  • Changes in lipid components of Gul-bi, salted-dried yellow corvenia, during processing and storage were studied. Raw fishes contained $6.7\%$ total lipid (TL) which consisted of $83.3\%$ neutral lipid (NL), $6.5\%$ glycolipid (GL) and $10.2\%$ phospholipid (PL), and salted-dried sample at 30 days storage contained $11.1\%$ TL which consisted of $86.1\% NL, $7.1\%$ GL and $6.8\%$ PL. The NL was mainly consisted of trigly ceride (TG), free sterol (FS), free fatty acid (FFA), and the main lipids in PL were phosphatidyl choline (PC), phosphatidly ethanolamine (PE) and sphingomyelin (SPM). The contents of TG, PC, PE decreased, while that of FFA, SPM increased during processing and storage. Total fatty acid contents of raw fish, salted sample, salted-dried sample (product) and product at 30/days of storage were 847.9, 764.0, 633.5 and 588.mg/g-lipid, respectively, taking $C_{23:0}$ as the internal standard material. Most fatty acid con tents of TL revealed a tendency to decrease during processing and storage, highly unsaturated fatty acid such as $C_{22:6},\;C_{20:5}$, decreased largely. The major fatty acids in TL, NL, GL and PL were generally $C_{16:0},\;C_{16:1},\;C_{18:1},\;C_{20:5},\;C_{22:6}. Percentage of $C_{16:1},\;C_{18:1}$ in NL and GL and GL are higher than those in PL, while $C_{22:6},\;C_{20:5},\;C_{20:4}$. Peroxide value in salted-dried sample is 121.6 meq/kg and decreased rapidly with storage period, carbonyl value and brown pigment formation showed tendency of increase during processing and storage.

  • We have compared(using the same series of experimental tissue samples) the levels of proteolytic enzyme activities and free radical-induced protein damage in synovial fluid from RA and CPH cases. Many protease types showed significantly increased (typically by a factor of approximately 2-3-fold) activity in RA, compared to normal rats. However, CPH significantly reduced the cytoplasmic enzyme activities of arginyl aminopeptidase, leucyl aminopeptidase, pyroglutamyl aminopeptidase, tripeptidyl aminopeptidase, and proline endopeptidase to almost about 1/10 each. For the Iysosomal proteases, synovial fluid samples from RA rats, CPH significantly reduced the enzyme activities of cathepsin B, dipeptidyl aminopeptidase I and dipeptidyl aminopeptidase II. In extracellular matrix degrading(collagenase, tissue elastase) and leukocyte as sociated proteases (leukocyte elastase, cathepsin G), CPH decreased these enzyme activities of collagenase, tissue elastase and leukocyte associated elastase in RA. In cytoplasmic and lysosomal protease activities in plasma from RA. CPH and normal plasma samples were not significantly different, suggesting that altered activity of plasma proteases (particularly those enzymes putatively involved in the immune response) is not a contributory factor in the pathogenesis of RA. In addition, the level of free radical induced damage to synovial fluid proteins was approximately twice that in RA, compared with CPH. CPH significantly decreased the level of ROS induced oxidative damage to synovial fluid proteins (quantified as protein carbonyl derivative). Therefore we conclude that both proteolytic enzymes and free radicals are likely to be of equal potential importance as damaging agents in the pathogenesis of inflammatory joint disease, and that the design of novel therapeutic strategies for patients with the latter disorder should include both protease inhibitory and free radical scavenging elements. In addition, the protease inhibitory element should be designed to inhibit the action of a broad range of protease mechanistic types (i.e. cysteine-, metallo- and serine- proteinases and peptidases). However, increased protein damage induced by ROS could not be rationalised in terms of compromised antioxidant total capacity, since the latter was not significantly altered in RA synovial fluid or plasma compared with CPH.

  • This study was designed to investigate the effects of butanol (BuOH) fraction of pine (Pinus densiflora Sieb et Zucc) needle extract on membrane fluidity (MF), basal and induced oxygen radicals (BOR and IOR), lipid peroxide (LPO) and oxidized protein (OP) as an oxidative stress, and lipofuscin (LF) in liver membranes of Sprague-Dawley male rats. The rats were fed basic diets (control group) and experimental diets (BuOH-25, BuOH-50 and BuOH-100) prepared with 25, 50 and 100 mg added to basic diet for 45 days. MFs were significantly increased (about 16∼22%) in mitochondria of BuOH-50 and BuOH-100 groups compared with control group (p<0.01∼0.001) BOR and IOR formations in mitochondria were significantly decreased (11∼17% and 11∼28%, respectively) in these three BuOH groups (p<0.05∼0.001), while BOR and IOR formations in microsomes were significantly decreased (11∼24%) in BuOH-50 and BuOH-100 groups, and (15∼24%) in these three BuOH groups compared with control group (p<0.05∼0.001; p<0.01-0.001). LPO levels were significantly decreased (9% and 9∼13%, respectively) in mitochondria of BuOH-100 and microsomes of BuOH-50 and BuOH-100 groups (p<0.05∼0.01), whereas OP levels were significantly decreased (10∼12%) in mitochondria of BuOH-50 and BuOH-100 groups compared with control group (p<0.05). LF formations were significantly decreased (8∼9%) in BuOH-50 and BuOH-100 groups (p<0.05). These results suggest that butanol fraction of pine needle extract may playa effective role in an attenuating an oxidative stress and increasing a membrane fluidity.

  • Acetolactate synthase activity inhibition and herbicidal activities were investigated with 2 sulfonylureas [chlorsulfuron{2-chloro-N-{{(4-methoxy-6-methyl-1,3,5-triazin-2-yl) amino} carboxyl} benzenesulfonamide}, metsulfuron-methyl{methyl-2{{{{(4-methoxy-6-methyl-1,3,5-triazin-2-yl)amino}carbonyl}amino}sulfonyl}benzoic acid}, and 2 imidazoli-nones [imazethapyr{2-{4,5-dihydro-4-methyl-4-(1-methyl)-5-oxo-1H-imidazol-2-yl}-5-ethyl-3-pyridinecarboxylicacid}, imazaquin{2-{4,5-dihydro-4-methyl-4-(1-methyl)-5-oxo-1H-imidazol-2-yl}-3-quinoline carboxylic acid} herbicides. A broad weeding spectrum was observed with the treated herbicides at low application rates. Both corn(Zea mays L.) and sorghum(Sorghum bicolor Moench) were very sensitive to the two herbicide groups. Although legumes, such as soybean(Glycine max Merr.), clover(Trifolium repense L.), and indian jointvetch(Aeschnomene indica L.) were sensitive to the sulfonylureas, they were tolerant to the imidazolinones. On the contrary, wheat(Triticum aestivum L.) and barley(Hoderum sativum Jess.) showed the reverse responses of the legumes to the two herbicide groups. Quackgrass(Agropyron repens(L.) P. Beauv.). however, was commonly tolerant to the two herbicide groups. Degrees of crop injury and acetolactate synthase inhibition also varied with the crops examined. The 50% inhibition concentrations of sulfonylureas on acetolactate synthase in vitro activity($IC_{50}$) from corn, wheat, and soybean did not relate to the greenhouse herbicidal activities ($GI_{50}$). With chlorsulfuron, for example, wheat had more than 100 times higher $GI_{50}$ than corn and soybean, but the $IC_{50}$ was 4 to 10 times lower. Similar observation was made with metsulfuron-methyl. However, closer relationships between $IC_{50}$ and $GI_{50}$ were found with the imidazolinones. When imazethapyr was applied, the order of $GI_{50}$ values against com, wheat, and soybean was the same as that of $IC_{50}$.

  • Purpose: The estrogen receptor (ER), which is over-expressed in ER-positive breast tumors, has been imaged by positron emission tomography (PET) using $[^{18}F]$ labeled estrogen ligands, especially $[^{18}F]FES$. However, $[^{18}F]$ has relatively short-lived half-life ($t_{1/2}$ =1.8 h) and the labeling yield of radio-fluorination is usually low compared with $^{64}Cu\;(t_{1/2}=12.7\;h)$. 1,4,7,10-tetraazacyclododecane (cyclen) is used to form stable metal complexes with copper, indium, gallium, and gadolinium. With these in mind, we prepared cyclen-based Cu complexes which mimic estradiol in aspect of two hydroxyl groups. Materials and Methods: 1.7-Protected cyclen, 1.7-bis (benzyloxycarbonyl)-cyclen was synthesized according to the reported procedure. After introducing two 4-benzyloxybenzyl groups at 4,10-positions, the benzyloxycarbonyl and benzyl groups were removed at the same time by hydrogenation on Pd/C to give 1,7-bis(4-hydroxybenzyl)-1,4,7,10-tetraazacyclododecane (1). Results: The prepared ligand 1 was fully characterized by $^1H,\;^{13}C$ NMR, and mass spectrometer. The synthesized ligand was reacted with copper chloride and copper perchlorate to give copper complexes $[Cu(1)]^{2+}2(CIO_4^-)\;and\;[Cu(1)Cl]^+Cl^-$ which were confirmed by high-resolution mass (FAB). Conclusion: We successfully synthesized a cyclen derivative of which two phenol groups are located on trans position of N-atoms. And, two Cu(ll) complexes of +2 and +1 overall charge, were prepared as a potential PET tracers for ER imaging.

  • Oxidized polyethylene wax (OPEW) is, one of the food additives, used as a coating agent in citrus fruits and nuts. OPEW is authorized to quantum satis in EU, USA, and is acceptable less than 250 mg/kg in Australia and New Zealand. But OPEW is unauthorized as a food additive in Korea. This study was to establish the analytical method of OPEW and demonstrate the effective application of various food samples. We first conducted to compare the various analytical method including acid value (AV), high temperature gel permeation chromatography (HT-GPC), matrix-assisted laser desorption ionization time-of-flight mass spectrometry (MALDI-TOF/MS), gas chromatography flame ionization detector (GC-FID) and fourier transform infrared spectroscopy (FT-IR). This result indicated that FT-IR spectrum of OPEW treated food sample displayed absorption bands for carbonyl group (C=O, $1714cm^{-1}$), ester group (C-O, $1463cm^{-1}$), aliphatic group (C-H, $2916cm^{-1}$). Furthermore, IR spectrum of OPEW treated food sample showed similar tendency with IR spectrum of OPEW standard. Therefore, it is confirmed that analytical method using FT-IR can be detected on analysis of OPEW in food. As a result of monitoring of 111 samples using established analytical method, OPEW was not detected in the food samples.

  • The current study examined the effects of radish loaves powder on hepatic antioxidative system in rats fed high-cholesterol diet. Sprague-Dawley male rats weighing 100$\pm$10 g were randomly assigned to normal group (N group), normal diet with 5$\%$ radish leaves powder supplemented group (NR group) and high-cholesterol groups, which were sub-divided into radish leaves powder free diet group (HC group) and 2.5$\%$ (HRL group), 5$\%$ (HRM group), 10$\%$ (HRH group) radish leaves powder supplemented groups. Hepatic super oxide dimutase activity was no significant differences. Hepatic glutathione peroxidase activity was sig-nificantly increased in 5$\%$, 10$\%$ radish leaves powder supplemented groups. Hepatic hydrogen peroxide contents in cytosol were no significantly differences Hepatic hydrogen peroxide contents in mitochondria were sig-nificantly reduced in radish leaves powder supplemented groups. Hepatic superoxide radical contents in mi-crosome were significantly reduced in radish leaves powder supplemented groups. Hepatic superoxide radical contents in mitochondria were significantly reduced in 5$\%$, 10$\%$ radish leaves powder supplemented groups. Hepatic TBARS values were significantly reduced in 5$\%$, 10$\%$ radish leaves powder supplemented groups. Hepatic lipofuscin contents were no significant difference in high-cholesterol groups. Hepatic carbonyl values were significantly reduced in 5$\%$, 10$\%$ radish leaves powder supplemented groups among high-cholesterol groups. The results indicate that radish leaves may reduce oxidative damage by activating antioxidative de-fense system of liver in rats fed high-cholesterol diets.

The structure was solved by direct methods and refined by cascade diagonal least-squares refinement. All the C-H bond lengths(= 0.96 ${\AA}$), the methyl groups and the methylene groups are fixed and refined as the rigid groups with ideal geometry. The macrocycle exists in the 1,3 alternate conformation (by Conforth) making the angles of 110.7, 684, 113.7 and 68.8$^{\circ}$ between the benzene rings and the methylenic mean plane, and four each acetoxy groups are twisted away from their own benzene rings with the angles of 68.2, 97.6, 78.9 and 71.3$^{\circ}$, respectively. The relative dihedral angles between two opposite side of the benzene rings are 135.6$^{\circ}$ for the rings (1) and (3) and 135.2$^{\circ}$ for (2) and (4). A water molecule which has nearly the same height of the methylenic plane of the macrocycle in the c-axis, is located within the distances of 2.942(5) ${\AA}$ from the O(8) atom of the carbonyl group and 2.901 ${\AA}$ from, another O(2)(1/2-x, -1/2+y, z). The shortest contact between the molecule is 3.193 ${\AA}$ from the O(4) to the C(3)(1/2+x, 1/2-y,-z).

  • The oxidative stability of lipids from eel (Anguilla japonica) fed diets containing different concentrations of conjugated linoleic acid (CLA) was studied. Eels, 3 weeks of age, with an average weight of 160 g, were randomly divided into 5 groups (5 fishes/group) by body weight, and assigned to one of the five CLA-supplemented diets at the following concentrations: 0, 0.5, 1.0, 2.5, and $5.0\%$ CLA. After 8 weeks of feeding, eels were sacrificed and the total lipid contents were extracted. The lipids from each treatment groups were stored at $37^{\circ}C$ for 5 weeks. Changes in the fatty acid profile, lipid class, weight gained, peroxide value (POV) and carbonyl value (COV) of the lipid from each treatment groups were analyzed weekly. The composition of CLA in the lipids of eels fed with 0.5, 1.0, 2.5, and $5.0\%$ CLA-supplemented diets were 0.5, 1.7, 3.3, and $6.2\%$, respectively After 4 weeks of storage, the proportion of polyunsaturated fatty acids (PUFAs) in the lipid of eels fed diets containing 1.0 and $2.5\%$ CLA were 15.3 and $14.8\%$, respectively. Whereas, lipid extracted from eels fed with 0.5 and $5.0\%$ CLA-supplemented diets contain 11.8 and $7.4\%$ PUFAs, respectively. Lipid from the control sample contained $9.0\%$ PUFAs. POV and COV were found to be the lowest in the lipids samples from 1.0 and $2.5\%$ CLA diets. These results indicate that lipids from diets containing 1.0 or $2.5\%$ CLA were more stable against oxidative rancidity relative to other concentrations, suggesting that these are the appropriate CLA concentrations for the production of stable eel lipids.
