Dietary Fiber Modulates Colon Cell Proliferation by Altering Luminal Concentrations of Short-Chain Fatty Acids

  • To compare the effects of various types of dietary fiber on microbial production of short-chain fatty acids (SCFA) and on colon cell proliferation which is used as an intermediate biomarker for colon carcinogenesis, groups of 10 male Sprague-Dawley rats were fed one of four fiber-supplemented diets (6% cellulose, 6% pectin, 6% polydextrose, and a mixture of 3% cellulose and 3% polydextrose) for three weeks. As a control, a fiber-free diet was fed to a separate group of 10 rats. Cell proliferation was measured by in vivo incorporation of bromodeoxyuridine into DNA in the proximal and distal colon, respectively. Luminal concentrations of SCFA were measured by gas chromatography. Dietary fiber significantly influenced microbial production of SCFA in the colon; pectin supplementation produced the highest concentrations of luminal SCFA in both the proximal and distal colon (p<0.05). The degree of individual SCFA production was characterized by a relatively higher increase in butyrate production by the pectin-supplemented diet, and in propionate production by the polydextrose-supplemented diet, resulting in alterations of the molar ratios of acetate, propionate and butyrate. There were significant differences in colon cell proliferation among the diet groups; the pectin-supplemented diet produced a significantly higher effect on cell proliferation of distal colonic epithelial cells (p<0.05), and the polydextrose-supplemented diet produced an intermediate effect compared to the fiber-free or cellulose-supplemented diet. Increased cell proliferation was correlated to increased luminal concentrations of butyrate in the proximal colon and to increased luminal concentrations of propionate and butyrate in the distal colon (p<0.05). Therefore, these data suggest that dietary fiber may modulate colon cell proliferation by altering luminal SCFA concentrations, particularly butyrate and perhaps propionate. In addition, the present study is the first finding that has demonstrated a relative increase in colon cell proliferation due to supplementation with polydextrose, suggesting that the overuse of this artificially synthesized polysaccharide in food processing technology needs to be carefully evaluated from the public health point of view.

  • Sodium butyrate (NaBt)는 장에서 탄수화물대사로부터 생겨나는 짧은 천연지방산 사슬로 다양한 인간 암세포들 에게서 강력한 항암효능을 나타냄이 보고된 바 있지만 자세한 기전은 아직 알려져 있지 않다. 이 논문에서 우리는 NaBt가 주요 세포부착분자이면서 종양억제인자의 일종인 E-cadherin의 발현을 세포-특이적으로 촉진하는 기전을 연구하였다. 또한 NaBt는 E-eadherin의 발현을 촉진하는 것으로 알려진 p21의 발현도 증가시켰지만, NaBt에 의하여 증가한 p21은 E-cadherin의 활성화와 관련이 없음이 밝혀졌다. 그 대신에 NaBt는 CCAAT-box를 통한 E-cadherin 유전자의 프로모터 활성을 증가시킴으로써 E-cadherin의 발현을 전사수준에서 촉진하는 것 같다. 이렇게 NaBt에 의하여 증가된 E-cadherin은 주로 세포간 접촉면에 위치하면서 Hep3B 세포를 더 분화된 형태로 유도하여 NaBt의 항암활성이 나타나는 것 같다.

잠재성 Herpes Simplex Virus의 재활성화에 대한 n-Butyrate의 효과 (Effect of n-Butyrate on the In Vitro Reactivation of Latent Herpes Simplex Virus)

  • 4-carbon fatty acid인 n-Butyrate(n-BTA)는 herpes virus의 일종인 Epstein-Bat virus(EBV)에 작용해서 잠복형인 EBV를 활성형태로 유도시키는 것으로 알려져 있다. 고로 본 실험에서는 mouse의 삼차신경절에 잠복하고 있는 HSV의 재활성화에 대한 n-BTA의 효과를 실험 관찰하였다. Pentobarbital로 마취시킨 mouse의 양쪽눈 각막을 30 gauge 주사바늘로 scarify한 후에 type I HSV(HSV-1) $10{\mu}1(1{\times}10^5$ plaque-forming units)를 각각 점안 감염시켰다. virus를 감염시킨 4주 후에 mouse의 삼차신경절을 적출하여 시험관 내에서 조직배양을 시행하였다. 조직배양시에 0. 1, 0. 25, 0.5. 1.0 그리고 2.0mM농도의 n-BTA를 첨가하였으며 1일, 2일, 3일간 각각 배양한 후 신경절을 연마하여 연마액내의 HSV-1 titer를Vero cell monolayer에서 plaque assay로 측정하였다. 1) n-BTA첨가군은 잠재성 HSV가 대조군에 비하여 현저하게 빨리 재활성화 되었고 재활성화되는 virus의 양도 현저히 증가되었다. 2) 24시간을 계속해서 n-BTA 각 농도를 첨가해서 배양할 군은 n-BTA 6시간 첨가 배양하고 새로운 배양액으로 갈아서 18시간 배양한 군에 비해 잠재성 virus의 재활성화가 현저히 증가되었다. 3) Gang1ionic latent HSV-1의 재 활성화에 영향을 미치는 각 농도의 n-BTA는 Vero cell의 monolayer에서의 HSV-1의 번식에는 아무런 영향을 미치지 않았다.

셀룰로오스를 이용한 전도성 체이스트의 개발 (Novel Conductive Paste based on Cellulose Acetate Butyrate)

  • 고분자를 바탕으로 한 전도성 페이스트는 전원과 기기를 연결하는 전도 통로를 제공하여 준다. 이러한 전도성 페이스트는 고분자 바인더와 전도성 금속의 두 부분으로 이루어져 있다. 본 연구에서는 낮은 점도와 우수한 금속 피막성을 가지는 셀룰로오스 아세테이트 부타이레이트를 바인더로 사용하여 새로운 전도성 페이스트를 개발하였다. 전도도 측정 결과 안정된 전도도를 보였을 뿐 아니라, 균일한 코팅성과 유연성을 나타내었다.

Correlation Between Enhancing Effect of Sodium Butyrate on Specific Productivity and mRNA Transcription Level in Recombinant Chinese Hamster Ovary Cells Producing Antibody

  • Sodium butyrate (NaBu) has been used to enhance protein expression levels in mammalian cell culture. To determine the clonal variability of recombinant Chinese hamster ovary (rCHO) cells in response to NaBu addition regarding specific antibody productivity $(q_{Ab})$, three rCHO clones were subjected to different concentrations of NaBu. For all three clones, NaBu addition inhibited cell growth and decreased cell viability in a dose-dependent manner. On the other hand, the enhancing effect of NaBu on $q_{Ab}$ varied significantly among the clones. NaBu addition enhanced the antibody production of only one clone. RT-PCR analysis revealed that the changes in $q_{Ab}$ correlated linearly with those of the mRNA transcription level. Thus, it was concluded that the different enhancing effects of NaBu on protein expression in rCHO cell clones resulted from their different mRNA transcription levels.

In Vitro Effects of Dietary Inulin on Human Fecal Microbiota and Butyrate Production

  • Administration of dietary fibers has various health benefits, mainly by increasing numbers of beneficial bacteria and enhancing production of short-chain fatty acids in the colon. There has been growing interest in the addition of dietary fiber to human diet, due to its prebiotic effects. This study aimed to evaluate the prebiotic activity of inulin using an in vitro batch fermentation system with human fecal microbiota. Fermentation of inulin resulted in a significantly greater ratio of Lactobacillus or Bifidobacteria to Enterobacteria strains as an index of healthy human intestine and elevated butyrate concentration, which are related to improvement of gut health.

산성 발효 산물과 배양 pH가 Clostridium acetobutylicum의 유지 에너지에 미치는 영향 (Effects of Acidic Fermentation Products and Culture pH on the Maintenance Energy of Clostridium acetobutylicum)

  • In order to elucidate the acid tolerance mechanism of Clostridium acetobutylicum against organic acid, the maintenance energy with added butyrate at different pH was determined. Maintenance coeffecient in acidogenic chemostat was higher at pH 6.5 than at pH 5.5 showing that this organism is an acidophile. The addition of butyrate at pH 5.5 and different dilution rate caused linear decrease of the cell concentration though $Y_{ATP}$ did not decrease with increasing undissociated organic acid. $Y_{ATP}$ decreased by increasing the concentration of undissociated organic acid at pH 5.0 by the addition of butyrate. From these results it is hypothesized that the ATP conaumption for pH stat of acidophile C. acetobutylicum is increased at the circumstance with over 30mM of undissociated organic acid.cid.

Anticancer Effects of Gut Microbiota-Derived Short-Chain Fatty Acids in Cancers

  • Short-chain fatty acids (SCFAs), such as butyrate, propionate, and acetate produced by the gut microbiota have been implicated in physiological responses (defense mechanisms, immune responses, and cell metabolism) in the human body. In several types of cancers, SCFAs, especially butyrate, suppress tumor growth and cancer cell metastasis via the regulation of the cell cycle, autophagy, cancer-related signaling pathways, and cancer cell metabolism. In addition, combination treatment with SCFAs and anticancer drugs exhibits synergistic effects, increasing anticancer treatment efficiency and attenuating anticancer drug resistance. Therefore, in this review, we point out the importance of SCFAs and the mechanisms underlying their effects in cancer treatment and suggest using SCFA-producing microbes and SCFAs to increase therapeutic efficacy in several types of cancers.

철 농도 변화에 대한 수소발효의 반응 (Reponse of Hydrogen Fermentation to Variation of Iron)

  • 철 농도변화에 따른 대사산물 생성에 대한 반응은 중온조건에서 회분식 실험으로 수해하였다. 철의 농도가 40mg/$\ell$까지는 철 농도의 증가에 따라 수소생성율도 증가하였다. acetate 생성율은 철의 농도가 증가함에 따라 감소하였다. Butyrate, ethanol 및 butanol은 각각 철 농도가 200,200 및 400mg/$\ell$에서 최대 생성율을 나타내었다. 철의 농도가 200mg/$\ell$이상의 농도엘 때에 propionate, i-butyrate 및 valerate 생성율이 철의 농도가 200mg/$\ell$ 이하의 경우 보다 낮았다. 미생물농도의 경우 철 농도가 300mg/$\ell$까지는 농도가 증가함에 따라 미생물농도도 증가하였다.

Butibufen의 합성 (Synthesis of Butibufen)

  • A new method for the synthesis of butibufen, which is a non steroidal anti-iriflammatory agent, is described. Friedel-Crafts reaction of isobutylbenzene with ethyl $\alpha$-chloro-.alpha.-(methylthio) acetate (1) gives ethyl $\alpha$-methylthio-(p-isobutylphenyl)acetate (2). Ethyl 2-methylthio-2-(4-isobutylphenyl)butyrate (3) is obtained from treatment of the compound (2) with NaH and Etl. Butibufen (5) is synthesized by reductive desulfurization of the compound (3) with zinc dust-acetic acid or Raney nickel, followed by hydrolysis of the resultant ethyl 2-(4-isobutylphenyl)butyrate (4).
