Anticancer Activity of the Branch Extracts from Vaccinium oldhamii through Cyclin D1 Proteasomal Degradation in Human Cancer Cells

  • In this study, we investigated the effect of the extracts from Vaccinium oldhamii on cell proliferation and the regulatory mechanisms of cyclin D1 protein level in human cancer cells. The branch extracts from Vaccinium oldhamii (VOB) showed higher inhibitor effect against the cell growth than leave extracts (VOL) and fruit extracts (VOF) in human colorectal cancer, breast cancer, prostate cancer, non-small lung cancer, pancreatic cancer and liver cancer cells. In addition, VOB decreased cyclin D1 level at both protein and mRNA level. MG132 treatment attenuated VOB-mediated cyclin D1 downregulation. A point mutation of threonine-286 to alanine attenuated cyclin D1 degradation by VOB. In addition, the inhibition of nuclear export by leptomycin B (LMB) attenuated cyclin D1 degradation by VOB. But, the treatment of PD98059 (ERK1/2 inhibitor), SB203580 (p38 inhibitor), SP600125 (JNK inhibitor), LiCl ($GSK3{\beta}$ inhibitor), LY294002 (PI3K inhibitor) or BAY 11-7082 ($I{\kappa}K$ inhibitor) did not affect VOB-induced cyclin D1 degradation. In conclusion, VOB induced cyclin D1 degradation through redistribution of cyclin D1 from the nucleus to cytoplasm via T286 phosphorylation of cyclin D1, which resulted in the inhibition of cancer cell proliferation.

  • There are some degradation factors for amorphous silicon solar cells. Light inducing is one of the factor that explained by Staebler-Wronski effect. Also, hotspot heating could be the reason that makes amorphous silicon solar cell degrade. Hotspot heating is occurred when a solar cell is shaded so this work is investigated into two types of shading condition and how these affect to solar cell differently. Reduced irradiance for whole cell and partially shaded as 0($W/m^2$) while the other part of cell is soaking as 1000($W/m^2$) of irradiance are two conditions that are experimented. The two types of shading show different characteristics of degradations. The result shows that partially shaded cell dropped maximum powerless and slower. Also sudden drop points have shown that should be concerned to decide the number of cells for a string. Otherwise, the current through a shaded cell might flow more than cell's capability. It makes cell and module damaged. This work would help to manufacture modules.

Histological Changes of Tissues and Cell Wall of Rice Straw Influenced by Chemical Pretreatments

  • Sodium hydroxide (SH) or ammonium bicarbonate (AB) were applied to rice straw to investigate the effects on histological change of stem tissue or cell wall before and after in sacco degradation using a scanning electron microscope (SEM) and a transmission electron microscope (TEM). The SEM revealed that, the parenchyma and vascular bundles were distorted by treatment with SH at 30 or 45 g/kg straw dry matter. Faultage between phloem of large vascular bundles and parenchyma occurred with further increasing SH to 60 or 75 g/kg. The cell wall in these stem tissues was crimped when observed by TEM. However, only parenchyma and large vascular tissues were slightly distorted in AB-treated stem. For untreated and AB-treated stems, the initiation of observable ruminal degradation of cell wall was prolonged from 12 h for inner parenchyma to 24 h for sclerenchyma and to 48 h for phloem of small vascular bundles, while the outer epidermis was intact even at 72 h. For SH-treated stem, however, the cell wall from all of the investigated tissues, epidermis, small vascular bundles, sclerenchyma, and parenchyma started to be degraded at 12 h incubation. These results indicate that SH treatment contracts rice straw stem leading to an improvement in rumen degradation, and that the degradation of SH-treated stem is bilateral from inner and outer surface simultaneously.

Effect of Ramping Rate on the Durability of Proton Exchange Membrane Water Electrolysis During Dynamic Operation Using Triangular Voltage Cycling

  • Proton exchange membrane water electrolysis (PEMWE) is an efficient method for utilizing renewable energy sources such as wind and solar powers to produce green hydrogen. For PEMWE powered by renewable energy sources, its durability is a crucial factor in its performance since irregular and fluctuating characteristics of renewable energy sources, especially for wind power, can deteriorate the stability of PEMWE. Triangular voltage cycle is well able to simulate fluctuating wind power, but its effect on the durability has not been investigated extensively. In this study, the performance degradation of the PEMWE cell operated with the triangular voltage cycling was investigated at different ramping rates. The measured current responses during the cycling gradually decreased for both ramping rates, and I-V curve measurements before and after the cycling confirmed the degradation of the performances of PEMWE. For both measurements, the degradation rate was larger for 300 mV s-1 than 30 mV s-1, and they were determined as 0.36 and 1.26 mV h-1 (at the current density of 2 A cm-2) at the ramping rates of 30 and 300 mV s-1, respectively. The comparison with other studies on triangular voltage cycling also indicate that an increase in the ramping rate accelerates the deterioration of the PEMWE performance. X-ray photoelectron spectroscopy and transmission electron microscopy results showed that the Ir catalyst was oxidized and did not dissolve during the voltage cycling. This study suggests that the ramping rate of the triangular voltage cycling is an important factor for the evaluation of the durability of PEMWE cells.

Protective Effects of Ukyium(우귀음, Yougui-yin) in Zinc-induced Apoptosis of C6 Glial Cells (우귀음이 Zinc에 의한 신경교세포의 고사(Apoptosis)에 미치는 영향)

  • Objectives : The objective of the current study is to determine the protective effect of Ukyium(Yougui-yin) on the apoptosis induced by zinc. Methods : Zinc is known to generate reactive oxygen species (ROS), including superoxide anion ($O_2$) and hydrogen peroxide ($H_2O_2$), which eventually contribute to cytotoxicity in a variety of cell types. We investigated the viablity of cells, $H_2O_2$ generation, chromatin condensation and nuclear fragmentation in Hoechst dye staining and $IkB-{\alpha}$ degradation in C6 glial cells of $ZnCl_2$ between pretreatment- and not pretreatment-group with Ukyium. The former methods were researched by Time- and Dose-dependent manners. Results : We demonstrated that pretreatment with Ukyium prevented zinc-induced cell death of C6 glial cells and apoptotic characteristics including chromatin condensation and nuclear fragmentation. Ukyium also prevented $H_2O_2-induced$ cell death. We further confirmed that Ukyium decreased zinc-induced generation of $H_2O_2$ and inhibited degradation of $IkB-{\alpha}$ by zinc in C6 glial ceHs. Conclusions : These data indicated that Ukyium (Yougui-yin) prevents zinc-induced apoptotic death of C6 glial cells via inhibition of ROS generation, such as $H_2O_2$ as well as inhibition of $IkB-{\alpha}$ degradation.

Lycorine induces apoptosis by enhancing protein degradation of survivin in human oral cancer cell lines (Lycorine의 사람 구강 암 세포주에서 survivin 단백질 분해 증진으로 세포자멸사 유도)

  • Lycorine, a natural alkaloid extracted from the Amaryllidaceae plant family, was reported to various physiological and pharmacological effects including anti-cancer activity. Nevertheless, there is no report of the anticancer effect of lycorine in oral cancer cells. The effects of lycorine on cell proliferation and apoptosis were examined through trypan blue exclusion assay, 4'-6-diamidino-2-phenylindole (DAPI) stain, Live/Dead assay, Western blot analysis and RT-PCR. Lycorine suppressed cell viability and induced apoptosis in MC3 and HSC-3 cell lines. Lycorine decreased survivin protein but did not affect its mRNA. It regulated survivin through accelerating protein degradation in a time-dependent manner although neither proteasome nor lysosome was not associated with lycorine-mediated protein degradation. Collectively, our results suggest that lycorine may be a potential therapeutic anti-cancer drug candidate for the treatment of human oral cancer.

EBP1 regulates Suv39H1 stability via the ubiquitin-proteasome system in neural development

  • ErbB3-binding protein 1 (EBP1) is a multifunctional protein associated with neural development. Loss of Ebp1 leads to upregulation of the gene silencing unit suppressor of variegation 3-9 homolog 1 (Suv39H1)/DNA (cytosine 5)-methyltransferase (DNMT1). EBP1 directly binds to the promoter region of DNMT1, repressing DNA methylation, and hence, promoting neural development. In the current study, we showed that EBP1 suppresses histone methyltransferase activity of Suv39H1 by promoting ubiquitin-proteasome system (UPS)-dependent degradation of Suv39H1. In addition, we showed that EBP1 directly interacts with Suv39H1, and this interaction is required for recruiting the E3 ligase MDM2 for Suv39H1 degradation. Thus, our findings suggest that EBP1 regulates UPS-dependent degradation of Suv39H1 to govern proper heterochromatin assembly during neural development.

Simvastatin Induces Avian Muscle Protein Degradation through Muscle Atrophy Signaling (Simvastatin이 메추리 근육 세포에 미치는 영향)

  • Many studies on poultry have been conducted in the poultry industry to improve their important economic traits, such as egg production, meat quality, and carcass yield. Environmental changes affect the poultry's economic traits, including muscle growth. The purpose of this study is to investigate the mechanisms by which simvastatin causes muscle injury in quail muscle cells. Following treatment with various doses of simvastatin, LD50 in the quail myoblast cells was determined using a cell viability test; cell death was caused by apoptosis and/or necrosis. Thereafter, the expression patterns of the atrophy marker genes were examined via quantitative reverse-transcription polymerase chain reaction (qRT-PCR). The results showed that the transcriptional levels of the muscle atrophy marker genes (Atrogin-1, TRIM63) and the upstream genes in their signaling cascade were increased by simvastatin treatment. This indicated that simvastatin induced myogenic cell death and muscle injury via protein degradation through muscle atrophy signaling. Further studies should focus on identifying the mechanism by which simvastatin induces the protein degradation signaling pathway in quail muscle..

  • The degradation of Nafion membrane by hydrogen peroxide was investigated in polymer electrolyte membrane fuel cell (PEMFC). Degradation tests were carried out in a solution of $10{\sim}30%$ hydrogen peroxide containing 4ppm $Fe^{2+}$ ion which is well known as Fenton's reagent at $80^{\circ}C$ for 48hr. Characterization of degraded membranes were examined through the IR, Water-uptake, Ion exchange capacity, mechanical strength and $H_2$ permeability. After degradation, C-F, S-O and C-O chemical bonds of membrane were broken by radical formed by $H_2O_2$ decomposition. Breaking of C-F bond which is the membrane backbone reduced the mechanical strength of Nafion membrane and hence induced pinholes, resulting in increase of $H_2$ crossover through the membrane. Also the decomposition of C-O and S-O, side chain and terminal bond of membrane, decreased the ion exchange capacity of the membrane.
