Since the epigenetic alteration in tumor cells can be reversed by the dietary polyphenol quercetin (Q) or butyrate (B) with chemopreventive activity, suggesting that Q or B can be used for chemopreventive as well as therapeutic agent against tumors. In this study the polyphenol flavonoid quercetin (Q) or sodium butyrate (B) suppressed human esophageal 9706 cancer cell growth in dose dependent manner, and Q combined with B (Q+B) could further inhibit Eca9706 cell proliferation than that induced by Q or B alone, compared with untreated control group (C) in MTT assay. The reverse expressions of global DNMT1, $NF-{\kappa}Bp65$, HDAC1 and Cyclin D1 were down-regulated, while expressions of caspase-3 and $p16INK4{\alpha}$ were up-regulated, compared with the C group in immunoblotting; the down-regulated HDAC1-IR (-immunoreactivity) with nuclear translocation, and up-regulated E-cadherin-IR demonstrated in immunocytochemistry treated by Q or B, and Q+B also displayed further negatively and positively modulated effects compared with C group. The order of methylation specific (MS) PCR of $p16INK4{\alpha}$: C>B/Q>Q+B group, while the order of E-cadherin expression level was contrary, Q+B>Q/B>C group. Thus, Q/B, especially Q+B display reverse effect targeting both altered DNA methylation and histone acetylation, acting as histone deacetylase inhibitor mediated via epigenetic-$NF-{\kappa}B$ cascade signaling.
For the ultimate goal of efficient retrovirus vector-mediated transgenic animal production, we tried to increase virus titer by employing three methods: boosting virus production by treating virus-producing cells with sodium butyrate, concentration of virus stock by either filtration or ultracentrifugation. Compared to the control, applications of sodium butyrate (5 mM) treatment and filtration resulted in only 3 and 3. 6 folds of titer increases on bovine EBTr target cells, respectively. However, concentration of virus-containing medium by ultracentrifugation showed 12.5 folds of titer increase compared to the control (10${\times}$10$^5$ LacZ$^+$ TU Im), indicating the best method which can enhance retrovirus vector-mediated transgenic animal production.

Sodium butyrate (NaBu) is used as an enhancer for the production of recombinant proteins in Chinese hamster ovary (CHO) cells. However, NaBu is well-known for its cytotoxic effect, thereby inducing apoptosis. CHO cells which had been engineered to express a humanised anti-HBV antibody were cultured using serum-free medium, Ex-cell 301. From a seeding density of $2{\times}10^5$ cells/ml, CHO cells grown with serum-free medium reached a maximum cell density of $1.3{\times}10^6$ cells/ml after 9 days in culture and produced a maximal antibody concentration of 130 mg/l after 13 days in culture. In the perfusion culture system, CHO cells producing anti-HBV antibody grown in an 7.5 1 bioreactor seeded with $2{\times}10^5$ cells/ml reached a maximal antibody concentration of 85 mg/1 after 720 h in culture. The addition of 0.3 mM NaBu and lowering culture temperature to $33^{\circ}C$ elongated the culture period to 60 days and increased the production yield by 2-fold, compared to control culture.
To investigate the effect of nitrogen fertilization on the quality of tropical grass silage, guinea grass grown with 3 types of nitrogen fertilizers, namely, urea, ammonium sulfate, and compound fertilizer 804, at 2 fertilization levels, 0.5 and 2.5 kg $Na^{-1}$ (0.5 N and 2.5 N, respectively), was subjected to silage fermentation. Silage fertilized with 0.5 N showed butyrate-dominant fermentation, irrespective of the type of fertilizer used. On the other hand, fermentation of silage fertilized with 2.5 N was significantly affected by the type of fertilizer used; fertilization with ammonium sulfate and compound fertilizer 804 resulted in silage that contained a large amount of butyrate and no lactate; this silage was considered to be of a significantly low quality as compared with silage fertilized with 0.5 N. Among silage fertilized with 2.5 N, the desirable butyrate-free fermentation was found only in urea-fertilized silage, which had the best quality. Grass material fertilized with a high level of urea accumulated a relatively high concentration of nitrate nitrogen (0.22% dry matter). Our results presented here suggest that nitrogen fertilizer management could affect the quality of tropical grass silage and that a relatively high concentration of nitrate in silage may promote butyrate-free fermentation even in tropical grass silage.
EoL-1 cells differentiate into eosinophils in the presence of n-butyrate, but the mechanism has remained to be elucidated. Because n-butyrate can inhibit histone deacetylases, we hypothesized that the inhibition of histone deacetylases induces the differentiation of EoL-1 cells into eosinophils. In this study, using n-butyrate and two other histone deacetylase inhibitors, apicidin and trichostatin A, we have analyzed the relationship between the inhibition of histone deacetylases and the differentiation into eosinophils in EoL-1 cells. It was demonstrated that apicidin and n-butyrate induced a continuous acetylation of histones H4 and H3, inhibited the proliferation of EoL-1 cells, and induced the expression of markers for mature eosinophils such as integrin ${\beta}7$, CCR1, and CCR3 on EoL-1 cells, while trichostatin A evoked a transient acetylation of his tones and induced no differentiation into eosinophils. These findings suggest that the continuous inhibition of histone deacetylases in EoL-1 cells induces the differentiation into mature eosinophils.
Hydrogen evolution from biomass-derived carbohydrates by some Clostridia and optimal culture conditions for hydrogen evolution were investigated. Among the organisms tested, Clostridium butyricum was efficient hydrogen producer with starch, xylan, pectin, cellobiose and xylose. In batch fermentation of Cl. butyricum, optimal conditions for hydrogen evolution were achieved at pH 7.0-8.5, 10-50 mM phosphate, and 2% (w/v) glucose. Total amount of molecular hydrogen evolved by the organism slightly increased at the presence of acetate (<150 mM) or butyrate (<20 mM) in the initial fermentation medium. Especially, in case of more than the above concentration of butyrate, growth and hydrogen evolution were dramatically inhibited. In the conditions were described here, 70 mmole of molecular hydrogen per mg of DCW was produced with 1%(w/v) glucose by the organism.
BACKGROUND/OBJECTIVES: Hyperglycemia is a major cause of diabetes and diabetesrelated diseases. Sodium butyrate (NaB) is a short-chain fatty acid derivative that produces dietary fiber by anaerobic bacterial fermentation in the large intestine and occurs in foods, such as Parmesan cheese and butter. Butyrate has been shown to prevent obesity, improve insulin sensitivity, and ameliorate dyslipidemia in diet-induced obese mice. Therefore, this study examined the effects and mechanism of NaB on the secretion of inflammatory cytokines induced by high glucose (HG) in THP-1 cells. MATERIALS/METHODS: THP-1 cells were used as an in vitro model for HG-induced inflammation. The cells were cultured under normal glycemic or hyperglycemic conditions with or without NaB (0-25 μM). Western blotting and quantitative polymerase chain reaction were used to evaluate the protein and mRNA levels of nuclear factor-κB (NF-κB), interleukin-6, tumor necrosis factor-α, acetylated p65, acetyl CREB-binding protein/p300 (CBP/p300), and p300 using THP-1 cells. Histone acetyltransferase (HAT), histone deacetylase (HDAC), and pro-inflammatory cytokine secretion activity were analyzed using an enzyme-linked immunosorbent assay. RESULTS: HG significantly upregulated histone acetylation, acetylation levels of p300, NF-κB activation, and inflammatory cytokine release in THP-1 cells. Conversely, the NaB treatment reduced cytokine release and NF-κB activation in HG-treated cells. It also significantly reduced p65 acetylation, CBP/p300 HAT activity, and CBP/p300 gene expression. In addition, NaB decreased the interaction of p300 in acetylated NF-κB and TNF-α. CONCLUSIONS: These results suggest that NaB suppresses HG-induced inflammatory cytokine production through HAT/HDAC regulation in monocytes. NaB has the potential for preventing and treating diabetes and its related complications.
Background : The antitumor effects of herpes simplex virus thymidine kinase (HSV-tk) and ganciclovir (GCV) strategies for cancer gene therapy have a the following advantages : 1) a direct cytotoxicity to HSV-tk modified cancer cells by GCV 2) a cell death by the local transfer of toxic metabolites from the HSV-tk modified cells to nearby unmodified tumor cells (bystander effect), and 3) in vivo bystander effect such as antitumor-immunity. Retroviral and adenoviral sequences can silence transgene expression in cells and mice. In this study, we investigated the above described advantages of HSV-tk/GCV strategy in Lewis lung cell and mouse lung cancer model using retroviral vector and adenoviral vector. Also, we observed whether the expression of a silenced gene can be reactivated by treating cells with butyrate. Methods : Retrovirus-HSV-tk and adenovirus-HSV-tk vectors were used for the transduction of Lewis lung carcinoma (LLC) cells. The change of HSV-tk expression by butyrate was measured by Western blol The antitumor activities containing bystander effect were observed in vivo (by MTT assay) and in vivo tumor models of various combinations of LLC and LLC-tk. Results : 1. Butyrate induced the enhancement of HSV-tk expression from adenovirally transduced cells but not from retrovirally transduced cells. 2. Both retrovirus-HSV-tk and adenovirus-HSV-tk vectors with GCV treatment were effective for killing of tumor cell in vitro and suppression of LLC tumorigenicity. Bystander effect was responsible for killing of mixture of LLC-tk and LLC in vitro and in vivo-tumorigenicity model. Conclusion : Butyrate could augment adenovirus-mediated HSV -tk gene expression. Cancer gene therapy with HSV-tk suicide gene by retroviral and adenoviral vector seems to be an effective approach for lung cancer therapy.
The immobilization of enzyme is one of the key issues both in the field of enzymatic research and industrialization. In this work, we reported a facile method to immobilize Candida Antarctica lipase B (CALB) in alginate carrier. In the presence of calcium cation, the enzyme-alginate suspension could be cross-linked to form beads with porous structure at room temperature, and the enzyme CALB was dispersed in the beads. Activity of the enzyme-alginate composite was verified by enzymatic hydrolysis reaction of p-nitrophenol butyrate in aqueous phase. The effects of reaction parameters such as temperature, pH, embedding and lyophilized time on the reactive behavior were discussed. Reuse cycle experiments for the hydrolysis of p-nitrophenol butyrate demonstrated that activity of the enzyme-alginate composite was maintained without marked deactivation up to 6 repeated cycles.
Hydrogen evolution by mixed fermentation of Clostn"dium butyn"cum and photosynthetic bacteria which were capable of consuming clostridial metabolites and evolving hydrogen was investigated. Acetate and butyrate formed from anaerobic clostridial fermentation were efficiently utilized by Rhodopseudomonas sPhaeroides K-7. For complete bioconversion of clostridial metabolites such as acetate and butyrate into hydrogen, mixed culture of both anaerobic organisms forming molecular hydrogen was performed. By the mixed culture, the yield of hydrogen production increased by 20 to 75% and the levels of clostridial metabolites such as acetate, butyrate decreased in the fermentation broth. Influence of cell mixing ratio. mixing time and inoculum level on hydrogen evolution by mixed culture were examined. And then cometabolic pattern compared with in pure culture was observed as time course.
