  • 상기제목의 화합물의 두가지 단결정이 n-hexane/CH2C12 과 n-hexane/Et2O 의 용매에 의하여 얻어졌으며, 그들의 서로 다른 공간군에도 불구하고 그들의 분자구조는 동일함이 밝혀졌다. C22H18N2O2 (I), Mr=343.70, Monoclinic, Pa, a=14.595(2), b=5.413(2), c=12.218(2)Å, β=96.86(1)°, V=958.3Å3, Z=2, Dx=1.19 Mgm-3, λ(MoKα)=0.71069Å, μ=0.072mm-1, F(000)=360.0, T=292K, 756 유일한 반사강도에 대하여 R=0.104이다. 한 비대칭단위는 분자간의 두 개의 N-H…O 수소결합으로 연결된 dimer이다. C11H9NO (II), Mr=171.85, Monoclinic, P21/a, a=14.611(2), b=5.423(6), c=12.191(2)Å, β=96.89(1)°, V=959.0Å3, Z=4, Dx=1.19 Mgm-3, λ(MoKα)=0.71069Å, μ=0.072mm-1, F(000)=360.0, T=293K, 824 개의 유일한 반사강도에 대한 R=0.066이다. 한 분자가 비대칭단위를 이루고 있으며 대칭중심관계에 있는 두 개의 N-H…O 수소결합으로 dimer 를 형성한다. 양쪽 구조에서 공히 선형인 diyne chain에 의하여 연결된 pyridyl 고리와 C(10)-C(11)-O가 만드는 평면간의 이면각은 수직에 가까우며, 그 분자들은 b-축을 반복주기로하여 b-축을 따라 쌓여있다.

숙신산 알킬 하프-에스테르 유도체의 합성 및 해수에 대한 방청성능 (Synthesis and Anti-corrosion Properties of Succinic Acid Alkyl Half-Ester Derivatives)

  • 숙신산 에스테르 유도체는 방청성능 및 윤활성능이 우수하여 금속가공유 및 유압작동유 등의 첨가제로 많이 사용되고 있다. 본 연구에서는 광유계 윤활기유의 방청제로 사용하기 위하여 알킬 무수 숙신산과 여러 가지 지방 알콜과의 개환반응을 행하여 카르복실 그룹과 에스테르 그룹을 동시에 함유하는 숙신산 알킬 하프-에스테르 유도체를 97% 이상의 수율로 합성하였으며 1 중량% 농도범위에서 광유계 오일에 잘 용해되었다. 합성 유도체는 $^1H-NMR$ 및 FT-IR 스펙트럼으로 구조를 확인하였으며, GC 분석을 통하여 화합물의 순도를 확인하였다. 또한, 합성 유도체의 해수에 대한 방청성능을 ASTM D665 표준방법과 무게 중량법으로 평가하여 카르복실산 그룹을 함유하지 않는 숙신산 알킬 에스테르 유도체와 비교하였다. 방청성능 평가 결과, 합성 유도체의 농도가 증가하고 분자 내에 포함된 알킬기의 사슬이 짧을수록 상대적으로 방청성능이 우수하였으며 비교물질인 숙신산 알킬 에스테르 유도체는 방청성능을 전혀 나타내지 않았다. 무게 중량법으로 평가한 80 중량ppm 농도를 첨가한 오일의 방청효율 % (IE%)는 알킬기의 사슬이 짧을수록 높은 값을 나타내어 최고 95% 이상이었다. 또한, 발청속도(Corrosion rate, CR)는 알킬기의 사슬이 짧을수록 낮은 값을 나타내어 80 중량ppm에서 최고 0.3 mm/year 이하를 나타내었다. 즉, 숙신산 알킬 하프-에스테르 유도체의 해수에 대한 방청성능은 분자 내에 함유되어 있는 카르복실산 그룹으로 인하여 나타나는 것으로 판단되었다.

방향족 고리 함량이 공중합 액정 폴리에스터의 성질에 미치는 영향 (Effect of Aromatic Ring Content on the Properties of Liquid Crystalline Copolyesters)

  • 단위체로서 4-[4-(4-hydroxyphenoxy)butoxy]benzoic acid (HBBA)와 p-hydroxy benzoic acid (HBA)를 사용하여 직접 중축합방법에 의해 액정 공중합체를 합성하였다. 합성된 중합체의 구조와 성질은 $^1H$-NMR, FT-IR, DSC, TGA, POM에 의하여 조사되었다. 연구결과에 의하면, 페놀/p-클로로페놀/1,1,2,2-테트라클로로에탄 (25/40/35=w/w/w) 내에서 측정된 중합체의 고유점성도 (${\eta}_{inh}$)는 0.77~1.60 dL/g로 측정되었으며, 중합체 P-80을 제외하고, 공중합체들의 전이온도와 액정상의 범위는 HBA의 양이 증가함에 따라 증가하였다. 중합체의 이러한 특성은 중합체 사슬의 불규칙성과 강직성이 증가하였기 때문으로 생각된다.

술포닐 우레아 유도체들의 형태분석 (Conformational Analysis of Sulfonylureas)

  • To determine the optimal conformation of sulfonylureas, the correlation between conformation and hypoglycemic activity of the two sulfonylureas of tolbutamide and chlorpropamide as hypoglycemic agent was studied using an empirical potential function (ECEPP/2) and the hydration shell model in the unhydrated and hydrated states. The conformational energy was minimized from several starting conformations with possible torsion angles in each molecule. The conformational entropy change of each conformation was computed using a harmonic approximation. To understand the hydration effect on the conformation of the molecules in aqueous solution, the contribution of water-accessible volume of each group or atom in the lowest-free-energy conformation was calculated and compared each other. From comparison of the computed lowest-free-energy conformations of two sulfonylureas, it could be suggested that the hydration of sulfonylurea moiety is related to increase the hypoglycemic activity. From the calculation results, it was known that the conformational entropy is the major contribution to stabilize the low-free-energy conformations of two sulfonylureas in unhydrated state. Whereas, in hydrated state, the hydration free energy largely contributes to the total free energies of low-free-energy conformations of tolbutamide and conformational entropy contributes to stabilize the low-free-energy conformations of chlorpropamide. The torsion angles from phenyl ring to urea moiety of the low-free-energy conformations of the two sulfonylureas were shown the nearly regular trend. On the basis of these results, the conformation exhibiting the optimal hypoglycemic activity of sulfonylureas and the binding direction to pancreatic receptor site A could be predicted. Also, according to the side chain lengthening of urea moiety, tolbutamide showed various conformational change. Therefore, steric effect may be important factor in the interaction between sulfonylureas and the putative pancreatic receptor.

위암에서 E-cadherin과 $\beta-catenin$ 발현과 유전자 돌연변이에 관한 연구 (E-cadherin and $\beta-catenin$ Expression and Mutation in Gastric Carcinomas)

  • Purpose: When cancer cels invade the stroma, they should be dissociated from the adjacent cells at first. E-cadherin and $\beta-catenin$ constitute an important protein complex associated with cellular adhesion, development, and differentiation, especially in epithelial cells. The role of E-cadherin and $\beta-catenin$ in gastric carcinogenesis were studied. Materials and Methods: The expression of E-cadherin and $\beta-catenin$ in gastric adenocarcinomas by using immunohistochemical staining and the mutation by using polymerase chain reaction- single stranded conformation polymorphism (PCR-SSCP) and sequencing were performed in 40 adenocarcinomas and 5 dysplasia of stomach. Thirteen cases, which had lymph node metastasis, were also included for immunohistochemical staining. Results: Inappropriate cytoplasmic and/or nuclear expression of a E-cadherin-$\beta-catenin$ complex was more frequent in poorly differentiated, diffuse type signet ring cell carcinomas than in well-differentiated, intestinal type adenocarcinomas (P<0.05). However, the expression was not related with clinical stage or lymph node metastasis. Mutation of E-cadherin was detected in 4 cases by using PCR-SSCP, whereas mutation of $\beta-catenin$ was detected in 2 cases. Conclusion: E-cadherin and $\beta-catenin$ seem to be important in gastric carcinogenesis, especially in poorly differentiated diffuse type.

알파술폰 고급지방산 폴리에틸렌글리콜 에스테르류의 계면물성 (Surfaces Properties of ${\alpha}-Sulfonated$ Fatty Acid Polyethylene Glycol Esters)

  • All the surface activities including surface tension, foaming power, foam stability, emulsifying power, dispersion effect, and detergency were measured and critical micelle concentration(cmc) was evaluated in dilute aqueous solution. The cmc evaluated by the Ring method was $10^{-3}{\sim}10^{-4}mol/L$ in case of monoesters, and $10^{-3}{\sim}5.0{\times}10^{-5}mol/L$ in case of diesters, respectively. Surface tension of the aqueous solution was decreased to $45{\sim}50dyne/cm$, showing the tendency that the ability of lowering the surface tension was dependent on increasing of carbon atom number in alkyl chain. Foaming power of all the monoesters was better than that of diesters. while foam stability of diesters was to the contrary. Emulsifying power of soybean oil or benzene was specially expected to be good for emulsifiers in industrial application fields. HLB values of monoesters and diesters evaluated by Griffin's method were in the range of 8 to 12. Dispersion property of ferric oxide was stable in the range of $4.5{\times}10^{-5}{\sim}5.0{\times}^{-4}mol/L$ in case of monoesters, and $10^{-5}{\sim}10^{-4}mol/L$ in case of diesters.

전기 자전거용 토크센서 개발 (The Development of Torque Sensor for Electric Bicycle)

  • 최근 환경오염, 지구 온난화, 화석 연료 고갈 등이 범지구적 문제가 됨에 따라 녹색 에너지 기술 개발이 주목을 받고 있다. 이런 추세에 따라, 자전거가 다양한 스마트 에너지 기술과 결합하여 친환경 근거리 이동수단으로 발전하고 있다. 스마트 자전거 기술과 관련하여 많은 기술이 개발되고 있는데 이중 PAS(Power Assist System)는 사람의 힘과 전기의 힘을 효과적으로 결합하여 배터리로 구동되는 전기 모터를 제어하는 기술이다. 본 논문은 PAS를 구성하는 핵심 기술인 새로운 토크 센서를 제안한다. 이 기술은 기존 기술들과는 달리 자전거 뒤축에 스트레인 게이지를 부착하여 슬립링의 필요성을 없애고 사람에 의해 가해지는 구동축 토크를 측정 할 뿐 아니라, 현재 작동되는 기어의 위치를 근사적으로 추정 가능하게 한다.

Role of IL-15 in Sepsis-Induced Skeletal Muscle Atrophy and Proteolysis

  • Background: Muscle wasting in sepsis is associated with increased proteolysis. Interleukin-15 (IL-15) has been characterized as an anabolic factor for skeletal muscles. Our study aims to investigate the role of IL-15 in sepsis-induced muscle atrophy and proteolysis. Methods: Mice were rendered septic either by cecal ligation and puncture or by intraperitoneal injection of lipopolysaccharide (LPS, 10 mg/kg i.p.). Expression of IL-15 mRNA and protein was determined by reverse transcriptase polymerase chain reaction and Western blot analysis in the control and septic limb muscles. C2C12 skeletal muscle cells were stimulated in vitro with either LPS or dexamethasone in the presence and absence of IL-15 and sampled at different time intervals (24, 48, or 72 hours). IL-15 ($10{\mu}g/kg$) was intraperitoneally administered 6 hours before sepsis induction and limb muscles were sampled after 24 hours of sepsis. Cathepsin L activity was determined to measure muscle proteolysis. Atrogin-1 and muscle-specific ring finger protein 1 (MuRF1) expressions in limb muscle protein lysates was analyzed. Results: IL-15 mRNA expression was significantly lower in the limb muscles of septic mice compared to that of controls. Cathepsin L activity in C2C12 cells was significantly lower in presence of IL-15, when compared to that observed with individual treatments of LPS or dexamethasone or tumor necrosis factor ${\alpha}$. Further, the limb muscles of mice pre-treated with IL-15 prior to sepsis induction showed a lower expression of atrogin-1 and MuRF1 than those not pre-treated. Conclusion: IL-15 may play a role in protection against sepsis-induced muscle wasting; thereby, serving as a potential therapeutic target for sepsis-induced skeletal muscle wasting and proteolysis.

방선균 분리주가 생산하는 Phospholipase C 저해물질인 MT-2617-2B의 분리 및 특성 (Isolation and Characterization of MT2617-2B, a Phospholipase C Inhibitor Produced by an Actinomycetes Isolate)

  • A phospholipase C (PLC) inhibitor (MT267-2B) was isolated from the culture broth of actinomycetes isolate MT2617-2 by the extraction with n-butanol and column chromatographic techniques. The molecular weight of the inhibitor was 1057, by the spectroscopic analyses of IR, $^{13}C$-and $^{1}H$-NMR and ESI-MS. The chemical structure of MT2617-2B was found to be a macrolide compound consisted of a hemiketal ring, polyhydroxyl and polymethyl groups, which had a malonate and guanidine group as its side chain. MT2617-2B produced its two isomers having the same molecular weight by standing in methanol solution at room temperature. Therefore, MT2617-2B was identified as copiamycin and niphithricin A, macrolide antibiotics. The values of $IC_{50}$ against PLC-${\gamma}$1 and PLC-${\beta}$1 were 25 and 50${\mu}$g/ml, respectively. MT2617-2B had antimicrobial activities against Staphylococcus aureus and Candida albicans, but not against Escherichia coli.

Possibility of Wound Dressing Using Poly(L-leucine)/poly(ethylene glycol)/poly(L-leucine) Triblock Copolymer

  • ABA-type block copolymers composed of poly(L-leucine)(PLL) as the A component and poly(ethylene glycol)(PEG) as the B component were synthesized by ring-opening polymerization of L-leucine N-carboxyanhydride initiated by primary amino group located at both ends of PEG chain. A silver sulfadiazine(AgSD)-impregnated wound dressing of sponge-type was prepared by the lyophilization method. Morphological structure of this wound dressing obtained by scanning electron microscopy(SEM) was composed of a dense skin layer and a macroporous inner sponge layer. Equilibrium water content(EWC) of wound dressing was above 10%. It increased with an increased of PEO content in the block copolymer due to the hydrophilicity of PEO. AgSD release from AgSD- impregnated wound dressing in PBS buffer(pH=7.4) was dependent on PEG composition in the block copolymer. Therefore, EWC and release of AgSD can be control by PEG composition. Antibacterial capacity of AgSD-impregnated wound dressing was examined in agar plate against Pseudmonas aeruginosa and Stapplococus aruous. Cytotoxicity of the wound dressing was evaluated by studing mouse skin fibroblast(L929). From the behavior of antimicrobial releasing and the investigation of the suppression of bacterial proliferation, it was supposed that the wound dressing containing antibiotics could protect the wound surfaces from bacterial invasion to suppress the bacterial proliferation effectively. In cytotoxicity observation, cellular damage was reduced by the control led released of AgSD from the LEL sponge matrix of AgSD-medicated wound dressing. In vivo test, granulous tissue formation and wound contraction or the AgSD and DHEA impregnated wound dressing were aster than any other groups.
