Human Amniotic Fluid Cells Support Expansion Culture of Human Embryonic Stem Cells (양수 세포를 이용한 인간배아줄기세포의 배양)

  • Objective: This study was performed to evaluate the possibility of prolonged culture of human embryonic stem cells (hESC; SNUhES2) on human amniotic fluid cells (hAFC), which had been storaged after karyotyping. Method: The hAFC was prepared for feeder layer in the presence of Chang's medium and STO medium (90% DMEM, 10% FBS) at $37^{circ}C$ in a 5% $CO_2$ in air atmosphere. Prior to use as a feeder layer, hAFC was mitotically inactivated by mitomycin C. The hESCs on hAFC were passaged mechanically every seven days with ES culture medium (80% DMEM/F12, 20% SR, bFGF). Results: The hAFC feeder layer support the growth of undifferentiated state of SNUhES2 for at least 59 passages thus far. SNUhES2 colonies on hAFC feeder appeared slightly angular and flatter shape as compared with circular and thicker colonies observed with STO feeder layer and showed higher level with complete undifferentiation in seven days. Like hESC cultured on STO feeders, SNUhES2 grown on hAFC expressed normal karyotype, positive for alkaline phosphatase activity, high telomerase activity, Oct-4, SSEA-3, SSEA-4, Tra-1-60 and Tra-1-81 and formed embryoid bodies (EBs). Conclusion: The hAFC supports undifferentiated growth of hESC. Therefore, these results may help to provide a clinically practicable method for expansion of hESC for cell therapies.

The study about operation condition of dental hospital and clinics used public data : focus on population of local autonomous entity (공공데이터를 활용한 치과병의원 운영실태 연구: 광역자치단체와 특별자치단체의 인구를 중심으로)

  • This study assayed regional distribution of dental hospital & dental clinics, the number of population & households per one dental hospital & clinic, operation condition & duration. This study used public data that display from 1946 years(the first dental clinic open in republic of korea) to 2016 years. We collected present condition of 21,686 dental hospital and clinics available in public data portal site on 28. Feb.2016. Data were classified by scale, location, permission year, operation duration of dental hospital & clinics and were analyzed using SPSS 20.0 program. Surveyed on Feb. 2016. Best top 10 regions of permission dental clinics are (1) Gangnam-gu, Seoul(1,337), (2) Seongnamsi, Gyeonggi-do(555), (3) Songpa-gu, Seoul(491), (4) Yeongdeungpo-gu, Seoul(472), (5) Suwon-si, Gyeonggi-do(443), (6) Seocho-gu, Seoul(428), (7) Nowon-gu, Seoul(417), (8) Goyang-si, Gyeonggi-do(413), (9) Jung-gu, Seoul(380), (10) Yongin-si, Gyeonggi-do(353). Whereas best top 10 regions of operating dental clinics are (1) Gangnam-gu, Seoul(581), (2) Seongnamsi, Gyeonggi-do(415), (3) Suwon-si, Gyeonggi-do(382), (4) Seocho-gu, Seoul(320), (5) Changwon-si, Gyeongsangnam-do(303), (6) Songpa-gu, Seoul(295) (7) Goyang-si, Gyeonggi-do(290), (8) Bucheon-si and Yongin-si, Gyeonggi-do(262), (9) Jeonju-si, Jeollabuk-do(224). Average population per one dental hospital & clinic by regional local government are 3,120 people. Best five region of population per one dental hospital & clinic are (1) Sejong-si(5,272), (2) Gangwon-do(4,653), (3) Chungcheongbuk-do(4,513), (4) Gyeongsangbuk-do(4,490), (5) Chungcheongnam-do(4,402). Average households per one dental hospital & clinic by regional local government are 1,316 households. Best three region of households per one dental hospital & clinic are (1) Sejong-si(2,126), (2) Gangwon-do(2,057), (3) Gyeongsangbuk-do(1,946). From 1946 to 1986, permission and operating dental hospital and clinics was steadily increasing. On 1986-1990, 1991-1995, permission, operation and closure of dental hospital and clinics increase rapidly. From the 2011-2015 to 2016(present), permission, operation and closure of dental hospital and clinics is decreasing. Average operating duration of closured dental hospital and clinics are 14.054 years. We need to map of dental hospital and clinics for open and operation of one, base on analyzed results. In an era of 30,000 dentist, we should to be concerned about operation of dental clinics in the light of past operating condition.

Prognostic Value of Tumor Regression Grade on MR in Rectal Cancer: A Large-Scale, Single-Center Experience

  • Objective: To determine the prognostic value of MRI-based tumor regression grading (mrTRG) in rectal cancer compared with pathological tumor regression grading (pTRG), and to assess the effect of diffusion-weighted imaging (DWI) on interobserver agreement for evaluating mrTRG. Materials and Methods: Between 2007 and 2016, we retrospectively enrolled 321 patients (male:female = 208:113; mean age, 60.2 years) with rectal cancer who underwent both pre-chemoradiotherapy (CRT) and post-CRT MRI. Two radiologists independently determined mrTRG using a 5-point grading system with and without DWI in a one-month interval. Two pathologists graded pTRG using a 5-point grading system in consensus. Kaplan-Meier estimation and Cox-proportional hazard models were used for survival analysis. Cohen's kappa analysis was used to determine interobserver agreement. Results: According to mrTRG on MRI with DWI, there were 6 mrTRG 1, 48 mrTRG 2, 109 mrTRG 3, 152 mrTRG 4, and 6 mrTRG 5. By pTRG, there were 7 pTRG 1, 59 pTRG 2, 180 pTRG 3, 73 pTRG 4, and 2 pTRG 5. A 5-year overall survival (OS) was significantly different according to the 5-point grading mrTRG (p = 0.024) and pTRG (p = 0.038). The 5-year disease-free survival (DFS) was significantly different among the five mrTRG groups (p = 0.039), but not among the five pTRG groups (p = 0.072). OS and DFS were significantly different according to post-CRT MR variables: extramural venous invasion after CRT (hazard ratio = 2.259 for OS, hazard ratio = 5.011 for DFS) and extramesorectal lymph node (hazard ratio = 2.610 for DFS). For mrTRG, k value between the two radiologists was 0.309 (fair agreement) without DWI and slightly improved to 0.376 with DWI. Conclusion: mrTRG may predict OS and DFS comparably or even better compared to pTRG. The addition of DWI on T2-weighted MRI may improve interobserver agreement on mrTRG.

Ethanol Electro-Oxidation and Stability of Pt Supported on Sb-Doped Tin Oxide (안티몬 도핑된 주석 산화물에 담지된 백금 촉매의 에탄올 산화 반응 및 안정성 연구)

  • Electrocatalytic activities and stabilities of Pt supported on Sb-doped $SnO_2$ (ATO) were examined for ethanol oxidation reactions. Pt colloidal particles were deposited on ATO nanoparticles (Pt/ATO) and the prepared electrocatalysts were characterized by X-ray diffraction, transmission electron microscopy (TEM), and cyclic voltammetry. Electrochemical activity of the Pt/ATO for ethanol electro-oxidation was compared to those of Pt supported on carbon (Pt/C) and commercial PtRu/C. The activitiy of the Pt/ATO was much higher than those of the Pt/C and commercial PtRu/C. The Pt/ATO exhibited much higher electrochemical stabilities than the Pt/C in 0.5M ${H_2}{SO_4}$ and in 0.5M ${H_2}{SO_4}$/1M ${C_2}{H_5}OH$. According to TEM, the growth rate of Pt particles was lower in the Pt/ATO than it was in the Pt/C. The ATO nanoparticle appears to be a promising support material that promotes electrochemical reactions and stabilizes catalyst particles in direct ethanol fuel cell.

A Recognition for community Based Rehabilitation by Public Health Center Physical Therapists in Seoul Province (서울시 보건소 물리치료사들의 지역사회중심재활에 대한 인식)

  • To survey the recognition of Community Based Rehabilitation(CBR) by Public Health Center(PHC) Physical Therapists in Seoul Province. This study analysed general characteristic of Physical Therapists and cognition, participation, education, problems and requirements for CBR. The subjects of this study were 31 therapists who work at PHC in Seoul Province and Seoul City Childerns Municipal Hospital. Data was collected for 18 days from April 13 to 31, 1998 and results computed from 25 responses. First, Female therapists outnumber males. The age category 30 to 35 old represented 44.0% of the total number studied. 48.0% of therapists had worked more than 7 years and therapists usually 84.0% worked from 3 to 4 years at the PHC. Junior colleage graduates were 72.0%, and respondents with family responsibility were 52.0%. Second, Recently pain control and therapeutic exercises has become the major requirement of patients at PHC(56.0%). If the offered expanded services, a rehabilitation programe at PHC(36.0%) was highly. When asked about a working guide book, nobody had one. Third, 64.0% of therapists were aware of the present level of CBR, 28.0% of them had gotten an information from newspaper and mass media. Fourth, For CBR, 72.0% had positive ideas in which they could participate. 56.0% wished to compose a team of various rehabilitation specialists. 52.0% wished for the meeting day to be flexible and 64.0% desired to meet once or twice a week. Fifth, Almost all(92.0%) desired more educational opportunity about CBR. Example, once or twice a year(48.0%) and training period of 4 to 7 day(44.0%). They wanted to learn theory and parctice together(88.0%) and 48.0% wanted national mutirehabilitation center as a educational managing organization. Sixth, The most important requirement for accomplishing CBR by Physical Therapists was job security and adequate number of therapists(48.0%). Required number of physical therapists in the PHC was 3(52.0%). The cooperative agency should be the distict govement(32.0%) and community welfare center of the disabled persons(32.0%). Factors inhibiting the execution of CBR were lack of physical therapists(56.0%), and equipments for pain control and for therapeutic exercises(68.0%).

Nano-emulsion Containing Parthenocissus tricuspidata Stem Extracts for Enhanced Skin Permeation and the Antibacterial Activity of the Extracts (피부 흡수 증진을 위한 담쟁이덩굴 줄기 추출물 함유 나노에멀젼 및 이의 항균활성 연구)

  • In a previous study, we investigated the antioxidative and cellular protective effects of Parthenocissus tricuspidata stem extracts. In this study, we prepared nano-emulsion containing P. tricuspidata stem extract to improve skin permeation. The particle size of the nano-emulsion using the microfluidizer was 302 nm. Its loading efficiency was over 86%. The size distribution of the nano-emulsion took a monodispersed form and the nano-emulsion was more stable than typical emulsion without using microfluidizer during a 2 week period. In vitro skin permeation study of nano-emulsion containing P. tricuspidata stem extracts was carried out using Franz diffusion cell. The 1,3-butylene glycol used as a control group had 32.59% skin permeation efficiency. The skin permeation efficiency of the nano-emulsion was 42.47%. Also, we observed the antibacterial activity of the ethyl acetate fraction on skin flora for prospective applications as a natural antimicrobial. The ethyl acetate fraction had antibacterial activities higher than methyl paraben on Staphylococcus aureus, and Bacillus subtilis. These results indicate that nano-emulsion containing P. tricuspidata stem extracts could possess valued applications in cosmetic formulations for improving skin permeation. Also, based on the antibacterial activities on skin flora, antioxidative and cellular protective effects shown in our previous study, we suggest that P. tricuspidata stem extracts could be used as functional cosmetic materials.

The Factors Affecting the Registration Rates for Emergency Department Based Post-suicidal Care Program (응급실 기반 자살 시도자 사후 관리사업의 등록률에 영향을 미치는 인자)

  • Purpose: The aim of this study was to investigate the independent factors associated with the registration rate for the community-based post suicidal care program in the emergency department (ED). Methods: This prospective observational study was conducted between March and December 2013 at the academic ED at the tertiary urban hospital. During the study period, the pre-designed registry was recorded. The variables examined included the following: patients' demographic data (Sex, age, address, type of insurance, marital status, level of education, and history of previous psychiatric disease), suicide-related data (suicidal methods, combined drink of alcohol and number of previous attempts), and management-related data (disposition at ED, physician's training level, etc.). Univariated and multivariated logistic regression analyses were performed for identification of factors affecting the registration rate for the community-based post suicidal care program. Results: A total of 163 suicides were included during the study period. Of these, 33 (20.2%) patients were registered in the post-suicide care program. Factors including a patient's address (OR: 14.92, 95% CI: 3.606-61.711), immediate intervention by psychiatric healthcare center (OR: 5.05, CI: 1.688-15.134), admissions in hospital (OR: 3.69, CI: 1.286-10.605), and history of previous psychiatric disease (OR: 3.52, CI: 1.216-10.201) showed significant association with registration for the program. Conclusion: The community-based post-suicidal care program, which is available 24 hours a day, should be operated in each district in order to increase the registration rate. Emergency physicians should actively consider the inpatient treatment program for suicidal patients and strongly recommend registration to the program, particularly for patients without previous history of psychiatric disease.

  • Background : Tumor necrosis factor(TNF) has been considered as an important candidate for cancer gene therapy based on its potent anti-tumor activity. However, since the efficiency of current techniques of gene transfer is not satisfactory, the majority of current protocols is aiming the in vitro gene transfer to cancer cells and re-introducing genetically modified cancer cells to host. In the previous study, it was shown that TNF-sensitive cancer cells transfected with TNF-$\alpha$ cDNA would become highly resistant to TNF, and the probability was shown that the acquired resistance to TNF might be associated with synthesis of some protective protein. Understanding the mechanisms of TNF-resistance in TNF-$\alpha$ cDNA transfected cancer cells would be an important step for improving the efficacy of cancer gene therapy as well as for better understandings of tumor biology. This study was designed to evaluate whether the levels of TNF receptor mRNA expression and soluble TNF receptor release from cancer cells are changed after TNF-$\alpha$ cDNA transfection. Method : We transfected TNF-$\alpha$ c-DNA to WEHI164(murine fibrosarcoma cell line), NCI-H2058(human mesothelioma cell line), A549(human non-small cell lung cancer cell line), ME180(human cervix cancer cell line) cells using retroviral vector(pLT12SN(TNF)) and confirm the expression of TNF with PCR, EUSA, MTT assay. Then we determined the TNF resistance of TNF-$\alpha$ cDNA transfected cells(WEHI164-TNF, NCIH2058-TNF, A549-TNF, ME180-TNF) and evaluated the TNF receptor mRNA expression with Northern blot analysis and soluble TNF receptor release with EUSA. Results : The TNF receptor mRNA expressions of parental cells and genetically modified cells were not significantly different. The soluble TNF receptor levels of media from genetically modified cells were lower than those from parental cells. Conclusion : The acquired resistance to TNF after TNF-$\alpha$ cDNA transfection may not be associated with the change in the TNF receptor and the soluble TNF receptor expression.

Oral Delivery of Probiotics Using pH-Sensitive Phthalyl Inulin Tablets

  • Probiotics show low cell viability after oral administration because they have difficulty surviving in the stomach due to low pH and enzymes. For the oral delivery of probiotics, developing a formula that protects the probiotic bacteria from gastric acidity while providing living cells is mandatory. In this study, we developed tablets using a new pH-sensitive phthalyl inulin (PI) to protect probiotics from gastric conditions and investigated the effects of different compression forces on cell survival. We made three different tablets under different compression forces and measured survivability, disintegration time, and kinetics in simulated gastric-intestinal fluid. During tableting, there were no significant differences in probiotic viability among the different compression forces although disintegration time was affected by the compression force. A higher compression force resulted in higher viability in simulated gastric fluid. The swelling degree of the PI tablets in simulated intestinal fluid was higher than that of the tablets in simulated gastric fluid due to the pH sensitivity of the PI. The probiotic viability formulated in the tablets was also higher in acidic gastric conditions than that for probiotics in solution. Rapid release of the probiotics from the tablet occurred in the simulated intestinal fluid due to the pH sensitivity. After 6 months of refrigeration, the viability of the PI probiotics was kept. Overall, this is the first study to show the pH-sensitive properties of PI and one that may be useful for oral delivery of the probiotics.
