and Angelica gigas Nakai. The SOD-like activity of Prunus persca Stokes showed 0.16 optical density (O.D), which means the most strong antioxidant activity among other herb extracts. The nitrite scavening effects tended to be different depending on pH. Pinus strobus and Angelica gigas Nakai showed 99.8% and 98.6% nitrite scavening effects at pH 1.2. And the effects were decreased as pH was increased. Especially, they didn't show the nitrite scavenging effect in pH 6.0. In conculsin, the Prinus strobus extract among herb extracts were the most effective antioxidant by evaluating several functional tests.

치자(Gardenia jasminoides Ellis) 과피의 용매별 추출물의 Flavonoid 함량 및 항산화 활성 비교 (Comparison of Flavonoid Content and Antioxidant Activities of Peel Extracts from Gardenia jasminoides Ellis by Various Solvents)

  • The purpose of this study was to measure the bioactivity and antioxidant activity of peel from Gardenia jasminoides fructus Ellis (GJE) in Namhae, Korea, following some established methods. CM (Chloroform:Methanol, 2:1, v/v), 70% ethanol, and n-butanol extracts were collected. Flavonoid content and value as a functional food ingredient of GJE peel was investigated through assessing antioxidant [DPPH (1,1'-diphenyl-2-picrylhydrazyl), ABTS (2,2-azino-bis-3-ethylbenzothiazoline-6-sulfonic acid)], and hydroxyl radical scavenging activities; superoxide dismutase like ability; ferrous ion-chelating capacity; and tannin content by solvent extraction. Solvent extract antioxidant activities significantly increased (p<0.05) at increasing concentrations (0.2, 0.4, 0.6 mg/mL). GJE peel extracts were less active than the positive control [ascorbic acid, BHA (butylated hydroxyanisole), and EDTA (ethylenediaminetetraacetic acid disodium salt dihydrate)]. Based on the results of this study, GJE peel could be used as a natural antioxidant source due to its high antioxidant activity and bioactive compound content.

Optimization of ultrasonic-assisted enzymatic hydrolysis conditions for the production of antioxidant hydrolysates from porcine liver by using response surface methodology

  • Objective: The objective of this study was to optimize ultrasonic-assisted enzymatic hydrolysis conditions, including enzyme-to-substrate (E/S) ratio, pH, and temperature, for producing porcine liver hydrolysates (PLHs) with the highest 1,1-diphenyl-2-picrylhydrazyl (DPPH) free radical scavenging activity by using response surface methodology (RSM). Methods: The study used RSM to determine the combination of hydrolysis parameters that maximized the antioxidant activity of our PLHs. Temperature ($40^{\circ}C$, $54^{\circ}C$, and $68^{\circ}C$), pH (8.5, 9.5, and 10.5), and E/S ratio (0.1%, 2.1%, and 4.1%) were selected as the independent variables and analyzed according to the preliminary experiment results, whereas DPPH free radical scavenging activity was selected as the dependent variable. Results: Analysis of variance showed that E/S ratio, pH, and temperature significantly affected the hydrolysis process (p<0.01). The optimal conditions for producing PLHs with the highest scavenging activity were as follows: E/S ratio, 1.4% (v/w); temperature, $55.5^{\circ}C$; and initial pH, 10.15. Under these conditions, the degree of hydrolysis, DPPH free radical scavenging activity, ferrous ion chelating ability, and reducing power of PLHs were 24.12%, 79%, 98.18%, and 0.601 absorbance unit, respectively. The molecular weight of most PLHs produced under these optimal conditions was less than 5,400 Da and contained 45.7% hydrophobic amino acids. Conclusion: Ultrasonic-assisted enzymatic hydrolysis can be applied to obtain favorable antioxidant hydrolysates from porcine liver with potential applications in food products for preventing lipid oxidation.

Evaluation of Antioxidative Effects of Lactobacillus plantarum with Fuzzy Synthetic Models

  • Numerous studies suggest that the effects of lactic acid bacteria (LAB) on oxidative stress in vivo are correlated with their antioxidative activities in vitro; however, the relationship is still unclear and contradictory. The antioxidative activities of 27 Lactobacillus plantarum strains isolated from fermented foods were determined in terms of 2,2-diphenyl-1-picrylhydrazyl, hydroxyl radical, and superoxide radical scavenging abilities, reducing activity, resistance to hydrogen peroxide, and ferrous chelating ability in vitro. Two fuzzy synthetic evaluation models, one with an analytic hierarchy process and one using entropy weight, were then used to evaluate the overall antioxidative abilities of these L. plantarum strains. Although there was some difference between the two models, the highest scoring strain (CCFM10), the middle scoring strain (CCFM242), and the lowest scoring strain (RS15-3) were obtained with both models. Examination of the antioxidative abilities of these three strains in $\text\tiny{D}$-galactose-induced oxidative stress mice demonstrated that their overall antioxidative abilities in vitro could reveal the abilities to alleviate oxidative stress in vivo. The current study suggests that assessment of overall antioxidative abilities with fuzzy synthetic models can guide the evaluation of probiotic antioxidants. It might be a more quick and effective method to evaluate the overall antioxidative abilities of LAB.
