It has been well known that the expression of S. typhimurium cadBA operon requires at least two extracellular signals: low pH and high concentration of lysine. To better understand the nature of pH-dependent and lysine dependent signal transduction, mutants were isolated in JF2238(cadA-lacZ) by Tn10 insertion, spontaneous mutagenesis, and EMS treatment. Two mutants were isolated from JF2238, expressed as a cadA-lacZ operon fusion in various growth conditions, and analyzed to have mutations in cadC, a gene encoding a function necessary for transcriptional activation of cadBA. One isolate (cadC6) conferred pH-independent and lysine-independent cadBA expression and the other(cadC4) showed pH-independent and lysine-dependent cadBA expression. cadR::Tn10 and cadR4 mutants were expressed in the absence of exogenously added lysine. They were also resistant to thiosine and complemented by lysP clone from E. coli. Thus, in the absence of exogenous lysine, cadR is a negative regulator of cadBA expression. Cadaverine, the product of lysine decarboxylation, was shown to inhibit expression of cadA-lacZ fusion in cad $C^+$ cell.
Objective: To assess shear bond strength and failure mode (Adhesive Remnant Index, ARI) of orthodontic brackets bonded to polymethylmethacrylate (PMMA) blocks for computer-aided design/manufacture (CAD/CAM) fabrication of temporary restorations, following substrate chemical or mechanical treatment. Methods: Two types of PMMA blocks were tested: $CAD-Temp^{(R)}$ (VITA) and $Telio^{(R)}$ CAD (Ivoclar-Vivadent). The substrate was roughened with 320-grit sandpaper, simulating a fine-grit diamond bur. Two universal adhesives, Scotchbond Universal Adhesive (SU) and Assure Plus (AP), and a conventional adhesive, Transbond XT Primer (XTP; control), were used in combination with Transbond XT Paste to bond the brackets. Six experimental groups were formed: (1) $CAD-Temp^{(R)}/SU$; (2) $CAD-Temp^{(R)}/AP$; (3) $CAD-Temp^{(R)}/XTP$; (4) $Telio^{(R)}$ CAD/SU; (5) $Telio^{(R)}$ CAD/AP; (6) $Telio^{(R)}$ CAD/XTP. Shear bond strength and ARI were assessed. On 1 extra block for each PMMA-based material surfaces were roughened with 180-grit sandpaper, simulating a normal/medium-grit ($100{\mu}m$) diamond bur, and brackets were bonded. Shear bond strengths and ARI scores were compared with those of groups 3, 6. Results: On $CAD-Temp^{(R)}$ significantly higher bracket bond strengths than on $Telio^{(R)}$ CAD were recorded. With XTP significantly lower levels of adhesion were reached than using SU or AP. Roughening with a coarser bur resulted in a significant increase in adhesion. Conclusions: Bracket bonding to CAD/CAM PMMA can be promoted by grinding the substrate with a normal/medium-grit bur or by coating the intact surface with universal adhesives. With appropriate pretreatments, bracket adhesion to CAD/CAM PMMA temporary restorations can be enhanced to clinically satisfactory levels.
Purpose: The purpose of this study is to compare three different CAD/CAM systems through the fracture strength of zirconia core and to evaluate the clinical availability of each system. Material and methods: The following three groups of 30 maxillary mesial incisor core for all-ceramics(each group 10) were fabricated as follows: group 1. $Adens^{(R)}$ CAD/CAM system, group 2. $Cerasys^{(R)}$ CAD/CAM system, group 3. 3M $Lava^{(R)}$ CAD/CAM system. All specimens were manufactured consistently thickness 0.5mm and relief $40{\mu}m$. Specimens were subjected to compressive loading on the lingual area by Z250/$SN5S^{(R)}$. Each group's mean and standard deviation were calculated and Kruskal Waillis test, Wilcoxon Rank Sum test were utilized to find out the relationship among the groups. Results: The results were as follows: 1. The mean fracture strength of $Adens^{(R)}$ system was $615.89{\pm}156.1N$, the $Cerasys^{(R)}$ system was $863.98{\pm}151.5N$, and the 3M $Lava^{(R)}$ system was $1143.1{\pm}286.6N$. 2. The fracture strength of the systems showed the significant statistical differences in order of 3M $Lava^{(R)}$ system, $Cerasys^{(R)}$ system, $Adens^{(R)}$ system. Conclusion: In this study, in spite of the differences among the groups, every group showed clinically useful results. It is necessary to study further clinical experiments on a long term basis.
Statement of problem: There have been many studies about marginal adaptation of Zirconia core fabricated with various CAD/CAM systems. But, the comparative study has not been conducted much. Purpose: This study was to compare marginal adaptations of zirconia cores fabricated with 3 different CAD/CAM systems. Material and methods: The in vitro marginal adaptations of zirconia cores fabricated with 3 different CAD/CAM systems($Adens^{(R)}$ system, $Lava^{(R)}$ system, and $Cerasys^{(R)}$ system) were evaluated and compared. In this study 30 zirconia cores were fabricated(10 zirconia cores per each system). The measurements of the marginal adaptatioin were carried out using microscope $L-150A^{(R)}$(Nikon, Japan). Measurements were recorded at 50 points. The measurements were analyzed using Kruskal-Wallis test and Wilcoxon rank sum test. Results: Within the limits of this study, the results were as follows. 1. Mean marginal gap values obtained were $284.5{\pm}152.9{\mu}m$ in Adens system, $99.0{\pm}73.0{\mu}m$ in Lava system, and $153.5{\pm}85.7{\mu}m$ in Cerasys system. 2. Zirconia core made by Lava system showed the smallest marginal gap(P<0.05). Conclusion: In the limitation of this study, we concluded that $Lava^{(R)}$ System was superior than other systems($Adens^{(R)}$ system and $Cerasys^{(R)}$ system) in the marginal adaptation.
The purpose of this study is to establish a CAD (Computer Aided Design) system for research and development(R&D) of a new product. In the R&D process of a new product, the design objects are frequently redesigned based on the experimental results obtained with prototypes. The CAD/CAE systems (which is based on computer simulation of physical phenomena) are effective in reducing the number of useless prototypes of a new product. These kinds of conventional CAD/CAE systems do not provide a function to reflect the experimental results to the redesign process, however. This paper proposes a methodology to establish the CAD system, which possesses the engineering model of a designed object in the model database, and refines the model on the basis of experimental results of prototype. The blackboard inference model has been applied to infer model refinement and redesign counterplan by using insufficient knowledge of R&D process of new products.

PURPOSE. To investigate the effect of accelerated aging on surface properties of glazed CAD/CAM resin blocks using a 2D surface profilometer and a 3D non-contact optical profilometer. MATERIALS AND METHODS. Three types of CAD/CAM resin restorative materials, LAVA Ultimate (3M ESPE, St Paul, MN, USA), VITA Enamic (Vita Zahnfabrik H. Rauter, Bad $S\ddot{a}ckingen$, Germany), and Cerasmart (GC Corparation, Tokyo, Japan) were used for this study. CAD/CAM blocks were cut in 3-mm thickness slabs and divided into three groups; Group 1: control group (specimens polished with 600 grit SCI paper); Group 2: specimens sandblasted, silanized, and glazed with Optiglaze Color (GC); Group 3: glazed specimens subjected to 5000 thermocycles (n=15). The surface roughness ($R_a$ and $R_z$) was evaluated using a profilometer and a 3D scanning instrument. Data were analyzed using two-way ANOVA and Tukey's post- hoc test (P<.05). RESULTS. LAVA, VITA, and Cerasmart exhibited statistically similar $R_a$ and $R_z$ values for each group (P>.05). For VITA and Cerasmart, the specimens in Group 1 exhibited significantly higher $R_a$ values than Group 2 (P<.05). Group 1 ($0.502R_a$), Group 2 ($0.384R_a$), and Group 3 ($0.431R_a$) exhibited statistically similar $R_a$ values for LAVA (P=.062). After 5000 thermocycles, surface roughness values did not change significantly for glazed LAVA, VITA, and Cerasmart (P>.05). CONCLUSION. Glaze material Optiglaze Color makes CAD/CAM resin surfaces smooth and glazed CAD/CAM surfaces seem resistant to deterioration under 5000 thermocycles.

Objective: To describe the experience of implementing a deep learning-based computer-aided detection (CAD) system for the interpretation of chest X-ray radiographs (CXR) of suspected coronavirus disease (COVID-19) patients and investigate the diagnostic performance of CXR interpretation with CAD assistance. Materials and Methods: In this single-center retrospective study, initial CXR of patients with suspected or confirmed COVID-19 were investigated. A commercialized deep learning-based CAD system that can identify various abnormalities on CXR was implemented for the interpretation of CXR in daily practice. The diagnostic performance of radiologists with CAD assistance were evaluated based on two different reference standards: 1) real-time reverse transcriptase-polymerase chain reaction (rRT-PCR) results for COVID-19 and 2) pulmonary abnormality suggesting pneumonia on chest CT. The turnaround times (TATs) of radiology reports for CXR and rRT-PCR results were also evaluated. Results: Among 332 patients (male:female, 173:159; mean age, 57 years) with available rRT-PCR results, 16 patients (4.8%) were diagnosed with COVID-19. Using CXR, radiologists with CAD assistance identified rRT-PCR positive COVID-19 patients with sensitivity and specificity of 68.8% and 66.7%, respectively. Among 119 patients (male:female, 75:44; mean age, 69 years) with available chest CTs, radiologists assisted by CAD reported pneumonia on CXR with a sensitivity of 81.5% and a specificity of 72.3%. The TATs of CXR reports were significantly shorter than those of rRT-PCR results (median 51 vs. 507 minutes; p < 0.001). Conclusion: Radiologists with CAD assistance could identify patients with rRT-PCR-positive COVID-19 or pneumonia on CXR with a reasonably acceptable performance. In patients suspected with COVID-19, CXR had much faster TATs than rRT-PCRs.

Most manufacturing enterprises create CAD data as a result of the product/part design process; however, the CAD data is being utilized only for production activities. Besides the processes directly related to manufacturing such as design and production, the CAD data is an important resource that can be used in variety of services (e.g., catalog production and production manuals) for manufacturing enterprises. This study proposes a software platform that can support a wide range of services for manufacturing companies in an efficient and productive way. The software platform was designed based on the functions identified by requirement analysis. The platform consists of four layers: data model layer to manage relevant data; library layer and common function layer to configure services; and application layer to install and run the software. Finally, this study evaluates the validity of the proposed platform architecture by applying it to the digital catalog system.

PURPOSE. This study was to evaluate the marginal fit of two CAD-CAM anatomic contour zirconia crown systems compared to lithium disilicate glass-ceramic crowns. MATERIALS AND METHODS. Shoulder and deep chamfer margin were formed on each acrylic resin tooth model of a maxillary first premolar. Two CAD-CAM systems (Prettau$^{(R)}$Zirconia and ZENOSTAR$^{(R)}$ZR translucent) and lithium disilicate glass ceramic (IPS e.max$^{(R)}$press) crowns were made (n=16). Each crown was bonded to stone dies with resin cement (Rely X Unicem). Marginal gap and absolute marginal discrepancy of crowns were measured using a light microscope equipped with a digital camera (Leica DFC295) magnified by a factor of 100. Two-way analysis of variance (ANOVA) and post-hoc Tukey's HSD test were conducted to analyze the significance of crown marginal fit regarding the finish line configuration and the fabrication system. RESULTS. The mean marginal gap of lithium disilicate glass ceramic crowns (IPS e.max$^{(R)}$press) was significantly lower than that of the CAD-CAM anatomic contour zirconia crown system (Prettau$^{(R)}$Zirconia) (P<.05). Both fabrication systems and finish line configurations significantly influenced the absolute marginal discrepancy (P<.05). CONCLUSION. The lithium disilicate glass ceramic crown (IPS e.max$^{(R)}$press) had significantly smaller marginal gap than the CAD-CAM anatomic contour zirconia crown system (Prettau$^{(R)}$Zirconia). In terms of absolute marginal discrepancy, the CAD-CAM anatomic contour zirconia crown system (ZENOSTAR$^{(R)}$ZR translucent) had under-extended margin, whereas the CAD-CAM anatomic contour zirconia crown system (Prettau$^{(R)}$Zirconia) and lithium disilicate glass ceramic crowns (IPS e.max$^{(R)}$press) had overextended margins.
