Background: Ionizing radiation causes cellular damage and death through the direct damage and/or indirectly the production of ROS, which induces oxidative stress. This study was designed to evaluate the in vivo radioprotective effects of a bio-active material coated fabric (BMCF) against ${\gamma}$-irradiation-induced cellular damage in Sprague-Dawley (SD) rats. Materials and Methods: Healthy male SD rats wore bio-active material coated (concentrations in 10% and 30%) fabric for 7 days after 3 Gy of ${\gamma}$-irradiation. Radioprotective effects were evaluated by performing various biochemical assays including spleen and thymus index, WBC count, hepatic damage marker enzymes [aspartate transaminase (AST) and alanine transaminase (ALT)] in plasma, liver antioxidant enzymes, and mitochondrial activity in muscle. Results and Discussions: Exposure to ${\gamma}$-irradiation resulted in hepatocellular and immune systemic damage. Gamma-irradiation induced decreases in antioxidant enzymes. However, wearing the BMCF-30% decreased significantly AST and ALT activities in plasma. Furthermore, wearing the BMCF-30% increased SOD (superoxide dismutase) and mitochondrial activity. Conclusion: These results suggest that wearing BMCF offers effective radioprotection against ${\gamma}$-irradiation-induced cellular damage in SD rats.
[6]-Gingerol, a major polyphenol of ginger (Zingiber officinale), exhibits a variety of biological properties including anti-oxidant, anti-inflammatory and anti-cancer activity. However, the radioprotective effect of [6]-gingerol is still unknown. The aim of this study was to investigate the radioprotective effect of [6]-gingerol against radiation-induced cell cytotoxicity and oxidative stress in HepG2 cells. [6]-Gingerol pretreatment attenuated radiation-induced cell cytotoxicity caused by 5Gy (half lethal dose, $LD_{50}$ of HepG2 cells). The measurements of superoxide dismutase (SOD) and catalase (CAT) activity were also performed. The results showed that [6]-gingerol pretreatment reduced increasing SOD and CAT activity after exposure of IR, indicating that [6]-gingerol protected oxidative stress by regulating cellular antioxidant enzyme (SOD and CAT) activity. These findings suggest that [6]-gingerol acts as a radioprotector by attenuating cell cytotoxicity and oxidative stress.
Effects of oxidative stress on the induction of apoptosis and the activity of antioxidant enzymes were investigated in HL-60 cells using $H_2O$$_2$and cisplatin which generate oxygen species in the cell. Various concentrations of oxidants were treated to cells and at different incubation time, cells were harvested for assays. Cell viability, morphology by propidium iodide staining and DNA fragmentation by agarose gel electrophoresis were observed to determine whether they induce apoptosis. The activity of antioxidant enzymes such as superoxide dismutase and catalase was also measured to evaluate the cellular response to the oxidative damage. The results are as follows: $H_2O$$_2$ induced apoptosis at 10 $\mu$M after 6h incubation, while it took 12h for cisplatin. Both oxidants induced the superoxide dismutase activity at a tolerable low concentration. However, at a concentration which causes apoptotic cell death, the enzyme level was dropped markedly at first and then recovered to the normal level after which it declined again, probably due to cell death. On the other hand, changes in the activity of catalase were not significant at most concentrations except the statistically significant decrease at 24h after 10 $\mu$M-$H_2O$$_2$treatment. In this study, $H_2O$$_2$- and cisplatintreated cells showed similar results in apoptotic response and enzyme activities, suggesting that anticancer activity of cisplatin may be related, at least in part, to the production of oxygen free radicals.
The biocide sodium hypochlorite (NaOCl) is widely used for controlling algal growth, and this application can be extended to marine environments as well. This study evaluates the biocidal efficiency and cellular toxicity of NaOCl on the harmful dinoflagellate Cochlodinium polykrikoides, with emphasis on pigment production and antioxidant enzyme activity. The test organism showed dose-dependent decrease in growth rate on exposure to NaOCl, and the 72 h $EC_{50}$ was measured to be $0.584mg\;L^{-1}$. NaOCl significantly decreased pigment levels and chlorophyll autofluorescence intensity, indicating possible detrimental effects on the photosystem of C. polykrikoides. Moreover, it significantly increased the activities of antioxidant enzymes, suggesting the production of reactive oxygen species in the cells. These data indicate that NaOCl exerted deleterious effects on the photosynthetic machinery and induced oxidative damage in the dinoflagellate and this biocide could be effectively used for the control of algal blooms.

The purpose of this study was to investigate the antioxidant, anti-inflammatory and anti-cellular adhesion molecules effects of Allii Macrostemonis Bulbus, Artemisiae Capillaris Herba, Curcumae Radix, Crataegi Fructus, Salviae Militiorrhizae Radix complex extract(AMCP) on the inhibition of atherosclerosis in HUVEC. We measured DPPH radical scavenging activity and ABTS radical scavenging activity of AMCP to evaluate its antioxidant effect. And we also measured the expression level of NF-κB, IκBα, ERK, JNK, p38 proteins to evaluate its anti-inflammatory effect. Lastly, we measured the expression level of MCP-1, ICAM-1, VCAM-1 mRNA and their level to evaluate its anti-celluar adhesion molecules. AMCP did not show any cytotoxicity in HUVEC within the concentraion tested except for a concentration of 400 ㎍/㎖. AMCP increased the DPPH radical scavenging activitiy and ABTS radical scavenging activity in HUVEC as the concentration of AMCP rises. AMCP significantly reduced NF-κB, IκBα, JNK, ERK and p38 protein expression in HUVEC compared to control group. AMCP significantly reduced MCP-1, ICAM-1, VCAM-1 gene expresion in HUVEC compared to control group. AMCP significantly decreased the levels of MCP-1, ICAM-1, VCAM-1 in HUVEC compared to control group. These results suggest that AMCP has effects on antioxidation, anti-inflammation and anti-cellular adhesion molecule, which helps the treatment and prevention of dyslipidemia and atherosclerosis.

Effects of mistletoe (Viscum album) extract and pueraria (Pueraria radix) extract on cellular NF-$textsc{k}$B activity were evaluated in human malignant keratinocytes (SCC-13) to elucidate the possible correlation of NF-$textsc{k}$B with antioxidant activity. The antioxidant activities of these natural extracts were examined in four different evaluation methods, i.e., lipid peroxidation value (POV) evaluation test, I,l-diphenyl-2-picrylhydrazyl radical (DPPH), nitric oxide (NO) scavenging test, and reducing power assay. Pueraria extract (0.5 mg) and mistletoe extract (5 mg) downregulated the cellular NF-$textsc{k}$B activation up to 35% and 10% compared to the control, respectively, although their effects were lower than the known NF-$textsc{k}$B downregulator, vitamin C (8.8 mg, 53%) in a cell-based NF-$textsc{k}$B activity assay system. In the POV test, relative antioxidant activities of mistletoe extract (86%) and pueraria extract (75%) were significantly higher than the known antioxidant, vitamin C (48%) at the same concentration (10 mg) and the degree of activity increased in a dose-dependent manner. Pueraria extract showed more potential radical scavenging activities than those of mistletoe extract evaluated in both DPPH and NO test. Especially, the NO radical scavenging activity of pueraria extract ($SC_{50}$/, 88 $\mu$g) was comparable to that of vitamin C ($SC_{50}$/, 77 $\mu$g). Even pueraria extract possessed a much less reducing power compared to vitamin C, it also revealed higher reducing power than that of mistletoe extract. These results indicate that mistletoe extract and pueraria extract may serve as an useful natural antioxidant agents, and led to suggest the hypothesis that compounds having an antioxidant activity, i.e., radical scavenging activity or reducing power may be correlated with the downregulation of NF-$textsc{k}$B activation in human keratinocytes.

As a part of an ongoing search for antioxidants from marine sources, antioxidant activities of 24 kinds of seaweeds (4 green algae, 8 brown algae, and 12 red algae) were investigated. The seaweeds were extracted by acetone/dichloromethane and methanol, respectively. The antioxidant properties of both extracts were evaluated using four different activity tests, including degree of occurrence of intracellular reactive oxygen species (ROS), NO, lipid peroxidation, and GSH (glutathione) in mouse macrophage Raw 264.7 cells. The levels of intracellular reactive oxygen species (ROS) and GSH were measured using 2',7'-dichlorofluorescin diacetate (DCFDA) and monobromobimane as fluorescence probe, respectively. Moreover, the generation of NO and lipid peroxidation products were determined by each method based on the Griess reaction and TBARS assay. Solvent extracts from seaweeds such as Scytosiphon lomentaria, Prionitis cornea, Laruencia okamurae, Callophyllis japonica, Sargassum horneri, Dictyopteris divaricata, Lomentaria catenata, Corallina confuse, Ishige okamurae, and Ahnfeltiopsis flabelliformi exhibited high antioxidant activities in cellular oxidizing systems.

The antioxidant activities of 29 components isolated from the aerial parts of Artemisia iwayomogi were evaluated in vitro and in cell culture. Among the tested compounds, 2, 6, 8, 10, 13, and 14 exhibited the greatest peroxyl radical-scavenging activities in the oxygen radical absorbance capacity (ORAC) assay, and 2, 10, and 14 also showed significant reducing capacities. However, all compounds showed weak metal chelating activities. Their cellular antioxidant activities were evaluated in HepG2 cells. At $10{\mu}M$, compounds 6, 8, and 14 exhibited stronger protection against 2,2'-azobis(2-amidinopropane) dihydrochloride (AAPH)-induced oxidative stress than compounds 2, 10, and 13. Moreover, Compounds 2 and 8 were more effective in protecting against $Cu^{2+}$-induced oxidative stress than compounds 6, 10, 13, and 14 at $10{\mu}M$. These results suggest that the phenolic compounds in A. iwayomogi have the potential to be developed as natural antioxidants for the treatment of oxidative stress-related diseases.
The purpose of this study was to purify antioxidant substances from steamed squid extract (SSE). The yield of SSE was 8% by dry weight. The approximate compositions of SSE proteins, lipids, moisture, carbohydrate and ash were 64.95%, 1.69%, 7.23%, 4.44% and 21.69%, respectively. The major amino acids in SSE were taurine (29.17%), glycine (20.33%), alanine (12.51%), and glutamic acid (9.83%). Antioxidant activities were evaluated using 1,1-diphenyl-2-picryl-hydrazyl (DPPH) radical scavenging activity, which was measured as 24.7% at 1.0 mg/mL. Four SSE fractions were isolated by Sephadex G-25 gel chromatography; the F2 fraction showed the highest DPPH radical scavenging activity. The F2 fraction was separated by reverse-phase high performance liquid chromatography (HPLC) using an octadecylsilane (ODS) column, yielding a purified antioxidant substance with a DPPH radical scavenging activity of 64.41% at 1.0 mg/mL, representing a 2.64-fold increase in the scavenging activity of SSE purified by the 3-step procedure. The amino acid compositions showed that purified SSE was rich in taurine, glycine, glutamic acid and alanine. The purified SSE significantly elevated 2',7'-dichlorodihydrofluororescein diacetate (DCFH-DA) fluorescence probe, which confirms its effective radical scavenging potential in cellular ROS. In addition, the SSE significantly inhibited oxidative damage of purified genomic DNA. These results suggest that a purified antioxidant substance from SSE can be used as a potential natural compound-based antioxidant in the functional food and pharmaceutical industries.
The protective activity of seolitae chungkukjang added with green tea against oxidative stress was investigated under the cellular systems using LLC-$PK_1$ cells. The treatment of 2,2'-azobis(2-aminopropane) dihydrochloride (AAPH) showed increase in lipid peroxidation, and decrease in endogenous antioxidant enzymes activity and cell viability. However, the methanol extract of seolitae chungkukjang inhibited lipid peroxidation by 58.3%, and increased cell viability up to more than 60%. In addition, it enhanced superoxide dismutase (SOD) and glutathione peroxidase (GSH-Px) activities. Seolitae chungkukjang improved oxidative stress-induced cellular injury through the radical scavenging activities. In particular, the addition of green tea in seolitae chungkukjang showed stronger effect against oxidative stress induced by AAPH. The more addition of green tea resulted in the greater antioxidative effect through elevation in activities of SOD and GSH-Px, and inhibition of lipid peroxidation, eventually leading to increase in cell viability. Theses results suggested that seolitae chungkukjang added with green tea have protective effects from cellular oxidative damage and could be considered as an application for the development of chungkukjang with functionality.
