Determination of Glucan Contents in the Fruiting Bodies and Mycelia of Lentinula edodes Cultivars

  • Shiitake mushrooms (Lentinula edodes) containing ${\beta}$-glucans may be beneficial for human health; they have been used in the treatment of cancer, hypertension, and high cholesterol levels. The objective of this study was to determine the ${\beta}$-glucan content in different sections of the fruiting bodies and mycelia of ten shiitake mushroom cultivars. The measured ${\beta}$-glucan content ranged from $20.06{\pm}1.76%$ to $44.21{\pm}0.13%$ in the pileus sections, and from $29.74{\pm}1.40%$ to $56.47{\pm}4.72%$ in the stipe sections. The results of this study indicate that the variance in ${\beta}$-glucan content dependent on the shiitake cultivar, and that the ${\beta}$-glucan content is higher in the stipe than in the pileus.

  • 상황버섯을 $90^{\circ}C$에서 환류추출 시 시간과 추출용매의 조건(물 및 에탄올 농도, pH)에 따른 추출물을 제조하여 ${\beta}$-glucan 함량과 항산화활성 및 항산화성분의 함량을 조사하였다. 상황버섯 추출물은 추출시간이 길어짐에 따라 수율과 ${\beta}$-glucan 함량이 증가하여 24시간 추출 조건을 실험에 사용하였다. 추출용매에 따른 상황버섯 추출물의 수율은 60% 에탄올, pH 4의 조건에서 가장 높았다. ${\beta}$-glucan 함량은 열수 추출물에서 높은 함량을 나타내었고, 산성과 중성 조건에서 높게 나타났다. 항산화활성은 60% 에탄올, pH 7 조건에서 추출한 것이 가장 높았다. 항산화성분의 함량 또한 항산화활성과 같은 경향을 보였으며, 수율, ${\beta}$-glucan 함량, 항산화활성 및 성분을 모두 고려하였을 때 60% 에탄올, pH 7 추출 조건이 적합하였다.

Effects of $\beta$-glucans from Coriolus versicolor on Macrophage Functions in Young and Aged mice

  • The immune system may play an important role in aging and the changes in the immune status are associated with treatment of various immunomodulators. This study examined the effects of $\beta$-glucans isolated from mushroom fungi, Coriolus versicolor on macrophages functions in young (8-weeks-old) and aged (82-weeks-old) male C57BL/6 mice. When peritoneal macrophages were treated with various concentrations of $\beta$-glucan ($1-100\;{\mu}g/ml$) for 24 hrs, tumoricidal activity, NO production and phagocytic activity were significantly increased in the young mice, whereas there are no effects in the aged mice. These results suggest that $\beta$-glucans has differential effects on the macrophage functions in young and aged mice and age nutrition might need to be considered to select proper immunomodulator. In addition, $\beta$-glucan could be used clinically for the treatment of diseases such as cancer therapy in the young.

Saccharomyces cerevisiae 변이주 세포벽 유래의 베타글루칸의 면역활성 및 Cisplatin과의 병용에 의한 항암 상승작용 (Immunostimulating Activity of Beta-Glucan Isolated from the Cell Wall of Mutant Saccharomyces cerevisiae, and Its Anti-Tumor Application in Combination with Cisplatin)

  • Cisplatin(cis-diamminedichloroplatium) is one of the most effective anti-cancer drugs being clinically used in the treatment of solid tumors. Despite its therapeutic benefits, its use in clinical practice is often limited because of dose related toxicity. It is known that yeast cell wall beta-glucans possess immuno-modulating properties, which allows for their application in antitumor therapy. IS2 is a kind of beta-glucan derived from the cell wall of mutated Saccharomyces cerevisiae, which exhibits anti-cancer activity in vitro and in vivo. The present study explored the possibility of combination therapy of IS2 with cisplatin. In experimental metastasis of colon26-M3.1 cells, prophylactic intravenous administration of IS-2 in combination with cisplatin effectively inhibited tumor metastasis compared with cisplatin alone or IS-2 treatment in vivo. IS-2 effectively enhanced Th1 type cytokines including IFN-$\gamma$, IL-2, IL-12 and GM-CSF. Simultaneously, this combined treatment inhibited production of Th2 type cytokines compared with control. These results suggested that IS-2 can be applied in combination therapy with anti-cancer drugs to minimize their side effects.

Ginseniside Rh2와 베타 글루칸의 암세포 전이억제효과 (Antimetastatic and Antitumor Effect of Ginsenoside Rh2 and ${\beta}-glucan$ in Mice)

  • We have investigated the antimetastatic and antitumor effects of Ginsenoside Rh2 and ${\beta}-glucan$ unsing an experimental metastatic mouse model intravenously injected with B 16 melanoma F 10 cells. Animal groups are divided into six groups according to the dosage of drug administration and the kind of drugs. The groups are control, ${\beta}-glucan$ with 50, 100 and 200 mg/kg, Geinsenoside Rh2 50 mg/kg, and ${\beta}-glucan$ 50 mg/kg + Ginsenoside Rh2 50 mg/kg. Oral administration of various concentration of ${\beta}-glucan$( 50, 100, and 200 mg/kg) were reduced the lung- metastatics induced by metastatic B16 melanoma F 10 cells injection with a dose dependent manner in the syngenic mice. At same dosage group, Ginsenoside Rh2 (50 mg/kg) has more antimetastatic effect than the ${\beta}-glucan$(50 mg/kg). The highest antimetastatic effects was observed in the ${\beta}-glucan$ 50 mg/kg + Ginsenoside Rh2 50 mg/kg group and has a similar tendency in the anti-tumor effects, including decrease of the average tumor weight and increase of the average survival rate. There are no differences of the average tumor weights were apparent in the ${\beta}-glucan$ groups, however there were little decrease of the average tumor weight in Ginsenoside 50 mg/kg group and ${\beta}-glucan$ 50 mg/kg + Ginsenoside Rh2 50 mg/kg group than that of the control group. The rate of average survival rate in the ${\beta}-glucan$ 50 mg/kg + Ginsenoside Rh2 50 mg/kg group, ${\beta}-glucan$ 200 mg/kg, ${\beta}-glucan$ 100 mg/kg and ${\beta}-glucan$ 50 mg/kg, and Ginsenoside 50 mg/kg groups were highly in order. These data suggest that antimetastatic and antitumor effect of combination of Ginsenodide Rh2 and ${\beta}-glucan$ be the highest in this study.

치마버섯균 유래의 베타글루칸에 대한 사료첨가제로서의 대식세포 기능 활성 및 유방암 세포주에서의 항암효능 효과 (Macrophage and Anticancer Activities of Feed Additives on β-Glucan from Schizophyllum commune in Breast Cancer Cells)

  • 치마버섯에서 추출한 베타글루칸을 Raw 264.7 세포에 처치한 후 MTT assay로 측정한 결과 베타글루칸 400 ${\mu}g$/mL까지에 의한 세포독성은 없었다. 대식세포의 활성능을 측정하기 위해서 Raw 264.7 세포에서 NO와 TNF-${\alpha}$의 생성을 측정하였다. 베타글루칸을 대식세포에 24시간 처리한 결과 대조군과 비교 시 NO와 TNF-${\alpha}$가 유의적으로 상승하였다. 이 결과 베타글루칸이 대식세포인 Raw 264.7 세포를 활성화 시키는 것으로 사료된다. 따라서 본 실험에 사용된 베타글루칸은 면역강화용 사료첨가제로서의 의미가 충분하다고 사료된다. MDA-MB-231 유방암 세포의 성장을 저해하는지 확인하기 위하여 MTT assay를 시행하였다. 400 ${\mu}g$/mL 베타글루칸을 48시간 처리한 결과 대조군과 비교 시 유방암 세포의 성장이 유의적으로 억제하였다. 그리고 베타글루칸이 유방암 세포에서 성장 억제 효과를 알아보기 위해 누드마우스에 MDA-MB-231 유방암 세포를 접종하였다. 베타글루칸의 용량은 대조군(0 ${\mu}g$/mouse), 저용량(200 ${\mu}g$/mouse), 고용량(400 ${\mu}g$/mouse)으로 설정하여 누드마우스에 경구투여 하였다. 이 결과 저용량군(200 ${\mu}g$/mouse)이 고용량군(400 ${\mu}g$/mouse)보다 항암효능이 더 많이 일어나는 것을 확인하였다. MDA-MB-231 유방암 세포주에서 베타글루칸이 종양 성장이 감소하였지만, 그 결과가 유의적인 차이를 보이지 않아 본 연구에서 사용한 용량에서는 베타글루칸이 MDA-MB-231 유방암 세포주에서 종양 성장을 유의적으로 억제하지 않는 것으로 사료된다. 조직병리학적 검사에서 독성이 무해한 것으로 판단되어 면역강화용 사료첨가제로 사용하여도 무방할 것으로 사료된다.

β-Gluconsan Calcium이 구강암 세포의 성장에 미치는 영향 (The Anti-Cancer Effect of β-Gluconsan Calcium on Oral Cancer Cell)

  • 이번 연구에서는 천연추출물인 ${\beta}$-gluconsan calcium이 구강암 세포의 증식과 사멸에 미치는 영향을 평가하고자 하였으며, 다음과 같은 결론을 얻었다. ${\beta}$-gluconsan calcium 0.5 mg/ml 처리군에서 세포수의 감소와 형태 변화가 관찰되었으며, 0.75, 1 mg/ml에서는 거의 모든 세포가 사멸하였다. ${\beta}$-gluconsan calcium을 농도별로 처리한 결과, YD-10B 세포수가 농도 의존적으로 감소하였으며(p<0.05), MTT 분석 결과, ${\beta}$-gluconsan calcium 처리한 군은 아무것도 처리하지 않은 군에 비해 모두 세포 활성이 저하되었다(p<0.05). 이상의 결과들을 종합해 보았을 때, polycan과 calcium gluconate 복합제인 ${\beta}$-gluconsan calcium은 구강암 세포의 성장 억제에 효과가 있는 것으로 생각된다.

Immunomodulatory Effects of Aureobasidium pullulans SM-2001 Exopolymers on Cyclophosphamide-Treated Mice

  • The immunomodulatory effects of Aureohasidium pullulans SM-2001 exopolymers containing $\beta$-1,3/1,6-glucan were evaluated in cyclophosphamide (CPA)-treated mice. To induce immunosuppression, 150 and 110 mg/kg of CPA were intraperitoneally injected 3 days and 1 day, respectively, before beginning administration of the test material. Exopolymers were delivered subcutaneously or orally, four times, in a volume of 10 ml/kg at 12-h intervals beginning 24 h after the second CPA treatment. Changes in thymus and spleen weights, splenic amounts of tumor necrosis factor (TNF)-$\alpha$, interleukin (IL)-$1{\beta}$, and IL-10, and numbers of CD3+, CD4+, CD8+, and TNF-$\alpha+$ thymus and spleen cells were monitored in CPA-treated mice. As a result of CPA treatment, dramatic decreases in the number of CD3+, CD4+, CD8+, and TNF-$\alpha+$ cells were detected in the thymus and spleen, along with decreases in thymus and spleen weights. In addition, splenic TNF-$\alpha$, IL-$1{\beta}$, and IL-10 contents were also decreased on observation with flow cytometry. However, oral and subcutaneous treatments with exopolymers effectively reduced the immunosuppressive changes induced by CPA. Therefore, it is concluded that exopolymers of A. pullulans SM-2001 can effectively prevent immunosuppression through, at least partially, the recruitment of T cells and TNF-$\alpha+$ cells or enhancement of their activity, and can provide an effective component of prevention or treatment regimens for immunosuppression related to cancer, sepsis, and high-dose chemotherapy or radiotherapy.

추출 및 침전조건에 따른 아가리쿠스버섯에서 분리한 조단백다당류의 특성 및 암세포 증식억제 효과 (Characteristics of Crude Protein-bound Polysaccharide from Agaricus blasei Murill by Extraction and Precipitation Conditions and Its Antitumor Effect)

  • 아가리쿠스버섯에 함유된 생리활성 물질인 조단백다당류의 효율적인 추출공정을 확립하기 위하여 다양한 추출조건에서의 화학적 특성을 조사하였으며, 추출 분리된 조단백다당류의 열분해 특성 및 SRB assay에 의한 인체 암세포 증식억제 효과를 조사하였다. 조단백다당류의 추출수율은 초음파추출이 13.0%로 가장 높았으며, 열수추출이 7.8%로 가장 낮았다. 추출된 조단백다당류의 당 조성은 약 80%의 glucose와 fucose, galactose 및 mannose로 구성되어 있었으며 가압조건에서 2시간과 3시간의 추출은 ${\beta}-glucan$ 함량이 각각 32.28%와 32.34%로 뚜렷한 차이가 발견되지 않아 가압추출 2시간이 생리활성물질의 추출조건으로 효과적이었다. 또한 침전조건에 따른 조단백다당류의 침전수율 및 ${\beta}-glucan$ 함량은 4배 첨가 시 각각 10.89%와 35.97%로 가장 많이 함유되어 있었다. 열분해 특성은 네 단계로 이루어져 있었으며, $240-365^{\circ}C$에서 67.8%의 무게 감소를 보여주어 높은 온도에서도 열적 안정성이 있었다. SRB assay에 의한 인체 암세포 증식억제 효과를 검토한 결과, A549의 경우 $1,000{\mu}g/mL$ 농도에서 72시간 처리 시에는 43.9%의 암세포 손상을 보여주어 MCF-7과 AGS에 비해 암세포 증식억제 효과가 높음을 확인할 수 있었다.

Agrocybe chaxingu polysaccharide prevent inflammation through the inhibition of COX-2 and NO production

  • The inhibition of nitric oxide (NO) and cyclooxygenase-2 (COX-2) production is considered to be a promising approach to the treatment of various diseases, including inflammation and cancer. In this study, we examined the effects of the Agrocybe chaxingu $\beta$-glucan (polysaccharide) on lipopolysaccaride (LPS)-induced nitric oxide (NO) and cyclooxygenase-2 (COX-2) expression in murine macrophage Raw 264.7 cells as well as 12-O-tetradecanoylphorbol 13-acetate (TPA)-induced ear edema in mice. The polysaccharide significantly inhibited (P < 0.01) LPS-induced iNOS and COX-2 expression levels in the cells. Furthermore, topical application of polysaccharide resulted in markedly inhibited (P < 0.01) TPA-induced ear edema in mice. These results suggest that this polysaccharide may be used for NO- and COX-2-related disorders such as inflammation and cancer.
