Protective effects of Opuntia ficus-indica on Glutathione and Antioxidative Enzyme (Opuntia ficus-indica가 Glutathione 및 항산화 효소에 미치는 보호효과)

  • The effects of Opuntia ficus-indica (OF) administration on the biochemical parameters of function in liver tissue and serum of $CCl_4$ treated rats were investigated. Opuntia ficus-indica (200 mg/kg) was administered into rats intraperitoneally for two weeks. $3.3m\ell$ of $CCl_4$$_4$ (50% $CCl_4$ : Olive oil = 1 : 1) was treated to rats on the 14th day and 15th day and they were operated on 15th day. We examined the antioxidative enzymatic activity by measuring the level of AST (Aspartate aminotransferase), ALT (Alanine aminotransferase), GSH (Glutathione reduced form), GSSG (Glutathione oxidezed form), GPx (GSH-peroxidase), SOD (Superoxide dismutase) and CAT (Catalase) in serum and liver tissue of rats. OFC administered group showed 24.8% of inhibitory effect in AST activity compared to $CCl_4$ -treated abnormal group (CTA). ALT level of OF administered group was decreased by 60.7% to the level of CTA. GSH, GSSG and GPx of OFC administered group were significantly higher than those of CTA group. SOD and CAT in OFC administered group were increased by 28.3% and by 16.9% respectively compared to those of CTA group.

Effect of Cyclohexanone Treatment on the Serum Levels of Glutathione S-Transferase Activities in Acute Liver Damaged Rats

  • To evaluate an effect of cyclohexanone (CHO) treatment on the serum levels of glutathione S-transferase (GST) activity in acute liver damaged animals, acute liver damage was induced in rats with pretreatment of 50% $CCl_4$ in olive oil (0.1 ml/100 g body wt) intraperitoneally 14 times every other day. To liver damaged rats, CHO (1.56 g/kg body wt, i. p.) was injected once and then rats were sacrificed at 4 hours after injection of CHO. Increasing rate of GST activity to the control in serum was higher in CHO-treated rats pretreated with CCL$_4$ than the $CCl_4$-pretreated those. All the more, the injection of CHO to the liver damaged rats led to more enhanced liver damage on the basis of liver functional findings, i. e., serum levels of alanine aminotransferase (ALT) activity, liver weight per body weight, and malondialdehyde content. The changing pattern of serum ALT activity was similar with that of GST activity, whereas that of liver in both enzymes differed more or less from each other; the liver GST activity in CHO-treated rats pretreated with $CCl_4$ being more increased tendency than that of $CCl_4$-pretreated rats. Concomitantly the injection of CHO showed a increasing tendency of liver GST activity compared with the control. Furthermore, CHO injection to the liver damaged rats showed somewhat higher Vmax in the kinetics of liver GST enzymes. In conclusion, injection of CHO to the liver damaged animals led to more increased activity of serum GST, and it may be chiefly caused by the alteration of membrane permeability.

Protective effects of four types of Taraxaci Herba water extract on carbon tetrachloride-induced acute liver injuries in mice (사염화탄소 유도성 급성 간 손상 모델에서 포공영(蒲公英) 열수 추출물 4종의 간 보호 효능 연구)

  • Objectives : Present study investigated hepatoprotective effects of four types of Taraxaci Herba water extract (TL-F, Taraxaci Herba leaf originated from foreign country; TR-F, Taraxaci Herba root originated from foreign country; TL-K, Taraxaci Herba leaf originated from Korea; TR-K, Taraxaci Herba root originated from Korea) on carbon tetrachloride (CCl4)-induced acute liver injury. Methods : Mice were administered orally with 200 mg/kg of TL-F, TR-F, TL-K, or TR-K for seven days, and intraperitoneally injected with 0.5 mL/kg of CCl4 1 h after last Taraxaci Herba treatment. Silymarin (100 mg/kg) was used as a positive drug. Body weight gain, relative liver weight, serum biochemistry, histopathological and immunohistochemical analyses, and hepatic antioxidant capacity were determined to investigate the hepatoprotective effect of four extracts. Results : Administration of four types of Taraxaci Herba extract increased body weight gain, and decreased relative liver weight in CCl4-injected mice. As compared to CCl4 group, TL-F and TR-F significantly decreased serum aspartate aminotransferase activity, while four extracts reduced CCl4-induced alanine aminotransferase activity. In addition, TL-F and TR-F significantly decreased the numbers of degenerated hepatocytes, infiltrated inflammatory cells, cleaved caspase-3 positive cells, and cleaved PARP positive cells in hepatic tissues. Moreover, TL-F, TR-F, and TR-K administration reduced the lipid peroxidation and nitrotyrosine, and increased glutathione, superoxide dismutase, and catalase activities in hepatic tissues. There were no statistical differences between TL-F- and silymarin-treated group. Conclusion : Of four extracts tested, present results suggest that TL-F is the most favorable candidate against CCl4-induced acute liver injury through enhancing antioxidant activity.

Oncolytic Vaccinia Virus Expressing 4-1BBL Inhibits Tumor Growth by Increasing CD8+ T Cells in B16F10 Tumor Model

  • Oncolytic viral vectors have shown good candidates for cancer treatment but have many limitations. To improve the therapeutic potential of oncolytic vaccinia virus, we developed a recombinant vaccinia virus expressing the 4-1BBL co-stimulatory molecule or CCL21. 4-1BBL and CCL21 expression was identified by FACS analysis and immunoblotting. rV-4-1BBL vaccination shows significant tumor regression compared to rV-LacZ, but rV-CCL21 shows rapid tumor growth compared to rV-LacZ in the poorly immunogenic B16 murine melanoma model. 4-1BBL expression resulted in the increase of the number of CD8+ T cells and especially the increase of effector (CD62L-CD44+) CD8+ T cells. These data suggest 4-1BBL may be the potential target for enhancement of tumor immunotherapy.

A Study on the Fluorination of Pentachloroethane (Pentachloroethane의 불소화 반응에 관한 연구)

  • Pentachloroethane($CHCl_2CCl_3$) was synthesized and reacted with hydrogen fluoride using antimony pentahalide catalyst($SbCl_xF_y$) in order to manufacture HCFC-123$(CF_3CHCl_2)$, a potential CFC-11$(CFCl_3$) substitute candidate. Products analyses showed the fluorination proceeds through fluorine-chlorine exchanges between $HF/SbCl_xF_y$ and $SbCl_xF_y/CCl_3CHCl_2$ respectively. The degree of fluorination of $CCl_3$ group in pentachloroethane was greatly affected on the reaction temperature, but the effect of catalyst concentration was relatively small. Mechanistic study was also performed to elucidate the pathway to the formation of side-products such as $CCl_3CFCl_2$, $CFCl_2CFCl_2$ and $CF_2ClCFCl_2$.

Protective Effects of Moschus Against Carbon Tetrachloride-Induced Acute Hepatotoxicity in ICR Mice (사향이 CCl4로 유발된 mouse의 간 손상에 미치는 영향)

  • Objectives : This study was aimed at investigating liver protection mechanism of Moschus by inducing liver toxicity through $CCl_4$ in mice and evaluated histological and serological findings. Methods : Experiment groups was categorized into untreated normal group, $CCl_4$ treated control group, and orally administered Moschus experiment group. At the termination of experiment, gross examination of the liver as well as histological findings, and Total protein, Total bilirubin, Direct bilirubin SGOT, SGPT, and ALP contents in the serum were evaluated. Results : 1. For gross examination and histological findings, $CCl_4$ treated control group showed destroyed lobular structure, increased fibrosis, as well as hepatic cirrhosis. For the group treated with Moschus, the lobular structure suffered less damage, and showed lower level of fibrosis and liver cirrhosis compared to the control group. 2. For serum analysis, Total protein were significantly increased in the Moschus experiment group than the control group. 3. Total bilirubin didn't show significant differences between the two groups. but direct bilirubin was significantly increased in the Moschus experiment group than the control group. 4. SGOT, SGPT, were significantly decreased in the normal and Moschus experiment groups compared to the control group. 5. ALP was significantly decreased in the normal group compared to the control group, but Moschus experiment group didn't show significant differences compared to the control group. Conclusion : Taken together, Moschus can be effectively used for recovering the liver functions and further researches must be conducted to verify the efficacies of Moschus bile juice.

Influence of Ganyeumilhobang on Acute and Chronic Liver Injury in Experimental Animal (간염1호방(肝炎1號方)이 실험동물(實驗動物)의 급만성(急慢性) 간손상(肝損傷)에 미치는 영향(影響))

  • This study was to investigate the hepatoprotective and anticirrhotic effects of Ganyeumilhobang(GIE) on the acute and chronic liver injury induced by various agents. Chronic liver injury induced by dimethylnitrosamine(DMN) ; a new experimental model for cirrhosis and the intraperitoneal injection of dimethylnitrosamine in the rat. Acute liver njury induced by carbon tetrachloride$(CCl_4)$ and D-galactosamine ; a experimental model for acute liver injury, the administration of $CCl_4$ and the intraperitoneal injection of D-galactosamine in the rat. The development of fibrosis and acute liver injury by the three prescriptions were examined by the chemical analysis of AST, ALT, prothrombin time and hydroxyproline. The results obtained were as follows. 1. The increasing level of hydroxyproline volume induced by DMN in mice was decreased by the oral administration of GIB. 2. The degree of histological fibrosis and hepatic inflammatory cell infiltration induced by $CCl_4$ decreased by the oral administration of GIB. 3. The increase of senun AST and ALT of mice with acute liver damage induced by $CCl_4$ and D-galactosamine was inhibited by the administration of GIB. 4. The prolongation of prothrombin time(seconds) of mice acute liver damage induced by $CCl_4$ was shortened by the oral administration of GIB. 5. The liver of mice was hepatectomized partial1y after the oral administration of GIB. The mitotic index(% of nuclei), weight of liver, contents of protein, RNA and DNA synthesis of the liver tissue were increased by the oral administration of GIB.

Effects of Injinsaryung-san on Experimental Liver Damage in Rats (인진사령산이 흰쥐 간손상(肝損傷)에 미치는 영향(影響))

  • The following results were made by observation GOT, GPT, ALP, total cholesterol and higlyceride in serum to research the effects of medicines which are Injinsaryungsan(Sample- A) and another medicine(Sample-B) of which Injin(Artemisiae Capillaris Herba)was increased in quantity on liver damaged by $CCl_4$ and d-galactosamine in rats. 1. The high concentrated extracts of Sample A group and Sample- B group showed significant inhibltory effects on the increase of serum GPT, ALP, LDH, total cholesterol and triglyceride levels induced by $CCl_4$ and d-galactosamine. 2. The high concentrated extracts of sample A group and Sample B group showed more significant inhibitory effects(P〈0.001) than the low ones' effects(P〈0.01) on the increase of serum triglyceride level induced by $CCl_4$ 3. Sample-A group showed significant inhibitory effects on the increase of serum GOT, GPT. ALP, total cholesterol and triglyceride levels, but no significance on the increase of serum LDH level induced by d-galactosamine. 4. Sample-B group showed very significant inhibitory effects on the increase of serum GOP, GPT, ALP, LDH, total cholesterol and triglyceride levels induced by d-galactosamine. 5. As compared with Sample-A group, Sample-B group of which Injin was increased in quantity showed more significant inhibitory effects on all items of this experiment induced by $CCl_4$ and d-galactosamine. As mentioned above. it seemed that both Injinsaryungsan and another medicine of which Injin was increased in quantity had effects protecting liver and anti-fatty liver induced by $CCl_4$ and d-galactosamine in rats. Specially Sample-B group had very significant effects on liver damage as compared with Sample-B group. Therefore it seems that more researches on variation according to the increase of Injin dose must be continued for curing liver diseases.

Antioxidant Effect and Liver Protection Effect of Spatholobi Caulis Water Extract (계혈등 물추출물의 항산화 및 간보호효과)

  • Objectives : This study investigated whether the water extract of Spatholobi Caulis (SCE) has the ability to protect hepatocyte against oxidative stress induced by tert-butylhydroperoxide (tBHP) in vitro and $CCl_4$ in vivo. Methods : In vitro, HepG2 cells pre-treated with Spatholobi Caulis water extract (1, 3, 10, $30{\mu}g$/ml) for 12h and further incubated with tBHP ($100{\mu}M$) for the next 12h. Cell viability was assessed by MTT assay. In vivo, rats were orally administrated with the aqueous extract of Spatholobi Caulis (SCE; 50, 100 mg/kg) for 4 days and then, injected with $CCl_4$ 1 mg/kg body weight to induce acute liver damage. Results : Treatment with SCE inhibited cell death induced by tBHP, as evidenced by alterations in the levels of the proteins associated with apoptosis:SCE prevented a decrease in $Bcl_2$, and cleavage of poly(ADP-ribose)polymerase and pro-caspase-3. Moreover, SCE inhibited the ability of tBHP to generate $H_2O_2$ production, thereby restoring GSH content. Moreover, SCE treatments in rats effectively decreased liver injuries induced by a single dose of $CCl_4$, as evidenced by decreases in hepatic degeneration and inflammation as well as plasma alanine aminotransferase and lactate dehydrogenase activities. Consistently, treatments of SCE also protected liver in rats stimulated by $CCl_4$, as indicated by restoration GSH and prevention of MDA in the liver. Conclusions : SCE has the ability 1) to protect hepatocyte against oxidative stress induced by tBHP and 2) to prevent $CCl_4$-inducible acute liver toxicity. Present findings may be informative not only in elucidating the pharmacological mechanism of Spatholobi Caulis, but in determining its potential application for oxidative cellular damage in the liver.

The Therapeutic Effect of Artemisia Capillaris Extract on Hepatic Damage Induced by Carbon Tetrachloride in Rats (흰쥐에서 사염화탄소 투여로 유발된 간 손상에 대한 인진쑥 추출물의 치료효과)

  • This experiment was conducted to find out the therapeutic effect of Artemisia capillaris extracts on hepatic damage in rats induced by carbon tetrachloride ($CCl_{4}$). In this experiment, 96 Sprague-Dawley rats were used as experimental groups, which were divided into 4 groups; control group(A), $CCl_{4}$-treated group(B), $CCl_{4}$+Artemisia extract-treated group(C) and $CCl_{4}$+silymarin-treated group(D). The B, C, D group were administrated single dose of $CCl_{4}$(2.5 ml/kg) to induce acute hepatic injury. C group was administrated with Artemisia capillaris extract(200 mg/kg/day) and D group treated with silymarin(50 mg/kg/day) for 7 days. Hematological, ultrasonographical, histological examinations and examination of antioxidant activity were also performed in all groups. AST and ALT activities of C group were significantly decreased compared with B group. The activities of AST and ALT in C and D groups returned to the normal range more rapidly than those of B group. In ultrasonographic examination, the echogenicity of liver in C group was significantly decreased compared with B group. Also C and D group had tended to recover faster than B group on liver histogram. Histologically, the percentage of degenerative regions and degenerative cell numbers in peri-central vein hepatic parenchyma of C and D group were significantly decreased compared with B group. In examination of lipid peroxidation, malondialdehyde of hepatic tissue in C group was decreased as compared with B group. In examination of antioxidant enzyme activity in liver, glutathione peroxidase and catalase activities were significantly increased compared with B group. As results of this study, it is thought that A. capillaris extract has therapeutic effects on hepatic injury induced by carbon tetrachloride, and has the similar therapeutic effects as silymarin in rats.
