Effects of Some Chemials on Ethylene Evolution and Abscission of Fruits and Leaves in Oriental Pera (수종(數種)의 약제(藥劑)가 배나무의 낙과(落果) 및 에틸렌 발생(發生)에 미치는 영향(影響))

  • In order to examine the relationship of fruit abscission and ethylene evolution in 'Jojuro' and 'Imamuraaki' pear trees, the thinning chemicals, ${\beta}$-naphthalene acetic acid (NAA), O, O-dimethyl-O-(3-methyl-4 nitrophenyl) posphorothioate (MEP), 1-naphthyl N-methyl carbamate (carbaryl), and 2-chloroethyl phosphonic acid (ethephon) were applied. Concurrently the effect of calcium acetate on the control of fruit abscission were examined when calcium acetate was added to ethephon and carbaryl. 1. All the applied chemicals (MEP, NAA, carbaryl and ethephon) was effective to abscise fruits in 'Jojuro', and MEP and carbaryl in 'Imamuraaki's. 2. The application of ethephon increased the ethylene evolution but other chemicals did not increase it. 3. There were no significant differences in total sugar contents of fruits by MEP, NAA, carbaryl and ethephon treatments. 4. When the calcium acetate was added to the ethephon and carbaryl, the thinning effects were offsetted. 5. The rate of defoliation due to ethephon treatment was controlled by addition of calcium acetate at 0.1~0.25 M. It was clarified that fruit abscission induced by thinning chemicals except ethephon was not directly related to ethylene production and that degree of fruit thinning can be regulated by addition of calcium acetate to thinning chemicals.

Analysis of Carbendazim and Carbaryl by Micro-HPLC (Micro-HPLC에 의한 Carbendazim과 Carbaryl의 분석)

  • A micro-high performance liquid chromatography(Micro-HPLC) techniquie with solid phase extraction was reported which detected carbendazim and carbaryl at picogram levels. They were separated on microbore packed $C_{18}$ column($1.0mm\;I.\;D{\times}150mm$, $d_f=5{\mu}m$) using a 50% methanol mobile phase and detected at UV 220nm(${\alpha}=2.94$, $R_s=4.71$), while they were not resolved on analtical HPLC system(${\alpha}=1.27$, $R_s=0.76$). The detection thresholds of carbendazim and carbaryl were 0.5ng and 0.1ng on Micro-HPLC, therefore Micro-HPLC system was 20~40 told more sensitive than anayltical HPLC system. Sep-Pak $C_{18}$ catridge was found to be efficient in enriching carbendazim and carbaryl from dilute aqueous solution with 97.0% and 97.8% recoveries of them. The Sep-Pak $C_{18}$ catridge followed by the Micro-HPLC had been applied to the quantitative analysis of carbendazim and carbaryl in spiked juices and a commercial drinking water.

`oxicological study of carbaryl in rats

  • The apparent effectiveness of 1-naphthyl-N-methyl carbamate (carbaryl) against a wide variety of insects motivated the study of its mammalian toxicity. In this toxicological study of carbaryl, mature male rats inhaled carbaryl at a mean concentration of 112mg, 168mg and 224 mg/$m^{3}$ for one hour. After inhalation, pentobarbital sleeping time, Nadph-cytochrome c reductase activity, cytochrome p-450 and protein content in liver microsomes, various tissue residues, cholinesterase inhibition in plasma and histopathological findings at autopsy were observed. The pentobarbital sleeping time was prolonged in rats inhaled with carbaryl for one day while the sleeping time was shortened in the 3 days inhaled group. The changes of cytochrome p-450 content and NADPH-cytochrome c reductase activity exhibited biphasic response showing the decrease in the one day inhaled group and the increase in the 3 days inhaled group. The marked depression of plasma ChE activity was observed in rats inhaled with carbaryl at 112 mg/$m^{3}$, however no more progressive effect was observed at the higher concentration of the compound. The main observations in histopathological finding were ciliary detachment, epithelial swelling and subepithelial inflammatory cellular infiltration in trachea due to the irritation.

Simultaneous Determination of Pesticides in Water Using a GC/MS Coupled with Micro Extraction by Packed Sorbent (MEPS-GC/MS를 이용한 농약류 동시 수질분석)

  • This study established an analytical method to simultaneously determine six organophosphorous pesticides [methyldemetone-S, diazinon, fenitrothion, parathion, phentoate, and O-ethyl O-(4-nitrophenyl) phenylphosphonothioate (EPN)] and carbaryl in water using a gas chromatography/mass spectrometry (GC/MS) system coupled with on-line micro extraction by packed sorbent (MEPS) and programmed temperature vaporizer (PTV) injector. Polystyrene divinylbenzene (PDVB) was used as a sorbent of MEPS. The effects of elution solvents, pH, elution volume and draw-eject cycles of samples on sample pretreatment process were investigated. Also, quality assurance and quality control (QA/QC) and the recovery of the pesticides in environmental samples were evaluated. The elution was performed using $30{\mu}L$ of a mixed solvent (acetone : dichloromethane = 80 : 20 (v/v)). Sample pretreatment processes were optimized with seven cycles of draw-eject of sample (1 mL) spiking an internal standard and sulfuric acid. At lower pH, the analytical sensitivity of diazinon decreased, but that of carbaryl increased. The method detection limit and the limit of quantification for this method were 0.02~0.18 and $0.08{\sim}0.59{\mu}g/L$, respectively. The method precision and accuracy were 1.5~11.5% and 83.3~129.8%, respectively, at concentrations of $0.5{\sim}5.0{\mu}g/L$. The recovery rates for all the pesticides except carbaryl in various environmental samples ranged 75.7~129.3%. The recovery rate of carbaryl in effluent sample was over 200% whereas carbaryl in drinking water, groundwater, and river water were in the acceptable range.

Determination of Bioconcentration Factor on Carbamates (Carbamate계 농약의 생체농축계수 측정)

  • It was reported that BCF's (Bioconcentration Factor) on Carbaryl and BPMC in concentration of 1, 2, 5 and 10 ppm, previously. Carassius auratus(goldfish) was chosen as test organism. Carbamates in fish and in test water were extracted with n-hexane and acetonitrile. GC-ECD was used to detecting and quantitating of Carbamates. Also, partition coefficients were determined with Stir flask method. To evaluate environmental toxicological profiles of tested compounds, experimental concentration were 0.05, 0.1 and 0.5 ppm in contrast to previous report. It was considered that higher BCFs of BPMC due to its higher partition coefficient compared to Carbaryl. The obtained results were as follows: 1. It was possible to determine short term BCF of Carbaryl and BPMC through relatively simple procedure in environmental concentrations. 2. BCF$_3$ of Carbaryl in concentration of 0.05, 0.1 and 0.5 ppm were 4.666 $\pm$ 0.002, 3.622 $\pm$ 0.004, 1.200 $\pm$ 0.002 and BCF$_5$ were 3.897 $\pm$ 0.005, 4.219 $\pm$ 0.017 and 1.186 $\pm$ 0.054, respectively. In the case of BPMC in same condition, BCF$_3$ were 4.077 $\pm$ 0.014, 4.900 $\pm$ 0.005, 4.750 $\pm$ 0.009 and BCF$_5$ were 3.465 $\pm$ 0.010, 4.612 $\pm$ 0.011 and 4.075 $\pm$ 0.012, respectively. 3. Carbaryl concentration in fish extract was increased as increasing test concentration, but BCF were decreased as prolonging test period, especially dropped at 0.5 ppm. 4. In the case of BPMC, BCF were decreased as increasing test concentration, but the concentration in fish extract of 3-day test group was slightly higher than that of 5-day test group. 5. Higher BPMC concentration in fish extract was due to its higher partition coefficient to compared with Carbaryl. 6. Determined logP of Carbaryl and BPMC were 2.200 and 3.180. But the calculated BCF using suggested equation was so different that predict BCF. It is suggested that BCF's of Carbamates have to be determined by experiment.

Confirmation of carbamate pesticides by liquid chromatography coupled with electrospray ionization mass spectrometry (LC/MS를 이용한 카바메이트계 잔류농약 확인방법)

  • A method was developed to separate, detect and qualify aldicarb, bendiocarb, carbaryl, carbofuran, ethiofencarb, methomyl, methiocarb, propoxur in meats and fruits. Experimental beef and fork samples were fortified with 0.05mg/kg of carbamate pesticides for analysis. Carbamate-detected pear by HPLC fluorescence detector(HPLC/FLS) are extracted with acetonitril and refined by solid phase extraction(SPE) filled with aminopropyl-bonded silca, In the following step, the injected materials into LC/MS are analyzed to result in the fact that bendiocarb, carbaryl, carbofuran, ethiofencarb, methomyl, methiocarb, propoxur presents several sorts of fraction ions following with; [M+H]$^{+}$, [M+Na]$^{+}$,[M-CONH$CH_3$]$^{+}$, [M-OCONH$CH_3$]$^{+}$. In addition, ethiofencarb presents [M-SCH$_2$$CH_3$]$^{+}$ ion distinctive and aldicarb presents [M+Na]$^{+}$ and [M-OCONH$CH_3$]$^{+}$ ion which is the most decisive fraction ion for pesticides such as bendiocarb, carbaryl, carbofuran, ethiofencarb, methiocarb, methomyl, propoxur excluding [M+H]$^{+}$ ion. However, [M-OCONH$CH_3$]$^{+}$ and [M-OCONH$CH_3$]$^{+}$ fraction ion charactering carbamate pesticides are detected most efficiently with fragment voltage 50ev. As a result, for rluantitative analysis, [M+Na]$^{+}$ ion is the most decisive ion for detection of aldicarb and [M+H]$^{+}$ ion is the most decisive fraction ion for Pesticides such as bendiocarb, carbaryl, carbofuran, ethiofencarb, methiocarb, methomyl, propoxur. Carbaryl-detected pear by HPLC/FLS are analyzed by L/MS and the result shows that [M+H]$^{+}$ and [M-CONH$CH_3$]$^{+}$ ions charactering carbaryl are detected.ering carbaryl are detected.

A Study on the Carbamate Pesticide Residues in Domestic and Imported Crops in Chonnam Province (전라남도에서 유통중인 국내산 및 수입산 곡류 중의 카바메이트제 농약 잔류 실태 연구)

  • 10 carbamate pesticides were surveyed in domestic and imported green peas, beans, red beans from March 1999 through December 1999. Samples were collected from Gwangju, Mokpo, Suncheon and Yosoo grain markets. This study was performed by post-column ο-phthalaldehyde and 2-mercaptoethanol prior to HPCL fluorophore detection. Pesticides were found in 14 of 80 samples(17.5%). The kinds of pesticide detected in green peas were methomyl, MTMC, carbaryl, those in sesames were oxamyl, carbofuran, carbaryl, 1-naphthol, MIPC, those in beans were oxamyl, MTMC, carbaryl and those in red beans were MTMC, carbaryl. The range of residues detected in green peas, sesames, beans and red beans were 0.002∼0.025ppm, 0.001∼0.469ppm, 0.005∼0.356ppm and 0.017∼0.125ppm, respectively. The most frequently detected pesticides were MTMC(6 times) and carbaryl(5 times), while aldicarb, PHC and BPMC were no detected in all samples. Detection frequencies of carbamate pesticides in imported crops were higher than those in domestic ones except red beans. Only one result of oxamyl residues determined in imported bean was higher than the proposed maximum residue limits of Codex, which was 0.356ppm.

  • A sensory element against carbaryl, as a widely used pesticide was prepared based on adsorbed acetylcholine esterase (AChE) from Torpedo california. Octadecyl was substituted on macro-porous silica, confirmed by infra-red (IR) spectroscopy and quantitatively estimated through thermo-gravimetric analysis (TGA). Immobilization of the enzyme was achieved by adsorption on this support. Activity of the immobilization product was measured as a function of the loaded enzyme concentration, and maximum binding capacity of the support was estimated to be 43.18 nmol.mg-1. The immobilized preparations were stable for more than two months at storage conditions and showed consistency in continuous operations. Possible application of the immobilized AChE for quantitative analysis of carbaryl is proposed in this study.

Comparison of 14C-radioactivity in rice-paddy soil exposed to atmospheric and elevated CO2 conditions after 14C-carbaryl treatment

  • This study was performed to investigate if elevated $CO_2$ affects the residue pattern of $^{14}C$ in the soil environment after $^{14}C$-carbaryl treatment $^{14}C$-carbaryl was applied on the rice plant-grown greenhouse soil exposed to atmospheric and elevated $CO_2$ conditions. $^{14}C$-radioactivity was measured in the rhizospheric soil and rice straw samples six months after $^{14}C$-carbaryl application. Significantly high radioactivity was observed in the soil exposed to atmospheric $CO_2$ as compared to that in the soil exposed to elevated C(h. Background level of radioactivity was observed in rice plant samples. These observations suggest the possibility that elevated $CO_2$ may affect residual radioactivity of $^{14}C$-carbaryl in the soil rather than that in the plant.

Chemical Control of Popillia quadriguttata(Coleoptera: Scarabaeidae) and Damage of Zoysia matrella and Newly Recorded Host Plants in Golf Courses (녹색콩풍뎅이(Popillia quadriguttata)의 화학적 방제와 골프장의 금잔디(Zoysia matrella) 및 새로운 기주의 피해)

  • Popillia quqdriguttata is a serious insect pest in Korean golf courses. The adult feeds on leaves of latifoliate trees and herbaceous plants while the larva feeds on roots of turfgrass. Thus, control of p. quadriguttata adults was tried with registered insecticides in golf courses in Korea. The insecticides used in the experiments were carbaryl + phosalone WP, chlorpyrifos + diflubenzuron WP, chlorpyrifos-methyl EC, deltamethrin EC, etofenprox WP, etofenprox + diazinon WP, etofenprox + tebufenozide EC, fenitrothion EC, imidacloprid SC, pyraclofos WP, and tralomethrin EC. Mortalities of P. quadriguttata were over 95% in carbaryl + phosalone WP, chlorpyrifos + diflubenzuron WP, etofenprox + diazinon WP, and fenitrothion EC at petri dish and in greenhouse. In the field test, corrected mortality of P. quadriguttata was 95.2% and 80.9% in carbaryl 40% +phosalone 20% WP and fenitrothion 50% EC, respectively. Spreading agent enhanced toxic effect of used insecticides to p. quadriguttata adults except carbaryl + phosalon 20% EC. p. quadriguttata adult was firstly observed to feed on leaves of Canna generalis at the Yongwon golf club in Jinhae, Gyeongnam and Prunus ameriaca var. ansu and p. salicina at the Anyang Honest golf club in Gunpo, Cyeonggi. Therefure, host plants off quadriguftata became 29 species of 26 genera in 19 families. Zoysia matreila at the fairway and roughs of Anyang Benest golf club was damaged by p. quadriguttata larvae in Octomber, 2002 and not recovered in April, 2003. Damaged turfgrass became yellowish and wilted and that retarded recovering in the following spring.
