Regulation of vascular smooth muscle cell (VSMC) phenotype plays an essential role in many cardiovascular diseases. In the present study, we provide evidence that $kr{\ddot{u}}ppel$-like factor 8 (KLF8) is essential for tumor necrosis factor ${\alpha}$ ($TNF{\alpha}$)-induced phenotypic conversion of VSMC obtained from thoracic aorta from 4-week-old SD rats. Stimulation of the contractile phenotype of VSMCs with $TNF{\alpha}$ significantly reduced the VSMC marker gene expression and KLF8. The gene expression of KLF8 was blocked by $TNF{\alpha}$ stimulation in an ERK-dependent manner. The promoter region of KLF8 contained putative Sp1, KLF4, and $NF{\kappa}B$ binding sites. Myocardin significantly enhanced the promoter activity of KLF4 and KLF8. The ectopic expression of KLF4 strongly enhanced the promoter activity of KLF8. Moreover, silencing of Akt1 significantly attenuated the promoter activity of KLF8; conversely, the overexpression of Akt1 significantly enhanced the promoter activity of KLF8. The promoter activity of SMA, $SM22{\alpha}$, and KLF8 was significantly elevated in the contractile phenotype of VSMCs. The ectopic expression of KLF8 markedly enhanced the expression of SMA and $SM22{\alpha}$ concomitant with morphological changes. The overexpression of KLF8 stimulated the promoter activity of SMA. Stimulation of VSMCs with $TNF{\alpha}$ enhanced the expression of KLF5, and the promoter activity of KLF5 was markedly suppressed by KLF8 ectopic expression. Finally, the overexpression of KLF5 suppressed the promoter activity of SMA and $SM22{\alpha}$, thereby reduced the contractility in response to the stimulation of angiotensin II. These results suggest that cross-regulation of KLF family of transcription factors plays an essential role in the VSMC phenotype.
In the present study, mouse follicular oocytes and 2-cell embryos(late -zygote stage embryos included) were cultured on the electric pad for electric stimulation in the culture incubator. In addition, follicular oocytes and embryos were tested for maturation and development under higher temperature condition($39^{\circ}C$).Mouse follicular oocyte maturation were not affected by anion electric stimulation and there is no significant difference in GBVD and MI between the control and experiment group after 4hr culture. In the embryo culture, it was found that more morula and blastocyst were found in the electric stimulation group rather than the control(96hr). This may seem to be caused with cytoplasmic $Ca^{2+}$ transient rise by electric stimulation(anion pad). On the other hand higher temperature incubation ($39^{\circ}C$) on the anion pad caused all the embryos degenerated within $12h{\sim}24hr$ culture. This was quite different from large animal embryos(bovine, pig, sheep), in which beneficial effect of high temperature incubation for oocyte maturation and embryo development were found.

Effects of TCDD and flavonoids on ethoxyresorufin deethylase in Hepa I cells and MCF-7 human breast cancer cells were examined. TCDD treatment have resulted in the stimulation of ethoxyresorufin deethylase activity based on fluorometry in Hepa I in dose and time dependent manner. 0.1 nM TCDD showed maximal stimulation of ethoxyresorufin deethylase activity and 24 hour treatment also showed maximal stimulation of ethoxyresorufin deethylase activity. In MCF-7 human breast cancer cells, untreated cells showed high basal level of ethoxyresorufin deethylase activity. TCDD treatment to MCF-7 cells resulted minor stimulation of ethoxyresorufin deethylase activity compared to that in Hepa I cells. Various chemicals were tested for ethoxyresorufin deethylase activity in both cell lines. Flavonoids, such as quercetin showed an inhibition of ethoxyresorufin deethylase activity that is stimulated with TCDD or 3-Methylcholanthrene. Estrogen and estrogen metabolites such as 16a-estriol also affects the ethoxyresorufin deethylase activity in MCF-7 cells.

Estrogen and estrogen metabolite such as 16 a-estriol and 2-hydroxyestradiol also affects the ethoxyresorufin deethylase activity in MCF-7 cells.
침자시 해당부위에서 발현되는 특수한 수용체에 대한 반응을 형태학적으로 연구하고자 흰쥐의 족삼리(足三里) 부위에 침자를 한 다음 경혈부위 조직내의 비만세포의 변화와 침자주변 세포의 미세구조 변화를 관찰하였다. 침자시 형성된 진피층의 동공주변에서는 비만세포들이 정상군에 비하여 유의성있는 종가(p<0.01)를 하였다. 비만세포의 세포질에는 전자밀도와 형태가 다른 과립들이 충만되어 있었을 뿐만 아니라 세포질 밖으로 분비된 과립들도 관찰할 수 있었다. 침자 결과 형성된 골격근육층의 근섬유들은 근원섬유 마디의 밝은띠가 정상군에 비하여 않게 관찰되는 것으로 보아 침자시 자극에 의하여 근육섬유들이 수축하였음이 밝혀졌다. 이상의 결과로 보아 침자에 의하여 유도되는 유효한 생체반응은 진피층에서 관찰되는 비만세포의 수와 미세구적인 변화는 침자에 의한 경혈기능과 밀접한 상관관계를 가지고 있을 것으로 사료되며, 또한 근섬유를 수축시키는 형태학적 변화가 유도되는 것으로 보아, 경혈 부위는 기계적 자극에 반응하는 특수한 수용체와 밀접한 관련이 있는 것으로 사료된다.

Nanotechnology, especially vertically grown silicon nanowires, has gotten great attentions in biology due to characteristics of one dimensional nanostructure; controllable synthetic structure such as lengths, diameters, densities. Silicon nanowires are promising materials as nanoelectrodes due to their highly complementary metal-oxide-semiconductor (CMOS) - and bio-compatibility. Silicon nanowires are so intoxicated that are effective for bio molecular delivery and electrical stimulation. Vertical nanowires with integrated Au tips were fabricated for electrical intracellular interfacing with PC-12 cells. We have made synthesized two types of nanowire devices; one is multi-nanowires electrode for bio molecular sensing and electrical stimulation, and the other is single-nanowires electrode respectively. Here, we demonstrate that differentiation of Nerve Growth Factor (NGF) treated PC-12 cells can be promoted depending on different magnitudes of electrical stimulation and density of Si NWs. It was fabricated by both bottom-up and top-down approaches using low pressure chemical vapor deposition (LPCVD) with high vacuuming environment to electrically stimulate PC-12 cells. The effects of electrical stimulation with NGF on the morphological differentiation are observed by Scanning Electron Microscopy (SEM), and it induces neural outgrowth. Moreover, the cell cytosol can be dyed selectively depending on the degree of differentiation along with fluorescence microscopy measurement. Vertically grown silicon nanowires have further expected advantages in case of single nanowire fabrication, and will be able to expand its characteristics to diverse applications.
Purpose: We tested whether repetitive transcranial magnetic stimulation (rTMS) improved recovery following spinal cord injury (SCI) in rats with transplantation of adipose tissue-derived stromal cells (ATSCs). Methods: Twenty Sprague-Dawley rats (200-250 g, female) were used. Moderate spinal cord injury was induced at the T9 level by a New York University (NYU) impactor. The rat ATSCs (approximately $5{\times}10^5$ cells) were injected into the perilesional area at 9 days after SCI. Starting four days after transplantation, rTMS (25 Hz, 0.1 Tesla, pulse width=$370{\mu}s$, on/off time=3 sec/3 sec) was applied daily for 7 weeks. Functional recovery was assessed using the Basso, Beattie, and Bresnahan (BBB) locomotor rating scale as well as pain responses for thermal and cold stimuli. Results: Both groups showed similar, gradual improvement of locomotor function. rTMS stimulation decreased thermal and cold hyperalgesia after 7 weeks, but sham stimulation did not. Conclusion: rTMS after transplantation of ATSCs in an SCI model may reduce thermal hyperalgesia and cold allodynia, and may be an adjuvant therapeutic tool for pain control after stem cell therapy in SCI.
경두개 직류전기자극(transcranial Direct Current Stimulation; tDCS)은 지각, 인지, 운동 등의 뇌기능 향상 및 발달 효과가 입증되며, 다양한 분야에서 활용 및 응용되는 비침습적 뇌자극술이다. tDCS 효과는 뇌의 해부학적 구조, 뇌의 노화 정도 등의 뇌신경활성화 특징에 따라 다르게 나타난다는 연구결과들이 보고되고 있다. 일주기 리듬(circadian rhythm)은 대략 하루 주기의 수면과 각성의 생리적 변화패턴을 의미하며 뇌신경활성화 상태는 일주기 리듬에 따라 다르게 나타난다. 일주기 유형(chronotype)은 하루 중에 발현되는 각성도의 크기에 따라 아침의 각성도가 큰 유형은 아침형으로 저녁의 각성도가 큰 유형은 저녁형으로 나누어진다. 본 연구는 일주기 리듬에 의해 변하는 뇌기능 특징이 tDCS 효과에 미치는 영향을 알아보고자 한다. 총 20명의 건강한 성인 대상으로 실험을 진행하였고, 참가자들은 일주기 유형을 분류하기 위해 아침형-저녁형 설문지에 의해 주간형(아침형, 중간형)과 야간형(저녁형)으로 분류했다. 본 실험은 Zoom 프로그램을 이용하여 참가자와 실험자가 온라인으로 만나서 실험을 진행했다. 실험이 확정된 참가자는 실험자로부터 뇌파 기기, 뇌파 데이터를 획득하는 앱이 있는 핸드폰, 핸드폰 거치대, 뇌자극 기기의 사용방법에 대한 설명을 듣고 기기를 테스트해보고 기기를 전달받았다. 기기사용의 어려움을 가진 2명의 참가자는 대면 실험을 진행하여, 실험자가 기기작동을 하여 실험에 참여했다. 일주기 리듬의 상태에 따른 뇌자극 효과를 알아보기 위해 1주일 간격으로 아침과 저녁에 실험했으며, tDCS 자극 전과 후의 신경활성화 반응의 차이를 뇌파를 이용하여 측정하였다. 뇌자극에 의한 뇌기능 변화를 확인하기 위해 자극 전의 뇌파와 자극 후 뇌파가 다른 패턴을 보이며 분류가 잘되는 지를 예측 정확도로 분석했으며, 뇌기능 특징 변화가 일주기 리듬과 일주기 유형에 따라 다르게 나타나는지 확인하기 위해 각 조건의 분류조건(아침/저녁, 주간형/야간형)에서 추출된 주요 EEG 특성을 비교했다. 54개의 뇌파 특성값을 추출하여 SVM(Support Vector Machine) 기계학습 알고리즘으로 분류 모델을 구축하였고, 구축된 모델을 Leave-One-Out 교차검증(Leave-One-Out Cross-Validation)을 사용하여 자극 전과 후의 뇌파 반응을 예측하는지 평가하였고, 분류예측모델의 주요 예측 인자를 확인하는 주요 특성 분석을 진행하였다. 아침과 저녁의 tDCS에 따른 뇌파 특징을 분류하는 예측 정확도는 모두 98%로 나타났으며, 주간형의 아침 자극 조건과 저녁 자극 조건의 예측 정확도는 92%와 96%이며, 야간형의 아침자극 조건과 저녁 자극 조건의 예측 정확도는 모두 94%로 나타났다. 아침 자극 전과 후의 뇌파를 분류하는 상위 3개의 주요 EEG 특성결과는 주간형과 야간형에 따라 다르게 나타났다. 주간형은 좌측 측두 두정엽과 전전두엽의 뇌파 특성값이 나타났으며, 야간형은 측두 두정엽의 뇌파 특성값들만 나타났다. 저녁 자극전과 후의 뇌파를 분류하는 상위 3개의 주요 EEG 특성 결과 또한 주간형과 야간형에 따라 다르게 나타났다. 주간형은 우측 측두 두정엽과 좌측 전두엽의 뇌파 특성값이 나타났으며, 야간형은 측두 두정엽과 전두엽의 뇌파 특성값이 나타났다. 이와 같은 연구결과는 일주기 리듬과 유형에 따라 아침과 저녁의 뇌기능 특징이 다르게 나타나서 뇌자극 효과가 다르게 나타날 수 있음을 확인한 결과이다. 본 연구의 결과는 효과적인 뇌자극을 위해 개인의 뇌신경 활성화 상태 및 특징에 따라서 뇌자극 프로토콜을 조정할 필요성을 제시한다는 데에 의의를 찾을 수 있다.
Stress fiber (SF) 변화는 세포외부의 결합인자와 세포 수용체와 결합후 리모델링을 위해 액틴골격에 신호를 전달하며 일어난다. 이 연관은 결합장소에서 기계적 활동과 신호전달활동을 조절하는 다양한 스케폴드들과 신호 전달자에 의해 매게된다. Heterotrimeric transmembrane lymphotoxin α1β2 (LTα1β2)는 용해성 homotrimeric LT α를 포함하는 tumor necrosis factor (TNF) 계로 림프조직을 구성하는데 중요한 역할을 한다. LTα1β2와 LTβR의 결합은 fibroblastic reticular cell (FRC)에서 신호전달을 촉발한다. Agonistic anti-LTβR antibody 단독 혹은 LTα 그리고 TNFα의 조합으로 LTβR 자극은 세포의 액틴과 형태적 변화를 보았다. Agonistic anti-LTβR antibody의 FRC에서 작용을 통한 세포골격 재배열이 myosin과의 관련성을 확인하기위해 myosin light chain kinase (MLCK)의 저해제인 ML-7과 myosin light chains (MLC)와 myosin phosphatase target subunit 1 (MYPT1)의 인산화에 대한 효과를 확인하였다. MLCK 저해는 액틴 세포골격 재배열과 세포형태 변화를 유도하였다. 또한, MLC와 MYPT1인산화가 LTβR 자극에 의해 줄어드는 것을 확인하였다. DNA chip 분석은 myosin and actin 구성선분이 전사체 수준에서도 줄어드는 것을 보였다. 결론적으로 LTβR 자극은 FRC에서 SF변화는 myosin과 관련되어 있다는 것을 제시한다.
