암을 치료하기 위한 양이온성 리포솜은 화학치료요법의 분야에서 발전되어 왔다. 양이온성 리포솜은 음이온성의 세포 표면에 정전기적 상호작용에 의해 결합이 된다. 본 연구의 목적은 음이온성 세포와 이온결합을 할 수 있는 양이온성 리포솜을 제조하는 것이다. 양이온성 리포솜은 지질인 1,2-디스테로일-sn-글리콜-3-포스포에탄올아민 (DSPE)과 양이온성 고분자인 폴리에틸렌이민 (PEI)을 그라프트 중합으로 합성된 물질로부터 제조하였다. 리포솜 표면의 이온 특성은 제타 포텐셜을 측정하여 확인하였다. 소 혈청 수용액에서 리포솜의 혈장 단백질 흡착 특성은 입자 크기와 탁도 변화를 측정하여 확인하였다. 완충 용액 속에서 리포솜 안정성을 예측하기 위하여 입자 크기를 7일 동안 상온에서 측정하였다. 양이온성 리포솜은 소 혈청 수용액에서 많은 양의 혈장 단백질이 흡착되었다. 이는 혈장 단백은 음전하를 가지고 있어서 양이온성 리포솜의 표면과 잘 붙기 때문이다. 양이온성 폴리에틸렌이민을 이용한 리포솜의 표면 변형은 소 혈청 수용액 내에서 단백질 흡착을 강화시킨다는 것을 예상하게 한다. 또한, 제조된 리포솜의 완충 용액 내에서 7일 동안 안정한 상태임을 관찰하였다.
To better understand the effects of PEGylation and biotinylation on the delivery efficiency of proteins, the cationic protein lysozyme (LZ) and anionic protein bovine serum albumin (BSA) were chemically conjugated with poly(ethylene glycol) (PEG) and biotin-PEG to primary amine groups of proteins using N-hydroxysuccinimide reactions. Four types of protein conjugates were successfully prepared: PEGylated LZ (PEG-LZ), PEGylated BSA (PEG-BSA), biotin-PEG-conjugated LZ (Bio-PEG-LZ), and biotin-PEG-conjugated BSA (Bio-PEG-BSA). PEG-LZ and Bio-PEG-LZ exhibited a lower intracellular uptake than that of LZ in A549 human lung cancer cells (in a two-dimensional culture). However, Bio-PEG-BSA showed significantly improved intracellular delivery as compared to that of PEG-BSA and BSA, probably because of favorable interactions with cells via biotin receptors. For A549/fibroblast coculture spheroids, PEG-LZ and PEG-BSA exhibited significantly decreased tissue penetration as compared with that of unmodified proteins. However, Bio-PEG-BSA showed tissue penetration comparable to that of unmodified BSA. In addition, citraconlyated LZ (Cit-LZ) showed reduced spheroid penetration as compared to that of LZ, probably owing to a decrease in protein charge. Taken together, chemical conjugation of targeting ligands-PEG to anionic proteins could be a promising strategy to improve intracellular delivery and in vivo activity, whereas modifications of cationic proteins should be more delicately designed.
The selective extraction behavior of lactoferrin (Lf) from whey protein mixture was examined using reverse micelles formed by the cationic surfactant, cetyldimethylammonium bromide (CDAB). The major whey proteins, including ${\beta}$-lactoglobulin, ${\alpha}$-lactalbumin and bovine serum albumin, were solubilized from aqueous phase to organic phase while Lf was recovered in the aqueous phase. The solubilization behaviors of the proteins were manipulated by the process parameters such as the pH and salt concentration of the aqueous phase and the surfactant concentration in the organic phase. Efficient forward extraction was achieved with sodium borate buffer (50 mM, pH 9) containing 50 mM KCl and organic phase containing 100 mM CDAB. Based on SDS-PAGE and densitometry, about 96% of the initial Lf remained in the aqueous phase after forward extraction. The dialyzed Lf fully maintained its bacteriostatic activity against E. coli O157:H7.
CHO/dhfr- 세포에 대해 LipofectAmine$^{TM}$을 이용한 유전자 이입 효율을 증가시키기 위하여 지질과 DNA의 최적 농도를 구하였다. Reporter 유전자로서 GFP 유전자를 이용하였으며, 여러 농도의 지질 DNA로 유전자 이입된 각 세포군에서 나타나는 green fluorescence intensity를 FACS 분석함으로써 유전자 이입 효율을 정량화 할 수 있었다. 그 결과 24-well plate에서 $2.0{\mu}L$LipofectAmine$^{TM}$과 $0.4{\mu}g$ DNA를 조합하여 사용했을 때 최적의 유전자 이입 효율이 나타남을 알 수 있었다. 또한, GFP는 유전자 이입 최적화를 수행하는 데에 여러가지 면에서 유용한 수단이 될 수 있음을 확인할 수 있었다.
나노 또는 마이크로 크기를 가지는 구형의 약물 전달체이다. 그러나 일반적인 리포솜은 혈류 순환시 혈장 단백질과의 흡착이 일어나 안정성이 떨어지고, 세망내피계의 대식세포에 의해 옵소닌작용이 일어나 혈중에서 쉽게 소실되는 단점이 있다. 이에 본 연구에서는 모델단백질로 소혈청 알부민(BSA)을 사용하였고, BSA의 등전점보다 높은 pH를 나타내는 수용액에 용해하여 BSA가 음이온성을 갖도록 제조하였으며 이를 양이온성 리포솜 표면에 정전기적 인력에 의해 결합시켰다. 그리고 리포솜 표면에 코팅된 알부민을 60oC 이상의 온도에서 변성시켜 알부민이 코팅된 리포솜을 제조하였다. 대조 리포솜과 양이온성 리포솜의 입자크기는 104±1nm를 나타내었고, 변성된 알부민이 결합된 리포솜은 109±1nm의 입자크기를 나타내었다. 모델약물로서는 독소루비신(doxorubicin, DOX)을 사용하였고, 이온구배에 의한 리모트 로딩 방법을 사용하여 리포솜 내부에 DOX를 봉입시켰다. 혈장 내에서 리포솜의 안정성을 평가한 결과, 알부민이 결합된 리포솜은 입자크기의 변화가 관찰되지 않았고, 대조 리포솜과 양이온 리포솜에 비해 단백질 흡착이 억제되어 변성된 알부민으로 코팅된 리포솜은 혈류 내에서 장기 순환이 가능한 약물전달체로서 유용할 것이라 사료된다.
Small multidrug resistance(SMR) family(TC #2.A.7.1)에 속하는 막단백질 중 하나를 coding하는 Bacillus subtilis의 yvaE유전자의 발현을 유도한 결과, 발현된 YvaE단백질에 의해 대장균세포내에서 살균제를 포함한 다양한 양이온 약제에 대한 efflux활성을 촉매하는 것으로 확인되었다. 같은 operon내에 인접한 yaD유전자의 동시발현을 유도한 결과, 이러한 efflux활성은 억제가 되는 것으로 나타났다. Ethidium bromide를 기질로 하여 fluorimeter를 이용한 efflux transport실험결과, YvaE 단백질이 발현된 대장균세포의 경우 vector만을 함유한 대조세포에 비해 현저히 빠른 efflux활성을 보여주었다 따라서 YvaE 막단백질은 multidrug export를 촉매하는 SMR-type efflux pump임을 확인하였다.
The prevalence of atopic dermatitis (AD) in school-age children has increased in industrialized countries. As diet is one of the main factors provoking AD, some studies have suggested that food additives in processed foods could function as pseudoallergens, which comprise the non-immunoglobulin E-mediated reaction. Eosinophil cationic protein (ECP) is an eosinophil granule protein released during allergic reactions to food allergens in patients with AD. Thus, serum ECP levels may be a useful indicator of ongoing inflammatory processes in patients with AD. The purpose of this study was to investigate the effect of consuming MSG in processed foods on serum ECP levels among children with AD. This study was performed with 13 patients with AD (age, 7-11 years) who had a normal range of total IgE levels (< 300 IU/ml). All participants ate normal diets during the first week. Then, six patients were allocated to a processed food-restricted group (PRDG) and seven patients were in a general diet group (GDG). During the second week, children in the PRDG and their parents were asked to avoid eating all processed foods. On the third week, children in the PRDG were allowed all foods, as were the children in the GDG throughout the 3-week period. The subjects were asked to complete a dietary record during the trial period. Children with AD who received the dietary restriction showed decreased consumption of MSG and decreased serum ECP levels and an improved SCORing score on the atopic dermatitis index (P < 0.05). No differences in serum ECP levels or MSG consumption were observed in the GDG. Serum total IgE levels were not changed in either group. In conclusion, a reduction in MSG intake by restricting processed food consumption may lead to a decrease in serum ECP levels in children with AD and improve AD symptoms.
백삼단백질의 가열에 의한 내열특성을 알아보기 위하여 가열시간에 따른 내열성 단백질의 함량과 전기영동패턴을 조사하고, CM-cellose 컬럼크로마토그라피 방법으로 얻어진 분획을 가열하여 내열성 단백질과 비내열성단백질의 이온성분의 단백질함량을 분석하고 분자량을 확인하였다. 그 결과 가열시간에 따른 내열성 단백질의 함량은 주근 중심이 주근 주피보다 전체적으로 높았으며 두부위 모두 가열시간이 90분 이후부터는 감소하는 경향이 커졌다, 가열시간에 따른 내열성 단백질량은 가열시간이 길어짐에 따라 단백질은 감소하였고, 가열시간 30분까지는 66, 45, 29, 24, 22, 20, 12 kD 등 7개의 밴드가 가열시간 60분부터는 분자량 22 kD는 소실되었음이 확인되었다. 단백질 함량은 내열성 단백질이 비내열성 단백질보다 높게 나타났으며, 내열성 단백질 함량은 양이온성 단백성분 분획이 28.24%로 음이온성 단백성분 0.80%보다 훨씬 높았다. 내열성 단백질의 분자량은 66, 55, 36 kD 임을 확인하였다.
Background : The level of serum eosinophil cationic protein(ECP) is elevated in Atopic Dermatitis patients. Objective : The aim of this study was to investigate the usefulness of serum ECP as a tool of evaluate the efficacy of herb medicine for atopic dermatitis. Material and Method : We investigated 20 patients suffering from atopic dermatitis and analyzed the relationship among the serum level of ECP, IgE, Eosinophil count, and clinical disease activity. Result: Significant elevation in the serum level of ECP, IgE, Eosinophil count is observed in Atopic Dermatitis. Conclusion : The serum level of ECP may be considered to be an useful tool in evaluate effect of herb medicine for atopic dermatitis.
Gene therapy is a method for the treatment of diseases with introducing the gene-engineered materials into a patient with gene-deficiency disease (e.g. cystic fibrosis) or cancer to produce a therapeutic protein in a patient's cells. Successful gene therapy requires establishing both gene expression systems and delivery systems. Viral and non-viral vectors have been used for gene delivery. Viral vectors have a high transfection efficiency, but are limited in relations to issues of safety, toxicity and immunogenecity. Non-viral vectors are easy to prepare and relatively safe. However, non-viral vectors have a low transfection efficiency. Cationic liposomes are the most available among non-viral vectors. Cationic liposomes have been used to transfect cells both in vitro and in vivo experiments. Besides, several formulations containing cationic lipid are being used in clinical trials in cases of cystic fibrosis or cancer. A crucial subject to the further development of gene delivery vectors will be a long-term gene expression with following characteristics; protecting and deliverying DNA efficiently, non-toxic and non-immunogenic, and easy to produce in large scale.
