Combined Genome Mapping of RFLP-AFLP-SSR in Pepper

  • We have constructed a molecular linkage map of pepper (Capsicum spp.) in an interspecific $F_2$ population of 107 plants with 320 RFLP, 136 AFLP, and 46 SSR markers. The resulting linkage map consists of 15 linkage groups covering 1,720 cM with an average map distance of 3.7 cM between framework markers. Most RFLP markers ($80\%$) were pepper-derived clones and these markers were evenly distributed all over the genome. Genes for defense and biosynthesis of carotenoids and capsaicinoids were mapped on this linkage map. By using 30 primer combinations, AFLP markers were generated in the $F_2$ population. For development of SSR markers in Capsicum, microsatellites were isolated from two small-insert genomic libraries and the GenBank database. This combined map provides a starting point for high-resolution QTL analysis, gene isolation, and molecular breeding.

  • Adult spinal deformity (ASD) is one of the most challenging spinal disorders associated with broad range of clinical and radiological presentation. Correct selection of fusion levels in surgical planning for the management of adult spinal deformity is a complex task. Several classification systems and algorithms exist to assist surgeons in determining the appropriate levels to be instrumented. In this study, we describe our new simple decision making algorithm and selection of fusion level for ASD surgery in terms of adult idiopathic idiopathic scoliosis vs. degenerative scoliosis.

  • X-chromosome inactivation is one of the most complex events observed in early embryo developments. The epigenetic changes occurred in female X-chromosome is essential to compensate dosages of X-linked genes between males and females. Because of the relevance of the epigenetic process to the normal embryo developments and stem cell studies, X-chromosome inactivation has been focused intensively for last 10 years. Initiation and regulation of the process is managed by diverse factors. Especially, proteins and non-coding RNAs encoded in X-chromosome inactivation center, and a couple of transcription factors have been reported to regulate the event. In this review, we introduce the reported factors, and how they regulate epigenetic inactivation of X-chromosomes.

Regulation of fpr Gene Encoding NADPH : Ferredoxin Oxidoreductase by the soxRS Locus in Escherichia coli

  • We isolated a promoter inducible by paraquat, a superoxide-generating agent, from Escherichia coli using a promoter-probing plasmid pRS415. From sequence analysis we found out the promoter is for fpr ENCODING nadph : ferredoxin oxidoreductase. We constructed on operon fusion of lacZ gene with fpr promoter to monitor the expression of the gene in the single-copy state. LacZ expression generators, menadione and plumbagin, also induced the expression of .betha.-galactosidase in the fusion strain. On the other hand, no significant induction was observed by treatment with hydrogen peroxide, ethanol, and heat shock. Induction of .betha.-galactosidase was significantly reduced by introducing a .DELTA. sox 8 :: cat of soxS3 :: Tn10 mutation into the fusion strain, indicating that fpr gene is a member of the soxRS regulon. The transcriptional start site was determined by primer extension analysis. Possible roles of fpr induction in superoxide stress were discussed.

  • Due to the many attractive aspects of DG in the future power distribution system, distribution automation will be a center hub of integration of the distribution system and resources to satisfy the various needs of customers in a competitive and deregulated environment. In this paper, operation strategies are presented which use network reconfiguration of the automated distribution systems with DG as a real-time operation tool for loss reduction and service restoration from the view of distribution operation. The algorithms and operation strategies of an automated distribution system with DG are introduced to achieve the positive effects of DG in distribution systems. A simple case study shows the effectiveness of the proposed operation strategies.

  • To investigate the biosynthetic pathway leading to artemisinin, the putative precursors, arteannuic acid and 11,12-dihydroarteannuic acid, were incubated in a cell-free system. For the experiment with dihydroarteannuic acid, artemisinin was generated in tumor homogenate. These results showed that dihydroarteannuic acid could be converted to artemisinin enzymatically. However, the experimental condition failed to convert arteannuic acid into 11,12-dihydroarteannuic acid.

  • Purpose: The purpose of this study was to describe hospital nurses' lived experience of being a preceptor in Korea. Methods: The data were collected in three focus group interviews with 15 nurses in one general hospital. The phenomenological analytic method suggested by Colaizzi was used to analyze the data. Results: Five theme clusters and 11 themes emerged from the analysis. The five themes include "inevitable rite of passage for an authentic nurse", "facing new role and situations that were difficult to handle", "preceptee-centered preceptorship", "healing connection with colleague nurses", and "enhanced status with competence". Conclusion: The findings of the study provide deep understanding on hospital nurses' positive as well as negative experiences of being a preceptor and should inform the development of a more integrative preceptorship program to empower both preceptees and preceptors.

  • The objective of this paper is to suggest structural design for the long-life housing apartment complex to save the construction cost. The key is to use unavailable underground space due to bearing walls or bad configuration of columns in apartments as the parking space. Therefore, the structural plan of apartment buildings considering the parking section in the underground should be designed. After analytical resutls of three cases, it is significant effect in saving construction cost.

  • Amylases catalyze the hydrolysis of starch material and play central roles in carbohydrate metabolism. The structure and a size exclusion column chromatography proved that the enzyme is a dimer in solution. The N -terminal segment of the enzyme folds into a distinct domain and comprises the enzyme active site together with the central (${\alpha}$/ ${\beta}$)$\sub$8/ barrel of the adjacent subunit.(omitted)

  • The voltage drop in distribution path of battery-reserved DC power system can affect the total of battery, cable and electricity costs. To determine an optimum voltage drop in distribution path for minimizing the total cost, battery, cable and electricity costs are represented as a function of the voltage drop, respectively, and are summed up to the total cost. An optimum voltage drop is selected as the value giving the minimum total cost. In this paper, a design technique of optimum voltage drop in distribution path of DC power system is proposed to minimize the total of battery, cable and electricity costs. The design procedure is described and design curve for selecting optimum voltage drop is also presented as a function of distribution distance.
