A Study on the Relationship between Autonomy and Group Cohesiveness Perceived by Nurs es and their Job Satisfaction, Organizational Commitment, Motivation and Intend to stay on jobs (간호사가 지각한 자율성, 그룹 결속력과 직무만족도, 조직몰입, 직무동기, 재직의도와의 관계)

  • The purpose of this study is analysis of the relationship between autonomy and group cohesiveness perceived by nurses and their job satisfaction, organizational commitment, motivation and intend to stay on jobs to develope the nursing staff management. The results are as follows: 1. Characteristics of subjects are not significantly different in perceived autonomy, but group cohesiveness, organizational commitment and intend to stay on jobs are different in nursing unit(p<.05). Educational background is significantly different in job satisfaction: nurses who has diploma show higher scores than who has baccalaureate degree(p<.05). 2. Perceived autonomy is related with job satisfaction, motivation and intend to stay on jobs but is not related with organizational commitment(p<.05). 3. Group cohesiveness perceived by nurses is significantly related with job satisfaction, organizational commitment motivation and intend to stay on jobs and their correlation coefficients are higher than that of perceived autonomy(p<.05). 4. The interaction effect of autonomy and group cohesiveness with all dependent variables is not significant(p>.05). The main effect on all dependent variables is presented as group cohesiveness. 5. The canonical correlation coefficient of independent variables and dependent variables is 0.503(p<0.000). The above results are important in the aspect of the development of strateges for retension of qualified nurses performing quality of nursing care and organizational efficacy. Autonomy and group cohesiveness perceived by nurses are important factors in nursing staff management; the further studys on job redesign and intervention strateges for enhancing group cohesiveness are needed.

  • The purpose of this study is to estimate the stochastic monthly runoff model for the Kunwi south station of Wi-stream basin in Nakdong river system. This model was based on the theory of Box-Jenkins multiplicative ARlMA and the state-space model to simulate changes of monthly runoff. The forecasting monthly runoff from the pair of estimated effective rainfall and observed value of runoff in the uniform interval was given less standard error then the analysis only by runoff, so this study was more rational forecasting by the use of effective rainfall and runoff. This paper analyzed the records of monthly runoff and effective rainfall, and applied the multiplicative ARlMA model and state-space model. For the P value of V AR(P) model to establish state-space theory, it used Ale value by lag time and VARMA model were established that it was findings to the constituent unit of state-space model using canonical correction coefficients. Therefore this paper confirms that state space model is very significant related with optimization factors of VARMA model.

Otolith microchemistry reveals the migration patterns of the flathead grey mullet Mugil cephalus (Pisces: Mugilidae) in Korean waters

  • Background: The flathead grey mullet Mugil cephalus has the widest distribution among mugilid species. Recent studies based on mitochondrial DNA sequences showed that the species comprises at least 14 different groups, three of which occur in the northwest Pacific. We analyzed the otolith microchemistry of M. cephalus at several locations in Korea to improve understanding of migration pattern and population origin. Results: We collected 123 sagittal otoliths from seven locations and determined their concentrations of eight elements (7Li, 24Mg, 55Mn, 57Fe, 60Ni, 63Cu, 88Sr, and 138Ba) using laser ablation inductively coupled plasma mass spectrometry. Mean otolith elemental ratios differed significantly among the locations. The Sr:Ca, Fe:Ca, and Ba:Ca ratios were significantly higher than others, and useful chemical signatures for investigating the habitat use of M. cephalus populations. We identified five diverse and complicated migration patterns using the otolith data that we collected: estuarine resident (type I), freshwater migrant (type II), estuarine migrant (type III), seawater resident (type IV), and seawater migrant (type V). A canonical discriminant analysis plot revealed separation of two groups (type II in the Yellow Sea vs. other types in remaining locations). Two locations on Jeju Island, despite their close proximity, had fish with quite different migration patterns, corroborating previous molecular studies that distinguished two groups of fishes. Conclusion: We successfully showed that the migration patterns of the Korean mullet varied by location. Only fish from the western sector of Jeju had a unique migration pattern, which is likely confined population in this area. Among the eight otolith elements measured, the Sr:Ca ratio was found to be the best indicator of migration pattern and population origin.

Sequence Analysis, Molecular Cloning and Restriction Mapping of Mitochondreal Genome of Domesticated Silkworm, Bombyx mori (누에 미토콘드리아 유전체의 제한효소 지도작성, 클로닝 및 염기서열 분석)

  • The mitochondrial genome of domesticated silkworm (Bombyx mori) was mapped with five restriction endonucleases (BamHI, EcoRI, HindIII, PstI and XbaI), the entire genome was cloned with HindIII and EcoRI. From the end sequencing results of 5$^1$and 3$^1$region for full genome set of eleven mitochondrial clones, the seven mitochondrial genes (NADH dehydrogenase 6, ATPase 6, ATPase 8, tRN $A^{Lys}$, tRN $A^{Asp}$, tRN $A^{Thr}$ and tRN $A^{Phe}$ of mori were identified on the basis of their nucleotide sequence homology. The nucleotide composition of NADH dehydrogenase 6 was heavily biased towards adenine and thymine, which accounted for 87.76%. On basis of the sequence similarity with published tRNA genes from six insect species, the tRN $A^{Lys}$, tRN $A^{Asp}$ and tRN $A^{Thr}$ were showed stable canonical clover-leaf tRNA structures with acceptible anticodons. However, both the DHU and T$\psi$C arms of tRN $A^{Phe}$ could not form any stable stem-loop structure. The two overlapping gene pairs (tRN $A^{Lys}$ -tRN $A^{ASP}$ and ATPase8-ATPase6) were found from our sequencing results. The genes are encoded on the same strad. ATPase8 and ATPase6 overlaps (ATGATAA) which are a single example of overlapping events between abutted protein-coding genes are common, and there is evidence that the two proteins are transcribed from a single bicistronic message by initiation at 5$^1$terminal start site for ATPase8 and at an internal start site for ATPase6. Ultimately, this result will provide assistance in designing oligo-nucleotides for PCR amplification, and sequencing the specific mitochondrial genes for phylogenetics of geographic races, genetically improved silkworm strains and wild silkworm (mandarina) which is estimated as ancestal of domesticated silkworm.sticated silkworm.

Genetic Evaluation of First Lactation Traits in Sahiwal Cattle Using Restricted Maximum Likelihood Technique

  • The data on 283 Sahiwal cows, sired by 16 bulls, maintained at Cattle Breeding Farm of Nagpur Veterinary College and Dairy Farm of Agricultural College, Nagpur, were considered for the estimation of genetic parameters. Variance and covariance estimates of first lactation traits were obtained using restricted maximum likelihood technique (REML). When first lactation milk yield (FLMY), first lactation length (FLL) and average daily yield (ADY) traits were considered for REML analysis, the heritabilities were $0.184{\pm}0.146$, $0.132{\pm}0.131$ and $0.141{\pm}0.133$, respectively. While, genetic and phenotypic correlations between them were medium to high except phenotypic correlations between FLL and ADY (-0.025). REML procedure considering FLMY, age at first calving (AFC) and first service period (FSP) combination exhibits heritabilities as $0.274{\pm}0.173$, $0.506{\pm}0.233$ and $0.274{\pm}0.172$, respectively. Genetic correlations were $-0.120{\pm}0.376$, $0.225{\pm}0.423$ and $0.365{\pm}0.331$ between FLMY and AFC, FLMY and FSP, AFC and FSP, respectively. Phenotypic correlations were 0.057, 0.289 and 0.123, respectively. Considering all five traits REML combination heritabilities estimated were $0.238{\pm}0.162$, $0.160{\pm}0.139$, $0.136{\pm}0.132$, $0.409{\pm}0.209$ and $0.259{\pm}0.168$ for FLMY, FLL, ADY, AFC and FSP, respectively. The genetic correlations were positive except FLMY and AFC. The phenotypic correlations were also positive except FLL and ADY, ADY and FSP. Almost all estimates were associated with high standard error.

Improving The Performance of Triple Generation Based on Distant Supervision By Using Semantic Similarity (의미 유사도를 활용한 Distant Supervision 기반의 트리플 생성 성능 향상)

  • The existing pattern-based triple generation systems based on distant supervision could be flawed by assumption of distant supervision. For resolving flaw from an excessive assumption, statistics information has been commonly used for measuring confidence of patterns in previous studies. In this study, we proposed a more accurate confidence measure based on semantic similarity between patterns and properties. Unsupervised learning method, word embedding and WordNet-based similarity measures were adopted for learning meaning of words and measuring semantic similarity. For resolving language discordance between patterns and properties, we adopted CCA for aligning bilingual word embedding models and a translation-based approach for a WordNet-based measure. The results of our experiments indicated that the accuracy of triples that are filtered by the semantic similarity-based confidence measure was 16% higher than that of the statistics-based approach. These results suggested that semantic similarity-based confidence measure is more effective than statistics-based approach for generating high quality triples.

Multi-decadal Changes in Fish Communities Jeju Island in Relation to Climate Change (기후변화에 따른 제주도 주변 해역 수산 어종 변화(1981-2010))

  • We compiled and analyzed long-term time-series data collected in Korea to evaluate changes in oceanographic conditions and marine ecosystems near Jeju Island ($33^{\circ}00^{\prime}-34^{\circ}00^{\prime}\;N$, $125^{\circ}30^{\prime}-127^{\circ}30^{\prime}\;E$) from 1981 to 2010. Environmental data included depth-specific time series of temperature and salinity that have been measured bimonthly since 1961 in water columns at 175 fixed stations along 22 oceanographic lines in Korean waters by the National Fisheries Research & Development Institute, and time series of estimated volume transport of the Tsushima Warm Current (TWC) and Korea Strait Bottom Cold Water (KSBCW) for the period from 1961 to 2008. We analyzed the species composition in terms of biomass of fish species caught by Korean fishing vessels in the waters near Jeju Island (1981-2010). Data were summarized and related to environmental changes using canonical correspondence analysis (CCA). The CCA detected major shifts in fish community structure between 1982 and 1983 and between 1990 and 1992; the dominant species were a filefish during 1981-1992 and chub mackerel from 1992 to 2007. CCA suggested that water temperature and salinity in the mixed layer and the volume transport of the TWC and the KSBCW were significantly related to the long-term changes in the fish community in the waters off Jeju Island. Fish community shifts seemed to be related to the well-established 1989 regime shift in the North Pacific. Further studies are required to elucidate the mechanisms driving climate change effects on the thermal windows and habitat ranges of commercial species to develop fisheries management plans based on reliable projections of long-term changes in the oceanographic conditions in waters off Jeju Island.

Production of a Recombinant Anti-Human CD4 Single-Chain Variable-Fragment Antibody Using Phage Display Technology and Its Expression in Escherichia coli

  • Single-chain variable fragment (scFv) is a fusion protein of the variable regions of the heavy (VH) and light (VL) chains of immunoglobulin, connected with a short linker peptide of 10 to about 20 amino acids. In this study, the scFv of a monoclonal antibody against the third domain of human CD4 was cloned from OKT4 hybridoma cells using the phage display technique and produced in E. coli. The expression, production, and purification of anti-CD4 scFv were tested using SDS-PAGE and Western blot, and the specificity of anti-CD4 scFv was examined using ELISA. A 31 kDa recombinant anti-CD4 scFv was expressed and produced in bacteria, which was confirmed by SDS-PAGE and Western blot assays. Sequence analysis proved the ScFv structure of the construct. It was able to bind to CD4 in quality ELISA assay. The canonical structure of anti-CD4 scFv antibody was obtained using the SWISS_MODEL bioinformatics tool for comparing with the scFv general structure. To the best of our knowledge, this is the first report for generating scFv against human CD4 antigen. Engineered anti-CD4 scFv could be used in immunological studies, including fluorochrome conjugation, bispecific antibody production, bifunctional protein synthesis, and other genetic engineering manipulations. Since the binding site of our product is domain 3 (D3) of the CD4 molecule and different from the CD4 immunological main domain, including D1 and D2, further studies are needed to evaluate the anti-CD4 scFv potential for diagnostic and therapeutic applications.

Relationship between the spatial distribution of coastal sand dune plants and edaphic factors in a coastal sand dune system in Korea

  • We conducted the study on the relationship between the distribution of coastal sand dune plants and edaphic factors from the shoreline to inland in sand dune ecosystem. The application of TWINSPAN classification based on 10 species, led to the recognition of three vegetative groups (A-C), which associated with their habitats (foredune, hummuck in semistable zone and stable zone). The associations were separated along soil gradient far from the seashore. The relationships between species composition and environmental gradients were explained by canonical correspondence analysis (CCA). Distance from the shoreline was an important indicator to determine soil properties (pH, total ion contents, sand particle sizes, organic matters and nitrogen contents) from the seaward area to inland area and distribution pattern of coastal sand dune plants. Group A is foredune zone, characterized by Calystegia soldanella; group included typical foredune species such as Elymus mollis, Carex kobomugi, Ixeris repens, C. soldanella and Glehnia littoralis. Group B on semi-stabilized zone was characterized by Vitex rotundifolia, a perennial woody shrub. This group was associated the proportion of fine sand size (100 to 250 μm). The results on the proportion of soil particle size showed a transition in sand composition, particularly with respect to the proportion of fine sand size that occurred from the foredune ridge at 32.5 m to the Vitex rotundifolia community at 57.5 m from the shoreline. Group C on stabilized zone was characterized by Zoysia macrostachya, Lathyrus japonicus and Cynodon dactylon and were associated soil organic matter and nitrogen contents. The spatial distribution of plants in the Goraebul coastal sand dune system may result from the interactions between the plant species and environmental heterogeneity.

Geographic Variation in Survival Rate and Height Growth of Pinus densiflora S. et Z. in Korea

  • This study was conducted to examine the geographic variation among provenances of Pinus densiflora in survival rate and height growth at four test plantations (Jungsun, Chungju, Naju, and Jeju). The plantations were parts of the eleven provenance trials of Pinus densiflora established by Korea Forest Research Institute in 1996. The survival rate and height growth were significantly different among test plantations at $p{\leq}0.01$. Latitude and longitude of test plantation were negatively correlated with survival rate and height growth. On the other hand, annual mean temperature, mean temperature (Nov.~Feb.), extremely low temperature (Dec.~Feb.), and annual mean growing days of test plantation were positively correlated with these two. The relationships between growth variables and geographic variables were analysed with canonical correlation analysis. A considerable amount of variation in survival rate and height growth was explained by latitude, annual mean growing days, extremely low temperature (Dec.~Feb.) and extremely high temperature (Nov.~Feb.) of provenances. It is estimated that up to 47.1% and 67.4% of the genetic variability in survival rate and height growth was attributable to the environmental variability of the provenances, respectively. The response surface curve of survival rate and height growth was plotted against latitude and longitude to examine growth performance of provenances for each test site. Generally, the local provenances showed better survival rate and height growth.
