  • In order to increase the oxygen transfer in a bioreactor for animal cell culture, a penluorocarbon, $Flutec^R$ ppll was used in a modified Celligen $Celligem^{TM}$. Also, the effects of ppll on the hybridoma cell growth and on the production of monoclonal antibody were investigated. There was no harmful effect of ppl1 on the cell growth and on the production of monoclonal antibody.

  • The medium compositions of Alcaligenes eutrophus were optimized for increasing PHB productivity. It is very important to optimize the concentrations of inorganic salts and trace eleme- nts as well as carbon and nitrogen sources to maximize cell growth rate and productivity. The fed-batch culture of Alcaligenes eutrophus by dual feeding of ammonia water and glucose under optimized initial medium concentrations was carried out. Glucose was fed manually according to glucose consumption rate and ammonia water by pH-stat. The final cell concentrations and PHB content in 30 hours were 122 g/l and 65% of dry cell weight(yielding 79 g of PHB/l), respectively and 2.64 g/l/hr of PHB production rate was obtained.

  • A process for efficient recycle fed-batch culture was carried out to increase cell mass and spore production by Lactic acid bacteria isolated from Kimchi. A large quantity of cell mass obtained by feeding concentration of sugar in recycle fed-batch culture. When the high density of salt was created that the cell mass was come-down. In this study, cultured in different feeding concentration of sugar conditions. Lactic acid bacteria by recycle fed-batch culture was investigated in 2L working volume of fermenter, obtained the maximum cell mass was 15.17g/L.

  • The effects of sucrose solution on cell growth and berberine production were studied in Thalictrum rugosum cell cultures. Application of sucrose solution without any other components enhanced berberine accumulation significantly in spite of slower cell growth. At high sucrose concentration, cells became very compact and fourfold increase in specific berberine content was achieved. Optimum concentration of sucrose in plain water to maximize the productivity was found to be 8%. Time course experiment in 8% sucrose solution showed that more than 5 days were required to utilize the advantage of this system efficiently. Addition of vitamins, growth regulators, and inorganic salts into the solution was not effective in increasing berberine productivity.

  • Effect of Pluronic F-68, a nonionic surfactant, on the extracellular production of hGM-CSF in transgenic Nicotiαna tabacum cell suspension culture was investigated. The addition of 5 g/L Pluronic F-68 did not affect the cell growth but increased the extracellular production of hGM-CSF by two-fold. This may be due to the enhanced permeability of the cell membrane by the interaction between the Pluronic F-68 and the cell membrane.

  • Thousand actinomycetes and 50 soil samples were used for the isolation of microorganisms producing yeast cell wall lytic enzymes. Among 493 strains producing large clear zones on autolysed washed yeast (AWY), 117 strains were selected on living yeast cell agar plates. With the method of lytic activity, one strain (St-1702) was selected, which was temporarily identified as Streptomyces eurythermus. The optimal condition for enzyme production of this strain was partially determined as follows: incubation of the strain for 3 days at 30$\circ$C in the medium containing 2% freeze dried yeast cell, 1% glucose, 1% K$_{2}$HPO$_{4}$, 0.01% MgSO$_{4}$'7H$_{2}$O, 0.5% peptone, and 0.2% (NH$_{4}$)$_{2}$CO$_{3}$ with pH 7.0. The protoplast formation of yeast by using the enzyme produced by this strain was compared with commercial enzymes.

  • A possibility of utilizing Chinese cabbage, a kind of renewable resources which is frequently overproduced in Korea, for the production of single cell protein was investigated. Saccharomyces cerevisiae and Candida utilis grew well in cabbage juice producing 4.3 and 5.1 g/l of dried yeast cells, respectively. Freezing fresh cabbage prior to juice extraction did not affect the growth of yeasts and the final cell yield.

  • To evaluate the anticarcinogenic effect and its mechanism of red ginseng, the mice were treated with red ginseng and received subcutaneous Bl6 melanoma cell line injection on the back. Tumor incidence was same (100%) both in water and red ginseng-treated groups, but tumor production was delayed in red ginseng-treated group. Survival time was somewhat longer in red ginseng-treated group. The histopathological findings were similar in both groups, but lymphocytic infiltration around the tumor and melanin production in the tumor cells were prominent in the red ginseng-treated group. Flow cytometric analysis on T lymphocytes and natural killer cells revealed increased $T_H$/$T_S$ ratio and increased NK cells in red ginseng-treated group. These results suggest that the anticarcinogenic effect of red ginseng may be exerted by the increased cell-mediated immunity and natural killer cell activity.

  • Ginseng root explants and calli induced from selected cell lines were cultured on modified Murashige and Skoog's media supplemented with different concentrations of organic or inorganic compounds and plant growth requlators to clarify the effects of chemical composition and plant growth regulators in the medium on the growth of ginseng calli and the production of ginseng saponin. For optimum growth of calli, the concentrations of 2, 4-D and sucrose were the range of 1 to 3 mg/${\ell}$l and 1 to $3\%,$ respectively. And it was clarified that sucrose, nitrogen, phosphate, calcium, magmesian plant growth regulators and their concentrations influcenced the relative biosynthesis of saponin in tissue cultures of Panax ginseng. The patterns of ginsenosides, pharmacologically useful component, were different among the cell lines and contents of ginsenosides were much higher in selected cell lines than in original cell line.
