  • 본 실험을 수행하기 위해 EPS를 생산하지 않는 Lactobacillus paracasei KLB58의 돌연변이주를 NTG mutagenesis 방법을 사용하여 선별하였다. 선별된 돌연변이주의 숙주에 대한 부착능을 연구하기 위해 세포 표면의 소수성의 변화에 관하여 연구가 진행되었다. 정량적인 측정을 위해 hydrocarbon의 한 종류인 hexadecane을 소수성층으로 사용하여 세포와 함께 섞이는 kinetics를 측정하였다. 실험결과 KLB58의 돌연변이주는 야생형에 비해 세포표면이 상대적으로 높은 소수성을 나타내었다.

태양전지 효율 향상용 졸-겔 법에 의한 반사방지 코팅막의 제조 (Preparation of the Anti-Reflective(AR) Coating Film by Sol-Gel Method to Improve the Efficiency of Solar Cell)

  • This study investigates the preparation of anti-reflective (AR) coating film to improve the efficiency of solar cell. The AR coating film was successfully obtained by dip-coating with AR coatings prepared by sol-gel method. Fluoroalkylsilane was additionally introduced into the coatings to give the self-cleaning effect of AR coating film. We performed the abrasion test, pencil scratch hardness test and cross-cut test to identify the mechanical strength of AR coating film. As the results, the transmittance of AR coating films with 9.07, 18.13 and 27.20 of IPA/MTMS molar ratios were 93.1, 93.6 and 95.3%, respectively. The water contact angle and transmittance of AR coating film increased by the introduction of hydrophobicity. The prepared AR coating film shows the high level of abrasion, hardness and adhesion. The IPA/MTMS molar ratio of 27.20 and the withdrawing speed range of 0.20 ~ 0.28cm/sec are the optimal coating condition in terms of the transmittance and mechanical strength of AR coating film.

Effect of Sub-minimal Inhibitory Concentration of Chlorhexidine on Biofilm Formation and Coaggregation of Early Colonizers, Streptococci and Actinomycetes

  • Chlorhexidine has long been used in mouth washes for the control of dental caries, gingivitis and dental plaque. Minimal inhibitory concentration (MIC) is the lowest concentration of an antimicrobial substance to inhibit the growth of bacteria. Concentrations lower than the MIC are called sub minimal inhibitory concentrations (sub-MICs). Many studies have reported that sub-MICs of antimicrobial substances can affect the virulence of bacteria. The aim of this study was to investigate the effect of sub-MIC chlorhexidine on biofilm formation and coaggregation of oral early colonizers, such as Streptococcus gordonii, Actinomyces naeslundii and Actinomyces odontolyticus. The biofilm formation of S. gordonii, A. naeslundii and A. odontolyticus was not affected by sub-MIC chlorhexidine. However, the biofilm formation of S. mutans increased after incubation with sub-MIC chlorhexidine. In addition, cell surface hydrophobicity of S. mutans treated with sub-MIC of chlorhexidine, decreased when compared with the group not treated with chlorhexidine. However, significant differences were seen with other bacteria. Coaggregation of A. naeslundii with A. odontolyticus reduced by sub-MIC chlorhexidine, whereas the coaggreagation of A. naeslundii with S. gordonii remained unaffected. These results indicate that sub-MIC chlorhexidine could influence the binding properties, such as biofilm formation, hydrophobicity and coaggregation, in early colonizing streptococci and actinomycetes.

RV-23, a Melittin-Related Peptide with Cell-Selective Antibacterial Activity and High Hemocompatibility

  • RV-23 is a melittin-related antibacterial peptide (MRP) with lower cytotoxicity than either melittin or AR-23, another MRP. The aim of this study was to explore the mechanism of RV-23's antibacterial selectivity and its hemocompatibility. The results showed that all the peptides exhibited lytic activity against Staphylococcus aureus and Escherichia coli, with RV-23 showing the highest potency. Moreover, RV-23 had lower cytotoxicity than melittin or AR-23 at their minimal inhibitory concentration. In addition, CD experiments showed that melittin, RV-23, and AR-23 all had a typical α-helical structure, and RV-23 had the lowest α-helix content. The structural information showed that RV-23 has the lowest hydrophobicity and highest hydrophobic moment. Because hydrophobicity and α-helix content are believed to correlate with hemolysis, the results indicate that the selective lytic activity against bacteria of RV-23 may be due to its low hydrophobicity and α-helicity, which lead to low cytotoxicity without affecting antibacterial activity. Furthermore, RV-23 did not affect the structure and function of blood components such as red blood cells, platelets, albumin, and the blood coagulation system. In conclusion, RV-23 is a cell-selective antibacterial peptide with high hemocompatibility due to its unique structure.

Fabrication of a Superhydrophobic Surface with Adjustable Hydrophobicity and Adhesivity Based on a Silica Nanotube Array

  • A superhydrophobic surface with a water contact angle > $150^{\circ}$ has attracted great interest from both fundamental and practical aspects. In this study, we demonstrated that hydrophobicity of a silica nanotube (SNT) array can be easily controlled by the SNT aspect ratio. In addition, the adhesive and anti-adhesive properties were controlled without modifying the hydrophobic surface. Various silica structures on a polydimethylsiloxane substrate were prepared using the desired alumina template. Bundle-arrayed and bowl-arrayed silica surfaces exhibited extraordinary superhydrophobicity due to the large frontal surface area and hierarchical micro/nanostructure. As the strategy used in this study is biocompatible and a wide range of hydrophobicities are capable of being controlled by the SNT aspect ratio, a hydrophobic surface composed of an SNT array could be an attractive candidate for bioapplications, such as cell and protein chips.

탄소성형 Bipolar Plate의 전기화학적 특성과 연료전지 성능 비교 (Electrochemical Characteristics of Home-Made Bipolar Plate and Its Relationship with Fuel Cell Performance)

  • 본 연구에서는 탄소성형 분리판의 물리화학적, 전기화학적 특성을 분석하여 수소 및 개미산연료 전지의 성능에 미치는 영향을 알아보았다. 기존 기계가공 탄소분리판과 탄소복합소재 탄소성형 분리판의 접촉저항, 부식특성, 소수성을 비교 평가하였다. 특히, 현재 연구개발 중인 탄소복합소재 성형분리판의 경우 계면접촉저항이 기계가공 분리판보다 1.5배 높게 나타났으며, 내식성 실험에서는 산에 취약하여 분리판 표면이 거칠어지고 결정성이 감소하였다. 연료전지의 성능은 분리판의 계면접촉저항에 크게 영향을 받았으며, 계면저항이 적은 기계가공 분리판이 수소 및 개미산 연료 전지에서 높은 성능을 나타냈다.

가자미식해에서 분리한 유산균의 프로바이오틱스 특성 (Probiotic Properties of Lactobacillus spp. Isolated from Gajami Sikhae)

  • 가자미식해에서 분리된 유산균 40종은 Lactobacillus plantarum spp., Leuconostoc mesenteroides spp., Lactobacillus brevis spp., and Weisella paramenteroides spp.로 확인됐으며, 40종 중 10종을 선별해 검사에 사용했다. 본 연구에서는 가자미식해에서 분리된 유산균을 프로바이오틱스로 활용하기 위해 산 및 인공위액 저항성, 담즙산 저항성, 자가 응집성, 공동 응집성, 세포 표면 소수성 등의 프로바이오틱 연구를 수행하였다. 분리한 유산균주는 산성 및 인공 위액에 대해 저항성을 보여 높은 생존율을 나타내었으며, L. plantarum GS11이 가장 뛰어난 저항성을 보였다. 또한 담즙산 저항성 측정 결과 모든 유산균주가 108~109 log CFU/ml의 생균수로, 100% 이상의 생존력을 보여주었다. 그리고 세포 표면 부착능을 간접적으로 측정하기 위해 응집력 평가를 한 결과, autoaggregation 능력을 46% 이상 나타내었다. 세포 표면 소수성 평가를 위해 Xylene 부착능을 측정한 결과 분리된 유산균주에서 32.2%의 소수성을 가지는 B. subtilis 보다 더 뛰어난 세포 부착률을 보여주었다. 이와같이 유산균은 프로바이오틱스로 활용될 만한 유의미한 결과를 보였으며, L. plantarum GS12와 L. plantarum GS13을 제외한 유산균에서 항균 활성이 나타났다. 따라서 가자미 식해에서 분리된 유산균은 다양한 프로바이오틱스 특성을 가진 프로바이오틱스로 활용할 수 있을 것으로 판단되어진다.

  • Proper water management is important to achieve high performance and durability of Polymer electrolyte fuel cell (PEFC). Among various stack components, gas diffusion layer (GDL) is considered as a core part to determine the gas and water transportation in a cell. To optimize the water management, the changes of properties as well as basic properties of GDLs were investigated before and after clamping of colls. Thickness, electric conductivity, porosity, hydroppobicity etc. were characterized by the same criteria. The amount of residual water after cell operation also was compared by direct measuring of weight. Based on the amount of residual water the endurance on the freeze condition was evaluated.

Isolation, Identification, and Characterization of Aero-Adaptive Campylobacter jejuni

  • Campylobacter is one of the emerging foodborne pathogens, and its worldwide incidence rate is extremely high. This study was undertaken to isolate and identify Campylobacter strains from chicken carcasses in the local markets, and analyze their characteristics regarding oxygen tolerance. They were isolated after aerobic enrichment and identified by biochemical, physiological, and morphological characteristics, PCR, and 16S rDNA sequencing. Their oxygen tolerances were analyzed in terms of the cell surface hydrophobicity, cell fatty acid composition, and oxidoreductase. Five strains of C. jejuni were isolated and identified from 61 isolates from 50 chickens. Among them, C. jejuni IC21 grew well in Brucella broth and commercial milk under aerobic condition. However, in the aerobic exposure, the cell surface hydrophobicity of C. jejuni IC21 was almost the same as the other isolates, even though its morphology changed from the spiral-bacilli form into the coccoid form. Fatty acid analyses showed that all Campylobacter strains had a high composition of $C_{19:1}$, cyclopropane fatty acid, and that the amount of the other fatty acids were very similar between them. Interestingly, however, only oxidoreductase activities of C. jejuni IC21 increased highly under aerobic exposure even though its activities were almost the same as the other C. jejuni strains just after microaerobic culture. It had 11.8 times higher catalase activity, 4.4 times higher for SOD, and 2.0 times higher for NADH oxidase activities. Therefore, in the case of the aero-adaptive C. jejuni IC21, expression of oxidoreductase significantly increased under oxidative stressed condition, which might allow it to survive for a longer time and grow on food under aerobic exposure. Such new strain might be one of the explanations for the increase of campylobacteriosis.

Screening of Probiotic Activities of Lactobacilli Strains Isolated from Traditional Tibetan Qula, A Raw Yak Milk Cheese

  • In this study, 69 lactobacilli isolated from Tibetan Qula, a raw yak milk cheese, were screened for their potential use as probiotics. The isolates were tested in terms of: Their ability to survive at pH 2.0, pH 3.0, and in the presence of 0.3% bile salts; tolerance of simulated gastric and intestinal juices; antimicrobial activity; sensitivity against 11 specific antibiotics; and their cell surface hydrophobicity. The results show that out of the 69 strains, 29 strains (42%) had survival rates above 90% after 2 h of incubation at pH values of 2.0 or 3.0. Of these 29 strains, 21 strains showed a tolerance for 0.3% bile salt. Incubation of these 21 isolates in simulated gastrointestinal fluid for 3 h revealed survival rates above 90%; the survival rate for 20 of these isolates remained above 90% after 4 h of incubation in simulated intestinal fluid. The viable counts of bacteria after incubation in simulated gastric fluid for 3 h and simulated intestinal fluid for 4 h were both significantly different compared with the counts at 0 h (p<0.001). Further screening performed on the above 20 isolates indicated that all 20 lactobacilli strains exhibited inhibitory activity against Micrococcus luteus ATCC 4698, Bacillus subtilis ATCC 6633, Listeria monocytogenes ATCC 19115, and Salmonella enterica ATCC 43971. Moreover, all of the strains were resistant to vancomycin and streptomycin. Of the 20 strains, three were resistant to all 11 elected antibiotics (ciprofloxacin, erythromycin, tetracycline, penicillin G, ampicillin, streptomycin, polymyxin B, vancomycin, chloramphenicol, rifampicin, and gentamicin) in this study, and five were sensitive to more than half of the antibiotics. Additionally, the cell surface hydrophobicity of seven of the 20 lactobacilli strains was above 70%, including strains Lactobacillus casei 1,133 (92%), Lactobacillus plantarum 1086-1 (82%), Lactobacillus casei 1089 (81%), Lactobacillus casei 1138 (79%), Lactobacillus buchneri 1059 (78%), Lactobacillus plantarum 1141 (75%), and Lactobacillus plantarum 1197 (71%). Together, these results suggest that these seven strains are good probiotic candidates, and that tolerance against bile acid, simulated gastric and intestinal juices, antimicrobial activity, antibiotic resistance, and cell surface hydrophobicity could be adopted for preliminary screening of potentially probiotic lactobacilli.
