Effect of volcanic ash on cell growth and production of exopolymers

  • Effect of volcanic ash on cell growth of Aspergillus sp. and production of exopolymers by Agrobacterium sp. and Aureobasidium pullualns was investigated. The volcanic ash contained various mineral salts such as $SiO_2$, $Al_2O_3$, CaO, $K_2O$. Maximal cell growth of Aspergillus sp. was obtained when 0.3% volcanic ash was added to medium. Cell growth of Aspergillus sp. increased with higher concentration of volcanic ash in medium. Amount of cell growth with 0.3 % volcanic ash was 6.7 times higher than that without volcanic ash. Volcanic ash also stimulated production of exopolymer as well as cell growth. Production of curdlan with 0.1% volcanic ash was 12.40 g/l whereas that without volcanic ash was 9.15 g/l. Production of pullulan with volcanic ash was also higher than that without volcanic ash.

Inhibitory Effect of Medicinal Plant Extract on Cell Toxicity and Interleukin-8 Production Induced by Helicobacter pylori (Helicobacter pylori에 의한 세포독성 및 Interleukin-8 생성에 미치는 생약혼합물의 영향)

  • The effects of Helicobacter pylori and medicinal plants extract (Leweifang) on the viability and interleukin(IL)-8 production of gastric epithelial cell were investigated. Cells viability was significantly decreased when they incubated with H. pylori or H. pylori toxin. Co-incubation with Leweifang increased H. pylori or H. pylori toxin-inhibited cell growth in a concentration-dependent manner. The production of IL-8 was greatly increased in H. pylori-infected KATO III gastric epithelial cells in a concentration- and time-dependent manner. The increased production of IL-8 was significantly inhibited by Leweifang $(1,000{\sim}5,000{\mu}g/ml)$. These results indicate that Leweifang has protective effect on H. pylori-inhibited cell growth and H. pylori-induced gastric mucosal cell inflammation by suppressing the production of inflammatory cytokine (IL-8) from gastric epithelial cells.

  • The purpose of this Study was to investigate effect of TakliSodokSan(TSS) on the anti-tumor, immunocytes and nitric oxide(NO) production from mice peritoneal macrophages. This Study estimated the proliferation of L1210 cell lines, A431 cell lines, Hep-G2 cell lines, K562 cell lines, 3T3 cell lines, mouse thymocytes and mouse splenocytes and NO production from pcritoneal macrophages in vitro, and estimated the proliferation of L1210 cells, thymocytes and splenocytcs, NO production from peritoneal macrophages and body weight in L1210 cells-transplanted mice in vivo. The results were obtained as follows; 1. TSS inhibited significantly the proliferation of L1210, A431, Hep-G2, K562 cell lines in vitro. 2. TSS accelerated the proliferation of mice thymocytes and splenocytes in vitro. 3. TSS was not increased the nitric oxide production from mice peritoneal macrophages in vitro. 4. TSS inhibited significantly the proliferation of L1210 cells in Ll210 cells∼transplanted mice. 5. TSS accelerated the proliferation of mice thymocytes and splenocytes In L1210 cells-transplanted mice. 6. TSS was increased significantly the nitric oxide production from mice peritoneal macrophages in L1210 cells-transplanted mice. 7. TSS was increased the body weight as comparing with control group in Ll210 cells-transplanted mice.

Roles of Glucose and Acetate as Carbon Sources in L-Histidine Production with Brevibacterium flavum FERM1564 Revealed by Metabolic Flux Analysis

  • The metabolic flux pattern for L-histidine production was analyzed when glucose and/or acetate were used as carbon sources. Total L-histidine production was enhanced when mixed substrate (glucose and acetate) was used, compared wish that when either glucose or acetate was used as the sole carbon source. Theoretical maximum carbon fluxes through the main pathways for L-histldine production, cell growth, and ATP consumption for cell maintenance were obtained by the linear programming (LP) method. By comparison of the theoretical maximum carbon fluxes tilth actual ones, it was found that a large amount of glucose was actually used for maintenance of cell viability. On the other hand, acetate was used for cell growth. After depletion of acetate in the mixed substrate culture, the flux for glucose to L-histldine synthesis was markedly enhanced. A strategy for effective L-histidine production using both carbon sources was proposed.

  • The purpose of this study was to investigate the anti-inflammatory effects of extract from Hwang lyun tang (HLT) on the THP-1 cell and HMC-1 cell. To evaluate of anti-inflammatory of HLT, we examined cytokines production in lipopolysacchride (LPS)-induced THP-1 cell and A23187, PMA-induced HMC-1 cell. Extract of HLT inhibit LPS-induced interleukin (IL)-8 production in human monocyte THP-1 cells. Extract of HLT inhibit A23187, PMA-induced IL-8, tumor necrosis factor-$\alpha$ (INF-$\alpha$) production in HMC-1 cells. HLT down-regulated LPS-induced IL-8 production and A23187, PMA-induced IL-8, TNF-$\alpha$ production, which may be provide a clinical basis for anti-inflammatory properities of HLT.

  • Objectives : The purpose of this study was to investigate the anti-inflammatory effects of extract from Jak-Yak Tang (JYT) on the THP-1 cell and HMC-1 cell. Method : To evaluate of anti-inflammatory of JYT, we examined cytokines production in lipopolysacchride (LPS)-induced THP-1 cell and A23187, PMA-induced HMC-1 cell.   • Amoving aeration-membrane (MAM) bioreactor was employed for the production of 2$\mu$g/mL of tissue type Plasminogen Activator (tPA)in serum free medium from normal human fibroblast cells. This system could maintain high cell density for long periods of steady state conditions in perfusion cultivation. Under normal operating condition, shear stress was as low as 0.65 dynes/$\textrm{cm}^2$ at the agitation speed of 80 rpm. Even though cell density gradually decreased with increasing agitation speed, tPA production increased linearly with increasing shear stress within a moderate range. This culture system allowed production of 2$\mu$g tPA/mL while maintaining a high cell denisty of 1.0$\times$107 viable cell/mL.

  • In order to obtain the basic data for the preparation of yogurt containing ginseng component, the effect of ginseng component on cell growth of Lactobacillus casei YIT 9018 and on lactic acid production were investigated. Initial cell growth and acid production were markedly inhibited by the addition of ethanol extracts in the level of 8% into 15% skim milk. Crude saponin did not show any inhibitory effect on cell growth and acid production, but ether layer fraction showed inhibitory effect. It was thought to be more advantageous to add ginseng extracts after the fermentation of milk than before. The addition of ginseng extract at 8% level into liquid yogurt was most suitable in organoleptic test. Cell viability was not affected by the addition of ethanol extracts up to 8% level during storage of liquid yogurt.

  • This paper presents an effective modeling scheme of SPE cell system for hydrogen production. As oxygen and hydrogen produced by water electrolysis using SPE are high purity, we can use oxygen in biomedical and hydrogen could be used in many ways. Recently, it is under the eye as a surplus power storage system. PSCAD/EMTDC model of SPE cell system for hydrogen production to efficiently utilize Solar cell energy which produces effectively hydrogen energy is showed in this paper. The simulated results are then verified by comparing them with the actual values obtained from the data acquisition system. Authors are sure that it is a useful method to the researchers who study SPE cell system for hydrogen production.

Bioreactor Cultures of Lithospermum erythrorhizon for Shikonin Production with In Situ Extraction (동시 추출을 겸한 생물반응기에서 Lithospermum erythrorhizon 배양에 의한 shikonin 생산)

  • Plant cell cultures of Lithospermum erythrorhizon were performed in stirred tank and packed-bed reactors with in situ extraction by n-hexadecane. The specific shikonin production and volumetric shikonin productivity of stirred tank reactor reached 1.5 mg shikoninlg cell and 400$\mu g$ shikonin/(L.day), respectively. In packed-bed reactor with calcium alginate-immobilized cells specific shikonin production and volumetric productivity reached 2.0 mg shikoninlg cell and 2857$\mu g$ shikonin/(L.day), which were 1.3 and 7.1 times higher than those of stirred tank reactor, respectively. The higher shikonin production and productivity of packed-bed reactor seemed to be due to high cell loading capacity of calcium alginate immobilized cells in packed-bed reactor and improved cell-cell contact.
