Distribution of Foodborne Pathogens from Garlic Chives and Its Production Environments in the Southern Part of Korea (남부지방 부추와 재배환경의 식품매개병원균의 분포)

  • Recently, foodborne illness outbreaks linked to fresh produce are being increasingly reported in the United States, the EU, and Korea as well. Some of this increase may be due to improved surveillance, increase in consumption, change in consumers' habits, and complex distribution systems. Garlic chive is a green, fresh-cut vegetable consumed year-round as a nutrition-rich herb in Korea. It is also prone to contamination with foodborne pathogens during pre-harvest, as amendment with high amounts of livestock manure or compost to soil is required in its cultivation. Our aim in this study was to evaluate microbial contamination of garlic chives, garlic chives cultivation soil, compost, and irrigation water in the southern part of Korea. Samples were collected in A, B, and C regions in 2019 and 2020, and 69, 72, 27, and 40 of garlic chives, soil, compost, and irrigated water, respectively, were analyzed for the presence of sanitary indicator bacteria (total aerobic bacteria, coliforms and Escherichia coli), Bacillus cereus, Staphylococcus aureus, pathogenic E. coli, E. coli O157:H7, Listeria monocytogenes, and Salmonella spp. In A, B, and C regions, levels of total aerobic bacteria, coliform, B. cereus, and S. aureus on all samples were between 1.14 and 8.83 log CFU/g, 0.43 and 5.01 log CFU/g, 0.41 and 5.55 log CFU/g, and 1.81 and 6.27 log CFU/g, respectively. B. cereus isolated from garlic chives and environmental samples showed β-hemolysis activity. Incidence of S. aureus in garlic chive and its production environments in 2020 was different from 2019. In this study, B. cereus and S. aureus were the only pathogenic microorganisms detected in all samples. As a result, this work suggests that continuous monitoring in the production and pre-harvest environment is required to improve hthe hygiene and safety of garlic chive.

Evaluation of Sanitation Management Practices and Microbiological Quality of Foods in Kindergarten Foodservice Settings (유치원 급식의 위생관리 실태조사 및 미생물적 품질평가)

  • This research aims to audit foodservice sanitation management practices and to assess microbiological quality of foods and their food contact environments in kindergartens. Sanitation auditing was conducted in 10 kindergartens in Seoul, Gyeonggi, and Incheon areas to assess the levels of safety practices. Results revealed that the surveyed kindergartens scored 41.4 out of 100 points, on average. The average scores of each category were 6.4/11 (58.1%) for facilities sanitation, 4.2/12 (35.0%) for equipment sanitation, 2.4/10 (24.0%) for personal hygiene, 5.1/10 (51.0%) for food ingredients management, 6.0/17 (35.3%) for production process, 5.4/10 (54.0%) for environmental sanitation, 2.0/6 (33.3%) for kitchen utensils sanitation, and 2.2/6 (96.7%) for safety management. Microbiological quality of raw, prepared foods, personal sanitation (hands), environmental sanitation, and drinking water were assessed. Total plate counts (TPC) of the following menus exceeded the critical limit: seasoned leek (5 log CFU/g), cucumber (5.0 log CFU/g), panbroiled fish paste (TNTC at $10^4$), tangpyeongchae (5.3 log CFU/g), egg rolls (6.1 log CFU/g), panbroiled sausage (TNTC at $10^4$), and soft tofu pot stew (TNTC at $10^4$). Coliform which exceeded the standard limit were detected from seasoned leek (2 log CFU/g), cucumber (2.5 log CFU/g), panbroiled fish paste (2.0 log CFU/g), egg roll (3.8 log CFU/g), tangpyeongchae (4.0 log CFU/g), panbroiled sausage (2.3 log CFU/g), and soft tofu pot stew (3.7 log CFU/g). For seasoned foods (muchim), S. aureus ranged 2.2~2.9 log CFU/g. In food workers' hands, microbial profiles ranged 3.8~7.9 log CFU/hand for TPC, ND~4.5 log CFU/hand for coliforms, ND~4.7 log CFU/hand for S. aureus, and ND~5.3 log CFU/hand for Enterobacteriaceae. Microbiological profiles of food contact surface of knives, cutting boards, dish-clothes, and trays showed possibilities of cross-contamination. General bacteria were 2.1~4.5 logCFU/ml in 4 purified water samples and E. coli were found in the kitchen of one kindergarten. These results suggested that environmental sanitation management practices need more strict improvement: effective sanitation education methods and practices were strongly required, and more strict sanitation management for cooking utensils and equipment were required.

Evauation of Hygienic Status of High School Foodservice using ATP Bioluminescence Assay & Microorganism Test Strips (ATP Bioluminescence와 간이미생물배지를 이용한 고등학교 급식시설의 위생상태 조사연구)

  • An investigation was conducted to evaluate the hygienic status of 33 high school foodservice systems in Yongin city by using hygiene management guide checklist, ATP bioluminescence assay and microbe inspection petrifilm (APC, coliform group, Staphylococcus aureus) of food utensils during use. The 22 hygiene management guide checklist items about facilities, personal hygiene, food control, distribution, washing and disinfection had good grade but there were some inadequate behaviors on observation. The inspection results showed their sanitary condition met the level B of the recommendation of Korea method, it means sanitary management system get settled but more practical CCP system was needed. ATP bioluminescence assay was conducted on surface of food facilities, ATP ranged 425~2,552 RLU on gloves, 541~70,251 RLU on apron, 1,596~88,490 RLU on working desk, 1,177~263,813 RLU on sterilizer grip, 715~32,814 RLU on sterilizer shelf, 114~619,725 RLU on refrigerator grip, 677~319,007 RLU on refrigerator shelf, 71~196,725 RLU on freezer grip, 1,535~233,375 RLU on freezer shelf. APC ranged $66.7{\pm}29.0CFU$ on freezer grip, $102.1{\pm}35.9CFU$ on refrigerator grip, $45.4{\pm}28.2CFU$ on heating cabinet grip, $58.8{\pm}40.4CFU$ on sterilizer grip, the number of coliform group ranged $5.6{\pm}4.9CFU$ on freezer grip, $9.1{\pm}8.7CFU$ on refrigerator grip, $1.2{\pm}1.1CFU$ on heating cabinet grip, $4.5{\pm}4.4CFU$ on sterilizer grip. S. aureus ranged $8.0{\pm}5.6CFU$ on freezer grip, $12.2{\pm}9.6CFU$ on refrigerator grip, $2.1{\pm}1.6CFU$ on heating cabinet grip, $11.6{\pm}6.4CFU$ on sterilizer grip.

Comparison of Common Enrichment Methods for Recovery of Yersinia Enterocolitica from Artificially Inoculated Swine Feed Samples

  • Five different enrichment methods were studied to find an optimal method to recover Yersinia enterocolitica from swine feed samples. When the recovery of Y. enterocolitica GER-C (serotype O:3) strain was studied at 1000 CFU/g feed, phosphate-buffered saline (PBS) enrichment at $4^{\circ}C$ and PBS plus sorbitol and bile salts (PSB) enrichment at $4^{\circ}C$ and $21^{\circ}C$ were not effective (< 22%). In contrast, both irgasan-ticarcillin-potassium chlorate (ITC) and tryptic soy broth plus polymyxin B sulfate and novobiocin (TSBPN) enrichment methods showed a full recovery (100%) at 100-1000 CFU/g feed. At 10 CFU/g feed, both ITC and TSBPN methods still recovered the strain (> 50%). In recovery of ATCC 9610 (serotype O:8) strain, TSBPN method was more sensitive than any other methods (P < 0.05) at 1000 CFU/g feed. Using TSBPN method, the strain was still recovered at 100 CFU/g feed, but not at 10 CFU/g feed. With its sensitivity and relatively simple recipe, TSBPN was most desirable method to recover Y. enterocolitica from swine feed samples.

The Effect of Phosphate Solubilizing Microorganism on Cucumber Productivity (미생물비료 인산가용화균이 오이의 생산량에 미치는 영향)

  • The effect of the Penicillium sp. PS-113 with high phosphate solubilizing activity and Lactobacillus sp. was studied on cucumber growing test in green house. Inoculation of Pemicillium sp. Ps113 $5.0{\times}10^6$cfu/ml and Lactobacillus sp. $6{\times}10^6$cfu/ml were longer than control about 3cm length of cucumber. The average weight was also increased 89.2g more than control. Total product of cucumber was 10,681.7g and it was increased 4517.3g as compared to control. Thus, Inoculations of high phosphate solubilizing microorganism increased in productivity on cucumber.

Indoor distribution characteristics of airborne bacteria in pig buildings as influenced by season and housing type

  • Objective: A concentration of airborne bacteria generated from swine houses is recognized to be relatively higher than other work places and it is essential to optimally manage it to prevent farmers' respiratory diseases. This study was conducted to assess the distribution characteristics of airborne bacteria in swine houses located at South Korea. Methods: A total 27 pig buildings of the enclosed type operated with mechanical ventilation system by a side wall fan and deep-pit manure system with slats were surveyed. Air samples were collected at 1.0 m above the middle floor in pig housing room. A six-stage viable particulate cascade impactor was used to identify the distribution of the sizes of particles in diameter. Results: Seasonal mean levels of airborne bacteria in the housing rooms of gestation/farrowing pigs, nursery pigs and growing/fattening pigs were 3,428(${\pm}1,244$) colony forming unit $(cfu)/m^3$, $8,325({\pm}3,209)cfu/m$, and $13,254({\pm}6,108)cfu/m^3$ for spring; $9,824({\pm}2,157)cfu/m^3$, $18,254({\pm}5,166)cfu/m^3$, and $24,088({\pm}9,274)cfu/m^3$ for summer; $1,707({\pm}957)cfu/m^3$, $4,258({\pm}1,438)cfu/m^3$, and $8,254({\pm}2,416)cfu/m^3$ for autumn; and $2,322({\pm}1,352)cfu/m^3$, $6,124({\pm}1,527)cfu/m^3$ and $12,470({\pm}4,869)cfu/m^3$ for winter, respectively. Conclusion: Concentrations of airborne bacteria according to pig housing type were highest in growing/fattening housing room followed by nursery housing room and gestation/farrowing housing room. In terms of seasonal aspect, the pig building showed the highest levels of airborne bacteria in summer followed by spring, winter and autumn. The respirable airborne bacteria which are ranged between 0.6 and $4.7{\mu}m$ accounted for approximately 60% compared to total airborne bacteria regardless of pig housing type.

키토산을 이용한 김치의 숙성 지연과 보존기간 연장에 관한 연구

  • 고분자 키토산(분자량 약 800,000)을 농도별(0%, 0.1%, 0.2%, 0.3%)로 김치를 제조하여, pH, 적정산도, 총균수, 젖산균 수, 대장균군 수 및 관능검사를 조사한 결과는 다음과 같이 나타났다. 1. 키토산 무첨가 김치는 발효 후 6일경에 pH가 초기 5.4에서 4.1로 급격히 떨어졌으며 그 후부터는 천천히 떨어져 발효 후 약 15일 경과 후에는 3.9로 나타났다. 그러나 0.1%, 0.2% 및 0.3% 고분자 키토산을 첨가한 김치에서는 초기 pH 5.38, 5.30 및 5.28에서 6일경에는 pH가 0.1%, 0.2% 및 0.3% 고분자 키토산에서 4.23, 4.34 및 4.47로 각각 나타났다. 15일 경과 후에는 0.1% 고분자 키토산 김치는 키토산 무첨가 김치와 거의 같은 pH인 3.9로 나타났으나 0.2%와 0.3% 고분자 키토산 김치의 pH는 3.90보다 높은 4.10, 4.10으로 각각 나타났다. 2. 각 김치 종류별 적정산도를 측정한 결과는 발효초기에 모든 구가 0.72%로 나타났으며, 발효 후 6일 경에서는 키토산 무첨가, 0.1%, 0.2% 및 0.3% 고분자 키토산 김치의 적정산도는 각각 2.16%, 2.00%, 1.70% 및 1.30%로 나타났다. 그리고 15일 경과 후에는 키토산 무첨가 김치의 적정산도가 2.25%이였으나 0.1%, 0.2% 및 0.3% 고분자 키토산 김치는 각각 2.26%, 2.24 및 2.22%로 나타났다. 3. 발효가 진행 중에 총균수를 측정한 결과 시간과 더불어 총균수가 모든 구에 있어서 서서히 증가하였고 발효초기의 모든 김치구의 총균수는 2.5X105∼5.4X106 cfu/g 범위였으며, 김치 맛이 들기 시작한 6일경에서의 각 구별 총균수는 키토산 무첨가 김치가 2.4X109 cfu/g이였구 0.1%, 0.2% 및 0.3% 고분자 키토산 김치의 총균수는 각각 1.2X109 cfu/g, 4.0X108 cfu/g 및 1.1X107 cfu/g으로 나타났다. 그리고 김치가 완전히 익은 15일 후에는 무첨가 김치, 0.1%, 0.2% 및 0.3% 고분자 키토산 김치의 총균수는 5.4X107 cfu/g, 3.3X107 cfu/g, 1.8X108cfu/g 및 4.2X108 cfu/g로 나타났다. 4. 발효가 진행 중에 젖산균 수를 측정한 결과 시간과 더불어 젖산균 수가 모든 구에 있어서 서서히 증가하였으며, 발효초기의 모든 김치구의 젖산균 수는 2.0X104∼2.7X106 cfu/g 범위였으며 김치 맛이 들기 시작한 6일경에서의 각 구별 젖산균 수는 키토산 무첨가 김치가 3.2X108 cfu/g이었고, 0.1%, 0.2% 및 0.3% 고분자 키토산 김치의 젖산균 수는 각각 1.6X108 cfu/g, 13X108 cfu/g 및 9.6X107 cfu/g으로 나타났다. 그리고 김치가 완전히 익은 15일 후에는 무첨가 김치, 0.1%, 0.2% 및 0.3% 고분자 키토산 김치의 젖산균 수는 5.4X107 cfu/g, 3.3X107 cfu/g, 8.6X106 cfu/g 및 2.6X106 cfu/g로 나타났다. 5. 발효가 진행 중에 대장균 군 수가 시간과 더불어 대장균군 수가 모든 구에 있어서 6일까지는 서서히 증가하다가 그 후부터는 대장균 군 수가 감소하였다. 즉, 발효초기에는 모든 구가 2.0X104∼4.0X105 cfu/g이었고, 6일 경에는 8.9X104∼4.5X105 cfu/g로 약간 증가하였으며 15일후에는 키토산 무첨가 김치가 2.0X102 cfu/g이었으며, 0.1%, 0.2% 및 0.3% 고분자 키토산 김치의 대장균군 수는 각각 2.0X102 cfu/g, 1.1X102 cfu/g 및 4.0X101 cfu/g로 점점 감소하였다. 6. 관능검사 결과는 키토산 무첨가 김치와 0.1% 첨가 김치는 유사한 선호도를 나타내어 0.1% 첨가 김치가 기능성과 보존성을 높여줄 뿐만 아니라 선호도도 높은 것으로 나타났다.

Analysis of Pathogenic Microorganism's Contamination on Cultivation Environment of Strawberry and Tomato in Korea

  • The purpose of this study was to analyze microbial hazards for cultivation environments and personal hygiene of strawberry and tomato farms at the growth and harvesting stage. Samples were collected from thirty strawberry farms and forty tomato farms located in Korea and tested for Staphylococcus aureus and Bacillus cereus. To investigate the change in the distribution of the S. aureus and B. cereus, a total of 4,284 samples including air born, soil or medium, mulching film, harvest basket, groves and irrigation water etc. were collected from eight strawberry farms and nine tomato farms for one year. As a result, total S. aureus and B. cereus in all samples were detected. Among the total bacteria of strawberry farms, S. aureus (glove: $0{\sim}2.1Log\;CFU/100cm^2$, harvest basket: $0{\sim}3.0Log\;CFU/100cm^2$, soil or culture media: 0~4.1 Log CFU/g, mulching film: $0{\sim}3.8Log\;CFU/100cm^2$), B. cereus (glove: $0{\sim}2.8Log\;CFU/100cm^2$, harvest basket: $0{\sim}4.8Log\;CFU/100cm^2$, soil or culture media: 0~5.3 Log CFU/g, mulching film: $0{\sim}4.5Log\;CFU/100cm^2$) were detected in all samples. The total bacteria of tomato farms, S. aureus (glove: $0{\sim}4.0Log\;CFU/100cm^2$, harvest basket: $0{\sim}5.0Log\;CFU/100cm^2$, soil or culture media: 0~6.1 Log CFU/g, mulching film: $0{\sim}4.0Log\;CFU/100cm^2$), B. cereus (glove: $0{\sim}4.0Log\;CFU/100cm^2$, harvest basket: $0{\sim}4.3Log\;CFU/100cm^2$, soil or culture media: 0~5.9 Log CFU/g, mulching film: $0{\sim}4.7Log\;CFU/100cm^2$) were detected in all samples. The contamination of S. aureus and B. cereus were detected in soil, mulching film and harvest basket from planting until harvest to processing, with the highest count recorded from the soil. But S. aureus and B. cereus were not detected in irrigation water samples. The incidence of S. aureus and B. cereus in hydroponics culture farm were less than those in soil culture. The amount of S. aureus and B. cereus detected in strawberry and tomato farms were less than the minimum amount required to produce a toxin that induces food poisoning. In this way, the degree of contamination of food poisoning bacteria was lower in the production environment of the Korea strawberry and tomato, but problems can be caused by post-harvest management method. These results will be used as fundamental data to create a manual for sanitary agricultural environment management, and post-harvest management should be performed to reduce the contamination of hazardous microorganisms.

Wahsing Effect of Micor-Bubbles and Changes in Quality of Lettuce (Lacutuca sativa L.) during Storage (마이크로버블에 의한 상추의 세척효과 및 저장 중 품질변화)

  • We assessed quality changes in and washing effects (time and method) on lettuce (Lactuca sativa L.) treated with micro-bubbles. Samples were treated with micro-bubbling for 1, 3, or 5 min, and the 5-min treatment yielded the best results in terms of reduced total microorganism counts, sensory aspects, and degree of washing. Total microorganism counts were 4.30 log colony-forming units (CFU)/g in unwashed lettuce(CT), 4.10 log CFU/g in hand-washed lettuce (HW), 3.98 log CFU/g in conventional, bubble-washed lettuce (BW) and 3.25 log CFU/g in micro-bubble-washed lettuce (MW). In comparison, total counts of samples examined after 10 days of storage were 7.00 log CFU/g for CT, 6.19 log CFU/g for HW, 6.02 log CFU/g for BW, and 5.89 log CFU/g for MW. The lowest counts were seen after micro-bubble treatment. BW and MW samples showed significantly higher counts than did CT and HW samples. In general, BW and MW samples did not vary significantly in count numbers. MW showed a 2.3-fold lower residual pesticide level compared with CT, and also had the lowest level of impurities. HW and BW samples were not well washed.

Microbial Contamination Levels of Ginseng and Ginseng Products Distributed in Korean Markets (국내 유통 중인 인삼 및 인삼 제품류의 미생물 오염도 평가)

  • The objective of this study was to investigate the microbial contamination levels of ginseng and ginseng products distributed in markets. A total of 81 ginseng and ginseng products samples (fresh ginseng 37, white ginseng 15, red ginseng 15, red ginseng beverage 14) were tested to analyze sanitary indicator bacteria (total aerobic bacteria, coliforms and Escherichia coli), major foodborne pathogens, and fungi. The contamination levels of total aerobic bacteria and coliforms were in the range of 3.19 to 7.02 log CFU/g for fresh ginseng, 0.25 to 7.31 log CFU/g for white ginseng, 0 to 2.89 log CFU/g for red ginseng and 0 to 1.70 log CFU/mL for red ginseng beverage. In case of major foodborne pathogens, B. cereus was detected at levels of 0.50 to 2.41 log CFU/g in samples except red ginseng beverage. Fungi was detected at levels of 2.41 log CFU/g in fresh ginseng, the contamination levels of the other ginseng products samples were lower than 1 log CFU/g or mL. These results indicate that the ginseng and ginseng products were comparatively safe with respect to microbiological hazard.
