The Functional Role of Phospholipase D Isozymes in Apoptosis (세포사멸에서 Phospholipase D 동위효소의 기능적 역할)

  • Phospholipase D (PLD) catalyzes the hydrolysis of phospholipid to phosphatidic acid (PA), a lipid secondary messenger. Two forms of PLD isozymes, phosphatidylcholine-specific PLD1 and PLD2, have been identified. PLD has emerged as a critical regulator of cell proliferation and survival signaling, and dysregulation of PLD occurs in a various illnesses, including cancer. PLD activity is essential for cell survival and protection from apoptosis. Overexpression of PLD isozymes or PLD-generated PA attenuates the expression of apoptotic genes and confers resistance to apoptosis. The apoptosis-related molecular mechanisms of PLD remain largely unknown. Recently, the dynamics of PLD turnover during apoptosis have been reported. The cleavage of PLD isozymes as specific substrates of caspase differentially regulates apoptosis. PLD1 is cleaved at one internal site, and PLD2 is cleaved two sites at the front of the N-terminus. The cleavage of PLD1 reduces its enzymatic activity, probably via the dissociation of two catalytic motifs, whereas the cleavage of PLD2 does not affect the catalytic motifs and its activity. Thus, PLD2 maintains antiapoptotic capacity, despite its cleavage. Therefore, the differential cleavage pattern of PLD isozymes by caspase affects its enzymatic activity and antiapoptotic function. Thus, PLD is considered a potential target for cancer therapy. We summarize recent studies regarding the functional role of PLD in apoptosis.

Catalytic Application of Metal-Organic Frameworks for Chemical Fixation of CO2 into Cyclic Carbonate (CO2로부터 5원환 탄산염의 화학적 고정화 반응을 위한 Metal-Organic Frameworks의 촉매적 응용)

  • The chemical fixation of CO2 into cyclic carbonates is considered to be one of the most promising way to alleviate global warming and produce fine chemicals. In this work, the catalytic applicability of metal-organic frameworks (MOFs) as porous crystalline materials for the synthesis of five-membered cyclic carbonate from CO2 and epoxides was reviewed. In addition, we have briefly classified the materials based on their different structural features and compositions. The studies revealed that MOFs exhibited good catalytic performance towards cyclic carbonate synthesis because of the synergistic effect between the acid sites of MOFs and nucleophile. Moreover, the effect of structure of designed MOFs and mechanism for the cycloaddition of CO2 were suggested.

  • Alkaline oxygen electrocatalysis, targeting anion exchange membrane alkaline-based metal-air batteries has become a subject of intensive investigation because of its advantages compared to its acidic counterparts in reaction kinetics and materials stability. However, significant breakthroughs in the design and synthesis of efficient oxygen reduction catalysts from earth-abundant elements instead of precious metals in alkaline media still remain in high demand. One of the most inexpensive alternatives is carbonaceous materials, which have attracted extensive attention either as catalyst supports or as metal-free cathode catalysts for oxygen reduction. Also, carbon composite materials have been recognized as the most promising because of their reasonable balance between catalytic activity, durability, and cost. In particular, heteroatom (e.g., N, B, S or P) doping on carbon materials can tune the electronic and geometric properties of carbon, providing more active sites and enhancing the interaction between carbon structure and active sites. Here, we focused on boron and nitrogen doped nanocarbon composit (BNC nanocarbon) catalysts synthesized by a solution plasma process using the simple precursor of pyridine and boric acid without further annealing process. Additionally, guidance for rational design and synthesis of alkaline ORR catalysts with improved activity is also presented.

Physico-chemical effects of cerium oxide on catalytic activity of CeO2-TiO2 prepared by sol-gel method for NH3-SCR (CeO2가 졸겔법으로 합성한 CeO2-TiO2계 SCR용 촉매의 활성에 미치는 물리화학적 영향)

  • The effects of $CeO_2$ on catalytic activity of $CeO_2-TiO_2$ for the selective catalytic reduction (SCR) of $NO_x$ were investigated in terms of structural, morphological, and physico-chemical analyseis. $CeO_2-TiO_2$ catalysts were synthesized with three different additions, 10, 20, and 30 wt% of $CeO_2$, by the sol-gel method. The XRD peaks of all specimens were assigned to a $TiO_2$ phase (anatase) and the peaks became broader with the addition of $CeO_2$ because it was dispersed as an amorphous phase on the surface of $TiO_2$ particles. The specific surface area of $TiO_2$ increased with the addition of $CeO_2$ from $60.6306m^2/g$ to $116.2791m^2/g$ due to suppression of $TiO_2$ grain growth by $CeO_2$. The 30 wt% $CeO_2-TiO_2$ catalyst, having the strongest catalytic acid sites ($Br{\Phi}nsted$ and Lewis), showed the highest $NO_x$ conversion efficiency of 98 % at $300^{\circ}C$ among the specimens. It was considered that $CeO_2$ contributes to the improvement of the $NO_x$ conversion of $CeO_2-TiO_2$ catalyst by increasing specific surface area and catalytic acid sites.

  • The formation of $C_2-C_{10}$ hydrocarbons from methanol over shape-selective ZSM-5 zeolite catalysts is studied. It seems that $C_2-C_5$ olefins formed from methanol via dimethylether are transformed further to higher hydrocarbons containing higher concentration of aromatics by the acid sites of ZSM-5. Unique cross linked channel structure and its hydrophobicity seems to be mainly responsible for its high activity of ZSM-5 catalyst for the conversion of methanol.

Nucleotide Sequence, Structural Investigation and Homology Modeling Studies of a Ca2+-independent α-amylase with Acidic pH-profile

  • The novel $\alpha$-amylase purified from locally isolated strain, Bacillus sp. KR-8104, (KRA) (Enzyme Microb Technol; 2005; 36: 666-671) is active in a wide range of pH. The enzyme maximum activity is at pH 4.0 and it retains 90% of activity at pH 3.5. The irreversible thermoinactivation patterns of KRA and the enzyme activity are not changed in the presence and absence of $Ca^{2+}$ and EDTA. Therefore, KRA acts as a $Ca^{2+}$-independent enzyme. Based on circular dichroism (CD) data from thermal unfolding of the enzyme recorded at 222 nm, addition of $Ca^{2+}$ and EDTA similar to its irreversible thermoinactivation, does not influence the thermal denaturation of the enzyme and its Tm. The amino acid sequence of KRA was obtained from the nucleotide sequencing of PCR products of encoding gene. The deduced amino acid sequence of the enzyme revealed a very high sequence homology to Bacillus amyloliquefaciens (BAA) (85% identity, 90% similarity) and Bacillus licheniformis $\alpha$-amylases (BLA) (81% identity, 88% similarity). To elucidate and understand these characteristics of the $\alpha$-amylase, a model of 3D structure of KRA was constructed using the crystal structure of the mutant of BLA as the platform and refined with a molecular dynamics (MD) simulation program. Interestingly enough, there is only one amino acid substitution for KRA in comparison with BLA and BAA in the region involved in the calcium-binding sites. On the other hand, there are many amino acid differences between BLA and KRA at the interface of A and B domains and around the metal triad and active site area. These alterations could have a role in stabilizing the native structure of the loop in the active site cleft and maintenance and stabilization of the putative metal triad-binding site. The amino acid differences at the active site cleft and around the catalytic residues might affect their pKa values and consequently shift its pH profile. In addition, the intrinsic fluorescence intensity of the enzyme at 350 nm does not show considerable change at pH 3.5-7.0.

Methanol-to-Olefin Reaction over MWW and MFI Zeolites: Effect of Pore Structure on Product Distribution and Catalyst Deactivation (MWW와 MFI 제올라이트에서 메탄올의 올레핀으로 전환 반응: 세공 구조가 생성물 분포와 촉매의 활성 저하에 미치는 영향)

  • Methanol-to-olefin (MTO) reaction was studied over MWW zeolite with independently developed two pores (circular and straight) and MFI zeolite with intercrossed sinusoidal and straight pores in order to investigate the effect of pore structure on their catalytic behavior. MWW and MFI zeolites with similar acidity exhibited commonly high conversion and slow deactivation in the MTO reaction, but their product selectivities were considerably different: linear hydrocarbons of $C_3-C_9$ were mainly produced on MWW, while the yield of $C_2{^=}$ and aromatics were high on MFI. Polyaroamatic hydrocarbons (PAHs) were accumulated on MWW, but a small amount of benzene and aromatics on MFI. The impregnation of phosphorous on MWW caused significant decreases in the catalytic activity and toluene adsorption, but the decreases were relatively small on MFI. Although the straight pores of MWW were inactive in the MTO reaction due to the accumulation of PAHs, its circular pores which suppressed the formation of PAHs sustained catalytic activity for the production of linear hydrocarbons. Therefore, the impregnation of phosphorous on the circular pores of MWW caused a significant decrease in catalytic activity. The phosphorous impregnation on the cross sections of MFI altered the product selectivity due to the neutralization of strong acid sites, but catalytic deactivation was negligible. The difference of MWW and MFI zeolites in the MTO reaction was explained by their difference in pore structure.

A Study on NH3-SCR Vanadium-Based Catalysts according to Tungsten Content for Removing NOx Generated from Biogas Cogeneration (바이오가스 열병합 발전에서 발생하는 NOx 제거를 위한 텅스텐 함량에 따른 NH3-SCR 바나듐계 촉매 연구)

  • In this study, a vanadium catalyst study was conducted on the various characteristics of the exhaust gas in the Selective-Catalytic-Reduction (SCR) method in which nitrogen oxides emitted from cogeneration using biogas are removed by using ammonia as a reducing agent and a catalyst. V/W/TiO2, a commercial catalyst, was used as the catalyst in this study, and the effect was confirmed according to the tungsten content under various operating conditions. As a result of the NH3-SCR experiment, the denitrification performance was confirmed at 380 ~ 450 ℃ more than 95%, and durability to trace amounts of SO2 was confirmed through the SO2 durability experiment and TGA analysis. As a result of H2-TPR analysis, the higher the tungsten content, the better the redox properties. Accordingly, enhanced oxidizing properties were confirmed in the oxidation test for a trace amount of carbon monoxide emitted from the cogeneration. In NH3-DRIFTs analysis, it was confirmed that the higher the tungsten content, the higher both the Bronsted/Lewis acid sites and the better the thermal durability when tungsten is added to the catalyst. Based on the experiments under various operating conditions, it is considered that a catalyst with a high tungsten content is suitable to be applied to cogeneration using biogas.

Dehydration Reaction of Fructose to 5-Hydroxymethylfurfural over Various Keggin-type Heteropolyacids (Keggin형 헤테로폴리산에 의한 과당의 5-하이드록시메틸퍼퓨랄로의 전환을 위한 탈수반응)

  • Four Keggin-type heteropolyacids, $H_nXM_{12}O_{40}$(X = P and Si, M = W and Mo) that were substituted with heteroatom and polyatom were applied to the dehydration reaction of fructose to 5-hydroxymethylfurfural (HMF). The results showed that the acid became stronger when the heteroatom and polyatom were substituted with P and W than the cases of Si and Mo, respectively. However, the amount of acidic sites increased with the decrease in the acid strength, resulting in the change of the catalytic activity of heteropolyacids in the dehydration reaction. The experimental results revealed that four different heteropolyacids produced similar amounts of HMF via the dehydration reaction of fructose due to the counterbalancing effect between the amount of active sites, which is related to the catalytic activity of heteropolyacids, and the softness of polyanion. In addition, it was observed that the prepared heteropolyacids showed good structural stability after heat treatment at $200^{\circ}C$.

Synthesis of High-energy-density Fuel through Dimerization of Bicyclo[2.2.1]hepta-2,5-diene over Co/HY Catalyst (Co/HY 제올라이트 촉매상에서 Bicyclo[2.2.1]hepta-2,5-diene 이량화를 통한 고에너지밀도 연료 제조)

  • The dimer of bicyclo [2.2.1] hepta-2,5-diene (norbornadiene) can be used as a high-energy-density fuel. The purpose of this study is to investigate the effect of Co loading on the acid properties of HY zeolite catalyst and the catalytic activity in norbornadiene dimerization. When the cobalt was loaded on the HY zeolite catalyst, the amount of acid sites did not change, but the acid strength weakened. This can be attributed to the decrease in $Br{\ddot{o}}nsted$ acid site and the increase in Lewis acid site. The norbornadiene conversion and yield of norbornadiene dimer over the Co/HY catalyst showed higher than those over the HY zeolite catalyst. The higher activity of the Co/HY catalyst can be ascribed to the higher amount of Lewis acid sites over the Co/HY catalyst. Density and calorific values of the norbornadiene dimer prepared by using the Co/HY catalyst agreed well with the known values in the literature. It was confirmed that the norbornadiene dimer prepared in this study can be used as a high-energy-density fuel.
