Examination of the Presence and Differential Expression of Connexin Isoforms in the Urinary Bladder of the Male Rat During Postnatal Period

  • The coordinated action of the detrusor muscle cells in the urinary bladder is governed by cell-cell communication through gap junction, consisted of connexin (Cx) molecules. Even though a number of researches have been mostly focused on expressional changes of a few Cx isoforms in clinically dysfunctional condition of the bladder, less attention has been paid for investigation of Cx isoforms present in the bladder. Using real-time PCR analysis, the present study examined Cx isoforms expressing in the male rat bladder during postnatal period. Also, expressional patterns of Cx isoforms were evaluated in the bladder at different postnatal ages. Of a total of 13 Cx isoforms tested in the present study, we were able to detect mRNAs of 6 Cx isoforms in the rat urinary bladder, including Cxs 31, 31.1, 32, 37, 40, and 45. The transcript levels of Cxs 31, 31.1, 37, 40, and 45 were gradually increased from 1 week of age until 25 days of age, followed by transient decreases at 45 days of age. However, abundance of Cx32 transcript was drastically increased at 15 days of age, followed by a sharp drop at 45 days of age. These results indicate that differential expression of Cx isoforms in the bladder during postnatal development would be necessary for maintaining proper function of the bladder. A question remains to be answered if significant decreases of transcript levels of some Cx isoforms at the elderly are associated with age-dependent dysfunction of the bladder.

Cervical Precancerous Lesions and Cancer among Patients in the Gynaecology Outpatient Department at a Tertiary Hospital in South Africa

  • Background: The purpose of this study was to determine proportional rates of cervical precancerous lesions and cancer, and associations with socio-demographic variables, HIV status, and CD4 cell count. Materials and Methods: A retrospective comparative study was conducted targeting the medical records of all women over age 18 that were diagnosed histologically through colposcopy with cervical precancerous lesions and cancer from 1 December 2011 to 30 November 2012. Results: A total of 313 patient records were used for data analysis. The average age was 39.1 (SD=2.04) years. More than a third (37.1%) of the patients had CIN III or cervical cancer. There was a significant association between age, CD4 cell count, HIV status, and cervical lesions (p<0.05). Age was found to be an influential predictor of cervical lesions (OR=0.67, 95%CI: 0.46-0.98). Conclusions: There exists an association between age and cervical lesions, with presentation of cervical precancerous lesions and cervical cancer at a younger age among HIV reactive patients. The National Guidelines should be brought in line with the National HIV Counseling and Testing Policy to offer Pap smears to all sexually active women that test HIV reactive during routine HIV Testing.

From cell senescence to age-related diseases: differential mechanisms of action of senescence-associated secretory phenotypes

  • Cellular senescence is a process by which cells enter a state of permanent cell cycle arrest. It is commonly believed to underlie organismal aging and age-associated diseases. However, the mechanism by which cellular senescence contributes to aging and age-associated pathologies remains unclear. Recent studies showed that senescent cells exert detrimental effects on the tissue microenvironment, generating pathological facilitators or aggravators. The most significant environmental effector resulting from senescent cells is the senescence-associated secretory phenotype (SASP), which is constituted by a strikingly increased expression and secretion of diverse pro-inflammatory cytokines. Careful investigation into the components of SASPs and their mechanism of action, may improve our understanding of the pathological backgrounds of age-associated diseases. In this review, we focus on the differential expression of SASP-related genes, in addition to SASP components, during the progress of senescence. We also provide a perspective on the possible action mechanisms of SASP components, and potential contributions of SASP-expressing senescent cells, to age-associated pathologies.

자돈뇌실(仔豚腦室) 주위의 subependymal layer의 형태(形態)에 관한 관찰(觀察) (Morphological observation on the subependymal layer of swine brain)

  • Normal swine brains at 1 to 70 days after birth were used to investigate the presence and morphology of the subependymal layer (SL) in the ventricle walls. The brain samples were taken from 27 pigs of 4 swine breeds. The results were summarized as follows: 1. SLs were observed on the walls of the lateral ventricle (LV) but none were observed on the walls of the 3th and 4th ventricles. 2. SLs of the LV walls were composed of mainly 3-to 10-cell layers in thickness. The thinest region of SLs was composed of only 1-to 2-cell thick on the dorsal and ventral walls, and the thickest region was composed of 250-to 300-cell thick on extension region of the SLs into the angle between the corpus callosum and caudate nucleus. 3. Of the LV parts observed, the SL thickness were 25-to 45-cell thick on the anterior horn, 3-to 10-cell thick on the body, 100-to 220-cell thick on the angle region between the corpus callosum and caudate nucleus, and 3-to 5-cell thick the superior walls of the posterior horn. Also the SL thickness was more thick on the anterior region than those on the posterior region. 4. SLs may be classified as three types by the cell distribution; one type of them is closely arranged cell region with the distinctive lateral margin from the periventricular white matter, the other type is loosely arranged cell region with the undistinctive lateral margin, and another type is two-subdivided region as the loosely and closely arranged cell layers in a layer. 5. SLs were extensively thick in young age but gradually decreased in size and cell number with age after 20-day age. SL layers were composed of mainly oligodendrocytes, astrocytes and immature cells of them. Morphological differences of SL in different breeds of pigs were not observed.

노인의 흡연상태와 나이가 SCE 빈도수로 본 임파구 DNA 손상에 미치는 영향 (Effects of Smoking and Age on SCE Frequency Reflecting DNA Damage of Human Lymphocytes in Elderly Koreans)

  • Sister chromatid exchange (SCE) has recently become a common cytogenic assay system for detecting exposure to chemical mutagens and carcinogens. One application of SCE is the monitoring of populations believed to have been exposed to such agents. A cross-sectional study of SCE frequency in peripheral blood lymphocytes from 45 Koreans aged 61 to 84 years was conducted. The effect of cigarette smoking and age on SCE was assessed by different degrees of smoking status such as smokers (n = 14), ex-smokers (n = 16) and non-smokers (n = 15). Mean spontaneous SCE per cell for the smokers (11.5 $\pm$ 1.1) was significantly higher (p < 0.05) than that for the non-smokers (8.8 $\pm$ 0.3). However, mean SCE frequencies per cell for the ex-smokers (10.3 $\pm$ 0.6) were not significantly different from those of the smokers or the non-smokers. The smokers showed an increased number of high SCE frequency cells (HFCs) when compared to the ex-smokers and non-smokers (p < 0.05). The mean SCE frequencies of the non-smokers showed a statistically significant increase (p < 0.05) with the subject's age. These results show that age and smoking habits contribute a great deal in setting a higher degree of basal DNA damage in elderly Koreans, and smoking appeared to be a more significant damaging factor than age.

급성 백혈병 아동의 동종 조혈모세포이식 후 신체적 및 사회적 적응 (Physical Conditions and Social Adaptation of Children Who Have Undergone Hematopoietic Stem Cell Transplantation)

  • Purpose: The purpose of this study was to investigate the physical conditions and social adaptation of children who have undergone hematopoietic stem cell transplantation. Methods: The participants in this descriptive survey were 37 children who had undergone hematopoietic stem cell transplantation at least one year ago. Data were collected through observation and questionnaires and processed with the SAS program. Results: Compared to the time of diagnosis, WBC, RBC and platelet counts had increased significantly. The children were taller than at the transplantation stage, but there was no change in weight and 73% of the children had physical symptoms. Boys who were between 6 to 11 years of age had lower rates of socialization and academic progress than normal Korean boys. Boys between 12 to 17 years of age had higher rates of socialization, but lower rates of academic progress. Girls between 6 to 17 years of age scored lower than normal Korean girls in both categories. Conclusion: The above findings indicate that children who have undergone hematopoietic stem cell transplantation try hard to participate in everyday activities. Therefore nursing interventions to improving normal growth and development and facilitate social adaptation should be provided for them.

The electrochemical properties is researched for the cell to be synthesised and crystallized the positive active material LiMnO2 of the secondary cell at 9250C to get a new improved data of the electric discharge for that the capacitance of the LiMnO2 thin film that is improving and researching with the properties and a merit and demerit in the this kind of asking.

  • This study was designed to investigate age-related changes in body composition and cell mediated immunity. Male Sprague-Dawley rats were fed stock diet ad libitum and 12 rats were sacrified at 1, 4, 6, 12 months of age. Body weight Increased sharply from 1 to 4 months. and Increased steadily thereafter. The weights of liver, epididymal fat pads, and kidney increased in similar pattern as body weight, but their relative ratio to body weight decreased with age. The ratio of epididymal fat pads to body weight increased with age. The weight of extensor digitorium longus, coleus and plantaris increased from 1 to 4 months, but it decreased at 6, 12 months. The protein content of muscles decreased or increased through- out 12 months. The T cell proliferation response to Con A stimulation was significantly lower at 6 months than 1 month and lower at 12 months than 6 months.

Variation of Transcribed X-linked Genes in Bovine Embryos Cloned with Fibroblasts at Different Age and Cell Cycle

  • The present study compared the developmental potential, telomerase activity and transcript levels of X-linked genes (ANT3, HPRT, MeCP2, RPS4X, XIAP, XIST and ZFX) in the bovine somatic cell nuclear transfer (SCNT) embryos derived from different age and cell cycle of female donor nucleus. In experiment 1, the fusion rate, cleavage rate to 2-cell stage, developmental rate to blastocyst stage, and the mean number of total and ICM cells was slightly increased in embryos cloned with fetal fibroblasts compared to those with adult fibroblasts, but there was no significantly (p<0.05) differences. Telomerase activity was also similar in blastocysts cloned with fetal and adult fibroblasts. Up-regulated RPS4X and down-regulated MeCP2, XIAP, and XIST transcript level were observed in blastocysts cloned with adult fibroblasts, compared to those with fetal fibroblasts. In experiment 2, the fusion rate, cleavage rate to 2-cell stage, developmental rate to blastocyst stage, and the mean number of total and ICM cells was significantly (p<0.05) increased in embryos cloned with fetal fibroblasts at early G1 phase of the cell cycle, compared to those of fetal fibroblasts at late G1 phase. DNMT1 transcript was observed to significantly (p<0.05) increased in the fetal fibroblasts at 3 hrs after trypsin treatment of confluent culture. Further, level of telomerase activity and transcribed X-linked genes was also significantly (p<0.05) higher in the early G1 SCNT blastocysts than those of late G1. The results imply that fetal fibroblasts at early G1 phase induces the enhanced developmental potential and up-regulated telomerase activity and X-linked gene, but aberrant transcript pattern of X-linked genes may be displayed in the SCNT embryos.

  • The values of blood composition were analyzed by age, season, and sex from Chejuhorse in Korea. The results obtained were summarized as follow; 1. The values of white blood cell(WBC), packed cell volume(PCV), mean corpuscular volume (MCV), and mean corpuscular hemoglobin concentration(MCHC) were significantly different between age. The WBC value was decreased with the progress of age and the PCV value was lower in the early age of 1,2 to 3 years than in the higher age of 4 to 5, and over 6 years. 2. The mean value of MCV was increased with the progress of age but the adverse was true in MCHC. 3. In the case of seasons, the values of red blood cell(RBC), WBC, hemoglobin(Hb) PCV, MCV, mean corpuscular hemoglobin(MCH), MCHC, globulin(Glo), albumin/globulin(A/G) ratio, inorganic phosphorus(Pi), cholesterol(Chol), and chlorine(Cl) showed significant differences. The number of RBC was not varied from winter to summer but decreased in autumn. The number of WBC was high in summer and autumn, but low in winter and spring. 4. The values of MCV were high in autumn and winter, but low in spring and summer. 5. The values of MCH and MCHC were similar in spring and summer, but strikingly increased in autumn. 6. The values of Glo and A/G ratio were negatively correlated with the season; The value of A/G ratio was lowest in summer but no differences were observed in spring, autumn and winter. On the other hand, the value of Glo was highest in summer but no differences were observed in spring, autumn, and winter. 7. The values of Pi, Chol, and CI were high in summer and winter, but low in spring and autumn. 8. The values of RBC, MCV, and MCH were significantly different between sex. The value of the RBC count in male was higer than the female, but the adverse is true in MCV, and MCH.
