Bacteriocin Production by Streptococcus sp. J-C1 Isolated from Kimchi (김치에서 분리한 Streptococcus sp. J-C1의 bacteriocin 생산)

  • Streptococcus sp J-C1 producing bacteriocin was isolated from Kimchi. The optimum conditions for bacteriocin production by Streptococcus sp. J-C1 were evaluated. For the maximum yield of bacteriocin production by Streptococcus sp. J-C1, the cell should be harvested at the late stationary phase and the temperature, pH and NaCl concentration should be 25$\circ$C, pH 8 and without the addition of NaCl, respectively. Sucrose should be used as a carbon source and organic nitrogen such as peptone should be used as a nitorgen source for the best yield. The production of bacteriocin is related to the cell growth of Streptococcus sp. J-C1. The bacteriocin from Streptococcus sp. J-C1 was active for gram positive microorganisms such as Lactobacillus sp., Leuconoctoc sp., Lactococcus sp., Streptococcus mutans, Staphylococcus aureus amd Bacillus subtilis and also active for gram negetive bacteria such as Acetobacter aceti. Antibacterial activity of the bacteriocin was completely disappeared by protease treatment.

Digoxin Production by Using Biotransformation in Digitalis lanata Cell Suspension Cultures (Digitalis lanata 현탁세포배양에서의 생물학적 변환을 이용한 Digoxin 생산)

  • For the production of digoxin by using biotransformation in suspension-cultured Digita- lis lanata cells, a two-stage culture process was optimized. Modified Murashige and Skoog medium was used for growth in the first stage and the cells were transferred to glucose solution for the production of digoxin from digitoxin via biotransformation in the second stage. When the cells were cultivated for 10 days in the growth period, 12$\beta$-hydroxylation capacity was the best. It was found that the optimum amount of digitoxin as substrate was 400 mg/l with initial cell density of 21%. Maximum productivity was achieved 5 days after transfer of cells to production medium. Sucrose and fructose provided similar digoxin yield as that in glucose, and 6% was proved to be the best glucose solution. Most of the components of modified MS medium except phosphate reduced the efficiency of digoxin formation. Besides, peptone and beef extracts inhibited 12$\beta$-hydroxylation, while promoting glucosylation. Finally, it was apparent that light enhanced the formation of digoxin significantly.

  • Callus was derived from the leaves of Scutellaria baicalensis Georgi. The productions of baicalin in the cultivated callus under various conditions, especially, the effects of the light sources and temperature were studied. In this experiment, the callus cultivated at $25^{\circ}C$ showed higer production of baicalin than the callus cultivated at $21^{\circ}C$ and $23^{\circ}C$. The illumination of light(fluorescence and UV) accelerated generally the growth of callus and the production of baicalin during the cultivation for three weeks. But, the illumination of light more than three weeks reduced the rate of production of baicalin in the callus.

  • In order to investigate the possible application of immobilized yeast cells in sparkling wine production instead of riddling puns by the traditional method, fermentation characteristics were tested during the sparkling wine fermentation in the bottle using immobilized yeast cells with alginate. The rates of sugar consumption and alcohol production were faster with free cells than those with immobilized cells during the fermentation. The higher concentration of yeast cells and the lower concentration of alginate in the cell immobilization resulted in the faster sugar consumption and alcohol production. It also resulted in the increase of yeast cell concentration released from immobilized beads during the fermentation. However, no differences were shown in the contents of alcohol, residual sugar and CO2 pressure after fermentation. In case concentration of yeast cells released from immobilized beads during bottle fermentation, the higher concentration of alginate had and the lower had.

Effect of Acetic Acid on Bacteriocin Production by Gram-Positive Bacteria

  • Acetic acid is indirectly involved in cell center metabolism, and acetic acid metabolism is the core of central metabolism, affecting and regulating the production of bacteriocin. Bacteriocin is a natural food preservative that has been used in the meat and dairy industries and winemaking. In this paper, the effects of acetic acid on bacteriocin produced by Gram-positive bacteria were reviewed. It was found that acetic acid in the undissociated state can diffuse freely through the hydrophobic layer of the membrane and dissociate, affecting the production, yield, and activity of bacteriocin. In particular, the effect of acetic acid on cell membranes is summarized. The link between acetic acid metabolism, quorum sensing, and bacteriocin production mechanisms is also highlighted.

  • Objectives : This experiment was designed to investigate the efficacy of DDTCMT hot water extract & ultra-fine powder on Alzheimer's Disease Model. Methods : The effects of the DDTCMT hot water extract on expression of IL-$1{\beta}$, IL-6, TNF-${\alpha}$, COX-2, NOS-II, IL-10, IL-1 receptor antagonist mRNA and production of IL-$1{\beta}$, IL-6, TNF-${\alpha}$ in BV2 microglial cell line treated by lipopolysacchaide(LPS) were investigated. Expression of NO, ROS in BV2 microglial cell line treated by LPS and AChE activity in PC-12 cell treated by NGF were investigated. anti-AChE was observed through Western blot analysis. The effects of the DDTCMT hot water extract & ultra-fine powder on the behavior of the memory deficit mice induced by scopolamine were investigated. Results : 1. The DDTCMT hot water extract significantly decreased the production of mIL-6, mNOS-II, mTNF-${\alpha}$, and increased the production of mIL-10, mIL-1 receptor antagonist. 2. The DDTCMT hot water extract significantly suppressed the production of IL-$1{\beta}$, IL-6, TNF-${\alpha}$ in BV2 microglial cell line treated by LPS. 3. The DDTCMT hot water extract significantly suppressed the NO and ROS production in BV2 microglial cell line treated by LPS. 4. The DDTCMT hot water extract groups showed inhibition of AChE activity in NGF treated PC-12 cell line. 5. The DDTCMT hot water extract suppressed anti-AChE expression in NGF treated PC-12 cell line was observed by Western blot analysis. 6. The DDTCMT hot water extract & ultra-fine powder groups showed significantly inhibitory effect on the scopolamine -induced impairment of memory in the experiment of Morris water maze. Conclusions : These results suggest that the DDTCMT hot water extract & ultra-fine powder may be effective for the prevention and treatment of Alzheimer's disease.

Studies on Microbial Utilization of Agricultural Wastes (Part IV) Cellulosic Waste Materials as Substrate on the Production of Cellulosic Single Cell Protein. (농산폐자원의 미생물학적 이용에 관한 연구 (제육보) 섬유소단세포단백 생산에서의 천연기질의 이용성)

  • Experiments were carried out to pursue the availability and the feasibility of utilizable cellulosic materials as substrate for the production of cellulosic single cell protein. The resuluts were obtained as fellows. 1. Effects of carbolydrates as a sole carton source on the growth of Cellulomonas flavigena KIST 321 were examined. The result showed that cellulose and xylose would be most utlizable for cell mass production. 2. Alkaline treated waste papers and clothes resulted in good growth of the organism than intact ones did. However the waste papers as substrate of cellulosic fermentation were not digestible, even if the meterial was treated with alkalies. 3. Rice straw, rape straw and panic grass appeared to be good substrates for the cell mass production. 4. Leaves were proved to be a good substrate for the cell mass production, but wood sawdust was hardly digested by merely alkaline treatment. 5. When cellulosic wastes as the substrate were examined into the concentration of alkaline solution, the result suggested that the best productivity of cell mass from cellulosic materials was obtained on treatment with 0.8∼1.0% NaOH solution. 6. The productivity of cell mass was increased by washing out with water after alkaline treatment of newspaper, pine sawdust, lime sawdust and pine leaf.

  • Bacteriorhodopsin in the purple membrane (PM) of halobacteria has recently been attracting much attention to be used as a component of molecular electron device and optical computers. In order to increase the productivity of bacteriorhodopsin in high cell density cultures of Halobacterium halobium R1, an internal membrane cell-retention bioreactor system was employed. As a result, the production of cell mass at OD660 of 12 and of bacteriorhodopsin at 125-130 mg/L were obtained using the internal membrane bioreactor system at a dilution rate of 0.066 hr-1. The productivity achieved by the internal membrane system (0.7 mg/L$.$hr) was 3.5-fold higher than that obtained by the corresponding batch cultivations (0.2 mg/L$.$hr).

Effect of Intracellular Calcium Level on the Hybridoma Cell Growth and Monoclonal Antibody Production (세포내 calcuim 농도가 하이브리도마 세포 성장 및 단일클론항체 생산에 미치는 영향)

  • The effect of intracellular Ca2+ level on the hybridoma cell growth and monoclonal antibody(MAb) production was examined. For the manipulation of intracellular Ca2+ concentration, the cells were treated with A23187, ryanodine, and thapsigargin at about 1x106 cells/mL. The treated cells were recultivated by using the Iscove's Modified Dulbecco's Medium(MDM) containing 1.49mM CaCl2. The ryanodine-treated cells showed better cell growth, MAb concentration, and specific MAb productivity than others. In comparison with control, the maximum cell concentration, MAb concentration, and specific MAb productivity were increased by 40.6%, 48.1% and 83.3%, respectively. Confocal microscopic images of Fura-2/AM loaded cells indicate that the increase in intracellular Ca2+ level can enhance the MAb productivity by allowing the calcium influx into the endoplasmic reticulumn.

  • In cultivating human neuroblastoma cells maximum number of neurites per cell and length of the neurite were estimated as 5.5 and 2.2 (nm), respectively It was found that there was correlation between growth and differentiation of nerve cells. Maximum specific BDNF production rate was also calculated as 2.5$\times $10$^{-5}$(ng/cell/day) at 7$\times $ 10$^{5}$ (viable cells/ml) of maximum cell density, corresponding to 100 (ng/mL) of BDNF. The secretion of BDNF was occurred most in the later peroids of the cultivation, yielding 75 (ng/mL) of BDNF. The production of rate of BDNF was elongated in adding 1 ($\mu $g/mL) of BDNF as well as 40% increase of the length of the BDNF. It proves that BDNF can be used as one of biopharmaceuticals to treat age-related diseases such as Alzheimer's disease and Prakinson's disease. It can also provide the information of scaling-up mammalian cell cuture system to economically produce BDNF.
