The morphology of filamentous fungi closely correlates with the productivity in submerged culture. Using itaconic acid (IA) production by Aspergillus terreus as a research model, the quantitative relationship between the growth form of A. terreus and IA production was investigated. IA fermentation was scaled up from shake flasks to a 7 L stirred tank bioreactor based on the quantitative relationship. Our results demonstrated the following: (1) Three morphologies of A. terreus were formed by changing the inoculum level and shape of the flask. (2) Investigation of the effects of the three morphologies on broth rheology and IA production revealed the higher yield of IA on dry cell weight (DCW, IA/DCW) and yield of glucose on DCW (consumed glucose/DCW) were achieved during clump growth of A. terreus. (3) By varying the $KH_2PO_4$ concentration and culture temperature, the relationships between clump diameter and IA production were established, demonstrating that the yield of IA on DCW ($R^2$ = 0.9809) and yield of glucose on DCW ($R^2$ = 0.9421) were closely correlated with clump diameter. The optimum clump diameter range for higher IA production was 0.40-0.50 mm. (4) When the clump diameter was controlled at 0.45 mm by manipulating the mechanical stress in a 7 L fermentor, the yield of IA on DCW and yield of glucose on DCW were increased by 25.1% and 16.3%, respectively. The results presented in this study provide a potential approach for further enhancement of metabolite production by filamentous fungi.
Objectives : The purpose of this study is to investigate characteristics of cell morphology of two different agar woods (Aquilaria agallocha Roxburgh (AAR) and imitative agar wood (IAW)) using microscopic techniques. Methods : Both light and scanning electron microscopes were used to observe morphological and histological features of the cell of agar woods collected from commercial products in the market. Results : The cell morphology and tissue arrangements of AAR depending on either the adsorption of resin into vessels or no adsorption of resin were observed to compare these features with those of both IAW and Aquilaria sinensis Gilb (ASG). Conclusions : As a result, the vessels of AAR with the resin adsorption was packed with lots of yellow-brownish materials, which were quite different from those of IAW and ASG. The cell walls were thick, and had well-developed traumatic parenchyma tissues with squared cells of the same height and width, which appeared as a clump of small grains packed.
This study was conducted to investigate the effects of quiescent treatment of donor cells and activation treatment time of recipient cytoplasm on nuclear remodeling and in vitro development of somatic cell-cloned bovine embryos. Serum starved, confluent and nonquiescent cycling adult skin cells were teansferred into enucleated oocytes. Nuclear transfer oocytes were activated at 30 min, 1 and 2 hrs after electrofusion. Some nuclear transfer embryos(23% to 35%) extruded a polar body, which was not affected by quiescent treatment of donor cells and activiation time of recipient cytoplasm. About 68% of nuclear transfer embryos fused with a serum starved cells has a chromatin clump, but which was not different from embryos fused with confluent(51%) and nonquiescent(47%) cells. The proportion of embryos with a single chromatin clump was sightly increased when nuclear transfer embryos were activated within 30 min after fusion(69%) compared to those were activated at 1 and 2 hrs after fusion, but there was not significantly different. Development rates to the blastocyst stage were 8.6% and 15.9% when serum starved and confluent cells were transferred, which were higher than that of control group. Developmental rate to the blastocyst stage was higher in embryos were activated within 30 min after fusion (17.3%) compared to those of embryos were activated at 1 and 2 hrs after fusion (P<0.05). From the present result, it is suggested that quiescent treatment of donor cells and activation time of recipient cytoplasm can affect the in vitro development. Quiescent plasm activation within 30 min after fusion could increase the number of embryos with a normal chromation structure, which results in increased in vitro development.
This study was conducted to investigate the effect of quiescent treatment of the donor cells on the nuclear remodeling and in vitro development of fetal fibroblast cell-cloned bovine embryos. Serum starved, confluent and nonquiescent cycling fetal fibroblast cells were transferred into the enucleated oocytes. About 20∼25% of nuclear transfer embryos fused with a serum starved or confluent cell extruded a polar body, which was slightly lower than that of nontreated control (36%). About 49∼51% of nuclear transfer embryos fused with a serum starved or confluent cell had a single chromatin clump, which was slightly higher than that of nontreated control (40%). The proportion of embryos with a single chromatin clump was significantly higher (P<0.01) in nuclear transfer embryos without showing a polar body (60.5%) than with a polar body (4.7%). Development rates to the blastocyst stage were 21.7% and 20.9% when serum starved and confluent cells were transferred, which were slightly higher than that of control (14.1 %). The result of this study suggests that quiescent treatment by serum starvation or growth to confluency of donor cells could increase the number of embryos with a normal chromatin structure, which results in increased in vitro development.
Developmental morphology of the red alga, Osmundea crispa (Hollenberg) Nam from California was studied on the basis of liquid-preserved and herbarium specimens. Vegetative axial segment of the species produces two pericentral cells and one trichoblast. Spermatangial filaments (branches) are derived from apical and epidermal cells in pocket-shaped apical pit with an ostiole-like upper opening. Procarp-bearing segment of female trichoblast produces five pericentral cells, of which the fifth functions as supporting cell of carpogonial branch. Tetrasporangial production occurs in random epidermal cells in apical pit of branchlets, and two presporangial cover cells show parallel arrangement to stichidial axis. As this vegetative and reproductive development is included in the generic delineation of Osmundea Stackhouse, O. crispa among the known Osmundea species is characterized by habit forming compact cushion-like clump with angular to terete thallus. It is also distinguished from O. hybrida (AP. de Candolle) Nam without the compressed thallus by the number of pericentral cells in procarp-bearing segment and shape of spermatangial pit. Taxonomic implication of the shape of spermatangial pit is also included.

Numerical simulations for large scale triaxial tests with large diameter rockfill materials are conducted using distinct element method. For generation of compacted assembly with specific grain size distribution and initial material porosity, the clump logic method and expansion of generated particles are adapted. With micro parameters which are chosen by calibration process, discrete particle modelling of triaxial test in case of other confining stress and cyclic loading condition were conducted. Also numerical simulations of fluid injection into particulate materials were conducted to observe cavity initiation and propagation using distinct element method. The fluid scheme solves the continuity and Navior-Stokes equations numerically, then derives pressure and velocity vectors for fixed grid by considering the existence of particles within the fluid cell.
To obtain a basic information of the development of Genus Viola, morphological and histological observation of in vitro calli and cells in Viola culture cells were investigated. There were two callus types obtained by long term subculture of wild viola (Viola partrinii DC. ) petiole callus. One was friable callus - soft and pale green in color and small cells in size, and the other was compact callus - compact and deep bluish green in color, large cells in size. In scanning electron microscopic observation, friable callus was composed of voculated cell around small. cell clump, while compact callus was composed of cells filled with protoplasm Somatic embryogenesis was observed from suspension culture of the compact callus.
Calli were induced from leaf explants of B.falcatum, and selected cell clumps of the calli (900-1, 000${\mu}{\textrm}{m}$) were cultured on MS medium supplemented with 0.1, 0.5, 1.0 or 2.0 mg/L 2, 4-D for 7 days, respectively: The clumps were subsequently transferred onto MS basal medium and subcultured for four weeks. In order to investigate the effect of 2, 4-D pretreatment, the selected clumps were cultured on MS medium supplemented with 0.1 mg/L 2, 4-D for 24, 48, 72, 96, 120 or 144 hours and then transferred to liquid MS basal medium, wherein they were cultured for 4 weeks. Histological observation showed that root initial cells were developed from cells on the surface of clumps or from cells in the inner region. Clumps on the basal medium produced mot within 5 days of culture. The rate of prutruding time was inversely proportional to the concentration of 2, 4-D. The number of adventitious roots per clump preheated with 0.1 mg/L 2, 4-D was an average of 5.2, which was the highest level. On MS medium as control, the clumps formed 3.3 adventitious roots each. As tile concentration of 2, 4-D increased, the number of adventitious roots were declined accordingly: The number of adventitious roots as the period of pretreatment increased upto 120 h.
This study was carried out to select cadmium resistant cell lines from leaf-derived calli of diploid and haploid of Nicotiana tabacum cv. BY4, for understanding adaptation mechanism of plants in cadmium contaminated environment. suspended cell clumps were plated onto selection medium containing 0 to 2,000 $\mu$M cadmium. Cadmium resistant colonies were formed on the selection medium after 3 or 4 weeks of culture. The minimum inhibition concentration (MIC) of cadmium on colony formation were 300 $\mu$M in diploid and 200 $\mu$M in haploid plants, respectively. In order to test the resistance to cadmium, selected cell line on MIC were transferred to medium containing high concentration of cadmium. The selected cell lines, especially haploid cell line, were resistant an the high concentration of cadmium. And dry weight, ash weight, and cadmium contents of cell were increased. These results indicated that the selected cell lines showed higher resistance of cadmium than control cells, and haploid plant is more resistant than diploid plant on medium with cadmium.
연구 목적: 본 연구는 인간배아줄기세포 동결에 minimum volume cooling (MVC) 초자화 동결방법이 유용하게 이용될 수 있는지의 여부를 조사하고자 실시하였다. 연구재료 및 방법: 인간배아줄기세포 콜로니는 0.05% collagenase 처리와 기계적 처리에 의해 작은 clumps로 자른 다음 동결 방법에 따른 효율을 비교 검토하고자, i) 대략 40-50개의 clumps를 10% DMSO가 들어있는 동결액에 $5{\sim}10$분간 처리하여 1ml cryo-vial에 넣고 slow-cooling용 cryo-module에 장착하고 -80C에서 overnight한 후 $LN_2$에 침지하여 완만동결을 실시하거나 ii) 10% ethylene glycol (EG)이 들어 있는 동결액에서 5$\sim$10분 처리하고 30% EG과 0.5 mol sucrose가 들어 있는 동결액에서 30초간 처리하여 본 연구를 위해 개발된 MVC straw에 10 clumps씩 적재한 다음 $4{\sim}5$ MVC straw를 $LN_2$가 들어있는 cryo-vial에 넣어 MVC 초자화동결을 실시하였다. 융해 후 생존율을 조사하였고 배아줄기세포의 특성을 유지하고 있는지의 여부를 조사하였다. 결 과: 융해 후, 인간배아줄기세포의 생존율은 완만동결을 실시했던 군 (20.0%) 보다 MVC 초자화동결을 실시했던 군에서 (76.0%) 유의하게 높게 나타났다. 또한 회복과정에서 완만동결을 실시했던 군에서는 세포성장이 매우 더디고 안 좋은 반면, MVC 초자화동결을 실시했던 군은 배아줄기세포의 증식이 동결을 실시하지 않은 세포와 같은 상태로 2주 이후부터 빠르게 전환되고 회복되는 것을 확인할 수 있었다. 이와 더불어 MVC 초자화동결-융해 후 회복된 배아줄기세포에서 정상 핵형, alkaline phosphatase acitivity, SSEA-4와 TRA-1-60 염색 및 Oct-4 발현을 확인하였으며 체외분화의 특성도 확인하였다. 결 론: 새로이 개발된 MVC 초자화동결을 이용하면 인간배아줄기세포는 고유의 특성을 잃지 않고 성공적으로 동결될 수 있다.
