Reconstruction with Extracorporeally Radiated Autogenous Bone Graft After Wide Resection of Bone Tumors (골종양 절제후 방사선 조사한 자가골을 이용한 재건술)

  • PURPOSE : For the reconstruction of large bone defect after tumor resection, it is possible to reuse the bone involved by tumor with some treatment to it. Several bone-reusing methods have been reported such as autoclaving, low-heat treatment(pasteurization) and intraoperative radiotherapy. We have used extracorporeally radiated autogenous bone graft for reconstruction after tumor resection, and analyzed the periods for junctional union, functional results and complications to know the indications of this method. METHODS : From Dec. 1993 to Sept. 1995, nine patients had taken autogenous bone graft with extracorporeal irradiation. Eight cases were osteosarcoma and 1 giant cell tumor. The graft sites were 5 in femur, 3 proximal tibia and 1 femur and tibia. Stage 3 was 1 case(GCT), Stage IIB 3 and Stage IIIB 5. After wide resection, surrounding soft tissue and intramedullary and extramedullary portion of the tumor were removed. Radiation was done in 5000cGy to the resected bone. Ender nails and bone cement were inserted and filled into the medulla to prevent fracture. RESULTS : Average follow-up period was 12.3(4 to 21) months. Average junctional union period in simple X-ray was 6.5 months in 4 cases. Average functional score following Enneking's criteria was 19(12-27). Complications were as follows ; condylar fractures and femur neck fracture in 4 cases, subluxation of the knee joint 3 and infection 1. Although local recurrence was detected in 1 case, the site of recurrence was not in the radiated bone but surrounding soft tissue. At final follow-up, no recurrence was found in one case(GCT), CDF 2, AWD 2, DOD 3, and died of chemotherapy related sepsis 1. CONCLUSIONS : Extracorporeally radiated bone autograft is considered to be a method for reconstruction of the large bone defect made by tumor resection, especially in the reconstruction around the joint.

Induction of Radiation Adaptive Response in Lymphocytes of patients Undergoing Bone Scintigraphy (골 신티그라피를 시행한 환자의 림프구에서 방사선 적응반응의 유도에 관한 연구)

  • Purpose: The purpose of this study was to ascertain whether radiation adaptive response could be induced by Tc-99m-methylene diphosphonate (Tc-99m-MDP) in peripheral lymphocytes of patients undergoing bone scintigraphy. Materials and Methods: Lymphocytes from 22 patients (6 males, 16 females, mean age $50{\pm}14$ years) were collected before and after bone scintigraphy using 740 MBq Tc-99m-MDP Lymphocytes from 10 controls (6 males, 4 females, mean age $43{\pm}7$ years) were also collected. They were exposed to challenge dose of 2 Gy gamma rays using a cell irradiator. Number of ring-form and dicentric chromosomes per 600 cells (chromosomal aberrations) was counted under the light microscope. Results: Chromosomal aberrations in patients before bone scintigraphy ($385.1{\pm}30.5$) was not different from that of controls ($367.8{\pm}36.6$). However, chromosomal aberrations in patients after bone scintigraphy was significantly decreased to $192.6{\pm}22.1$ (p=0.0001). Conclusion: Low dose gamma-irradiation by Tc-99m-MDP used for bone scintigraphy induces a cytogenetic adaptive response in peripheral lymphocytes.

Effects of Low-Level Laser Irradiation on the ALP Activity and Calcified Module Formation of Rat Osteoblastic Cell (저수준레이저(GaAs 반도체)조사가 골모세포의 알칼리성 인산분해효소의 활성과 석회화결절의 형성에 미치는 영향)

  • 저수준레에저요법에 대해서는 지난 10여년간 의학계 및 치과계에서 임상적으로 사용하여 좋은 결과가 있다고 많은 보고가 발표되고 있다. 특히 치근의 골결손에 관한 연구에서는 전기요법, 초음파요법, 전자장요법 등 뿐만아니라 저수준레이저를 사용하여 골절부내 Callus형성이 촉진되었음을 보고하고 새로운 치료법의 하나가 될 수 있음을 제안한 바도 있다. 본 연구에서는 칼륨비소를 다이오드로 사용한 저수준레이저조사가 골결손의 치유에 어떠한 영향을 미치는지 확인하고자 골모세포의 알칼리성 인산분해효소의 활성화와 석회화결절의 형성을 평가함으로 골모세포의 기능을 조사코저하였다. 실험은 첫째, 9개군으로 나누어 레이저 조사기간에 따른 알칼리성 인산분해효소의 활성화를 조사하였고, 둘째, 이를 근거로 9일간 계속 매일 1회 1.3 J/cm2의 레이저를 조사한 후 펄스의 종류별 차이를 비교하였으며, 세째,레이저펄스별 석회차 결절의 형성 정도를 광학현미경으로 관찰하여 비교분석하였다. 결과, 7일 계속 레이저를 조사한 경우 다른 군에 비해 서서히 ALP의 활성이 증가하였으나 유의한 차이는 없었으며. 따라서 9일동안 레이저를 계속 조사한 경우에는 전체 에너지량이 5.895 J/cm2 인 펄스13과 15가 뚜렷하게 유의한 증가를 보여주었다. 그러나 석회화결절의 형성은 전체 에너지량이 2.546 J/cm2 인 펄스11에서 가장 많았다. 결론적으로 골형성이나 알칼리성 인산분해효소의 활성을 촉진하는 데에는 적절한 레이저 조사조건이 필요하나, 알칼리성 인산분해효소의 활성을 촉진한 펄스와 석회화결절의 형성을 촉진하는 펄스가 서로 다르게 나타난 것은 골형성을 촉진하는 여러요인 들이 저수준레이저에 자극받았을 가능성이 높음을 보여준다 이러한 결과들로 보아 저수준레이저는 골모세포의 기능을 자극하여 골결손의 치유를 개선하는 데 도움될 것이라 사료된다.다. 각 백서의 양측 창상중 하나는 1,3,5,7일 마다 각 실험의 방법에 따라 레이저를 조사하고 실험동물의 다른 창상은 대조군으로서 사용하였다. 모든 창상의 면적은 실험 1,3,5,7 일째에 일정한 거리에서 사진촬영하여 면적계를 이용, 측정한 후 통계적인 의의를 조사하였다. 본 연구의 결과는 저수준레이저는 특정 조건하에서 S. aureus의 증식을 촉진하였다. 그러나 S. aureus에 감염된 창상을 저수준레이저로 조사시 치유가 촉진되었다. 중앙 조사법고 주변조사법에 의한 창상치유효과는 통계적인 의의가 보이지 않았다. 따라서 결론적으로 S. aureus 에 감염된 창상에 직접 또는 간접적이든 pulse의 종류에 관계없이 조사하는 경우 치유효과가 나타나는 것은 정사주위 조직의 LLLI 자극효과가 염증의 확산을 억제한다고 말할수 있다.4/1$0^{\circ}C$에서는 Shoa-Nan-Tsan과 Lenkwang이 가장 높았으며 백앙벼는 3 온도 조건 모두에서 활성이 낮았다. 발아소요일수와 amylase 활성과는 유의적인 정의 상관관계를 보였다., 다다조, 미국의 건답직파재배 품종 등이었으며 우리 나라 육성종들은 모두 지중에서 신장이 멈추어 제1본엽이 지중에서 추출하였으며, Scm파종심에서 불완전엽이 지면을 뚫고 나오는 품종은 Chinsura Boro뿐이었고 Nato, Labelle, Weld Pally, Italliconaverneco 등도 지면 가까이 까지 신장하였다. 6. 50% 출아일수는 제2절간장을 제외 한 모든 유아 형질의 신장도와 유의한 부의 상관을 보였는데 가장 높은 상관을 보인 것은 중배축장+제1절간장+불완전옆장이었으며, 다음이 불완전엽장이 었다. 7. 출아율은 중배축장+제1절간장+불완전엽장, 중배축장+초엽 장과 모든 파종심에서 높은 정의 상관을 보여 제1본엽의 추출 위치가 높을수록

Modification of Gamma-radiation Response in Mice by Green Tea and Diethyldithiocarbamate (마우스에서 방사선 영향에 대한 녹차와 Diethyldithiocarbamate의 조절효과)

  • We performed this study to determine the effect of green tea on jejunal crypt survival, endogenous spleen colony formation, and apoptosis in jejunal crypt cells of mice irradiated with high and low dose of gammairradiation. The radioprotective effect of green tea was compared with the effect of diethyldithiocarbamate (DDC). Jejunal crypts were protected by pretreatment of green tea (p<0.01). Green tea administration before irradiation resulted in an increase of the formation of endogenous spleen colony (p<0.05). The frequency of radiation-induced apoptosis in intestinal crypt cells was also reduced by pretreatment of green tea (p<0.05). The radioprotective effect on jejunal crypts and apoptosis in the DDC treated group appeared similar to those in the green tea treated groups. Treatment with DDC showed no significant modifying effects on the formation of endogenous spleen colony. These results indicated that green tea might be a useful radioprotector, especially since it is a relatively nontoxic natural product. Further studies are needed to characterize better the promotion nature of green tea and its components.

Effects of Oral Intake of Kimchi-Derived Lactobacillus plantarum K8 Lysates on Skin Moisturizing

  • Skin is the soft outer covering of vertebrates that provides protection from pathogenic infection, physical damage, or UV irradiation, and controls body temperature and water content. In this study, we examined the effects of oral intake of kimchi-derived Lactobacillus plantarum K8 lysates on skin moisturizing. In an in vitro study, we observed that the hyaluronic acid content increased in HaCaT cells treated with L. plantarum K8 lysates. Oral administration of L. plantarum K8 lysates effectively attenuated the horny layer formation and decreased epidermal thickening in DNCB-treated SKH-1 hairless mice skin. The damage to barrier function was reduced after 8 weeks of oral administration of L. plantarum K8 lysates as compared with that in the atopic dermatitis mice. For the test with volunteers, we manufactured experimental candy containing 2.1% L. plantarum K8 lysates, while control candy did not contain bacterial lysate. A significant increase in hydration in the experimental candy-administered group as compared with the control candy-administered group was observed on the face after 4 and 8 weeks, and on the forearm after 4 weeks. Decreases in horny layer thickness and TEWL value were observed on the face and forearm of the experimental group. Together, the in vitro cell line and in vivo mouse studies revealed that L. plantarum K8 lysates have a moisturizing effect. A clinical research study with healthy volunteers also showed an improvement in barrier repair and function when volunteers took L. plantarum K8 lysates-containing candy. Thus, our results suggest that L. plantarum K8 lysates may help to improve skin barrier function.

The Effect of Melatonin on Mouse Jejunal Crypt Cell Survival and Apoptosis (멜라토닌이 생쥐 소낭 세포 재생과 아포토시스에 미치는 영향에 대한 연구)

  • Purpose :To evaluate protective mechanism of melatonin against radiation damage and its relationship with apoptosis in mouse jejunum. Materials and Methods: 168 mice were divided into 28 groups according to radiation dose and matatonin treatment. To analysis crypt survival, microcolony survival assay was done according to Withers and Elkind's method. To analysis apoptosis, TUNEL assay was done according to Labet-Moleur's method. Results : Radiation protection effect of melatonin was demonstrated by crypt survival assay and its effect was stronger in high radiation dose area. Apoptosis index with 8 Gy irradiation was 18.4$\%$ in control group and 16.5$\%$ in melatonin treated group. After 18 Gy, apoptosis index was 17.2$\%$ in control group and 15.4$\%$ in melatonin treated group. Apoptosis index did not show statistically significant difference between melatonin treated group and control group. Conclusion : Melatonin shows clear protective effect in mouse jejunum against radiation damage but its protective effect seems not to be related with apoptosis protection effect.

In Vitro Selection and Characterizations of Gamma Radiation-Induced Salt Tolerant Lines in Rice (방사선을 이용한 내염성 계통의 기내선발 및 특징)

  • The combination of radiation technique with an in vitro culture system was initiated to develop salt tolerant rice. We established an in vitro culture system to select tolerant lines against salt stress. NaCl tolerant cell lines were selected from the callus irradiated with gamma ray on N$_{6}$ medium with 1.5% NaCl and 2 mg/L 2,4-D. Regenerants (M$_1$) were obtained from the tolerant callus which was cultured for 30 days auxin-free medium. The M$_2$seeds were harvested from M$_1$plants on an individual plant basis. Thirty seedlings from each 450 M$_2$lines were transplanted in a field and total 5,000 M$_3$lines were harvested with an average 90 percent of fertile grain. M$_3$lines were utilized for selection of salt tolerance. Salinity-tolerant lines (225) were selected among 5,000 M$_3$lines. Of the 225 lines tested, the morphological traits of two lines (120-10 and -11) were far superior to control (Donagjinbyeo) in agromomic traits such as plant height, root length and no. of roots. Control and tolerant lines were analyzed by RAPD markers. Three polymorphic bands were presented in only tolerant lines, demonstrating a genetic difference between control and the tolerant lines. Such tolerant lines could be used as genetic resources to improve salt tolerance.e.

  • Redox flow secondary battery have been studied actively as one of the most promising electrochemical energy storage devices for a wide range of applications, such as electric vehicles, photovoltaic arrays, and excess power generated by electric power plants. In all-vanadium redox flow battery using solution of vanadium-sulfuric acid as a active material, the difficulty in developing an efficient ion selective membrane can still be identified. The asymmetric cation exchange membrane(M-30) as a separator of all-vanadium redox flow battery which were obtained by the reaction of chlorosulfonation for 30 minutes under the irradiation of UV, showed its superiority in the transport number of 0.94 and electrical resistivity of $0.5{\Omega}{\cdot}cm^2$. The base membrane were prepared by lamination a low density polyethlene film of $10{\mu}m$ thickness on polyolefin membrane(HIPORE 120). The electrical resistivity of M-30 membrane in real solution of vanadium-sulfuric acid was $3.79{\Omega}{\cdot}cm^2$ and it was similar to that of Nafion 117 membrane. Also the cell resistivity was $6.6{\Omega}{\cdot}cm^2$and lower than that of Nafion 117. In considertion of electrochemical properties and costs of membranes, M-30 membrane was better than that of Nafion 117 and CMV of Asahi glass Co. as a separator of all-vanadium redox flow battery.

Grape Pruning Stem Extract (GPSE) Suppresses Allergy and Skin Proliferation Inhibition against UVB Induced Skin Damage (포도전정가지 추출물이 UVB에 유도된 피부 손상에 대한 알레르기와 피부증식 억제효과)

  • This study examined the effects of polyphenols in grape pruning stem extracts (GPSE) using grape stems discarded after harvest. The inhibitory effects on allergy, proliferation, and apoptosis in UVB-induced HR-1 hairless mice were analyzed. The applicability as a material for functional food and functional cosmetics was evaluated. The contents of the active ingredients of GPSE were analyzed by HPLC. After UVB irradiation on the dermis of HR-1 hairless mice, the number of mast cells was determined by toluidine blue staining to confirm that the skin allergic reaction was caused by GPSE. The results were similar to the normal group in the group applied GPSE, and there was no allergic reaction in the GPSE application group and a significant decreased compared to the sun cream control. PCNA immunohistochemical staining of the epidermal proliferation factor had an inhibitory effect on epidermal epithelial cell proliferation in all concentrations of GPSE and serum base mixture as an average of 42% compared to the control group. The mixture of GPSE and serum base suppressed the apoptosis inhibition rate by 27% on average compared to the control. These results confirmed the inhibitory effects GPSE on the allergic, proliferation, and apoptosis activities by with a serum base on UVB-induced skin damage. GPSE is a functional ingredient with potential skin protection effects, and has a high utilization as an ingredient for functional cosmetics.

An Experimental Study on the Effectiveness of Microwave Hyperthermia Combined with Radiation on the Small and Large Intestine in rats (흰쥐의 장조직에 X-선 조사와 마이크로파 온열요법의 효과에 관한 실험적 연구)

  • The synergistic effect of combining radiation therapy and hyperthermia kills significantly more cells than using either modality alone. The reason for enhanced cell killing from the combined treatment is that the two modalities are complementary. For histopathological exmination, 102 rats were divided into 4 groups as hyperthermia, radiation, hyperthermia combined with radiation and normal control groups. The effect of prior irradiation (6-15 Gy of X-ray) on the response of small and large bowel of rats to $40^{\circ}C-44^{\circ}C$ (for 30 minutes) microwave (2450 MHz) hyperthermia was investigated. The musculature of the small and large intestine remained intact and the circumference of the histological sections were not significantly altered by the heated at $43^{\circ}C$ for 30 minutes. Thermal enhancement ratios of normal tissue is 1.0 Thermal enhancement ratio was not increased in combination therapy by evaluation of histopathologic changes in small and large intestine.
