This study was performed to compare the growth rates, cellular fatty acid compositions and morphology by using electron microscope of Escherichia coli (E. coli) grown in various conditions including different concentrations of phenol, CdCl₂ and HgCl₂. Ninety eight E. coli strains were isolated from Naktong river and human feces. The content of unsaturated fatty acids, especially 16:1ω7c and 18:1ωc increased as the concentration of phenol and CdCl₂ increased. The content of unsaturated fatty acid increased up to 50 ppb of HgCl₂, but decreased at 75 ppb of HgCl₂. There were more unsaturated fatty acids than saturated fatty acid in the presence of toxic substances. However, the ration was reversed when the affected E. coli was transferred to toxic substance free fresh trypticase soy broth medium. Also, by using transmission electron microscope these cells were observed to various morphological deformation by heavy metals and their deposition on the surface. From these results, we suggested that the changes of major fatty acids composition and morphology of E. coli may be considered to indicate contaminated levels of heavy metals or organic solvents. The information presented here may be useful in predicting effects of heavy-metal and organic solvent contamination in streams and provides a basis for further studies of metal or organic solvent effects on microbial communities.
The present study was designed to investigate cellular regulation of phosphate metabolism between catabolically repressed and derepressed states in yeast (Saccharomyces uvarum). The activities of various phospatases and the contents of phosphate compounds were detected according to the culture phase and various phosphate concentrations. As the results, Saccharomyces uvarum derepressed many phosphate metabolizing enzymes such as alkaline phosphatase, acid phosphatase and ATPase more than ten fold simultaneously during catabolic repression (phospgate and sugar starvation). At the same state, the amounts of orthophosphate, nucleotidic labile phosphate and acid soluble polypgosphate were increased, compared to basal levels of normally cultivated cells. $Mg^{++}-stimulated$ type among all phospatases was appeared to have most of the enzyme activity. It could be postulated that $K^+ -stimulated$ alkaline phosphatase was directly or indirectly correlated with the synthesis of acid insoluble polyphosphate $Mg^{++}-stimulated$ phosphatase with the degradation of polyphosphates. In case of cultivation in the medium supplemented with sugar and phosphate (catabolic derepression), phospgatase activities except for alkaline phosphatase were decreased rapidly through the progressive batch culture, After 12 hrs culture, at early exponential phase, the cellular accumulation of acid insoluble polyphosphate increased about 5 fold, compared to those of the starved cells. Under catabolic repression, it could be postulated that intracellular phosphate metabolism was regulated by derepressions of phosphatases. The function of polyphosphate system was shown to compensate the ATP/ADP system as phosphate donor and energy source especially during catabolic repression.

Quinolone antibiotics are DNA gyrase inhibitors, but their bactericidal action seems to involve more than the inhibition of DNA gyrase activity. Hence, the potentially crucial factors among possible mechanisms of quinolone action; cleavable complex formation, inhibition of DNA synthesis, and induction of SOS response were investigated. These parameters were measured in an Escherichia coli strain exposed to quinolones in the logarithmic growth phase, and correlated with the bactericidal activity of quinolones. Cleavable complex formation proved to be the factor most related to bactericidal action. Inhibition of DNA synthesis was substantially correlated with bactericidal activity, but induction of SOS response was least correlated with bactericidal activity. Therefore, it was concluded that quinolones exert bactericidal action primarily through cleavable complex formation, and subsequent unknown cellular processes together with inhibition of DNA synthesis contribute to the bactericidal activity of quinolones.
Paljintanggagmbang has been used for cure of tumor as a traditional medicine without any experimental evidence to support the rational basis for its clinical use. This study was carried out to evaluate. the possible therapeutic or antitumoral effects of Paljintanggagmbang extract against tumor, and to carry out some mechanisms responsible for its effect. Experimental studis were performed for measurement of Humoral and Cellular Immune Response and Phagocytosis in Mice treated with mitomytion C(MMC) and Paljintanggagmbang alone and combination. The results obtained in this study were as follows 1. The adminstration of Paljintanggagmbang extract decresed size of tumors cell which MCA induced. 2. The adminstration of Paljintanggagmbang extract decresed growth of the tumors which S 180 transplant. 3. The adminstration of Paljintanggagmbang extract decresed reproduction of A549, Hep3b, 3LL cell and S 180 in vivo. 4. The adminstration of Paljintanggagmbang extract incresed activity of the NK cell. These results also suggested that effect of Paljintanggagmbang might be chiefly due to nonspecific enhancement of Humoral and Cellular Immune Response and Phagocytosis in Mice treated with MMC and Paljintanggagmbang alone and combination.
The non-viral delivery of genes into macrophages, known as hard-to-transfect cells, is a challenge. In this study, the microporation of a CpG-free and small plasmid (pCGfd-GFP) showed high transfection efficiency, sustainable transgene expression, and good cell viability in the transfections of Raw 264.7 and primary bone marrow-derived macrophages. The non-viral method using the pCGfd vector encoding anti-EGFR single-chain Fv fused with Fc (scFv-Fc) generated the macrophages secreting anti-EGFR scFv-Fc. These macrophages effectively phagocytized tumor cells expressing EGFR through the antibody-dependent mechanism, as was proved by experiments using EGFR-knockout tumor cells. Finally, peri-tumoral injections of anti-EGFR scFv-Fc-secreting macrophages were shown to inhibit tumor growth in the xenograft mouse model.
Gammaherpesvirus (${\gamma}HV$) infection of the central nervous system (CNS) has been implicated in diverse neurological diseases, and murine ${\gamma}HV$-68 (MHV-68) is known to persist in the brain after cerebral infection. The underlying molecular mechanisms of persistency of virus in the brain are poorly understood. Here, we characterized a unique pattern of MHV-68 persistent infection in neuroblastoma cells. On infection with MHV-68, both murine and human neuroblastoma cells expressed viral lytic proteins and produced virions. However, the infected cells survived productive infection and could be cultured for multiple passages without affecting their cellular growth. Latent infection as well as productive replication was established in these prolonged cultures, and lytic replication was further increased by treatment with lytic inducers. Our results provide a novel system to study persistent infection of ${\gamma}HVs$ in vitro following de novo infection and suggest application of MHV-68 as a potential gene transfer vector to the brain.
Design of settlement space is a complicated process while reasonable spatial layout bears great significance on the development and resource allocation of a settlement. The study proposes a weighted L-system generation algorithm based on CA (Cellular Automation) model which tags the spatial attributes of cells through changes in their state during the evolution of CA and thus identifies the spatial growth mode of a settlement. The entrance area of the Caidian Botanical and Animal Garden is used a case study for the model. A design method is proposed which starts from the internal logics of spatial generation, explores possibility of spatial rules and realizes the quantitative analysis and dynamic control of the design process. Taking a top-down approach, the design method takes into account the site information, studies the spatial generation mechanism of settlements and further presents a engine for the generation of multiple layout proposals based on different rules. A optimal solution is acquired using GA (Genetic Algorithm) which generates a settlement spatial layout carrying site information and dynamically linked to the surround environment. The study aims to propose a design method to optimize the spatial layout of the complex settlement system based on parametric generation.
Bone morphogenetic proteins (BMPs), belonging to the transforming growth factor-${\beta}$ superfamily, regulate many cellular activities including cell migration, differentiation, adhesion, proliferation and apoptosis. Use of recombinant human bone morphogenic protein-2 (rhBMP-2) in oral and maxillofacial surgery has seen a tremendous increase. Due to its role in many cellular pathways, the influence of this protein on carcinogenesis in different organs has been intensively studied over the past decade. BMPs also have been detected to have a role in the development and progression of many tumors, particularly disease-specific bone metastasis. In oral squamous cell carcinoma - the tumor type accounting for more than 90% of head and neck malignancies- aberrations of both BMP expression and associated signaling pathways have a certain relation with the development and progression of the disease by regulating a range of biological functions in the altered cells. In the current review, we discuss the influence of BMPs -especially rhBMP-2- in the development and progression of oral squamous cell carcinoma.
A key to the species which have been isolated from the Korean forest soils was provided, and three unrecorded species found in the forest soils of Mt. Halla were described and illustrated. Twenty dictyostelid cellular slime molds had been found in Korean forest soils. Among these, nine species were PG positive, and eleven species were PG negative. Three species described in this study were all PG positive. These were Dictyastelium aurea-stipes Cavender, Raper et Norberg vaL aurea-stipes (new strain HL-5), Dictyastelium monachasiaides Hagiwara, PalysPhandylium tenuissimum Hagiwara. Morphological features and growth habits of these species were examined and compared, and their distribution patterns were illustrated.trated.

Signal transducer and activator of transcription 3 (STAT3) is a transcription factor that regulates various cellular processes such as cell survival, angiogenesis and proliferation. In the present study, we examined that Cryptotanshione(CT), a tanshinone from oriental traditional medicinal herb Danshen (Salvia miltiorrhiza Bunge), had the inhibitory effects on hypoxia-mediated activation of STAT3 in androgen independent human prostate cancer PC-3 cells. CT inhibited the protein expression of hypoxia-inducible factor-1alpha (HIF-$1{\alpha}$) under hypoxic condition. Consistently, CT blocked hypoxia-induced phosphorylation and nuclear accumulation of STAT3. In addition, CT reduced cellular of vascular endothelial growth factor (VEGF), a critical angiogenic factor and a target gene of STAT3 induced under hypoxia. Of note, chromatin immunoprecipitation (ChiP) assay revealed that CT inhibited binding of STAT3 to VEGF promoter. Taken together, our results suggest that CT has anti-angiogenic activity by disturbing the binding STAT3 to the VEGF promoter in PC-3 cells.
