The Mechanism of Proteasome Inhibitor-Induced Apoptosis in Lung Cancer Cells (폐암 세포에서 Proteasome Inhibitor에 의한 Apoptosis의 기전)

  • Background : Proteasome inhibitors can promote either cell survival or programmed cell death, depending on both the specific type and proliferative status of the cell. However, it is not well known whether inhibition of proteasome activity is related to apoptosis in lung cancer cells. In addition, the exact mechanisms responsible for apoptosis induced by proteasome inhibition are not well understood. In the present study, we have examined the effect of proteasome inhibition on lung cancer cells and tried to test the mechanisms that may be associated with the apoptosis of these cells. Methods : We examined the effect of proteasome inhibition with MG132 or PS-341 on cell survival in A549 and NCI-H157 lung cancer cells using MTT assay, and analyzed the cleavage of PARP by Western blot analysis to find evidence of apoptosis. Next, we evaluated the activation of caspase 3 by Western blot analysis and the activity of JNK by immunocomplex kinase assay. We also examined the changes in anti-apoptotic pathways like ERK and cIAP1 by Western blot analysis after inhibition of proteasome function. Results : We demonstrated that MG132 reduced cell survival by inducing apoptosis in A549 and NCI-H157 cells. Proteasome inhibition with MG132 or PS-341 was associated with activation of caspase 3 and JNK, reduced expression of activated ERK, and downregulation of cIAP1. Conclusion : Apoptosis induced by proteasome inhibition may be associated with the activation of pro-apoptotic pathways like caspase 3 and JNK and the inactivation of anti-apoptotic pathways in lung cancer cells.

Population Learning with Coercive Isomorphism in the Korean Business Groups: A Comparison between the Samsung and Hyunda Group (한국 기업진단의 강제적 동질성(isomorphism)을 수반한 모집단학습: 삼성그룹과 현대그룹의 비교)

  • The Purpose of the current study is to understand population learning with coercive isomorphism in the Korean Business Groups. Can we observe isomorphic phenomena by coercive isomorphism? Why do these happen? What mechanisms are embedded in the coercive isomorphism. To answer these questions this study focused on the two Korea's largest Chaeblos the Samsung and Hyundai. An empirical study to compared daily routines used in these two Chaeboles and contents analysis on the founders' characteristics were performed. Three major mechanisms were suggested as main processes to impact the formation of the coercive isomorpism in the two Korean Chaebols. Research implications were discussed at the end of study.

Roles of the Community Facilities for Foreigners as a Platform for Urban Globalization - Focused on the Seoul Global Centers - (도시 국제화를 위한 플랫폼으로서 외국인 커뮤니티 시설의 역할에 관한 연구 - 서울시 글로벌센터를 중심으로 -)

  • Globalization has diversified ethnic composition thus increased the risk of conflicts and socio-political instability in global cities. However, still the status of community facility in a global city is unclear despite its critical role to build sustainable community in global era. In Seoul, as a reaction of globalization, 19 community facilities known as the 'Seoul Global Center' have been established since 2007 by Seoul Metropolitan Government. These facilities have started to provide basic foreigner services but been transformed to foreigner service hub with multiple functions in terms of a living, a business, a labour, etc. This study explores the role of the community facility for foreigners by using the Seoul Global Center as a case study, conducting a site observation, an interview(13 staff) and a questionnaire(148 visitors). The findings are the community facility functions as a 'global platform' in forms of a policy tool for implementing the urban globalization strategy, a mediator connecting foreign migrants with local communities, an applicant for successful settlement of foreign residents, an incubator that grows human and social capital, and a base for collecting and aggregating information on foreign migrants and forming new local identities.

Pentacene-based Thin Film Transistors with Improved Mobility Characteristics using Hybrid Gate Insulator

  • Hybrid insulator pentacene thin film transistors (TFTs) are fabricated with thermally grown oxide and cross-linked polyvinylalcohol (PVA) including surface treatment by dilute ploymethylmethacrylate (PMMA) layer on $n^+$ doped silicon wafer. Through the optimization of $SiO_2$ layer thickness in hybrid insulator structure, carrier mobility is increased to more than 35 times than that of the TFT which has only a gate insulator of $SiO_2$ at the same electric field. The carrier mobility of $1.80cm^2$/V-s, subthreshold swing of 1.81 V/decade, and $I_{on}/I_{off}$ current ratio> $1.10{\times}10^5$ are obtained less than -30 V bias condition. The result is one of the best reported performances of pentacene TFTs with hybrid insulator including cross-linked PVA layer as a gate insulator at relatively low voltage operation.

Numerical Classification of Actinomycetes Isolated from Volcanic Soil

  • Of actinomycetes isolated from volcanic compost soils, 115 representative strains which showed distinctive morphologicla features were numerically classified, compared with reference strains of Streptomyces. One hundred and twenty unit characters were tested and the average probability of error was 4.27%. The cluster analysis resulted in two groups: group A included strains of actinomycetes except streptomycetes. Group A was divided into 2 major clusters (over 5 strains), 10-diaminopimelic acid. Group B was divided into 5 clusters, of which 4 clusters contained mesodiminopimelic acid and 1 cluster LL-diaminopimelic acid. The major clusters of group A showed higher abilities of substrate utilization and degradation, and higher resistance to inhibitors, whereas the minor and single member clusters of group A showed relatively higher antimicrobial activities. On the other hand, all clusters of group B showed relatively lower abilities of substrate utilization and degradation and lower resistance to inhibitors.

An FMN-containing NADH-quinone reductase from streptomyces sp (An FMN-Containing NADH-Quinone Reductase from Streptomyces sp.)

  • NADH-quinone reductase was purified 22-fold from the cytosolic fraction of Streptomyces sp. Imsnu-1 to apparent hemogenity, with an overall yield of 9%, by the purification procedure consisting of ammonium, sulfate precipitation and DEAE Sephacryl S-200 and DEAE 5 PW chromatographies. Thes molecular mass of the enzyme determined by gel filtration chromatography was found to be 110 kDa. SDS-PAGE revealed that the enzyme consists of two sugunits with a molecular mass of 54 kDa. The enzyme contained 1 mol of FMN per subunit as a cofactor. The $A_{272}$ A$_{457}$ ratio was 6.14 and the molar extinction coefficients were calculated to be 20, 800 and 25, 400M$^{-1}$ $cm^{-1}$ / AT 349 AND 457 nm, respectively. The N-terminal sequence of the enzyme contained the highly conserved fingerprint of ADP-binding domain. The enzyme used NADH as an electron donor and various quinones as electron acceptors. Cytochrome c was practically inactive. Air-stable flavin semiquinone was produced by the addition of NADH to the enzyme. Also, naphthosemiquinone was detected in the reaction mixture containing the enzyme.

Diagnostic aids for the detection of oral cancer (구강암의 간편 진단 기법)

  • Historically, the screening of patients for signs of oral cancer and precancerous lesions has relied upon the conventional oral examination. A variety of commercial diagnostic aids and adjunctive techniques are developed to potentially assist in the screening of healthy patients for evidence of occult cancerous change. This paper is reviewing the literature associated with current oral cancer screening aids such as spectroscopy, chemoiluminescence, exfoliative cytopathology, vital staining and saliva as a diagnostic tool. Despite the increased public awareness of oral cancer, no technique or technology to date has provided definitive evidence to suggest that it improves the sensitivity or specificity of oral cancer screening beyond clinical oral examination alone.

  • 최근 익명성이 보장되는 네트워크와 인터넷이 생기며 이를 이용한 악성코드가 증가하고 있다. 이를 막기 위한 방안 중 하나로 코드의 저자를 밝혀내는 연구인 코드 저자 식별이 있다. 이에 관해 최근 연구들은 소스 코드와 바이너리에서 높은 정확도로 저자를 식별해낼 수 있다는 것을 밝혀냈다. 하지만 스트립 바이너리와 관련해서는 연구가 많이 이루어지지 않았다. 이에 본 연구에서는 최근 연구에 사용되는 방법을 스트립 바이너리에 적용하여 실험을 진행하여 그 결과가 좋지 않음을 보였다. 그리고 이를 바탕으로 스트립 바이너리에서 저자 식별이 어려운 이유를 분석하였다.
