1) The hydroxy-substitution in the simple phenolic compounds follows an additivity rule in the chemical shifts of their aromatic carbon atoms. In para-and ortho-effects is a good agreement between calculated and measured values, but the meta-effect is not certain. 2) The additivity rule was applied to assign the chemical shifts of catechins. 3) The nuclear overhauser effect was applied to assign the chemical shifts of C-8 and C-6 atoms of catechins and their polymer. The signal of C-8 is lower in intensity and appear in lower field than C-6. 4) The results of the NOE were applied to determine the bonding positions of catechin units in the catechin dimer and trimer. The bonding positions are C-8a and C-8b atoms of the second and third catechin units. 5) It was tried to determine the conformation of the catechin dimer and trimer by analysing the signal shapes of C-3' and C-4' atoms in the catechol moieties. The catechol moieties lie in opposite side in the dimer and trimer structure. A combined analysis of $^{13}C-and\;^1H-NMR$ results lead to the suggestion that such a catechin polymer is a zigzag planar form.
This study was conducted to determine changes in phenolic compounds of astrigent persimmons before and after softening process and evaluate discoloring properties of major phenolic compounds. Phenolic compounds in soft persimmons were mainly composed of catechins and chlorogenic acid. Although contents of phenolic compounds were reduced during the softening process, little change in the ratio of catechins to total phenolic compounds was observed. Most of phenolic compounds in damaged astringents persimmons were existed in the high molecular weight fraction and more phenolic compounds were extracted at the temperature higher than room temperature. To evaluate discoloring abilities of phenolic compounds, phenolic compounds were dissolved separately into water or 80% methanol. With presence of various amounts of anti-discoloring agents such as vitamin C, citric acid, and L-cystein, (+)catechin was significantly reduced.
Over-consumption of alcohol leads to many side-effects such as malnutrition, liver disease, and neuronal disorders and many investigators have tried to identify methods for preventing the side-effects of drinking. This study was carried out to investigate the effect of the beverage contained green tea catechins (GTC) on the alcohol administered rats. We observed that blood alcohol concentration level decreased significantly in plasma. GTC (200 mg/kg) also reduced the aspartate aminotransferase (AST) and alanine aminotransferase (ALT) level of the intoxicated rats. These results suggest that GTC may be useful for the prevention and therapy of hepatotoxic pathogenesis.

In the course of searching for BACE1 (${\beta}$-secretase) inhibitors from natural products, the ethyl acetate soluble fraction of green tea, which was suspected to be rich in catechin content, showed potent inhibitory activity. (-)-Epigallocatechin gallate, (-)-epicatechin gallate, and (-)-gallocatechin gallate ware isolated with IC$\_$50/ values of 1.6${\times}$10$\^$-6/ M, 4.5${\times}$10$\^$-6/ M, and 1.8${\times}$10$\^$-6/ M, respectively. Seven additional authentic catechins were tested for a fundamental structure-activity relationship. (-)-Catechin gallate, (-)-gallocatechin, and (-)-epigallocatechin significantly inhibited BACE1 activity with IC$\_$50/ values of 6.0${\times}$10$\^$-6/ M, 2.5${\times}$10$\^$-6/ M, and 2.4${\times}$10$\^$-6/ M, respectively. In this study, we evaluated the antioxidant activity of pine needle extracts prepared with hot water, ethanol, hexane, hot water-hexane (HWH), and hot water-ethanol (HWE), using the DPPH (1,1-diphenyl-2-picrylhydrazyl) radical method. The hot water extract possessed superior antioxidant activity than the other extracts. We also compared the antioxidant activity of pine needle extracts through ROS inhibition activity in a cellular system using MC3T3 E-1 cells. The hot water extract exhibited the lowest ROS production. The pattern of HPLC analysis of each extract indicated that the hot water extract contained the highest proanthocyanidin level. The pine needle hot-water extract was then isolated and fractionated with Sephadex LH-20 column chromatography to determine the major contributor to its antioxidant activity. The No.7 and 12 fractions had high antioxidant activities, that is, the highest contents of proanthocyanidins and catechins, respectively. These results indicate that the antioxidant activity of procyanidins from the hot water extract of pine needles is positively related to not only polymeric proanthocyanidins but also to monomeric catechins. Moreover, the antioxidant activity of the pine needle hot water extract was similar to well-known antioxidants, such as vitamin C. This suggests that pine needle proanthocyanidins and catechins might be of interest for use as alternative antioxidants.
국내산 녹차의 채엽시기가 녹차 중의 카테킨, 카페인, 아미노산 및 무기질 함량과 조성에 미치는 영향에 대하여 조사하였다. 녹차의 총 카테킨 함량은 40.61-52.04mg/g으로 채엽시기가 늦을수록 증가하였으며, 카테킨 조성은 채엽시기에 관계없이 EGC, EGCg, EC, CC, ECg, C, GCg 및 Cg의 순으로 함유되어 있음을 알 수 있었다. 카테킨류 중 기능성 작용이 우수한 EGC 및 EGCg는 전체의 50% 이상을 차지하였고 온화한 떫은맛으로 후미에 감미를 남기는 유리형 카테킨인 EC와 EGC는 전체의 53.21%에서 57.09%를 차지하였다. 카페인 함량은 4월차 17.56mg/g, 5월차 15.71mg/g, 6월차 14.92mg/g 및 7월차 14.61mg/g으로 채엽시기가 늦을수록 감소하는 경향을 나타냈다. 총 아미노산 함량은 4월차 45.84mg/g, 5월차 22.64mg/g, 6월차 12.22mg/g 및 7월차 10.45mg/g으로 채엽시기가 늦을수록 감소하였으며 특히, 중요한 생리적 기능을 갖는 theanine의 감소 폭이 큰 것으로 타났다. 무기질 함량은 채엽시기가 늦어질수록 Fe, Mn 및 Mg는 증가하였으며, Cu는 감소함을 알 수 있었다.
HPLC에 의한 카테킨류 5성분의 정량분석법을 검토한 결과, 종래의 이동상을 acetonitril, acetic acid 및 methanol 수용액에서 acetontrile, N,N-dimethyl formamide, ethyl acetate의 0.06% 인산수용액으로 전환하였을 때 컬럼에서의 용출시간을 45분에서 28분으로 줄일 수 있었으며, 특히 (-)-epigallocatechin-3-gallate(EGCG)나 (-)-epicatechin gallate(ECG)의 peak를 종래의 넓은 형태에서 예리한 chromatogram을 얻으므로써 분석값의 오차를 최소화 할 수 있었고 머므름시간$(0.32{\sim}3.97%)$이나 peak 면적$(1.61{\sim}7.02%)$의 변동계수도 신뢰할 수 있는 값을 얻었다. 또한 시판되고 있는 수종의 녹차류와 홍차 및 대만산 오룡차를 분석한 결과 녹차류의 카테킨 함량은 $120.3{\sim}153.7\;mg/g$ 이었고 홍차는 녹차류의 1/4수준 이었다.
저자들은 녹차 카테킨(GTC)이 강한 항 혈전 작용을 나타내며, 이는 항 혈소판 활성에 의한 것임을 보고한 바 있다. 본 연구에서는, 8가지 녹차 카테킨 성분들의 항 혈소판 활성을 비교하였다. 실험결과, 갈레이트(gallate) 구조를 갖는 카테킨들(EGCG, GCG, ECG, CG)은 콜라겐$(5{\mu}g/ml)$으로 유도한 토끼 혈소판 응집능을 강하게 억제하였으며, 50% 억제농도$(IC_{50})$는 각각 79.8, 63.0, 168.2, $67.3{\mu}M$이었다. 또한 EGCG, GCG, CG는 아라키돈산(AA, $100{\mu}M$)으로 유도한 토끼 혈소판 응집능을 억제하였고, 50% 억제농도$(IC_{50})$는 각각 98.9, 200.0, $174.3{\mu}M$이었다. 반면에, 갈레이트 구조를 가지지 않는 카테킨들은 혈소판 응집능 억제 효과가 매우 약했다. 이 결과는 항 혈소판 활성에서 카테킨들의 C-3 위치의 갈레이트 구조의 존재가 매우 중요하다는 것과 카테킨들과 B-ring 갈레이트 구조의 존재 또한 녹차카테킨의 항 혈소판 활성에 중요한 작용한다는 것을 의미한다. 그리고, EGCG는 농도 의존적으로 세포내 칼슘 생성과 아리키돈산의 생성을 억제시켰는데, 이는 혈소판 응집능의 억제와 일치하였다. 반면에, EGC는 세포 내 칼슘 및 다른 혈소판 활성 기전에 아무런 영향이 없었다. 이들 결과는 EGCG의 항 혈소판 활성은 C-ring에서 carbon 3 자리의 에스테르화 된 갈레이트 구조의 존재에 의해서 강화된 항 혈소판 작용으로, 아라키돈산 생성과 세포 내 칼슘 생성을 억제하는 효과에 기인한 것이라 사료된다.
본 연구는 tannase를 이용하여 녹차의 생물학적 전환에 대한 최적 추출 조건을 확립하고 최적 추출 조건으로 마련된 녹차 추출물에 대해 라디칼 소거능 검증을 통해 항산화력의 향상 정도를 평가하고자 하였다. 그 결과 tannase의 반응은 0.5시간 내에 대부분 이루어지며 처리 1시간까지도 일부 작용이 이루어져 최적 추출물을 획득하기 위한 tannase의 반응 시간을 1시간으로 결정하였다. 기질 농도 1% 이상의 경우에서는 EC 및 EGC 전환율이 오히려 낮아져 tannase 작용을 위한 기질 농도는 1%가 적당한 것으로 판단되었다. 또한 tannase 농도가 증가함에 따라 EC, EGC 및 gallic acid는 증가되었으며 일정 농도(30 U/mL)에서 급격한 증가를 나타내었고, 이 농도부터는 통계적으로 EC와 EGC 전환율이 증가하지 않는 것으로 보아 tannase의 적정 반응 농도는 30 U/mL으로 판단되었다. 위의 조건으로 마련된 최적의 tannase 처리 녹차 추출물의 라디칼 소거능은 tannase 처리 전 녹차 추출물에 비해 ABTS와 DPPH 라디칼에 있어 모두 유의하게 소거능이 증가되는 것으로 나타나 tannase 처리 최적 추출 조건에 의해 녹차의 항산화력이 향상됨을 알 수 있었다.
녹차, 부분발효차, 발효차 및 후발효차 등 다양한 차 제품 분말을 NIRSystem를 이용하여 전 대역(400-2500 nm)에서 스펙트럼을 얻어 중회귀분석에 의해 색상 관련 특성 및 카테킨류 성분에 대한 검량식을 유도하였다. Hunter color scale에서 발효정도의 변이를 a/b로 98.7% 설명될 수 있어 a/b값으로 차제품의 발효정도를 추정할 수 있다. MPLS를 이용하여 작성된 검량식의 결과 L값의 검량식 작성시 결정계수는 0.997, a값의 결정계수는 1.00, b값의 결정계수는 0.996, a/b값의 결정계수는 0.999로 매우 높았다. MPLS를 이용하여 작성된 검량식의 결과 EGC, EC, EGCg, ECg 및 총 카테킨의 결정계수는 각각 0.919, 0.896, 0.978, 0.905 및 0.983으로 매우 높았다. 차 제품 분말의 색상관련 특성들(L, a, b, a/b) 및 카테친류 (EGCg, EGC, EC, ECg 및 총 카테킨) 의 검량식 정확도가 매우 높아서 NIRS의 가시광선 대역(400~700 nm)및 근적외선 대역(780~2500 nm)에서 동시에 두 특성의 성분을 1~2분 내로 측정이 가능하다.
