Studies on genetic monitoring of inbred mice in conventional breeding unit (일반사육시설 마우스의 유전적 오염에 대한 실태조사 연구)

  • These studies were carried out to survey the genetic contamination of six inbred mice (A, BALB/c, C3H, C57BL/6, CBA and KK) produced and supplied from the conventional breeding unit for improving the quality of mice as experimental animal. We examined alleles of five loci (Akp-1, Car-2, Hbb, Es-1 and Trf) by the use of biochemical markers with celluose acetate electrophoresis. As the results of test, BALB/c, A, C3H, C57BL/6, CBA and KK showed standard alleles in Akp-1, Car-2 and Hbb. But Es-1 of A and C57BL/6 and Trf of A, C3H, C57BL/6 and CBA did allelic divergence in loci. These results suggest that the colonies of A, C3H, C57BL/6 and CBA were genetically contaminated. Therefore, we recommend to eliminate the genetically contaminated mice thoroughly, to check on genetic monitoring regularly and to consider a counterpaln for improving the quality control as soon as possible.

Modified Cellulose for Heparin Binder (헤파린 제거제용 셀룰로오스의 개질)

  • Heparin binders (Cell-PALA) used for selective heparin removal from blood, were prepared by immobilizing a cationic polymer, poly(allylamine) (PALA), onto cellulose substrate by a novel method. Their absorbing capacity for heparin was compared with untreated cellulose control using heparin solution in vitro. The surface areas of obtained heparin binders and untreated cellulose were 1.36 and ($2.56{\mu} g$/$cm^2$, respectively. The amount of bound heparin to PALA immobilized celluloses was determined to be 0.16 - $0.30{\mu}g$/cm, which is much higher than that of untreated cellulose ($0.03{\mu} g$/$cm^2$). These results suggest that Cell-PALA materials can be utilized for a heparin removal system.

  • In order to successfully meet the environmental and recycling problems, natural polymer and their derivatives are recognized as a promising biodegradable material. In this study, the biodegradable composites of cellulose diacetate and starch were prepared, and their physical and thermal properties were investigated. For the melting processing, triacetine was added as a plasticizer into the composites. The processability of cellulose diacetate was further enhanced by increasing the amount of starch in the composites. The tensile stress and Young's modulus were decreased and elongation was increased with increasing the amount of starch in them. A $T_g$ value was decreased with increasing the amount of starch in the composites. Also, the morphology of the composites were observed with the SEM.

  • Four Taro isolectins (I, II, III, IV) were purified by ammonium sulfate, chromatography on CM-celluose and isoelectric focusing. I and IV lectins proved homogeneous by disk polyacrylamid gel electrophoresis and densitometric patterns. But in the presence of urea, IV lectin further dissociated into two different subunits. These lectinis had different hemagglutinating activities and inhibition in their activities after mixed with pepsin particuclary, but not with carbohydrates, heating pH, urea, guanidine, trypsin, pronase and $Ca^{2+}$.

  • The dyeing on cellulose fibers such as cotton and viscose viscose rayon was carried out by the use of solution extracted from natural fresh leaves of indigo plant under several dyeing conditions. The dyeing affinity of viscose rayon was higher than that of cotton. The total K/S value of dyed fabrics was considerably increased with repeated dyeings, while a dyeing time has a little influence on it. Both fibers were hardly dyed by indigo at $60^{\circ)$ and cotton was dyed better at $40^{\circ)$ than at $20^{\circ)$, but in viscose rayon, a little difference of total K/S was shown between $20^{\circ)$ and $40^{\circ)$. The color change of dyed fabric according to dyeing conditions was evaluated by the CIELAB color system. viscose rayon had a lower $b^*$ so that it looked bluer than cotton and when the celluose fibers were dyed by indigo plant at the lower temperature, the bluer it looked. By repeated dyeings the $b^*$value of dyed fabrics was much increased but the $a^*$ value was little influenced and in case of viscose rayon the change was considerable.

  • The biodegradable Properties of various polyester resins with different chemical structures have been studied by applying the controlled compost test and soil burial test. Celluose was taken as a fully biodegradable reference resin while PVC and PE were empolyed as non-biodegradable reference chains or ester group were rather easily degraded by hydrolase, meanwhile copolymer type polyesters which contain aromatic rings showed relatively low biodegradability. According to the results from controlled compost test, cellulose(the positive reference) showed 70.6% degradation after 45 days, whereas synthetic poly(butylene adipate-co-succinate), poly(butylene succinate), poly(butylene adipate-co-succinate-co-terephthalate) showed 44.0%, 32.0% and 23.4% degradation respectively. In this regard, it was concluded that biodegradable properties of polymers are largely dependant on the chemical structures constituting the polymers.

Antioxidative Materials in Domestic Meju and Doengjang 3. Separation of Hydrophilic Brown Pigment and Their Antioxidant Activity (재래식 메주 및 된장중의 항산화성 물질에 관한 연구 3. 수용성 갈변물질의 항산화력)

  • Hydrophilic brown pigments(HBPs) produced during fermentation and aging the domestic Meju and Doenjang were separated by dialysis and chromatography , and their antioxidant activity were measured . The chemical properties of HBP were determined by UV and IR spectrophotometry. HBPs contents were found to be 93.1 mg/g and 183.2mg/g in Meju fermentated for 30 days and 80 days , respectively. The ratio of dialysate to diffusate of the HBPs were appeared to be 70 : 30 and 87 :13 in the Meju fermented for 30 days and 80 days, respectively. and the rtio in the Doenjang aged for 60 days was 91 :9 , indicating that dialysate slowly days, respectively, and the ratio in the Doenjang aged for 60 days was 91 : 9, incidatin that dialysate slowly increased by the fermentation . Both portion exhibited strong antioxidant activity, but more stronger antioxidant activity was found in the dialysate. DEAE-celluose column chromatography showed that dialysate contained more materials eluted by 0.01-0.03M HCI solution than 0.01M acetate buffer, but diffusate showed opposite results. The degree of browning reaction and antioxdiant activity found in the fraction eluted by HCI solution was stronger than that of the fraction eluted by acetate buffer. The fraction eluted from DEAE-cellulose column chormatogrphy was further fractionaged by TLC and found that strong antioxidant activity was present in the fractions which did not possess fluorescenece and showed a negative ninhydrin reaction. TLC fractions of HCI eluant in Meju exhibited a strong absorbance at 260-280nm, but most of other fractions did not show any absorbance at UV region. TLC fractions from dialysate and diffusate showed fairly identical IR spectrum with absorbance at 3400cm-1, 2800cm-3000cm -1, 1600cm, -1 1400cm-1 and 1100 cm-1 , however, in addition to these absorbances, the spectrum from HCI eluant of Meju exhibited a strong absorbance at 1750cm , indicating the carbonic acid or carbonate ester.

Purification and Characterization of a Fibrinolytic Enzyme form Bacillus sp. KDO-13 Isolated from Soybean Paste

  • A microorganism producing fibrinolytic enzyme was isolated from Korean traditional soybean paste and identified as Bacillus sp. KDO-13. The fibrinolytic enzyme was purified to homogeneity by ammonium sulfate fractionation, ion-exchange chromatography on DEAE-celluose, and gel chromatography on Sephadex G-100 of the culture supernatant of Bacillus sp. KDO-13. The molecular weight of the purified enzyme was estimated to be 44,000 by SDS-PAGE. The optimum pH and temperature for the enzyme activity were pH 8.0 and $50{\circ}C$, respectively. The enzyme activity was relatively stable at pH 7.0-9.0 and temperature below $50{\circ}C$. the activity of the enzyme was inhibited by $AI^{3+}$ and $Hg^{2+}$, but activated by $Co^{2+}$\;and\;Ni^{2+}. In addition, the enzyme activity was potently inhibited by EDTA and 0-phenanthroline. The purified enzyme could completely hydrolyze a fibrin substrate within 6 h in vitro, and had a low $K_m$ value for fibrin hydrolysis. It was concluded that the purified enzyme was a metalloprotease with relatively high specificity for fibrinolysis, and thus, could be applied as an effective thrombolytic agent.

  • The fed-batch process which combinded high selectivity of affinity chromatography and membrane process was developed. The mixture of trypsin and chymotrypsin, having almost the same molecular weight and the chemical structure, were used as model enzymes. The water soluble polymer having more affinity for trypsin and celluose acetate membrane gelated in 50vol.% ethanol for removing free enzymes and retentating trypsin-affinity polymer complex simutaneously were used in this system. The membrane pore size was controlled by ethanol concentration in the gellation bath, and the affinity polymer was prepared by polymerization of acrylamide with N-acryloyl-m-aminobenzamidine at $4^{\circ}C$. The trypsin could be effectively concentrated by utilizing an affinity polymer and a prepared UF-50 ultrafiltration membrane. As a result, 86% purity trypsin was recovered by the current purification process.

  • A cellulose-degrading enzyme from Ganoderma lucidum was partially purified by ammonium sulfate precipitation and its enzymatic properties were studied. The enzyme had an optimum pH for activity at 4.0, and its stability range was pH $4.0{\sim}7.0$. The optimum temperature was $55^{circ}C$ and the enzyme retained 80% original activity after heated at $50^{\circ}C$ for 60 min. The activation energy of the enzyme for CMC degradation was caculated and found to be 6.2 Kcal/mole. The enzyme was activited by the addition of $Co^{++},\;Mn^{++}$, but slightly inactivated by $Hg^{++}$. Various enzyme inhibitors and chemical reagents did not affect the enzyme activity. The enzyme acted on native celluose as well as CMC. The Michaelis constant for CMC was calculated to be 2.4 mg glucose ep/ml.
