Two Ethylene Signaling Pathways in Senescing Carnation Petals: Exogenous Ethylene-induced Expression of Genes for 1-Aminocyclopropane-1-Carboxylate (ACC) Synthase and ACC Oxidase is Different from That of the Gene for Cysteine Proteinase

  • Carnation petals exhibit autocatalytic ethylene production and wilting during senescence. The autocatalytic ethylene production is induced by the expression of 1-aminocyclopropane-1-carboxylate (ACC) synthase and ACC oxidase genes, whereas the wilting of petals is related to expression of the cysteine proteinase (CP) gene. Until recently, it has been believed that these two phenomena, autocatalytic ethylene production and wilting, are regulated in concert in senescing carnation petals, since the two phenomena occurred closely in parallel. Our studies with petals of a transgenic carnation harboring a sense ACC oxidase transgene and petals of carnation flowers treated with 1,1-dimethyl-4-(phenylsulfonyl) semicarbazide showed that the expression of ACC synthase and ACC oxidase genes and that of CP are regulated differently in carnation psanetals. Interestingly, in the petals of transgenic carnation, the transcript for CP was accumulated but the transcripts for ACC synthase and ACC oxidase were not accumulated in response to exogenous ethylene. Based on these results, we hypothesized that two ethylene signaling pathways, one leading to the expression of ACC synthase and ACC oxidase genes and the other leading to the expression of CP gene, are functioning in senescing carnation petals.

Effect of Planting Date and Substrate on the Growth and Flowering of Hydroponically-grown Carnation (정식시기와 배지의 종류가 양액재배 카네이션의 생장과 개화에 미치는 영향)

  • This study was conducted to investigate growth and flowering of hydroponically-grown carnation as affected by substrate and planting date, Three substrates, coir, perlite, and coir+perlite(1:1. v/v), and two planting dates. May 1 and September 1 were used. Plant height and stem diameter at harvesting time of cut flowers were greater for the September 1 planting than for the May 1 planting. The plants planted on May 1 produced flowers with weak stems and short stem lengths. In addition, flower weight and blossom width were gloater for the September 1 planting than for the Mar 1 planting. The planting date had no significant effect on the number of petals, The carnation planted on May 1 flowered 50 days earlier compared to those Planted on September 1. Plant height and number of petals were the greatest in the plot of coir substrate. The results indicated that for commercial production of cut carnations in a hydroponic system, planting on September 1 is better than May 1. In addition. the results confirm that coir is the superior substrate for the production of cut carnations in a hydroponic system compared to either Perlite or coir+perlite mixture.

Effect of the Treatment Methods of Ethanol and Glycerine on Preserved Flower Quality of Carnation 'Desio' (에탄올 및 글리세린 처리방법이 카네이션 '데지오' 보존화의 품질에 미치는 영향)

  • This study was carried out to examine the optimal treatment conditions of ethanol and glycerine for processing technology development of preserved flowers in carnation (Dianthus caryophyllus) 'Desio' commonly used for flower design. For this purpose, effects of dipping duration of ethanol solution and treatment duration and concentration of glycerine on preserved flower quality were evaluated. Ethanol treatment resulted in perfect dehydration and decoloration of petals and it was proper at 24~48 hours under high brightness and low chroma. Appropriate concentration and time of glycerine treatment was 30% at 36 hours because it resulted in Munsell value of 4.0R in Hue, 6.49 in Value, and 19.8 in Chroma (4.0R 6.49/14) representing the most approximate value to that of fresh petals. Decreasing rate in weight after desiccation tended to reduce by longer time of immersing and higher concentration. Weight after 12 hours of immersing reduced up to 86~90% according to treatment time in non-treatment group of glycerin, meanwhile, it reduced up to 51~69% under higher concentration of 40%. However, weight after 48 hours of immersing reduced up to 90% regardless of desiccation time in non-treatment group of glycerine, to the contrary, decreasing rate reduced by 46~54% through glycerine treatment of 40%. Time for desiccation required 24 hours in glycerin concentration of 10~20% except 6 hours of immersing time, however, higher concentration resulted in increased time up to 48 hours.

Characterization of flavonoids specific gene expression in the petals of Dianthus caryophyllus (carnation) (카네이션 (Dianthus caryophillus)의 색소 발현체계 분석)

  • This study aimed to develop carnation cultivars with new coloring system. We used four genes of Petunia hybrida - chalcone synthase (CHS), flavanone 3-hydroxylase (FHT), dihydroflavonol 4-reductase (DFR), and anthocyanidin synthase (ANS) - as probes, in order to isolate four genes from carnations (Dianthus Caryophyllus). The isolated genes were used as probes in order to select mutants out of collected carnations, using Northern blot analysis. The Northern blot analysis revealed 10 DFR mutants - Gumbyul, Eunbyul, Ballatyne, Crystal, Eugenia, Koreno, Imp. White Sim, West Crystal, White Alpine, and White Charotte. Six among the selected 10 cultivarswere excluded from the target cultivars, because Eugenia, Imp. White Sim, and White Alpine were proved to be double mutants of DFR and ANS, Koreno was considered to be a double mutant of DFR and CHS, and Gumbyul and Ballatyne were proved to be double mutants of DFR and CHI (Chalcone isomerase). Consequently, we selected five DFR mutants, including Virginie, which was already selected as a DFR mutant. Finally, we measured DFR activities in order to confirm the selection, and the results showed that all of the five cultivars - Eunbyul, Crystal, West Crystal, White Charotte, and Virginie - had got no DFR activity.

Breeding of Vivid Pink Standard Carnation 'Pink Beryl' with Round Petals for Cut Flower (둥근 꽃잎에 화색이 선명한 분홍색 스탠다드 카네이션 'Pink Beryl' 육성)

  • A new standard carnation Dianthus caryophyllus 'Pink Beryl' was bred from a cross of red standard cultivar 'Francesco' and purple standard cultivar 'Saturn' in 1996 at the National Institute of Horticultural & Herbal Science. After investigation of the characteristics for 4 years from 1997 to 2000 and registered to the official breeding variety of Rural Development Administration in 2000. 'Pink Beryl' is pink standard cultivar with round edge of petal for cut flower. It has medium resistance to Fusarium wilt. To avoid calyx splitting caused by high temperature, this cultivar is recommended to grow over $8^{\circ}C$ at night and under $25^{\circ}C$ at days during flower bud differentiation period.
