Lucigenin (Lg)- and luminol (Lm)-dependent chemiluminescence (CL) was used to compare the respiratory burst of rockfish (Sebastes schlegeli) phagocytes after stimulation with phorbol myristate acetate (PMA). To establish which reactive oxygen species (ROS) contributes to the observed CL, the modulators of ROS metabolism, such as superoxide dismutase (SOD), catalase, and sodium azide $(NaN_3)$ were used. Although LgCL responses were inhibited significantly by the addition of either SOD or catalase, in comparison to the control, significantly lower LgCL responses were recorded by SOD than catalase. LmCL also showed significantly decreased responses by the addition of SOD and catalase. However, there were no statistical differences in CL responses between SOD and catalase additions. More profound and significant decrease of LmCL responses were recorded by simultaneous addition of SOD and catalase. Sodium azide markedly enhanced LgCL responses, while it significantly inhibited LmCL responses. These results indicate that LgCL and LmCL can be used to measure extracellular $O_2$ production and myeloperoxidase (MPO)-mediated ROS production in fish phagocytes, respectively. Furthermore, LmCL can be used for analyzing intracellular ROS production by simultaneous addition of both SOD and catalase.
Five different types of catalases from photosynthetic bacterium Rhodospirillum rubrum S1 grown aerobically in the dark were found in this study, and designated Catl (350 kDa), Cat2 (323 kDa), Cat3 (266 kDa), Cat4 (246 kDa), and Cat5 (238 kDa). Analysis of native PAGE revealed that Cat2, Cat3, and Cat4 were also produced in the cells anaerobically grown in the light. It is notable that only Cat2 was expressed much more strongly in response to the anaerobic condition. Enzyme activity staining demonstrated that Cat3 and Cat4 had bifunctional catalase-peroxidase activities, while Catl, Cat2, and Cat5 were typical monofunctional catalases. S1 cells grown aerobically in the presence of malate as the sole source of carbon exhibited an apparent catalase Km value of 10 mM and a Vmax of about 705 U/mg protein at late stationary growth phase. The catalase activity of Sl cells grown in the anaerobic environment exhibited a much lower Vmax of about 109 U/mg protein at late logarithmic growth phase. The catalytic activity was stable in the broad range of temperatures (30$\^{C}$-60$\^{C}$), and pH (6.0-10.0). R. rubrum S1 was much more resistant to H$_2$O$_2$in the stationary growth phase than in the exponential growth phase regardless of growth conditions. Cells of stationary growth phase treated with 15 mM H$_2$O$_2$for 1 h showed 3-fold higher catalase activities than the untreated cells. In addition, L-glutamate induced an 80-fold increase in total catalase activity of R. rubrum S1 compared with magic acid. Through fraction analyses of S1 cells, Cat2, Cat3, Cat4 and Cat5 were found in both cytoplasm and periplasm, while Catl was localized only in the cytoplasm.
In order to examine the antioxidant activities of SJDBT(Sibjeondaebo-tang), the study was done through measurement of parameters such as LPO(lipidperoxidation), GSH(glutathione), SOD(superoxidation dismutase), catalase, GOT, GPT, ALP. The results were obtained as follows: For the weight changes, the kidney and testis of group given SJDBT showed decrease in the weight compared to the control group, and the left cerebrum, right cerebrum, cerebellum and liver of group given SJDBT showed increase. But the changes were not significant. The left cerebrum and right cerebrum of group given SJDBT showed significant decrease in the content of LPO compared to the control group, and in the activity of SOD and catalase showed significant increase. The cerebellum of group given SJDBT showed significant decrease in the content of LPO and GSH compared to the control group, and in the activity of SOD and catalase showed significant increase. For the changes of LPO, GSH in the liver, the group given SJDBT showed significant decrease in the content of LPO and GSH compared to the control group, and in the activity of catalase showed significant increase. For the changes of LPO in the kidney, the group given SJDBT showed significant decrease in the content of LPO compared to the control group. For the changes of LPO, GSH and the activity of SOD, catalase in the testis, the group given SJDBT showed significant decrease in the content of LPO and GSH compared to the control group, and in the activity of SOD and catalase showed significant increase. From above results, the antioxidant action of SJDBT is effective. And it is expected to be necessary to the study of the mechanism in the antioxidant of SJDBT.

This study was undertaken to evaluate the effects of catalase using xanthine (X) - xanthine oxidase (XO) system on in vitro maturation and fertilization in pig. When follicular oocytes were cultured in maturation medium with X and/or XO, the maturation rates were not significantly different between in medium with and without catalase despite of different culture periods. However, significantly (P<0.05) higher maturation rates were obrained in culture with X-XO system. The rates of degenerated oocytes were increased with culture periods prolonged, and were significantly (P<0.05) higher in medium without than with catalase at 120 h of culture. On the other hand, the parthenogenetic oocytes were observed with high proportions at 72 h of culture, hut were not different in medium with and without catalase at various times of culture. In another experiment, the frozen-thawed boar spermatozoa treated with X-XO system for in vitro fertilization. The penetration rates were higher in medium with that than without catalase during the in vitro fertilization with, none (P<0.05), XO and X+XO. On the other hand, when sperm were treated with none, X, XO and X+XO, lipid peroxidation were higher in medium without that than with catalase. However, the changes in sperm penetration and lipid peroxidation showed opposite patterns. The sperm suspensions were also treated with X and/or XO for assay of sulfhydryl (-SH) group content. Under the above all conditions, sperm-SH group were higher detected In medium with that than without catalase. The activity of sperm binding to zona pellucida was also evaluated through binding to salt-stored porcine oocytes. In control group, sperm binding to zona pellucida were higher than in medium with X, XO and X+XO groups. No significant differences, however, were observed between medium with and without catalase. In conclusion, the exposure of follicular oocytes and spermatozoa to X-XO system may be caused stimulating in vitro maturation and fertilization in pig. It has been believed that the increased release of free oxygen radicals ($O_2^-,H_2O_2$, and $OH^-$) might be a factor in the pathogenesis of periodontal diseases. Antioxidant enzymes such as glutathione peroxidase(GSH-PX) and catalase can protect the tissue damage from the $H_2O_2$. In order to investigate the GSH-PX and catalase activity in the blood plasma and red blood cells(RBCs) of the patients with periodontitis, 19 patients who had good general health, attachment loss more than 6 mm and bone loss were selected as periodontitis group, 7 patients who had severely inflamed gingiva were selected as gingivitis group, and 15 volunteers with good general and periodontal health were selected as normal group. 17 of 26 patients were performed scaling and root planing to reduce the gingival inflammation for gingivitis and periodontitis groups, and were selected as posttreatment group. After blood plasma and RBCs were collected and separated 1 ml of peripheral blood from each subject, GSH-PX activity in blood plasma and RBCs was measured by the same method that Stefan et al. did, and catalase activity in RBCs was measured by the same method that Beers et al. did. The difference of GSH-PX and catalase activity between normal, gingivitis, and periodontitis groups was statistically analyzed by ANOVA with SPSS/PC+ program, and the difference between pretreatment and posttreatment groups was analyzed by Student t-test. The results were as follows : 1. GSH-PX activity in blood plasma was significantly lower in the gingivitis group($0.8683{\pm}0.0658$), periodontitis group($0.7130{\pm}0.1333$) than in the normal group($1.0241{\pm}0.0801$)(p<0.05), and GSH-PX activity in RBCs was significantly lower in the gingivitis groupt. $0.8156{\pm}0.1167$), periodontitis group($0.7533{\pm}0.1185$) than in the normal group($l.1963{\pm}0.2044$)(P<0.05), but there was no statistical significance in the difference of GSH-PX activity in RBCs between the gingivitis group and periodontitis group(p>0.05). 2. Catalase activity in RBCs was siginficantly lower in the periodontitis group($117.34{\pm}35.01$) than in the normal group($l52.38{\pm}32.09$)(p<0.05). 3. GSH-PX activity in blood plasma was significantly increased in the posttreatment groupe $1.0376{\pm}0.2820$) compared to the pretreatment group(0.7608 0.1600) (p<0.05), and GSH-PX activity in RBC was significantly increased in the posttreatment group($1.0421{\pm}0.2330$) compared to the pretreatment group($0.7728{\pm}0.1210$)(p<0.05). 4. There was no statistical significance in the difference of catalase activity in RBCs between the pretreatment group($112.04{\pm}43.65$) and posttreatment group($l33.41{\pm}39.16$)(p>0.05).The results, within the limits of the present experiment, suggest that the lowered activity of GSH-PX and catalase in blood plasma and RBCs may be related with periodontopathogenesis.
본 실험은 벌꿀이 항균활성에 미치는 영향을 규명하기 위해 국내산 벌꿀인 밤꿀, 잡화, 아카시아, 재래종 벌꿀과 외국산 벌꿀인 마누카, 클로버, 캐놀라 벌꿀 그리고 인공벌꿀을 각각 12.5%, 25.0%, 50%의 희석액으로 조제하여 catalase무첨가 또는 첨가한 경우에 있어서 벌꿀의 Staphylococcus aureus에 대한 항균활성을 agar well diffusion assay로 비교한 바 다음과 같은 결과를 얻었다. Catalase 무첨가의 경우 12.5%희석한 벌꿀은 마누카꿀 > 밤꿀이, 25.0%로 희석한 벌꿀은 마누카 꿀 > 밤꿀 > 잡화꿀 > 재래종꿀 > 클로버 꿀 > 아카시아꿀이, 50.0%로 희석한 벌꿀은 마누카꿀 > 밤꿀 > 캐롤라꿀 > 재래종꿀 > 잡화꿀>클로버꿀 > 아카시아꿀 순으로 항균활성이 인정되었다(p>0.01). Catalase 무첨가의 경우 12.5%, 25.0%, 50.0%로 희석한 벌꿀의 생육억제환은 각각 5.85∼6.60mm, 4.26∼8.27 mm, 5.24∼11.49mm 범위였다. Catalase 첨가의 경우 12.5%로 희석한 벌꿀은 마누카꿀에서 만 항균활성을 나타냈다. 25.0%로 희석한 벌꿀은 마누카꿀이 밤꿀보다 항균활성이 더 높게 나타냈다.(p > 0.01). 50.0%로 희석한 벌꿀은 마누카꿀 > 밤꿀>클로버꿀)캐롤라꿀>재래종꿀 순으로 항균활성이 높았으며 마누카꿀, 밤꿀, 클로버꿀, 캐롤라꿀, 재래종꿀 사이에서 유의성이 인정되었다(p > 0.01). Catalase 첨가의 경우 12. 5%, 25.0%, 50.0%로 희석한 벌꿀의 생육억제환은 각각 5.89mm, 5.01∼6.84mm, 3.10 ∼8.28mm범위였다.
본 연구에서는 담배식물의 생장과 glutathione reductase와 catalase의 활성에 미치는 p-CA, BA 및 SA의 영향을 연구하였다. 담배식물의 생장에 미치는 p-CA, BA 및 SA의 영향을 알아보기 위하여, $10^{-5}$ mM p-CA, BA 및 SA가 각각 함유된 MS배지에서 배양한 후 20일부터 50일까지 10일 간격으로 담배의 생장을 비교하였다. 3가지 물질 모두에 의해 50일에서 생장이 가장 잘 되였다. Glutathione reductase의 활성에 미치는 효과를 측정한 결과, BA의 활성은 대조구보다 감소하였으며, p-CA 역시 활성이 감소하였고, SA의 활성은 증가하였다. 또한 모두 40일에서 가장 활성이 높았다. Catalase의 활성에 미치는 효과를 측정한 결과, SA의 활성은 증가하였으며, BA와 p-CA의 활성은 감소하였다. 또한 모든 실험구에서 40일에 활성이 가장 양호하였다. 결론적으로 p-CA와 BA는 담배식물의 생장을 촉진시켰으며, 50일에서 생장이 가장 양호하였으나 항산화 효소의 활성을 억제하였다. SA는 담배식물의 생장을 억제시켰으나 항산화 효소의 활성은 촉진시켰으며, 40일에서 가장 활성이 양호하였다.
Oxygen radical은 생물체의 산소대사 과정의 부산물로 생성되어 세포내 여러 성분을 불환성화시키거나 미생물에 대한 방어기전으로 작용한다. 기생충에 존재하는 antioxidant enzyme은 숙주의 방어기전에서 유리하는 oxygen radical의 독성을 제거하므로 이 효소의 활성도는 기생충의 생존에 영향을 미친다고 생각하고 있다. 폐흡충은 피낭유충이 종숙주에 침입한 다음 숙주내 조직이행 시기를 거쳐서 폐에 도달하여 성충이 되므로 이 과정에서 각 시기별 산소독성과 이에 대항하는 효소의 환성이 다를 것으로 추정하였다. 폐흡충의 피낭유충과 감염후 4주, 8주, 12주에 얻은 충체의 추출물을 조효소(조효소)로 하여 SOD, catalase, peroxidase, glutathione peroxidase의 활성도를 측정하였다. 각 효소의 비환성도(specific activity) 중 catalase는 피낭유충에서 최고치였으며, SOD 와 peroxidase는 4주 충체에서 가장 높았고, glutathione peroxidase는 8주 충체에서 높았다. 이들 4가지 antioxidant효소의 비환성도는 감염 12주인 성충에서 모두 낮게 측정되어 조직 이행시기의 충체에서 더 높은 효 소 활성도를 지니고 있음을 알 수 있었다.
녹차카테킨(GTC)의 항산화 작용을 알아보고자 in vitro와 in vivo에서 지질과산화와 superoxide dismutase(SOD), catalase 활성에 미치는 영향에 대한 실험을 행하였다. In vitro 시험계에서의 항산화활성 실험결과, GTC는 peroxide value와 과산화지질 생성을 유의성 있게 억제시켰고, SOD 활성이 매우 높았다. 또한 GTC를 rat에 경구투여 한 후 항산화활성실험 결과, GTC는 $CCl_4$로 유도된 rat의 간 microsome의 지질과산화를 유의성 있게 억제시켰으며, SOD와 catalase 활성을 유의성 있게 증가시켰다. 따라서 GTC는 암과 노화의 예방과 관련이 되는 항산화 활성이 있는 것을 알 수 있었다.
