Organogenesis from Callus Derived from In Vitro Root Tissues of Wild Prunus yedoensis Matsumura

  • In vitro organogenesis system of the valuable ornamental species, Prunus yedoensis which is native to Korea, was established through callus culture derived from root tissues. Callus were induced on the medium supplemented with 2,4-D and BA or NAA and kinetin. Organogenesis was differ from the callus type, and NAA and kinetin combination was effective to induce organogenic callus. Growth of callus was influenced by sucrose concentrations. High level of sucrose (over 5%) had adverse effects such as decreased fresh weight and increased mortality of callus. Shoots developed from the callus when $GA_3$ was treated with BA in the medium. Results showed that $GA_3$ is essential for shoot development and elongation from callus in this species.

Effects of Medium Components and Growth Regulators on Callus Development and Shoot Regeneration from Shoot Explants of Black Locust (Robinia pseudoacacia)

  • Effects of growth regulators and medium components were tested for shoot multiplication and callus growth from shoot explants of black locust. During shoot multiplication, callus growth at the cut end of shoot explants proceeded shoot development. The basal callus growth seemed to be a function of both mineral components and cytokinin supplemented in the medium. Maximum callus growth was induced by 0.5${\mu}$M BAP and the callus growth decreased as the level of BAP increased. Positive correlations were found between basal callus growth, and shoot multiplication and growth. Shoot multiplication was greatest on BSM medium (black locust shoot culture medium) supplemented with 1 $\mu$M BAP. With medium containing high nitrogen content, both shoot multiplication and growth were significantly enhanced. A new BRM medium was the most effective for rooting of black locust among three rooting media tested.

High Efficiency of Plant Regeneration from Seed-Derived Callus of Zoysiagrass cv. Zenith (Zoysiagrass japonica의 효율적인 재분화체계에 관한 연구)

  • The development of a protocol for high efficiency of embryogenic callus separation, maintenance and plant regeneration from the seeds of zoysiagrass cv. Zenith was studied. Embryogenic callus ratio is absolutely determined by genotype, but by adding high concentration of copper into medium, changing light condition and maintaining callus on initial induction medium for 8∼10 weeks, embryogenic callus can be easily distinguished and its growth can be promoted. There were significant differences among selected callus lines (each from one seed) according to their growth rates and regeneration percentages. Callus pre-treatment with activated charcoal inhibited callus growth, increased the level of precocious germination during culture and promoted shoot cluster formation after transfer to regeneration medium. For long-term callus maintenance, N6AA medium was better than MS medium, because the former inhibited non-embryogenic callus formation and kept vigor of embryogenic callus. The best callus lines Z-(5) has been successfully used for transformation and somaclonal variation selection in our laboratory.

Effect of Embryogenic Callus Conditions on Plant Regeneration in Satsuma Mandarin (Citrus unshiu Marc.)

  • The ability to form embryiod from callus in satsuma mandarin is low and unstable. In this study, the conditions of embryogenic calli induced from nucellar tissue for promotion of plant regeneration in satsuma mandarin were investigated. The calli of I, II and III line were divided into two sizes of 0.5 mm and 1.0 mm in diameter and two weight gradients of percoll at 40% and 50% though the filter mesh. The frequency of embryo formation from $\phi$ 1.0mm-40% was slightly higher than callus that from others. Adventitious embryoids developed to a globular stage were transferred to regeneration medium. In 'Miyagawa Wase', the embryos from I and II line developed into a heart stage from most of $\phi$ 0.5 mm- 40% and $\phi$1.0 mm-40% calli, but it failed in 'Sugiyama Unshu'. In the cultivar of 'Miyagawa Wase', 63% of adventitious embryos transferred to the regeneration medium developed into the heart stage from the most $\phi$ 1.0 mm-40% calli of I line, but of 'Sugiyama Unshu' failed in some calli condition. The embryoids from two callus lines developed further to shoots and plantlets, while the embriods from III line abnormal failed to regenerate in the cultivar. From these results, it is suggested that the plant regeneration from embryogenic callus in satsuma mandarin could be affected by callus conditions.

Hormonal Effect and Cytokinin Autonomy in callus Culture of Phaseolus vulgaris L. (식물 Hormone의 영향과 Cytokinin Autonomy)

  • The activities of auxins and cytokinins have been examined in the growth of callus tissue derived from Phaseolus vulgaris L. cv. Damyang. The synthetic auxin, picloram was the most effective in promoting callus growth and the range of effective concentrations (0.1$\mu{M}$ to 32$\mu{M}$) was broad. 2, 4-D also enhanced callus growth at the optimal concentration of 3.2$\mu{M}$. NAA promoted callus growth at relatively higher concentrations than other auxins tested. IAA was less effective in supporting callus growth. Cytokinin bearing saturated side chain ($N^6$-isopentyladenine) was approximately 30 times more active than the corresponding unsaturated compound, $N^6$-($\D^2$-isopentenyl) adenine. The abilities of cytokinin-autonomous growth were also examined. Callus tissues previously grown on concentrations lower and/or higher than optimal concentrations of cytokinins were better habituated in the subsequent passage. It was suggested that the development of cytokinin autonomy may be related to dosage-concentrations of cytokinin in the previous passage.

  • To obtain a basic information of the development of Genus Viola, morphological and histological observation of in vitro calli and cells in Viola culture cells were investigated. There were two callus types obtained by long term subculture of wild viola (Viola partrinii DC. ) petiole callus. One was friable callus - soft and pale green in color and small cells in size, and the other was compact callus - compact and deep bluish green in color, large cells in size. In scanning electron microscopic observation, friable callus was composed of voculated cell around small. cell clump, while compact callus was composed of cells filled with protoplasm Somatic embryogenesis was observed from suspension culture of the compact callus.

Callus Induction and Plant Regeneration Efficiency According to Tissue Culture Conditions in Teff grass (Eragrostis) (테프그라스 조직배양을 통한 캘러스 형성 및 식물체 재분화 효율)

  • Teff grass is a warm season C4 annual grass that is used for dry hay, silage and haylage. We have developed a high-frequency plant regeneration system for teff grass via callus culture using mature seeds. It was revealed that mature seeds cultured on MS medium supplemented with 2 mg/l 2,4-D, 0.5 g/L proline, 0.5 g/L casamino acid and 3 g/L Gelrite under light condition produced the highest percentage of callus formation (91.9%). Addition of cytokinins (BA) at 0.0~0.5 mg/L to media containing 2 mg/l 2,4-D enhanced callus growth. The most suitable medium for plant regeneration from dehydrated calli was MS agar medium supplemented with 0.1 mg/l NAA, 1 mg/l BA, 0.5 g/L proline, 0.5 g/L casamino acid 3 g/L Gelrite which induced the highest percentage of calli forming shoots (47.0%). The shoots were rooted at the highest rate (100%) when transferred onto 1/2 MS medium and acclimated in greenhouse conditions.

Esterase isozyme patterns in developing plant regeneration from calli of citrus junos Sieb.

  • The callus from the hypocotyl region of immature embryo of Citrus junos Sieb. was efficiently induced in the $\frac{1}{2}$ MS medium containing 45uM BA after 8 weeks culture. The callus was developed into the two callus type, embryogenic callus and nonembryogenic callus, which can be distinguished by visual examination depending on color and appearence. In vitro regeneration of callus established efficiently in the hormone-free MS medium from the embryogenic callus. In order to investigate the physiological changes depending on the developmental stage of embryo, the embryo was formed in the MS medium. The embryogenic and nonembryogenic callus, and the various stages of the somatic embryo were examimed the changes of esterase activity, and their isozyme patterns as well. The protein content and esterase activities was gradually increased on the developmental stages of embryo. Total protein pattern were different by the SDS-PAGE and were appeared strong band of 23 KD in the torpedo stage. The pattern of the esterase isozyme was exhibited a difference between embryogenic callus and nonembryogenic callus. It was appered pI 6.0, 8.0, 8.2 in the embryogenic callus. Also the new band of pI 4.75 was appeared in the cotyledon. These results suggest that the changes of esterase activities and isozyme patterns are importent factor in the differentiation and development of citrus.

Cytohistological Study of Development of Callus and Adventitious Shoots from Cultured Stem of Vigna radiata (녹두 줄기 조직배양에서 캘러스와 부정아 형성에 관한 세포조직학적 연구)

  • This study was carried out to establish a reproducible culture system for callus formation and adventitious shoot development from young stem segments of Vigna radinta, and histological work for orgin of callus tissue and adventitious shoot. Induction of callus from young stem explants of Vigna radiata was very effective on MS inorganic salts supplemented with 0.5 mg/L 2,4-D and 1.0 mg/L kinetin. For the adventitious shoot regeneration from the callus tissues, the hormone combination of 0.75 mg/L NAA, 1.5 mg/L kinetin and MS salts resulted in about 21% efficiency. Histological examination showed that callus tissues originated from out-growths by callus cambium rings with do novo meristematic activities, which were localized at the outside of the vascular cambium. Adventitious shoots were developed from shoot apical meristem originated from the surface of callus masses. The shoot apical meristem produced leaf primordium, which then became leaf.

Variation of Essential Oil Content and Its Composition during Callus Subculture of Peppermint (Mentha piperita) (페퍼민트 캘러스 계대배양 기간 중 정유함량과 성분변화)

  • Peppermint (Mentha piperita L.) belongs to a member of the mint family (Lamiaceae) and is widely used in food, cosmetics and medicines. This study was carried to investigate the variation of essential oil content and its composition during callus subculture of M. piperita. For callus induction from the leaf explant of peppermint, the basal medium was supplemented with various concentrations of 2, 4-D. The best callus induction rate (93%) of M. piperita. was obtained in MS medium containing 2 mg/l 2, 4-D. The induced peppermint callus maintained on Lin-Staba medium were studied during a period of 20th subcultures for the stability of essential oil production. Growth rates of peppermint callus increased during prolonged subculture. However, there was a progressive decrease of essential oil content and unstability of monoterpene productions when callus cultures were serially subcultured.
