Purpose: The triglyceride-to-high-density lipoprotein-cholesterol (TG/HDL-C) ratio is one of the main predictive indices for cardiovascular disease. This study was examined the relationship between TG/HDL-C ratio and metabolic syndrome (MetS) in male office workers. Methods: Secondary analysis was conducted to determine the risk between the TG/HDL-C ratio and MetS in male office workers. A total of 765 people underwent the 'regular workplace health checkups in 2014'. Among the subjects who were male and responded to the questionnaire and health lifestyle survey, 470 (61.4%) excluding those with missing and/or abnormal values were analyzed. The association between MetS, MetS components, and the TG/HDL-C ratio was examined by a Chi-square test, One-way ANOVA, Turkey post-hoc test and Logistic regression analysis. Results: The number of males with MetS was 70 (14.9%) and the number of MetS components increased with increasing TG/HDL-C ratio (p<.001). Logistic regression analysis with an adjustment for potential confounders revealed a 31.8 times higher odds ratio of the Quartile4 group for MetS than that of the Quartile1 group (p<.001). Conclusion: These results show that the likelihood of MetS, particularly the risk of MetS in the Quartile4, increases with increasing TG/HDL-C ratio.
BACKGROUND/OBJECTIVES: Several previous studies have investigated whether regular walnut consumption positively changes heart-health-related parameters. The aim of this study was to investigate the effects of daily walnut intake on metabolic syndrome (MetS) status and other metabolic parameters among subjects with MetS. SUBJECTS/METHODS: This study was a two-arm, randomized, controlled crossover study with 16 weeks of each intervention (45 g of walnuts or iso-caloric white bread) with a 6 week washout period between interventions. Korean adults with MetS (n = 119) were randomly assigned to one of two sequences; 84 subjects completed the trial. At each clinic visit (at 0, 16, 22, and 38 weeks), MetS components, metabolic parameters including lipid profile, hemoglobin A1c (HbA1c), adiponectin, leptin, and apolipoprotein B, as well as anthropometric and bioimpedance data were obtained. RESULTS: Daily walnut consumption for 16 weeks improved MetS status, resulting in 28.6%-52.8% reversion rates for individual MetS components and 51.2% of participants with MetS at baseline reverted to a normal status after the walnut intervention. Significant improvements after walnut intake, compared to control intervention, in high-density lipoprotein cholesterol (HDL-C) (P = 0.028), fasting glucose (P = 0.013), HbA1c (P = 0.021), and adiponectin (P = 0.019) were observed after adjustment for gender, age, body mass index, and sequence using a linear mixed model. CONCLUSION: A dietary supplement of 45 g of walnuts for 16 weeks favorably changed MetS status by increasing the concentration of HDL-C and decreasing fasting glucose level. Furthermore, consuming walnuts on a daily basis changed HbA1c and circulating adiponectin levels among the subjects with MetS. This trial is registered at ClinicalTrials.gov as NCT03267901.
새로운 NK-2 antagonist이며 고리상 펩티드인 cycIo-($Gln^1-Trp^2-Phe^3-{\beta}Ala^4-Leu^5-Met^6$)의 DMSO 용액 중에서의 형태를 2차원 핵자기공명과 분자동력학적인 계산에 의하여 결정하였다. 조건을 만족시키는 25개의 구조는 모든 아미노산 잔기의 골격원자들 (N,$C^{\alpha}$, C')의 자승 평균 평방근이 $0.02{\AA}$이내로 수렴하였다. 이 고리상 펩티드의 구조는 $Met^6NH$와 $Ala^4CO$, $Ala^4NH$와 $Met^6CO$, $Phe^3NH$와 $Met^6CO$의 사이에 분자내 수소결합이, Gln과 Trp에 type-I ${\beta}$-turn, 그리고 Leu에서 ${\gamma}$-turn을 취하고 있었다. 알려져 있는 고리상 펩티드인 cyclo-($Gln^1-Trp^2-Phe^3-Gly^4-Leu^5-Met^6$)의 Gly이 ${\beta}$Ala으로 바뀜에 따라 ${\beta}$Ala의 여분의 메틸렌이 고리의 골격의 반발력을 완화시키고 수소결합들이 형태의 안정에 기여하고 있다고 생각된다.
Objective: To evaluate whether the combination of magnetic resonance spectroscopy (MRS) and 11C-methionine positron emission tomography (11C-MET PET) could increase accurate diagnostic sensitivity for non-enhancing supratentorial gliomas. Materials and Methods: Between February 2012 and December 2017, 109 patients with non-enhanced supratentorial lesions on contrast-enhanced MRI were enrolled. Each patient underwent MRS and 11C-MET PET before treatment. A lesion was considered to be a glioma when either the MRS or 11C-MET PET results reached the diagnostic threshold. The radiological diagnosis was compared with the pathological diagnosis or medical diagnostic criteria. Results: The sensitivity and specificity were 60.0% and 50.0% for MRS and 75.8% and 50.0% for 11C-MET PET, respectively. Upon combining the two modalities, the sensitivity and specificity of the imaging-based diagnosis prior to surgery reached 89.5% and 42.9%, respectively. Statistically significant differences in the sensitivities were observed between the combined and individual approaches (MRS alone, 89.5% vs. 60.0%, p < 0.001; 11C-MET PET alone, 89.5% vs. 75.8%, p = 0.001). However, no significant differences in specificity were observed between the combined and individual modalities. Conclusion: The combination of MRS and 11C-MET PET findings significantly increases accurate diagnostic sensitivity for non-enhancing supratentorial gliomas without significantly lowering the specificity. This finding suggests the potential of the combined MRS and 11C-MET PET approach in clinical applications.
Purpose: E-cadherin is an adhesion molecule essential for tight connection between cells, forming the cadherin/catenin complex. Truncated $\beta$-catenin disrupts the interaction between E-cadherin and $\alpha$-catenin, leading to the loss of intercellular adhesion. Met protein, the hepatocyte growth factor receptor, plays important roles in signal transduction. We investigated the relationships between the expressions of E-cadherin, $\beta$-catenin, and c-met protein and the clinicopathological and prognostic parameters in gastric adenocarcinomas. Materials and Methods: The patterns of E-cadherin, $\beta$-catenin, and c-met protein expression were studied using immunohistochemistry in formalin-fixed, paraffin-embedded archival tissues from 76 surgically resected gastric adenocarcinomas. Results: Increased expressions of E-cadherin, $\beta$-catenin, and c-met were more significantly correlated in early gastric cancers (EGC) than in advanced gastric cancers (AGC) (P=0.002, P=0.003 and P=0.026). The positive immunoreactivities of all three markers were markedly lower in signet ring-cell type and poorly differentiated type lesions than in intestinal-type lesions. Decreased expression of the $\beta$-catenin protein correlated well with increased tumor invasion depth (P=0.039), and increased lymph node metastasis correlated well with reduced expression of c-met (P=0.046). Conclusion: In gastric cancers, reduced expressions of the E-cadherin, $\beta$-catenin, and c-met proteins may play some role in poorer tumor differentiation, deeper tumor invasion, and increased lymph node metastasis. Also, the c-met gene is thought to play a specific role in the mechanism of the yet unknown catenin action.
Corynebacterium glutamicum에서 promoter-probe vector인 pSK1Cat을 이용해 분리된 프로모터를 함유하는 단편들 중 가장 높은 활성을 나타낸 $P_{19}$ 단편에 대한 심도 있는 분석을 수행하였다. Subcloning을 실시하여 프로모터 활성을 지닌 DNA 영역을 180 bp로 압축할 수 있었고 $(P_{180})$, 이를 C. glutamicum의 균주개량 측면에서 그 활용성을 분석하였다. C. glutamicum에서 메치오닌 생합성에 관여하는metX유전자의 메치오닌에 의한 repression을 해제시키기 위하여 metX유전자의 promoter를 $P_{180}$ promoter로 교체하였고 $(P_{180}-metX)$, $P_{180}-metX$를 C. glutamicum에 도입하여 발현되는 homoserine acetyltransferase 활성을 다양한 성장조건에서 측정하였다. MB 영양배지에서 배양하는 경 우 $P_{180}-metX$를 함유는 균주는 wild type보다 약 24배 높은 homoserine acetyltransferase 활성을 나타내었다. Tac 프로모터에 연계하는 경우 $(P_{tac}-metX)$, 약 13배의 활성 증가만이 관찰되었다. 최소배지에서 배양한 후 분석한 결과, $P_{180}-metX$에서의 발현양상은 배지에 첨가된 methionine에 의해 영향받지 앓음을 확인하였는데, 이는 $P_{180}$ 단편이 생합성 유전자의 derepression에 의한 아미노산 생산균의 개량에 효율적으로 이용될 수 있음을 의미한다. $P_{180}-metA$를 라이신 생산균에 도입하는 경우 최대 약 0.8g/l의 메치오닌이 생산됨을 확인하였다.
The Arabidopsis thaliana S-Adenosylmethionine decarboxylase (AdoMetDC) cDNA ($GenBank^{TM}$ U63633) was cloned. Site-specific mutagenesis was performed to introduce mutations at the conserved cysteine $Cys^{50}$, $Cys^{83}$, and $Cys^{230}$, and $lys^{81}$ residues. In accordance with the human AdoMetDC, the C50A and C230A mutagenesis had minimal effect on catalytic activity, which was further supported by DTNB-mediated inactivation and reactivation. However, unlike the human AdoMetDC, the $Cys^{50}$ and $Cys^{230}$ mutants were much more thermally unstable than the wild type and other mutant AdoMetDC, suggesting the structural significance of cysteines. Furthermore, according to a circular dichroism spectrum analysis, the $Cys^{50}$ and $Cys^{230}$ mutants show a higher a-helix content and lower coiled-coil content when compared to that of wild type and the other mutant AdoMetDC. Also, the three-dimensional structure of Arabidopsis thaliana AdoMetDC could further support all of the data presented here. Summarily, we suggest that the $Cys^{50}$ and $Cys^{230}$ residues are structurally important.
This study was performed to investigate the characteristics of nutrient intake according to metabolic syndrome in Korean elderly. Data on 1,200 elderly (men 545, women 655) over 65 years was obtained from the Korea National Health and Nutrition Examination Survey 2010. The subjects were divided into metabolic syndrome (MetS) and normal groups based on NCEP-ATP III guidelines and by applying the Asia-Pacific definition with waist circumference. The prevalence of MetS was 48.9% in all subjects, 35.6% in men and 58.9% in women, respectively. All nutrient intake was significantly lower in MetS than normal subjects, except iron, vitamin A, carotene, and retinol. In the carbohydrate: protein: fat (C:P:F) ratio, MetS showed a higher rate for carbohydrate, but a lower rate for protein and fat than normal. MetS was lower in calcium, vitamin B1, and vitamin C by the nutrient adequacy ratio than normal. Mean adequacy ratio of MetS (0.69) was lower than that of the normal group (0.72). These results demonstrated that undernutrition, not excess intake of specific nutrients - energy, fat, or cholesterol was associated with the risk for MetS in Korean elderly with MetS. Therefore, it is recommended to have a healthy dietary pattern with diverse nutrients for the prevention of MetS.
Glucose ($C_6H_{12}O_6$)의 이론적인 최대 메탄수율은 표준상태(1 atm, $0^{\circ}C$)를 기준으로 0.35 L $CH_4/g$ COD이지만, 전통적인 혐기성소화조에서 유기물이 메탄으로 전환되는 양은 연구의 방법이나 유기물의 종류에 따라 매우 다양하게 보고되고 있으며, 대부분의 연구실 규모 실험에서 안정화 후 메탄 수율은 0.35 L $CH_4/g$ COD 이하로 나타난다. 최근, 미생물 전기화학 기술(Microbial Electrochemical Technology, MET)은 지속가능한 신재생에너지 생산 기술로서 큰 주목을 받고 있으며, MET를 혐기성소화조에 적용할 경우 고농도의 유기성폐기물의 빠른 분해가 가능할 뿐만 아니라 전기화학적인 반응에 의해 휘발성지방산(VFAs)이나 독성물질, 생분해 불가능한 물질까지도 분해가 가능하며, 소화조 내 미생물의 활성을 높이고 바이오가스의 생산량을 극대화 할 수 있다고 알려져 있다. 본 연구에서는 MET가 혐기성소화의 메탄발생에 미치는 영향에 대하여 연구하기 위해 음식물 탈리액과 하수슬러지의 원소조성에 따른 이론적인 최대 메탄수율을 분석하였으며, BMP (Biochemical Methane Potential) 실험과 연속식 실험을 통한 메탄수율의 특성을 평가하였다. 그 결과, MET가 적용된 혐기성소화에서의 메탄수율은 일반적인 혐기성소화조에 비하여 기질에 따라 2-3배 정도 높았으며, 이론적인 최대 메탄수율에 미치지는 못하였으나 일부는 거의 근접한 결과가 도출되었다. 또한, 일반적인 혐기성소화조와 MET가 적용된 혐기성소화조의 안정화 후 바이오가스의 조성은 거의 유사하게 나타났다. 결과적으로, MET가 혐기성소화조의 유기물 제거효율을 향상시켜 메탄발생량을 증가시킨 것으로 나타났으며, 향후 추가적인 연구를 통하여 MET에서 메탄발생 메카니즘이 명확히 규명되어야 할 것이다.
