Inverase was entrapped in calcium alginate gel. The immobilized beads had excellent uniform size and configuration. To screen the optimal conditions possessing high enzyme activity, effects of sodium alginate concentration, $CaCl_2$ concentration, and the incubation time of beads in $CaCl_2$ solution during the immobilization procedure were investigated. Immobilized beads prepared from the optimal conditions had 18.8 units per ml of gel, which is equivalent to 68% of the activity of soluble enzyme. Several kinetic parameters were determined: Km value. 143 mM; optimum pH, 4.5; optimum temperature, $50^{\circ}C$. Enzyme loading capacity was 150 mg per 20 ml gel. No significant decrease in sugar conversion was observed during the column operation for 6 days.
The adsorption behavior of $Pb^{2+}$ was compared between calcium alginate beads and capsules, which have different structures of alginate-gel core beads and liquid core alginate-membrane capsules, respectively. In terms of adsorption kinetics and isotherms, adsorption characteristics depending on pH and hardening time were compared for both adsorbents and also released calcium ion during the adsorption process was monitored. The adsorption of $Pb^{2+}$ on both adsorbents was caused by surface complexation and ion exchange mechanisms, both of which have similar effects on adsorption process regardless of the amount of adsorbed $Pb^{2+}$. The dependence of $Pb^{2+}$ adsorption upon pH was also similar for both adsorbents indicating the existence of similar functional groups on the surface of adsorbents. However, a different $Pb^{2+}$ adsorption behavior was observed considering the adsorption kinetics. The adsorption kinetic of $Pb^{2+}$ on alginate beads was slower than on alginate capsules and the maximum adsorption loading ($Q_{max}$) onto alginate beads was also less than onto alginate capsules by 49%. This drawback of alginate beads compared to capsules were ascribed to a diffusion limitation due to solid gel-core structure of alginate beads.
Alginic acid is a hydrophilic , colloidal polysaccharide obtained from cell wall of seaweed or brown algae and has a broad range of applications. Alginlc acid becomes alginate gel bead due to its cation-induced gelation. Dried alginate beads can be reswollen according to environmental pH. The purpose of this paper is to explore the possible applicability of alginate beads as an oral controlled release system of ibuprofen. In this experiment ibuprofen was incorporated in alginate beads and alginate beads were treated with various methods. Ibuprofen release from alginate beads in phosphate buffer (pH 7.4) was laster than in distilled water and dilute HCl. The release of ibuprofen was more sustained in bead than simple mixture and coprecipitate of ibuprofen and sodium alginate. The dissolution rate of ibuprofen was decreased in using of bead that hardened with formaldehyde. The dissolution rate of the drug from the bead was the fastest in 12 hour dried beads, 1.5%-sodium alginate concentration and 1%-calcium chloride concentration. Sodium alginate bead can be used as a sustaind release drug delivery system of water-insoluble drugs.
The Pseudomonas sp. strain NGK1 (NCIM 5120) was immobilized in calcium alginate, agar, and polyacrylamide gel matrices. The salicylic acid-producing capacity of freely suspended cells was compared with immobilized cells in batches with a shake culture and continuous culture system in a packed bed reactor. Freely suspended cells ($4\times10^{10}cfu/ml$) produced 12 mM of salicylic acid, whereas cells immobilized in calcium alginate ($1.8\times10^{11}$cfu/g beads), agar ($1.8\times10^{11}$cfu/g beads), and polyacrylamide ($1.6\times10^{11}$cfu/g beads) produced 15, 11, and 16mM of salicylic acid, respectively, from naphthalene at an initial concentration of 25 mM. The continuous production of salicylic acid from naphthalene was investigated in a continuous packed bed reactor with two different cell populations. The longevity of the salicylic acid-producing activity of the immobilized cells from naphthalene was also studied in semi continuous fermentations. The immobilized cells could be reused 18, 13, and more than 20 times without losing salicylic acid-producing activity in calcium alginate-,agar-, and polyacrylamide-entrapped cells, respectively. The study reveals a more efficient utilization of naphthalene and salicylic acid production by the immobilized Pseudomonas sp. strain NGK1 as compared to the free cells.
Sodium alginate was used to immobilize Bifidobacterium breve ATCC 15700 cells. The ability of the Ca-alginate beads to protect the B. breve ATCC 15700 was evaluated under different conditions including alginate concentration, bead size, pH, hydrogen peroxide, and storage period. The survival of the B. Breve ATCC 15700 was estimated in pasteurized yogurt, containing either the immobilized or free cells, throughout the storage period. The survival cells in bead after exposure to acidic solution (pH 3.0) increased with increase of both the alginate gel concentration and bead size. Also, immobilized cells in alginate bead were more resistant than the free cells to hydrogen peroxide, storage period, and the environment inside yogur. When retreated beads with skim milk and nonretreated beads were tested in acidified pH 3.0 TPY media including acetic and lactic acid, the number of viable cells in the retreated bead was approximately 10-fold higher than that of nonretreated beads. This suggests that the skim milk operated as a material decreasing the diffusion of acid and hydrogen perosicde into alginate gels. From this research, it was found that yogurt itself supported immobilized cells with an improved protection from the extreme acidity in yogurt.
This study is to investigate for obtaining the operating conditions of continuous vinegar production using fluidized bed reactor by Acetobacter aceti cell immobilized in Ca-alginate gel. The optimum conditions obtaining by batch fermentation using fluidized bed reactor were as follows; The fermentation temperature and aeration rate were 3$0^{\circ}C$ and 1.0VVM and the initial concentration of ethanol and acetic acid in medium were 33g/l and 27g/l respectively. The amount of bead used was 25%(w/v). The overall acetic acid productivities of batch fermentations by free cell and immobilized cell were 0.31g/l-hr and 0.48g/l-hr, respectively, at the final acetic acid concentration of 50g/l. In the continuous vinegar production using fluidized bed reactor by immobilized cell under optimum conditions, it was possible to produce 23g/l acetic acid continuously up to 90 days with maximum acetic acid productivity of 2.76g/l-hr at dilution rate 0.12hr-1.
Primary hepatocytes of small animals such as rat and rabbit were often used for the study of extracorporeal liver support systems. Freshly isolated rat hepatocytes form spheroids within tow days when cultivated as suspension in spinner vessels. These spheroids showed enhanced liver specific functions and more differentiated morphology compared to hepatocytes cultured as monolayers However, shear stress caused by continuous agitation deteriorated spheroids gradually. In this work we immobilized spheroids to prolong liver specific activities. First, hepatocyte spheroids were suspended in collagen solution containing calcium chloride and then dropped into alginate solution. A thin layer of calcium alginate was formed around the droplet and then was removed after the inner collagen was gelled by treatment of sodium citrate buffer. Spheroids embedded in collagen-gel bead maintained liver specific functions such as albumin secretion rate longer than hepatocyte spheroids exposed to shear stress. Therefore, we suggest that this immobilization technique may offer an effective long-term hepatocyte cultivation and facilitase the development of a bioartificial liver support device.
The effective diffusion coefficients of glucose, sucrose and fructo-oligosaccharides in Ca-alginate gel beads at high concentration of sucrose solutions were investigated at $50^{\circ}C$. A mathematical model for the kinetics of fructo-oligosaccharide production using immobilized cells was proposed and compared with experimental results varying the bead size, the substrate concentration and the bead ratio. Very low values of diffusion coefficients ranging $1.2-7.6\times10^{-7}\textrm{cm}^2$/sec were obtained, and the predicted results were in good agreement with experimental ones in all cases tested.
