Effects of $\beta$Carotene on the Browning of Phenolics Extracted from Potato and Polyphenol Oxidase Characteristics (감자에서 추출한 phenolics의 갈변화에 미치는 $\beta$carotene의 영향과 polyphenol oxidase 특성)

  • The effects of synthetic and carrot carotene on the browning of polyphenolics extracted from potato for eliminating the effects of other components in potato which inhibit the browning of potato juice and the potato polyphenol oxidase (PPO) activity were investigated. Total phenolics content in potato sample was 1.854mg/g D.W. and PPO activity was 100 unit. Delta 'L'value of polyphenolics extracted from potato decreased makedly, but that of potato with carrot decreased little by little. Potato with P-carotene showed a little decrease after 50 min. At the same time, polyphenolics extracted from potato were mixed with carotene extracted from carrot or with syntetic $\beta$-carotene. As the results, the delta 'L'value of the former increased but decreased similarly to the latter after 1 hour. The effects of enzyme and substrate concentration on the browning of PPO extracted from potato were investigated. Optimum enzyme concentration was 10% and optimum substrate concentration was 13.3% $\beta$-carotene concentration did not appear to influence significantly on PPO activity.

In vitro Fermentation Characteristics of Juice Pomaces Using Equine Fecal Inoculum (말 분변을 이용한 주스박의 in vitro 발효 특성)

  • This study was conducted to evaluate the changes of pH, dry matter digestibility (DMD), $NH_3-N$ concentrate, gas production and volatile fatty acid (VFA) through in vitro fermentation by adding horse feces to various juice pomaces fermented with Bacillus, yeast and lactic acid bacteria. The pH range of fermented fluid with juice pomaces was 6.4-7.1, indicating that the digestion by microbial fermentation was normal. Juice pomaces adopted will be helpfully used to assist with digestion by microbes in intestines because approximately $10^9CFU/m{\ell}$ microbes were grown after 48 hours in fermented fluid. DMD rate gradually increased from 12 hours. It was 39.19% in pomaces of apple, 38.22% in grape, 37.02% in carrot, 36.2% in citrus and 34.35% in mixture respectively after 48 hours. $NH_3-N$ concentrate was not changed significantly as it was maintained at $1.5mg/100m{\ell}$ level in the entire treatment group from beginning of fermentation until 12 hours, but increased rapidly from 24 hours. Amount of gas produced was lowest in the mixture and increased rapidly after 12 hours. Total VFA increased from 24 hours and was highest at 48 hours. It was suggested that dry matter digestion was processed while fermented juice pomaces kept proper pH during in vitro digestion, and cellulose degrading microorganisms could act actively in the caecum and colon of horses.

Microbial Safety and Quality of Fresh Carrot Juice Prepared with Different Environmentally-Friendly Washing Methods (친환경 세척제의 처리 방법을 달리하여 착즙한 당근 주스의 미생물 안전성 및 품질)

  • The aim of this study was to evaluate the microbial inhibitory activity and physicochemical quality of fresh carrot juice prepared with different environmentally-friendly washing methods during low temperature storage. Individual and combined treatments with sodium bicarbonate (baking soda, $NaHCO_3$) and citric acid were applied to carrots for 10 min. Tap water and 50 ppm of sodium hypochlorite (NaOCl) were used as the control. Combined treatment of 1% $NaHCO_3$ and 1% citric acid significantly reduced total aerobic counts and coliforms. In addition, combined treatment of 1% $NaHCO_3$ and 1% citric acid inhibited microbial growth for 7 days at $4^{\circ}C$ and $10^{\circ}C$ in a shelf-life study. There were no significant differences among the sanitizers in terms of $^{\circ}Brix$, acidity, pH, and color. Changes in physicochemical quality were not significantly different by sanitizer but were affected by storage temperature. These results indicate that washing with combined treatment of 1% $NaHCO_3$ and 1% citric acid is an effective method to inhibit the microbial population and maintain physicochemical quality. Therefore, combined treatment of 1% $NaHCO_3$ and 1% citric acid can be effectively used to sanitize and prepare carrot juice without affecting other properties.

Development of Regional Noodles Using Agricultural and Fishery Products of Cheju Island (제주특산물을 이용한 향토국수의 개발)

  • Acceptable fish noodles of better nutritional and sensory values than conventional noodle made only wheat flour were prepared by mixing wheat flour and ground tile fish (Branchioste gus japonicus). In oder to mask a distinctive fish odor, various herbs and spices, such as ginger (Zingiber officinale), curry (Chalcas koenigii), nutmeg (Myristica Pagrans), garlic (Ailium sativum), black pepper (Piper nigrum), lemon (Cirtus limon) or sodachi (Citrus sudachi) were added to the basic tile fish noodle. Also, for the purpose of enhancing eating qual ify and nutritional value of basic noodle were combined some agricultural or fishery products of Cheju island, citrus fruits; danyooja (Citrus danyooga), hagul (C. natfudaidai), medical plants; angelica utilis (Angelica keiskei), ginseng (Panax ginseng), cactus (Opuntia dillenii), vegetable; carrot (Daucus carota), dropwort (Oenanthe jnvanica), seaweeds; fusiforme (Hizkia fusiforme), gulfweed (Sargaceae hlvelium). The optimal mixing ratio for preparing the basic tile fish noodle amounted to ground tile fish 135 g: tile fish stock 139 $m\ell$: wheat flour 450 g: salts 10 g. The mixture of curry powder (2.5 g) and nutmeg powder (2.5 g) was proved to be the most effective combinations for masking unfavorable fish odor. The optimal amounts of materials to be added to the prepared basic noodle were 25 g citrus zest and 80m1 citrus juice for citrus fruits noodles, and ca. 140 g puree for noodles from medical plants, vegetables and seaweeds, respectively. The preference score obtained from consumer preference test, on a 9-point scales, were in oder of i) danyooja > carrot, angelica, ginseng > fusiforme, ii) hagul > dropwort, cactus > gulfweed. The shelf-life of tile fish noodle based on bacterial counting was estimated to be 7-days at 5$^{\circ}C$.

Effects of Various Polysaccharides on the Physicochemical Properties of the Dextran Culture Containing Carrot Juice Residue Obtained from Submerged Culture Using Leuconostoc citreum S5 (다당류 첨가에 따른 당근박 함유 Leuconostoc citreum S5 발효물의 물성변화)

  • The physicochemical properties of dextran culture fermented by Leuconostoc citreum S5 were evaluated by the addition of various polysaccharides and heat-treatment. The consistency of dextran culture increased with the addition of carrot juice residue (CJR) in submerged culture, resulting in the highest consistency of 150 $Pa{\cdot}s^n$ and viable cell counts of $2.36{\times}10^9$ CFU/mL at 20% level of CJR. The dextran culture showed the pseudoplastic behavior and its consistency was greatly increased with the addition of various polysaccharides at 2% level. Addition of glucomannan indicated the highest consistency of 1275 $Pa{\cdot}s^n$ and their heat-treatment resulted in the increase of consistency except for glucomannan. After heat-treatment, the fermented dextran culture containing CJR fortified with gellan gum and carrageenan showed great change in rheology, indicating the highest consistency and hardness value resulted in the great increase of elastic and viscous moduli. The dynamic viscoelastic properties of dextran culture were greatly increased by the heat-treatment after fortification of various polysaccharides. Thus, the consistency and viable cell counts of dextran culture were increased with the addition of CJR. The rheological properties of dextran culture were manipulated by the fortification of various polysaccharides and heat-treatment.

A Survey on the Sensory Preference for Making Summer Kimchi by Nationwide Region (여름철 배추김치 담금시 지역별 관능적 선호도 조사)

  • The purpose of this study was to investigate the preferred methods for making summer kimchi as a basic research of making commercial kimchi. Questionnaire were collected from 590 housewives nationwide in Korea, and the data were analyzed by SPSS program. The results were as follows: (1) The average salting time of Chinese cabbage was 3~5 hrs when the combined method of dry and brine salting was used, regardless of the region. (2) Seven jeotkals (salt-fermented fishes) including anchovy, anchovy juice, shrimp, northern sand lance juice, hair-tail viscera, flatfish and yellow corvenia were mainly used in kimchi. Among them, anchovy and anchovy juice jeotkals were preferred to all others in Southern area (Busan, Gwangju, Gyeongnam, Gyeongbuk, Jeonnam), while shrimp jeotkal in Seoul, Gwangju, Jeonbuk, Chungnam and Jeju, and northern sand lance juice jeotkal in Daegu, Daejeon and Gyeonggi, respectively. In most regions, however, blending type of 2 jeotkals was used in kimchi. (3) Eleven ingredients such as red pepper, garlic, ginger, green onion, radish, leek, onion, carrot, sugar, sesame and MSG were used as basic components for making kimchi. In particular, MSG was used as a basic ingredient regardless of region and age. However, a standard taste for making kimchi was depended on housewife in this study.

Fermentation Characteristics of Juice Pomace Feed by Horse Feces Microbes (말분변 미생물을 이용한 주스박 사료의 발효 특성)

  • In vitro fermentation was conducted to figure out alternative fiber sources for horse feed. For the development of value-added products as a horse feed resource, the pomaces from apple, carrot, grape, and citrus were fermented under solid-state conditions in the presence of 60% soybean meal with 40% of each fruit pomace at 60% of moisture content. Lactobacillus plantarum SK3873, Lactobacillus plantarum SK3893, Weissella cibaria SK3880, and Bacillus subtilis SK3889 were isolated from the fermented fruit pomace by inoculation of horse feces. For the growth of Bacillus subtilis, Saccharomyces cerevisiae, and Lactobacillus plantarum, they were inoculated in 3-step order at 0, 12, and 24 h, respectively. The fruit pomace was fermented for 48 h at $35^{\circ}C$. The pH of the apple, carrot, grape, citrus and all mixed pomaces decreased from 5.45~6.25 to 4.40~4.77. Microbial growth was maintained at $10^8{\sim}10^9cfu/g$. After 12 and 24 h incubation, dry matter of carrot pomace were highest at 54.84 and 56.66%, respectively (P<0.05) and that of grape pomace was lower than others during fermentation (P<0.05). Dry matter was generally reduced by about 20%. NDF decreased gradually or maintained after 24 h, indicating the fiber degradation. Ash content tended to decrease during fermentation. After 48 hours fermentation, Bacillus, yeast and Lactobacillus showed an excellent growth by using juice by-products. These results suggest that fermented juice pomace has a potential as horse feedstuff with probiotics to maintain beneficial microflora in horse gut.

Monitoring and Risk Assessment of Pesticide Residues in Agricultural Products for Raw Juice in Gyeonggi-Do, Korea (경기도내 유통 생즙 원료 농산물의 잔류농약 실태조사 및 위해성 평가)

  • This study was conducted to monitor the current status of pesticide residues in a total of 297 agricultural products for raw juice from January 2017 to September 2017. Pesticide residues in samples were analyzed by multiresidue method for the determination of multiclass pesticide with an aim to detect 220 pesticides using GC/ECD, GC/NPD, TOF/MS, LC/PDA, LC/FLD, and LC/MS-MS. The analysis revealed the presence of nine pesticides in 14 samples. The detected pesticides were chlorpyrifos, cyflufenamid, chlorothalonil, pendimethalin, azoxystrobin, pyridalyl, imazalil, fludioxonil and procymidone. The kinds of detected crops were mandarin, carrot, apple, celery, chicory, orange and grape. Risk assessment evaluated human health exposure with the ratio of EDI (Estimated daily intake) to ADI (Acceptable daily intake) of pesticides detected and the range of %ADI (the ratio of EDI to ADI) values was 0.0011~6.7087% and safety level.

Estimated Carotenoids Intake in Korean Adults Using Food-frequency Questionnaire: Association with Smoking, Drinking and Other Life-style Factors

  • The aim of this study was to determine the association between, smoking, exercise, sex, and dietary carotenoids ($\alpha$-carotene, $\beta$-carotene, $\beta$-cryptoxanthin, lutein + zeaxanthin, lycopne) intake in Korean middle-aged adults. Food-frequency questionnaire were analyzed from 304 healthy adults (115 men, 189 women) aged 20 - 59. The self-administered questionnaire contained subject s habitual diet and alcohol intake were the previous 3 months. Data on frequency of 102 foods, including vegetables, fruits, beverages and legumes were analyzed. Total dietary carotenoids intake were 27.13 $\pm$ 3.09 mg/d for men and 26.71$\pm$ 2.82 mg/d for women. It was found that smoking had no significant contribution to the dietary intake of carotenoids. Among other lifestyle factors that had significant correlation was the amount of exercise time. The increases in exercise time was associated with increase in carotenoids intake (r= 0.121, p : 0.04). The major contributors of $\alpha$-carotene and $\beta$-carotene were carrots consumed as single-food item or carrot juice. Lutein and Zeaxanthin intake mainly came from spinach and most lycopene intake was derived from tomato products not fresh tomatoes. Persimmon was the major contributor of $\beta$-cryptoxanthin. These findings provide valuable information on understanding the unique pattern of dietary intake of Korean, which might help identify the risks for developing various diseases.

Development of Enzyme-Linked Immunosorbent Assay for Rapid and Sensitive Analysis of Biotin (Biotin의 분석을 위한 효소면역측정법(ELISA)의 개발)

  • In order to develop more rapid and reproducible analysis of biotin known as vitamin H, attempts were made to establish the condition for enzyme linked immunosorbent assay(ELISA) compared with traditional microbiological assay(MBA). Antibiotin and antiserum were obtained from the immunized rabbits injected with emulsion of biotin KLH conjugate and Freund's adjuvant. The antiserum showed cross reactivity on biocytin, a derivative of biotin, which is converted to biotin in intestine, at the rate of 177%(median inhibitory concentration(IC50)=12.58ppb), but not on other derivatives such as desthiobiotin, diaminobiotin and 2 imino biotin. Specific antibody for biotin was purified from the antiserum through protein A column and desalting column. The conditions of competitive direct ELISA (cdELISA) were established. Detection range of biotin concentration by cdELISA was 0.01∼300ng/ ml(ppb). In the spike test with milk, fruit flake and pine carrot juice, the correlation coefficient between two methods of MBA and ELISA was reliably consistent at the value of r=0.992. But detection of biotin by microbiological assay(MBA) was rather restricted in range and nonspecific. Detection range of biotin by MBA was 0.1∼0.5ng/ml(ppb). It showed cross reactivities on biocytin and desthiobiotin at the rate of 80.1% and 66.7%, respectively. In conclusion, ELISA revealed a significant improvement compared with MBA for the biotin detection in terms of sensitivity, detection range and cross reactivity. In addition, a variety of samples could be analyzed rapidly and conveniently at one time by using ELISA. These results strongly suggest that the ELISA is very promising for the practical application to detect biotin contents in a wide range of food stuffs.
