  • This paper reports experimental excess molar volumes $V^E_m$ using a digital vibrating-tube densimeter and excess molar enthalpies $H^E_m$ by means of an isothermal microcalorimeter with a flow mixing cell for the binary mixtures{1,2-dichloropropane + 2-(2-methoxyethoxy)ethanol} and {1,2-dichloropropane + 2-(2-ethoxyethoxy)ethanol} at 298.15 K under atmospheric pressure. All the $V^E_m$ and $H^E_m$ of the two binary mixtures showed S-shaped forms, being negative for poor and positive for rich 1,2-dichloropropane mole fractions. These show that the excess properties were shown to be negative deviation from ideality due to the strong self-association effect among 2-(2-alkoxyethoxy)ethanol molecules at an early stage of mixing, a relatively high energy then is needed to break hydrogen bonds of 2-(2-alkoxyethoxy)ethanol with an increase ofhalogenated hydrocarbon molecular at high mole fraction of 1,2-dichloropropane. The values of excess molar properties($V^E_m$ and $H^E_m$) were fitted by the Redlich-Kister equation using Nelder-Mead's simplex pattern search method. The Wilson, NRTL, and UNIQUAC models were used to correlate the $H^E_m$ values.

  • In previous works, we found that solvent extract of Opuntia humifusa Raf., a member of the lactaceae family, displayed potent anti-oxidative and anti-inflammatory activities. Thus, all solvent fractions, except for the water layer, showed potent scavenging effects. According to activity-guided fractionation, one of active radical scavenging principles in the ethyl acetate fraction was found to be taxifolin. In this study, we investigated whether taxifolin showed anti-oxidative activity. In addition, taxifolin modulated nitric oxide (NO) release and the expression of pro-inflammatory cytokine mRNA such as interleukin-$1{\beta}$ (IL-$1{\beta}$), IL-6, granulocyte-macrophage colony-stimulating factor (GM-CSF), and TNF-${\alpha}$. Taxifolin showed potent anti-oxidant activity with the $IC_{50}\;of\;8.5{\pm}1.4\;and\;9.3{\pm}1.0{\mu}M$ using xanthine/xanthine oxidase (XO) assay and 2,2-Diphenyl-lpicrylhydrazyl radical (DPPH) assay, respectively. We next determined the role of taxifolin on the immunomodulating activity using murine macrophage cell line RAW264.7 cells. Taxifolin dose-dependently inhibited NO production in lipopolysaccharide (LPS)-activated RAW264.7. It also significantly blocked the expression of inducible NO synthase (iNOS) mRNA in the LPS-stimulated RAW264.7 cells. In addition, taxifolin potently suppressed the expression of IL-$1{\beta}$, IL-6 and GM-CSF mRNA in LPS-activated RAW264.7 cells, but not that of TNF-${\alpha}$ Moreover, taxifolin significantly inhibited the transcriptional activity of nuclear factor-${\kappa}B$ (NF-${\kappa}B$) and activator protein -1 (AP-1). These results suggest that taxifolin may downregulate inflammatory iNOS, IL-$1{\beta}$, IL-6 and GM-CSF gene expressions through inhibition of NF-K and AP-1 activation in LPS-stimulated RAW264.7 cells.

  • (E)-3-Phenyl-1-(2-pyrrolyl)-2-propenone (PPP) is a pyrrole derivative of chalcone, in which the B-ring of chalcone linked to ${\beta}$-carbon is replaced by pyrrole group. While pyrrole has been studied for possible Src inhibition activity, chalcone, especially the substituents on the B-ring, has shown pharmaceutical, anti-inflammatory, and anti-oxidant properties via inhibition of NF-${\kappa}B$ activity. Our study is aimed to investigate whether this novel synthetic compound retains or enhances the pharmaceutically beneficial activities from the both structures. For this purpose, inflammatory responses of lipopolysaccharide (LPS)-treated RAW264.7 cells were analyzed. Nitric oxide (NO) production, inducible NO synthase (iNOS) and tumor necrosis factor-${\alpha}$ (TNF-${\alpha}$) mRNA expression, and the intracellular inflammatory signaling cascade were measured. Interestingly, PPP strongly inhibited NO release in a dose-dependent manner. To further investigate this anti-inflammatory activity, we identified molecular pathways by immunoblot analyses of nuclear fractions and whole cell lysates prepared from LPS-stimulated RAW264.7 cells with or without PPP pretreatment. The nuclear levels of p50, c-Jun, and c-Fos were significantly inhibited when cells were exposed to PPP. Moreover, according to the luciferase reporter gene assay after cotransfection with either TRIF or MyD88 in HEK293 cells, NF-${\kappa}B$-mediated luciferase activity dose-dependently diminished. Additionally, it was confirmed that PPP dampens the upstream signaling cascade of NF-${\kappa}B$ and AP-1 activation. Thus, PPP inhibited Syk, Src, and TAK1 activities induced by LPS or induced by overexpression of these genes. Therefore, our results suggest that PPP displays anti-inflammatory activity via inhibition of Syk, Src, and TAK1 activity, which may be developed as a novel anti-inflammatory drug.

  • A study was conducted to compare the feeding value of urea treated and untreated mustard straw (MS) for sheep. Treated MS was prepared by adding urea-N at 1.84% and followed by packing in a pit silo for 21 days. Two groups of six empty Avikaline ewes were fed untreated (UTMS) and treated (TMS) mustard straw along with 200 g concentrate per head daily for 90 days. Untreated MS had 0.41% N and the urea treatment increased its N value to 1.58 %. The cell wall constituents were decreased in the TMS except for cellulose which remained unaffected. Dry matter intake of TMS was consistently higher than that of UTMS. Digestibility of DM, OM and fibre fractions of MS improved by the urea treatment. Ewes in both groups were in positive N balance while % N retention was lower in UTMS (26.30%) than in TMS (52.14%). The TMS fed group on average consumed 30.2 g DM, 2.9 g digestible crude protein and $0.2MJ\;DE\;per\;kg\;BW\;day^{-1}$ and maintained their weight whereas, the UTMS fed ewes lost weight. The VFA concentration in rumen liquor was higher in TMS than in UTMS. Total-N, ammonia-N and TCA-precipitable-N were also higher in TMS fed ewes. Blood glucose concentrations in the two groups were similar at initiation of the study. However the glucose concentration of UTMS fed group was significantly (p<0.01) lower than those fed UTMS at the termination of the study. Urea-N concentration was also higher in TMS fed group after 90 days of feeding period. It is concluded that urea treatment of MS improved N value of MS from 0.41% to 1.58% along with sizable improvement in nutritive value and in conjunction with 200 g concentrate, TMS can serve as maintenance ration for sheep. ($ME_{lakt}/ME_{m}=1.46$).

  • DNA ploidy is an important prognostic parameter in paediatric B-ALL, but the significance of the S-phase fraction is unclear. In present study, DNA ploidy was assessed in 40 pediatric B-ALL cases by flow cytometry. The DI (DNA index) and percentage of cells in S-phase were calculated using Modfit software. Aneuploidy was noted in 26/40 (65%) cases. A DI of 1.10-1.6 (hyperdiploidy B) was noted in 20/40 (50%) and 6/40 (15%) had a DI>1.60 (triploid and tetraploid range). Some 14/40 (35%) cases had a diploid DI between 0.90-1.05. None of the cases had a DI <0.90 (hypodiploid) or in the 1.06-1.09 (hyperdiploid A) range. The mean S-phase fraction was 2.6%, with 24/40 (60%) having low and 16/40 (40%) high S-phase fractions. No correlation was noted with standard ALL risk and treatment response factors with DI values or S-phase data, except for a positive correlation of low S-phase with high NCI risk category (p=0.032). Overall frequency of hyperdiploidy in our cohort of B-ALL patients was very high (65%). No correlation between hyperdiploidy B and low TLC or common B-phenotype was observed in our study as 42% cases with DI 1.10-1.6 had TLC> $50{\times}10^9$ and 57.1% CD 10 negativity. The study also highlighted that S-phase fraction analysis does not add any prognostic information and is not a useful parameter for assessment in ALL cases. However, larger studies with long term outcome analysis are needed to derive definitive conclusions.

  • This study was carried out to investigate functional activities of the free radical scavenging and germ of Glycin max. Merrill fur cytotoxicity toward P338 and L1210 cells derived from mouse. Effect of crude saponin were examined to oxygen radicals and their scavenger enzymes in liver fractions of Spragae-Dawley(SD) rats. Male rats were fed basic diets of control and experiment diets of 0.5∼1.0% crude saponin. There were no significant differences in hydroxy radical($.$OH) formation of liver mitochondria and microsomes in 1.0% group, while $.$OH formations were significantly decrease in 0.5% and 1.0% saponin compared with control group. Their oxygen radical(O$_2$$\^$$.$/) scavenging activities were significantly decrease in liver cytosol of 0.5% and 1.0% saponin group compared with control group. Soybean germ saponin was isolated purified by the method of HPLC to investigate the cytotoxicity of mouse cells by using the MTT assay. SA-1 saponin fraction of soybean germ showed to inhibit toward growth cell of P338 and L1210 cells and its showed less than 50% cytotoxicity These results suggest that the saponin may play a effective role in attenuating a oxygen radical formations and increasing a scavenger enzyme activities.

  • Samples of eleven brown midrib (ICSU 96031, ICSU 93046, ICSU 96082, ICSU 96078, ICSU 96075, ICSU 95101, ICSU 96034, ICSU 96063, ICSU 45116, ICSA 93-3 and ICSA 3845 X 3816) and nine white midrib genotypes (ICSU 96050, ICSU 96030, ISU 95082, SSG 59-3, FSHI 93-1, FSHI 2219A X 3211, HC 171, ICSA 93-2 and ICSA 93-1) based on their phenotypic appearance were collected at 50 per cent flowering from the sorghum germplasm grown at Research farm of IGFRI, Jhansi. These genotypes were evaluated with respect to crude protein, fiber composition, in-sacco dry matter, OM, cell wall components disappearance/digestibility besides the fodder yield, total phenolic and availability index values. Brown midrib genotypes were lower (p<0.05) in NDF, ADF, cellulose and acid detergent lignin concentration than white midrib genotypes. Mean NDF, ADF, cellulose and lignin contents were 69.4, 42.1, 35.4 and 5.7% in brown mid rib vis-a vis 75.8, 47.5, 39.6 and 7.3% in white mid rib genotypes. Nonsignificant (p>0.05) differences were observed in dry matter, crude protein and organic matter contents between brown midrib and white midrib genotypes. Phenolic contents were significantly (p<0.05) lower in browm mid rib (0.2) than white mid rib (0.3%) sorghum. Brown midrib genotypes exhibited significantly (p<0.05) higher in-sacco DM, OM and CP disappearance than normal (white midrib) genotypes. The mean degradability of DM, OM and CP was 64.1, 62.6 and 79.6% in brown mid rib and 53.1, 54.0 and 76.6% in white mid rib genotypes, respectively. There were no significant (p>0.05) differences between genotypes in extent of fiber fraction degradability though in-sacco NDF and ADF degradability was more by 5 and 4 units, respectively in brown midrib genotypes vis-a-vis white midrib genotypes. Average fodder yield (green and dry g/plant) and availability index (%) values were significantly (p<0.05) higher for brown midrib (474.2, 129.8 and 80.4) genotypes than white midrib (375.0, 104.8 and 69.2) genotypes. Lignin contents had significant negative correlation with DM, OM, NDF and ADF degradability. The results of the study revealed that brown midrib genotypes are superior not only with regard to chemical entities and disappearance of DM and fiber fractions but also better in respect of fodder yield and availability index values. Thus, brown midrib sorghum strains may be useful in increasing digestibility, intake, feed efficiency and animal performance.

  • The nutritive value of lentil straw (Lens culinaris) relative to 4% urea treated wheat straw (w/w) was assessed in adult bucks and milking buffaloes on-station and on-farm, respectively. A digestion-cum-nitrogen balance experiment was conducted with three bucks (24.35 kg) fed ad libitum lentil straw (LS), urea treated wheat straw (UTS) and a 1:1 (w/w DM basis) mixture of LS and UTS (LS:UTS) in a 3$\times$3 Latin square design. Though daily dry matter intake (g kg $W^{0.75}$ or % LW) of bucks fed LS and UTS did not differ significantly, that of LS:UTS was significantly (p<0.05) low. Similarly, digestibility of nutrients (DM, OM and CP) and nitrogen balance (g/day) did not differ significantly for bucks given LS or UTS although a significant positive improvement was evident in these parameters for LS:UTS. The digestibility of fibre fractions (NDF and ADF) was significantly (p<0.05) lower in LS as compared to UTS or LS:UTS. Except for the significantly (p<0.05) lower intake of DCP (g kg$W^{0.75}$), the bucks fed LS had statistically similar nutrient intake as compared to UTS. Feeding of LS:UTS resulted in significantly higher nutrient (DCP, TDN) intake by bucks. The important blood-biochemical parameters (haemoglobin, packed cell volume, glucose, urea nitrogen, total protein) and activity of serum enzymes (alanine aminotransferase and aspartate aminotransferase) did not differ significantly in goats irrespective of dietary treatment. In the onfarm study, eighteen multiparous milking buffaloes were randomly assigned into three equal groups to assess the relative effect of feeding untreated wheat straw (WS), UTS and LS:UTS ad libitum as basal feed with a supplement. The study continued for 3 months and revealed that daily dry matter intake and average milk production (liters/day) by buffaloes offered LS:UTS and UTS was statistically similar but it was significantly higher compare to WS. However, milk composition did not differ significantly among the dietary treatments. It may be concluded that the nutritive value of lentil straw appeared to be no different to UTS, however, a positive synergistic effect was evident by feeding a mixture of LS and UTS on performance of goats and lactating buffaloes.

  • The strength of particle-reinforced metal matrix composites is, in general, known to be increased by the geometrically necessary dislocations punched around a particle that form during cooling after consolidation because of coefficient of thermal expansion (CTE) mismatch between the particle and the matrix. An additional strength increase may also be observed, since another type of geometrically necessary dislocation can be formed during extensive deformation as a result of the strain gradient plasticity due to the elastic-plastic mismatch between the particle and the matrix. In this paper, the magnitudes of these two types of dislocations are calculated based on the dislocation plasticity. The dislocations are then converted to the respective strengths and allocated hierarchically to the matrix around the particle in the axisymmetric finite-element unit cell model. The proposed method is shown to be very effective by performing finite-element strength analysis of $SiC_p$/Al2124-T4 composites that included ductile failure in the matrix and particlematrix decohesion. The predicted results for different particle sizes and volume fractions show that the length scale effect of the particle size obviously affects the strength and failure behavior of the particle-reinforced metal matrix composites.

  • Nitric oxide (NO) is a free radical synthesized from L-arginine by nitric oxide synthase (NOS). It is believed that NO is an important mediator in numerous physiological and inflammatory responses. Particularly, a large amount of NO released from the inducible nitric oxide synthase (iNOS) is mostly associated with inflammatory processes. Overproduction of NO in these processes including sepsis and autoimmune diseases can have deleterious consequences and pathophysiologic relevance. Therefore, for the discovery of new inhibitory agents against iNOS activity, we have evaluated about 100 kinds of natural products after partition into three layers (n-hexane, ethyl acetate and aqueous) from 100% methanol extracts to study inhibitory effects on iNOS activity induced by lipopolysaccharide (LPS) in RAW264.7 cells culture system. As a positive control, curcumin, which is known as an anti-tumor promoter, anti-inflammatory agent as an iNOS inhibitor, was used and showed the dose-dependent inhibitory effect $(IC_{50},\;2.5\;{\mu}g/ml)$. Among tested fractions, the n-hexane fraction of Cimicifuga heracleifolia $(IC_{50}:\;9.65\;{\mu}g/ml)$, Forsythiae fructus $(IC_{50}:\;6.36\;{\mu}g/ml)$, Saposhnikovia divaricata $(IC_{50}:\;5.92\;{\mu}g/ml)$, and the ethyl acetate fraction of Chrysanthemum sibiricum $(IC_{50}:\;2.56\;{\mu}g/ml)$, Gastrodia elata $(IC_{50}:\;3.46\;{\mu}g/ml)$, and the aqueous fraction of Dianthus chinensis $(IC_{50}:\;6.73\;{\mu}g/ml)$, Euonymus alatus $(IC_{50}:\;6.78\;{\mu}g/ml)$, Mechania urticifoloria $(IC_{50}:\;8.01\;{\mu}g/ml)$ showed strong inhibitory activity against LPS-stimulated iNOS. Especially, the ethyl acetate fraction of Chrysanthemum sibiricum $(IC_{50}:\;2.56\;{\mu}g/ml)$, which exhibited the strongest inhibition against iNOS, was fractionated with silica-gel column chromatography. These subfractions exhibited dose-dependent inhibition against iNOS activity in the range of $2.59-5.6\;{\mu}g/ml$ except for fraction No. 3, 4, 5, 6, 8, 9, and 16. Our study shows that Chrysanthemum sibiricum has the strongest inhibitory effect against iNOS activity and has similar effect to curcumin. Therefore, further studies for the identification of active principles from Chrysanthemum sibiricum and investigation for the mechanism of the inhibition of iNOS by active principles will be performed.
