Encapsulated Animal Cell Culture for the Production of Monoclonal Antibody(MAb)

  • Biopolymer membrane was prepared using two oppositely charged natural biopolymer. The biopolymer membrane was used for the encapsulation of two hybridoma cell lines(ATCC CRL-1606, ATCC BH-8852) to produce monoclonal antibodies. In order to reduce the down stream steps, the pore size of the membrane was controlled to retain the monoclonal antibodies in the capsules based on the diffusion experiments with standard proteins. T-flask culture showed cell densities of 8$\times$107cells/mL 3$\times$107cells/mL, and MAb concentrations of 506 $\mu\textrm{g}$/mL and 109$\mu\textrm{g}$/mL for encapsulated ATCC CRL-1606 and HB-8852, respectively. Two liter perfusion culture with encapsulated ATCC HB-8852 was performed to enhance the MAb production. The MAb production of the encapsulated hybridoma increased considerably comparing to the culture using silicone tubing for oxygen transfer.

Morphological Variation of Enterobacter sp. BL-2 in Acetate-mediated pH Environment for Excretive Production of Cationic Microbial Polyglucosamine Biopolymer

  • Enterobacter sp. BL-2 excretively produced a unique cationic polyglucosamine biopolymer PGB-1 comprised of more than 95% D-glucosamine in an acetate-mediated culture condition. The excretion of the biopolymer PGB-1 was closely associated with the cellular morphology of Enterobacter sp. BL-2, a feature highly dependable on the pH of the medium. The initially formed uneven and irregular surface cells were aggregated into the cell-biopolymer network structure connected by the adhesion modules of the cell-bound biopolymer. The excretive production of the biopolymer PGB-1 coincided with the disruption of the cell-biopolymer network, most actively at the medium pH of 8.0.

Production of Essential Oils by Cell Culture of Codonopsis lanceolata (세포배양에 의한 더덕 정유의 생산)

  • The essential oils from the roots of Codonopsis lanceolata and the cultivated callus were analysed and compared by gaschromatography-mass spectrometry. In the experimental study of cell culture, it appeared that 2,4-dichlorophenoxy acetic acid in the culture medium induced higher production of essential oils in the callus than indole acetic acid. The growth of callus was inhibited by illumination of the light. The production of essential oil in cultured cells was increased by the addition of biosynthetic precursors. The essential oils from the roots of Codonopsis lanceolata and the cultured callus showed different compositions. Tetradecanoic acid, 1,1,-dimethoxyl 4-methoxy phenol, 9,12-octadecanoic acid and hexadecanoic acid were identified as main components of the cultured callus oil.

  • HCPV(High Concentrated PV) systems have well known for CPV market all over the world. Low concentration type silicon based modules have been introduced in the market. But low cost of standard flat silicon modules made them useless nowadays. High cost of compound semiconductor solar cell reduced cost effective cpv module production than that of recently silicon solar cell. In order to overcome increasing cost of CPV module, we study middle concentration type fresnel lens simulation using concentrated type silicon based solar cell. Linear type fresnel lens made production of CPV module without secondary optics such as light pipe or light tunnel. This type of fresnel lens design makes more cost effective solution for cpv niche market.

  • In this study, the control and scheduling of the flexible manufacturing cell (FMC) is discussed, which can perform the mixed production and relieve the effect of machine failure. The control of the FMC isvery complex task due to the property of multiple jobs and the dynamically changing states. For effective control of proposed FMC, the hierarchical scheme is introduced and the functions of each levels are defined. Especially for the control functions of shop floor level and cell level, the intelligent scheduler is implemented. To show the efficiency of the intelligent scheduler, the production method fo the existing assembly lines was evaluated and compared with the proposed intelligent FMC method. The results from the production performance show that the proposed method is superior to the existing method in various performance indices.

  • Effects of various elictitors were investigated to enhance the production of berberine in plant cell suspension cultures of Thalicrtrum rugosum. Treatments of yeast elicitor, 15 different types of abiotic elicitors, 16 kinds of fungal elicitors from three species of fungi were performed. Cell growth and berberine production were examined and compared for both normal and elicitor treated cultures. No distinguished increases in berberine yield by the addition of elicitors could be attained.

  • Immunositmulation effects of the mice fed with the cell lysate of Lactobacillus plantarum isolated from Kimchi were studied. The mice group fed with cell lysate was different from the control group on the degree of immune responses, e.g. 1) proliferation of splenocytes and Peyer's patch cells, 2) production of nitric oxide (NO) by peritoneal macrophages, 3) production of intestinal secretory lgA (slgA), 4) variation of TNF-${\alpha}$ and IL-2 concentration in blood, 5) production of specific lgG against sheep red blood cells. A general enhancement in enteric and systemic immune responses was observed with a simple oral administration of immunostimulators. With the oral feeding of L. plantarum, not only the total amount of gut secretion antibody, but also the binding capacity of antibodies to the enteric microorganisms including L. plantarum was increased. These experimental results clearly showed that the oral feeding of immunostimulators gave multifunctional effects on the mucosal and systemic immune systems of mice.

  • This paper deals with the development of a software module for production planning and scheduling activities of an existing Flexible Machining and Assembly System (FMAS). The Production Planning Module uses the hierarchical and sequential scheme based on "divide and conquer" philosophy. In this module, routes are determined based on the production order, orders are screened, tools are allocated, and order adjustments are executed according to the allocated tools. The Scheduling Module allocates the resources, determines the task priority and the start and completion times of tasks. Re-scheduling can be done to handle unforeseen situations such as lumpy demands and machine breakdowns. Since all modules are integrated with a central database and they interface independently, it is easy to append new modules or update the existing modules. The result of this study is used for operating the real FMAS consisting of a machining cell with 2 domestic NC machines and a part feeding robot, an assembly cell with a conveyor and 3 robots, an inspection cell, an AGV, an AS/RS, and a central control computer.

The Effect of Light on Champagne Yeast Cell Growth and Ethanol Production Under Variable pH Conditions

  • The effect of wtlitc light on unaeraten growth of Baker's yeast and the accompanying ethanol production has been studied in a batch process at 27$^{\circ}C$. Over the 80-hour period of the Champagne yeast process without pH control, the cull growth was inhibited by the fluorescent light. Another observed difference between the runs is that the drop and subsequent rise in redox potential occurred much sooner in the fermentation with light than in the fermentation without light. This preliminary study indicated that ethanol production could be enhanced by light as the cell concentration is repressed. The possible pathway, shift of the sugar substrate toward ethanol and away from cells was manifested by another difference as well. As observed under the microscope, many of the yeast cells grown under light budded without dividing by the normal fission process as they did in the dark. Furthermore, the undivided and branched (light grown) cell did not agglutinate at the end of the fermentation process as did the distinct spherical (dark grown) cells.

  • This Paper presents a heuristic algorithm for machine-part group formation considering part production information (Production volume, roution sequence, unit operation time, facility size) in cellular manufacturing logistics system. In general, factory space is restricted within limited size when cells are located. A twofold heuristic algorithm is developed for considering factory space restrictions of located cells. The first phase is a aggregation procedure to minimize inter cell movement for satisfactoring space restriction. The second phase is a rearrangement procedure to maximize line balancing efficiency between machines within the cell and non assigned machine during first phase. Numerical example is presented to verify the efficiency of proposed algorithm.
