Eucheuma cottonii 추출물에 의한 인체 골육종암 Saos-2 세포의 자가사멸 유도 (Extract from Eucheuma cottonii Induces Apoptotic Cell Death on Human Osteosarcoma Saos-2 Cells via Caspase Cascade Apoptosis Pathway)

  • 본 연구에서는 인간 골육종암세포주인 Saos-2 세포를 이용하여 Eucheuma cottonii 추출물(Extract of Eucheuma cottonii, EE)의 항암 활성 및 분자적 작용기전을 분석하였다. 먼저 EE가 세포증식에 미치는 영향을 WST-1® assay를 통해 확인한 결과 EE는 인간 위암세포 AGS, 인간 간암세포 SK-Hep 1, 인간 뇌교모세포종 U87MG, 인간 정상 신장세포 HEK-293의 생존율에는 영향을 미치지 않고 Saos-2 세포주의 생존율만을 농도의존적으로 감소시킴을 확인 하였다. 또한 처리 농도가 증가함에 따라 Saos-2 세포의 외형적 변화가 나타남을 도립현미경을 통해 확인할 수 있었다. 그리고 DAPI staining을 통해 apoptosis classical hall marker라고 할 수 있는 DNA fragmentation이 EE 처리 농도 의존적으로 나타남을 관찰할 수 있었다. 이를 토대로 EE가 Saos-2 세포에서 세포 내의 어떠한 기작을 통해 apoptosis를 유도하는지 Western blot analysis를 통해 확인한 결과, Fas-Associated Death Domain(FADD)에 의한 caspase cascade signal pathway 발현이 증가하였고, apoptosis의 key protein 이라고 할 수 있는 cleaved caspase-3와 하위 인자인 cleaved PARP가 증가 함을 확인할 수 있었다. 이와 관련된 분자적 기전 분석을 위한 immunofluorescence staining과 Flow cytometry analysis를 추가로 수행한 결과, EE 처리시 caspase cascade signal pathway의 시발점인 FAS와 cleaved caspase-3의 발현증가가 실제 세포 내에서 일어남을 관찰 할 수 있었으며 apoptosis 유발군인 sub G1기가 증가 함을 알 수 있었다. 이상의 결과를 통해 EE는 인간 골육종암세포에서 FADD에 의한 caspase cascade signal pathway 발현증가가 유도되어 apoptosis를 유발시킨다는 것을 증명하였으며 새로운 골육종암 치료제로서의 개발 가능성과 기전연구를 위한 중요한 기초자료가 될 수 있음을 시사한다.

영지버섯균 발효 꾸지뽕나무 가지 톱밥 추출물의 항염증 활성 (Anti-inflammatory effects of Cudrania tricuspidata twig sawdust fermented with Ganoderma lucidum mycelium)

  • 염증은 외부 자극으로부터 보호하는 중요한 면역반응이다(Jeong et al., 2012). 그러나 NO 및 inflammatory cytokine과 같은 염증 매개 인자의 과도한 생성은 비정상적인 염증 반응을 초래할 수 있다(Paradise et al., 2010). 염증 매개 인자의 생성과 염증 신호 전달을 조절하는 중요한 역할을 하는 대식세포는 LPS 자극에 의해 NF-κB가 활성화 되어 inflammatory cytokine 등의 염증 매개 인자들이 분비가 증가되며 염증 반응을 심화시킨다(Lee et al., 2012). 본 연구에서는 영지버섯균 발효 꾸지뽕나무 가지 톱밥 추출물을 이용하여 LPS로 염증을 유도한 Raw264.7세포에서 염증 관련 인자들의 생성 및 발현에 미치는 영향을 분석하여 항염증 효과를 확인하였다. NO는 염증에서 중요한 역할을 하며 대식세포의 염증 반응 조절 평가시 대표적인 지표로 사용되며 iNOS에 의해 생성이 유도되며 PGE2는 염증 매개 인자로 inflammatory cytokine 생성에 관여하며 COX-2에 의해 생성이 유도된다(Paradise et al., 2010; Posadas et al., 2000). 발효추출물이 RAW264.7세포에서 세포독성 없이 NO의 생성과 PGE2의 생성을 감소시켰으며 이 결과는 발효 추출물 처리에 의해 iNOS와 COX-2의 발현이 감소한 것과 일치하였다. 또한 염증 반응을 조절하는 대표적인 inflammatory cytokine으로 알려진 IL-1β, TNF-α의 생성이 감소되었다. 따라서 발효추출물은 NO, PGE2, inflammatory cytokines 생성을 감소시켜 항염증 효과를 나타낸다고 볼 수 있다. NF-κB는 iNOS, COX-2 및 inflammatory cytokine의 발현을 조절하는 전사인자로 IκB와의 결합에 의해 NF-κB의 핵 내 이동이 억제되며 불활성화상태로 존재한다. LPS 자극에 의해 IκB가 인산화 및 분해되면 NF-κB가 활성화되어 핵 내로 이동하여 염증성 매개인자들의 발현을 유도하고, 염증성 질환 뿐만 아니라 다양한 질환을 유발시키는 것으로 알려져 있다 (Kawai and Akira, 2006; Ghosh and Ksarin, 2002). 본 연구에서는 LPS에 의해 자극된 RAW264.7 세포에서 IκB의 분해 및 NF-κB의 전이가 발효추출물에 의해 감소됨을 확인하였으며 발효추출물에 의한 iNOS, COX-2 및 inflammatory cytokine 감소는 NFκB 활성화 감소에 의해 조절되는 것으로 사료된다. 이러한 결과들을 통해서 발효추출물이 NF-kB 활성화 억제를 통해 염증 매개 인자들의 생성 및 발현을 감소시킴으로서 염증 반응 억제 효과를 나타냄을 확인하여, 염증 관련 질환에 대해 예방 및 개선을 위한 항염증 기능성 소재로 사용될 수 있음을 시사한다.

기장 종자 유래 추출물의 인간 유방암 MDA-MB-231 세포에 대한 세포독성에 관련된 미토콘드리아 손상-의존적 아폽토시스 유도 효과 (Apoptotic Effect of Proso Millet Grains on Human Breast Cancer MDA-MB-231Cells Is Exerted by Activation of BAK and BAX, and Mitochondrial Damage-mediated Caspase Cascade Activation)

  • 잡곡 류에 속하는 기장(Panicum miliaceum L.)의 항암 효과를 알아보기 위해, 기장의 종자를 80% 에탄올(EtOH)로 추출하였으며, 이를 감압 농축하여 건조시키고 재차 물에 녹인 후 4가지 유기용매(헥산, 메틸렌크로라이드, 에틸아세테이트 및 부탄올)로 순차적으로 추출 분획하였다. 다양한 인간 암세포에 대하여 80% 에탄올 추출물의 세포독성을 조사한 결과, 인간 유방암 세포주 MDA-MB-231에 대한 세포독성 효과가 가장 강하게 나타났다. 또한 에탄올 추출물 유래 각 유기용매 분획들 중에서 세포독성이 가장 높게 나타난 부탄올 분획을 사용하여, 유방암 세포주 MDA-MB-231에 대한 아폽토시스성 세포사멸 유도 효과를 조사하였다. 그 결과로서, BAK/BAX 활성화, 미토콘드리아 막 전위(Δψm) 손실, 미토콘드리아 시토크롬 c 방출, 카스파아제-8/-9/-3의 활성화, PARP의 분해, 그리고 TUNEL-양성 아폽토시스성 DNA 단편화와 같은 아폽토시스 반응들이 검출되었다. 한편, 인간 급성백혈병 Jurkat T 세포의 A3 클론(야생형), I2.1 클론(FADD-결손형) 및 I9.2 클론(카스파아제-8 결손형)은 부탄올 분획의 세포독성에 대해 유사한 감수성을 나타내었는데, 이는 부탄올 분획의 아폽토시스 유도 활성에는 외인성 아폽토시스 기전이 관련되지 않음을 시사한다. 흥미롭게도, 인간 정상 유방 상피세포 MCF-10A는 유방암 MDA-MB-231세포에 비해 부탄올 분획의 세포독성에 대하여 훨씬 낮은 감수성를 보였다. 이러한 연구결과는 기장 종자 유래 부탄올 분획의 인간 유방암 세포주 MDA-MB-231에 미치는 세포독성효과는 BAK/BAX 활성화에 따른 미토콘드리아 외막 손상 및 시토크롬 c 방출, 이에 수반되는 카스파아제 활성화에 의해 매개됨을 보여준다.

Ginsenoside compound K protects against cerebral ischemia/ reperfusion injury via Mul1/Mfn2-mediated mitochondrial dynamics and bioenergy

  • Background: Ginsenoside compound K (CK), the main active metabolite in Panax ginseng, has shown good safety and bioavailability in clinical trials and exerts neuroprotective effects in cerebral ischemic stroke. However, its potential role in the prevention of cerebral ischemia/reperfusion (I/R) injury remains unclear. Our study aimed to investigate the molecular mechanism of ginsenoside CK against cerebral I/R injury. Methods: We used a combination of in vitro and in vivo models, including oxygen and glucose deprivation/reperfusion induced PC12 cell model and middle cerebral artery occlusion/reperfusion induced rat model, to mimic I/R injury. Intracellular oxygen consumption and extracellular acidification rate were analyzed by Seahorse multifunctional energy metabolism system; ATP production was detected by luciferase method. The number and size of mitochondria were analyzed by transmission electron microscopy and MitoTracker probe combined with confocal laser microscopy. The potential mechanisms of ginsenoside CK on mitochondrial dynamics and bioenergy were evaluated by RNA interference, pharmacological antagonism combined with co-immunoprecipitation analysis and phenotypic analysis. Results: Ginsenoside CK pretreatment could attenuate mitochondrial translocation of DRP1, mitophagy, mitochondrial apoptosis, and neuronal bioenergy imbalance against cerebral I/R injury in both in vitro and in vivo models. Our data also confirmed that ginsenoside CK administration could reduce the binding affinity of Mul1 and Mfn2 to inhibit the ubiquitination and degradation of Mfn2, thereby elevating the protein level of Mfn2 in cerebral I/R injury. Conclusion: These data provide evidence that ginsenoside CK may be a promising therapeutic agent against cerebral I/R injury via Mul1/Mfn2 mediated mitochondrial dynamics and bioenergy.

이중확장칼만필터(DEKF)를 기반한 건설장비용 리튬이온전지의 State of Charge(SOC) 및 State of Health(SOH) 추정 (State of Health and State of Charge Estimation of Li-ion Battery for Construction Equipment based on Dual Extended Kalman Filter)

  • 전기자동차와 신재생에너지에 관한 관심이 높아지면서 건설장비 산업분야에서도 리튬이온 배터리를 접목하려는 요구가 높아지고 있다. 건설중장비는 건설 현장의 다양한 작업으로 인해 전류 용량의 감소가 급속히 진행되기 때문에 SOC(State of Charge) 및 SOH(State of Health) 같은 배터리의 상태를 더욱 정확하게 추정할 필요가 있다. 본 논문에서는 SOC와 SOH를 동시에 추정이 가능한 적응제어 기법 기반 이중확장칼만필터(Dual Extended Kalman Filter, DEKF) 알고리즘을 이용하여 실제 측정데이터와의 오차를 비교하였다. 배터리 충전 상태 예측을 위해 배터리 셀을 완전 충전 후 0.2C-rate조건에서 SOC 5% 간격으로 OCV를 측정하였고, 배터리의 열화를 판단할 수 있는 건전성 지표 확보를 위해 다양한 C-rate(0.2, 0.3, 0.5, 1.0, 1.5C rate) 조건에서 50 Cycle 동안 노화 실험을 수행하였다. DEKF를 이용한 SOC 및 SOH 추정 오차는 C-rate이 커질수록 커지는 경향을 보였으며 특히 SOC 추정결과, 0.2, 0.5 및 1C-rate에서 6%이하로 나타남을 확인하였다. 또한 SOH 추정 결과는 0.2 와 0.3C-rate에서 각각 최대오차 1.0% 및 1.3% 이내로 좋은 성능을 보이는 것으로 확인하였다. 다만, C-rate가 0.5C-rate에서 1.5C-rate으로 증가함에 따라 추정오차도 1.5%에서 2%로 다소 증가하는 것을 확인할 수 있었으나, 모든 C-rate 조건에서 DEKF를 사용한 SOH의 추정 성능은 약 2% 이내인 것으로 나타났다.

  • 연구배경 : 염증매개 사이토카인은 염증성 폐질환의 중요한 매개물질이다. 폐 상피세포는 염증세포에서 분비되는 사이토카인에 의해 interleukin, chemokines, colony stimulating factors와 growth factor등을 생산 및 분비함으로써 국소 염증 부위에서의 사이토카인 network에 중요한 역할을 한다. 따라서 폐 상피세포에서 염증매개 사이토카인의 발현 기전에 대한 이해는 염증성 폐질환의 기전규명과 이에 기초한 새로운 치료법의 개발에 생각된다. 대부분의 사이토카인은 NF-${\kappa}B$전사인자에 의해 발현되는데 폐 상피세포에서 염증매개 사이토키인의 발현과 NF-${\kappa}B/I{\kappa}B$ 경로와의 관련성에 관한 연구는 부족한 실정이다. 방법 : BEAS-2B, A549, NCI-H157, NCI-H719 세포에서 IL-1$\beta$와 TNF-$\alpha$ 자극에 의한 IL-8과 TNF-$\alpha$ mRNA의 발현 양상을 평가하였고 이들의 발현과 관찰하였고 NF-${\kappa}B/I{\kappa}B$ 경로와의 관련성을 평가하기 위하여 IL-l$\beta$와 TNF-$\alpha$ 자극에 의한 NF-${\kappa}B$의 활성화 및 $I{\kappa}B{\alpha}$$I{\kappa}B{\beta}$의 분해 양상을 관찰하였다. 폐 상피세포의 종류에 따른 NF-${\kappa}B/I{\kappa}B$ 경로 조절의 기전을 규명하고자 IL-1$\beta$와 TNF-$\alpha$ 자극에 의한 $I{\kappa}B{\alpha}$의 인산화와 기저상태에서 IKK$\alpha$의 발현을 평가하였다. 결과 : BEAS-2B, A549, NCI-H157 세포에서는 IL-1$\beta$와 TNF-$\alpha$ 자극으로 $I{\kappa}B{\alpha}$$I{\kappa}B{\beta}$가 분해되었고 NF-${\kappa}B$의 활성화가 관찰되었으며 IL-8과 TNF-$\alpha$mRNA의 발현이 유도되었다. NCI-H719 세포에서는 IL-1$\beta$와 TNF-$\alpha$ 자극으로 $I{\kappa}B$ 분해에 의한 NF-${\kappa}B$의 활성화 및 염증매개 사이토카인의 발현이 관찰되지 않았다. BEAS-2B, A549, NCI-H157 세포에서는 IL-1$\beta$와 TNF-$\alpha$ 자극으로 ${\kappa}B$의 인산화가 관찰되었지만 NCI-H719 세포에서는 관찰되지 않았다. 기저상태의 IKK$\alpha$ 발현은 세포간에 차이가 관찰되지 않았다. 결론 : 폐 상피세포에서 NF-${\kappa}B/I{\kappa}B$ 경로는 염증매개 사이토카인 발현에 매우 중요한 역할을 하고, 일부 세포에서는 NF-${\kappa}B/I{\kappa}B$ 경로 조절의 차이를 보이는데 이는 IKK보다 상위 단계의 세포내 신호전달체계의 이상에 기인한 것으로 생각된다.

Proinflammatory Cytokines과 TGF-beta가 섬유모세포의 증식에 미치는 영향 (The Effects of Proinflammatory Cytokines and TGF-beta, on The Fibroblast Proliferation)

  • 연구배경: 조식의 손상에의한 염증반응후 회복과정에서 정상조직으로 환원되지 않고 일부는 결체조직으로 대체되어 섬유화가 일어난다. 이 비정상적 섬유화에 의해 섬유화증이 일어나 조직의 원래 기능을 상설하기도 한다. 이때 염증에 관계되는 세포로부터 분비된 여러종류의 cytokine들이 섬유모세포의 증식과 교원질 합성을 조절한다. 이에 관계된 cytokine들의 상호 관련성과 섬유모세포증식에의 영향을 알아내어 조절함으로써 섬유화증을 예방하고 치료하는데 도움을 줄 수 있다. 본 연구에서는 proinflammatory cytokine인 TNF-$\alpha$, IL-1$\beta$, IL-6와 섬유모세포의 증식에 큰 영향을 미치는 TGF-$\beta$의 상호 관련성을 알아보고자 하였다. 방 법: 섬유모세포는 사람 태아의 섬유모세포주인 MRC-5를 사용하였으며, 0, 0.5, 1, 5, 10, 20%의 우태아 혈청을 첨가하여 1, 2, 3, 4, 5일째의 MRC-5 증식을 측정하여, 최소한으로 MRC-5의 증식을 억제하면서 생존을 유지시킬 수 있는 배양액중 혈청농도를 먼저 확정한 후, TGF-$\beta$, TNF-$\alpha$, IL-1$\beta$, IL-6를 각각 RPMI 배지에 첨가하여 $37^{\circ}C$, 5% $CO_2$ 보온기에서 96 시간동안 배양하여 0.5 % naphthol blue black으로 염색한 뒤, 50mM의 NaOH로 naphthol blue black을 유리시켜 630nm에서의 분광흡수를 잼으로써 MRC-5의 수를 측정하였다. 또한 TGF-$\beta$와 다른 cytokine의 관련성을 알아보기위해 TGF-$\beta$와 함께 IL-1$\beta$, IL-6, TNF-$\alpha$를 각각 배지에 첨가하여 96 시간 배양 후, 위와 동일한 방법으로 MRC-5의 수를 측정하였으며, TNF-$\alpha$와 IL-1$\beta$, IL-6의 상호 관련성을 알아보기 위해 TNF-$\alpha$와 함께 IL-1$\beta$, IL-6를 각각 배지에 첨가하여 96 시간 배양한 뒤 위와 동일한 방법으로 MRC-5의 수를 측정하였고, 마지막으로 TGF-$\beta$와 TNF-$\alpha$의 섬유모세포 증식에 미치는 상호 관련성을 알아보기 위하여 TGF-$\beta$와 TNF-$\alpha$를 함께 배지에 첨가하여 96 시간 배양 후, MRC-5의 수를 역시 통일한 방법으로 측정하였다. 결 과: 1. 최소한으로 MRC-5의 증식을 억제하면서 생존을 유지할 수 있는 배양액중 혈청 농도를 측정한 결과, 0.5% 혈청 농도에서 MRC-5의 증식이 50% 증가된 상태로 배양 6일째 까지 유지되었다. 따라서 이후의 실험에서 사용된 배양액에는 0.5%의 우태아혈청을 포함시켰다. 2. 각 cytokine이 MRC-5 증식에 미치는 영향 0.5%의 우태아혈청만이 있는 상태에서의 MRC-5 증식 정도를 100%로 기준하였을 때, IL-1$\beta$는 50 ng/ml의 농도에서만 45%의 증식 촉진효과를 보였고, IL-6는 영향이 없었으며, TGF-$\beta$와 TNF-$\alpha$는 농도에 의존성을 보이며 MRC-5의 증식을 최고 160% 까지 증가시켰다. 3. TGF-$\beta$에 의한 MRC-5 증식능증가에 IL-1$\beta$, IL-6, TNF-$\alpha$가 미치는 영향 50 ng/ml의 TGF-$\beta$ 단독 자극에 비하여, IL-1$\beta$ 혼합배양시 농도에 의존성으로 보이며 최고 64% 까지, TNF-$\alpha$ 혼합배양시 농도에 의존성을 보이며 최고 159% 까지 MRC-5의 증식을 증가시켰다. IL-6는 약한 억제효과를 보였으나 TGF-$\beta$에 의한 MRC-5 증식능 증가에 영향을 미치지 못하였다. 4. TNF-$\alpha$에 의한 MRC-5 증식능 증가에 IL-1$\beta$, IL-6가 미치는영향 50 ng/ml의 TNF-$\alpha$ 단독자극에 비하여, IL-1$\beta$을 혼합 배양하였을때 농도에 의존성을 보이며 최고 50%까지 MRC-5의 증식을 억제하였고, IL-6는 영향을 미치지 못하였다. 5. TNF-$\alpha$와 TGF-$\beta$의 MRC-5 증식능 증가에서의 상호관련성 50 ng/ml의 TGF-$\beta$와 TNF-$\alpha$는 MRC-5의 증식을 각각 89%, 135% 증가시켰으며, TGF-$\beta$와 TNF-$\alpha$ 공존시에는 MRC-5의 증식을 222% 증가시켰다. 결 론: TNF-$\alpha$ TGF-$\beta$, IL-1$\beta$의 순서로 MRC-5에 대한 증식 자극효과가 있으며 IL-6는 증식을 억제하는 효과가 있었다. TGF-$\beta$의 MRC-5에 대한 증식 자극효과는 IL-1과 TNF-$\alpha$에 의해 첨가효과를 관찰할 수 있었고, TNF-$\alpha$의 MRC-5 증식 자극효과는 IL-1$\beta$에 의하여 억제되었다.

  • The wall shear stress in the vicinity of end-to end anastomoses under steady flow conditions was measured using a flush-mounted hot-film anemometer(FMHFA) probe. The experimental measurements were in good agreement with numerical results except in flow with low Reynolds numbers. The wall shear stress increased proximal to the anastomosis in flow from the Penrose tubing (simulating an artery) to the PTFE: graft. In flow from the PTFE graft to the Penrose tubing, low wall shear stress was observed distal to the anastomosis. Abnormal distributions of wall shear stress in the vicinity of the anastomosis, resulting from the compliance mismatch between the graft and the host artery, might be an important factor of ANFH formation and the graft failure. The present study suggests a correlation between regions of the low wall shear stress and the development of anastomotic neointimal fibrous hyperplasia(ANPH) in end-to-end anastomoses. 30523 T00401030523 ^x Air pressure decay(APD) rate and ultrafiltration rate(UFR) tests were performed on new and saline rinsed dialyzers as well as those roused in patients several times. C-DAK 4000 (Cordis Dow) and CF IS-11 (Baxter Travenol) reused dialyzers obtained from the dialysis clinic were used in the present study. The new dialyzers exhibited a relatively flat APD, whereas saline rinsed and reused dialyzers showed considerable amount of decay. C-DAH dialyzers had a larger APD(11.70$\pm$1.32mmHg/min)compared to CF dialyzers(4.32$\pm$0.55mmHg/min)(p<0.05). However, there was no observable difference in the UFR between the two dialyzers. Neither APD nor UFR showed any significant increase with an increasing number of reuses for up to more than 20reuses.   • The wall shear stress in the vicinity of end-to end anastomoses under steady flow conditions was measured using a flush-mounted hot-film anemometer(FMHFA) probe. The experimental measurements were in good agreement with numerical results except in flow with low Reynolds numbers. The wall shear stress increased proximal to the anastomosis in flow from the Penrose tubing (simulating an artery) to the PTFE: graft. In flow from the PTFE graft to the Penrose tubing, low wall shear stress was observed distal to the anastomosis. Abnormal distributions of wall shear stress in the vicinity of the anastomosis, resulting from the compliance mismatch between the graft and the host artery, might be an important factor of ANFH formation and the graft failure. The present study suggests a correlation between regions of the low wall shear stress and the development of anastomotic neointimal fibrous hyperplasia(ANPH) in end-to-end anastomoses. 30523 T00401030523 ^x Air pressure decay(APD) rate and ultrafiltration rate(UFR) tests were performed on new and saline rinsed dialyzers as well as those roused in patients several times.
