Improved Method for the Preparation of Crude Ginseng Saponin (인삼 조사포닌의 조제 방법 개선)

  • This stuffy was carried to establish a new efficient method for the preparation of edible crude ginseng saponin. The conventional butanol extraction and resin adsorption methods were compared for the contents of total crude ginseng saponin and major ginsenosides. Seventy- percent methanol extract was applied to Diaion HP-20 column and the resin was washed with Hn and eluted with absolute methanol. The methanol elute was dried in vivo and analyzed for its ginsenosides. Use of ethanol instead of methanol to make edible crude ginseng saponin gave a similar result. Butanol extraction was performed by the conventional method. The final aqueous layer from butanol extraction was passed through Diaion HP-20 column followed by elution with methanol and Diaion HP-20 passed fraction was extracted with butanol to recover remaining components, respectively, in order to determine saponin loss. TLC and HPLC qualitatively and quantitatively monitored Ginsenosides, respectively. Loss of ginsenosides was higher in butanol extraction method than in Diction HP-20 adsorption method. In addition, saponin fractions prepared by Diction HP-20 adsorption method showed higher content of each ginsenoside, showing 8.2% higher purity than that of butanol extracted fraction. From these results, we propose the resin adsorption method as a new efficient measure for the preparation of crude ginseng saponin, which is edible by using spirit instead of methanol.

Effect of Injin Butanol Fraction with Thin Layer Chromatography on Fas-mediated Apoptosis (인진butanol 분획의 TLC추출성분이 Fas-mediated Apoptosis에 미치는 영향)

  • Objective: The aim of this study is to investigate the effect of Injin butanol fractions with Thin Layer Chromatography on Fas-mediated Apoptosis. Method: Injin-butanol fraction separated by TLC. MIT assay, cell cycle analysis, Caspase-3 protease assay, DNA fragmentation assay and quantitative RT-PCR were performed to evaluate the effects of TLC extraction of lnjin-butanol fraction on cell viability, cell cycle progression and apoptosis. Results: Scopoletin, luteolin, apigenin and unknown powder was isolated by TLC. Fas-mediated apoptosis analysis shows that scopoletin has inhibiting function on apoptosis. Caspase- 3 protease assay analysis shows that scopoletin inhibits activity of caspase-3. Quantitative RT-PCR analysis shows that no activity on caspase-3, but apoptosis inhibition cytokine -Bcl-2- is activated, and apoptosis activating cytokine -Bax- is unactivated. Conclusion: These results show that each fraction of Injin-butanol TLC extraction, especially scopoletin, acts as a protective function on liver cell viability, and inhibitory function on apoptosis.   • Antimicrobial activities, extraction yields and chemical compositions of the extracts from the powder of vacuum dried Houttuynia cordata root on various solvents were investigated. Antimicrobial activity was investigated by disc diffusion method against several microorganisms, nine species of Gram positive and thirteen Gram negative bacteria. Ethanol, ethylacetate and butanol extracts exhibited potent antimicrobial activities against B. megaterium C. xerosis, S. aureus, S. epidermidis and E.coli. Chloroform and aqueous extracts from H. cordata showed weak antimicrobial activity against the tested bacteria. Ethylacetate and butanol extracts exhibited strong antimicrobial activity against the tested microorganisms. The extraction yields were 15.7%, 3.7%, 0.13%, 0.5% and 5.9% in ethanol, chloroform, ethylacetate, butanol and aqueous fractions, respectively. To further elucidate the effective components, solvent extracts was analyzed by GC/MSD.

Effect of Solvents of Extraction on the Biological Activities of Phyllostachys Nigra Munro (추출 용매에 따른 오죽(Phyllostachys nigra Munro) 추출물의 생리활성 효과)

  • In order to research the effect of solvents of extract on biological activities of Phyllostachys nigra Munro, antioxidative activity, superoxide dismutase (SOD)-like activity, nitrite scavenging activity, tyrosinase inhibitory activity, angiotensin-converting enzyme (ACE) inhibibitory activity in vitro were investigated. When ethyl acetate and hexane as solvents were used, the total phenols concentrations were 44.1 and 47.3 mg/kg, respectively, which was about 2.0 fold higher than that of water. The antioxidive activity was affected by solvents of extraction. The antioxidative activity was increased in order of hexane > ethyl acetate > n-butanol > methanol > water extraction. On the other hand, in the case of SOD-like activity, it was in the order of methanol > n-butanol > hexane > ethyl acetate extraction. The nitrite scavenging ability showed the most remarkable activity at pH 1.2 irrespective of solvents. Especially, when pH was increased from 1.2 to 6.0 using ethyl acetate extraction, it was decreased from 69.2 to 7.8%. The tyrosinase inhibitory activity was in the range of 15.2~21.3% and was increased in order of water > methanol > n-butanol>hexane > ethyl acetate extract. These results suggest that hexane and ethyl acetate extraction of Phyllostachys nigra Munro can be used in bioactive and functional materials.

Evaluation of Extractants for Bio-butanol Extraction Fermentation Using Organic Solvents and Ionic Liquids (유기용매와 이온성액체를 이용한 바이오 부탄올 추출발효 용매 선정 평가)

  • Oleyl alcohol, butyl butyrate, and two different ionic liquids were evaluated for the extraction of butanol from culture broth without toxic effect to cells. The tested solvents showed more than 50% extraction efficiency, and oleyl alcohol was chosen as the best extractant for butanol among the used extractants with a partition coefficient of 2.89. When oleyl alcohol was used as an extractant, more than 80% of butanol was extracted in the wide range of butanol concentrations (1-20 g/L) and pH values (pH 4-5.5). In extractive fermentation using oleyl alcohol only, there was 11% more butanol production and glucose consumption when compared to that without extractive fermentation, implicating a reduced inhibitory effect of butanol due to butanol removal to the oleyl alcohol phase. In addition, oleyl alcohol did not inhibit cell growth, while a mixture of oleyl alcohol and butyl butyrate with the volume ratio of 9:1~7:3 inhibited either butanol production or cell growth significantly due to the toxicity of butyl butyrate to cells. In conclusion, oleyl alcohol can be used as an efficient and non-toxic solvent for extractive fermentation for butanol production.

Antimicrobial Activity and Composition of Extract from Smilax china Root (청미래덩굴 뿌리에서 추출한 순차분획물의 항균활성과 성분 분석)

  • Antimicrobial activities, extraction yields and chemical compositions of the extract from the powder of vacuum dried Smilax china root on various solvents were investigated. Antimicrobial activities against. A rhizogenes, A. tumefaciens, C. utilis, S. cerevisiae, B. megaterium, B. subtilis and E. coli were measured by disc diffusion method. Methanol, chloroform, ethylacetate and butanol extracts exhibited potent antimicrobial activities against B. megaterium and B. subtilis. Also, methanol, ethylacetate and butanol extracts had the activities against A. rhizogenes, and chloroform and ethylacetate extracts against A. tumefacciens. The extraction yields were 13.10%, 8.71%, 2.73%, 0.83%, 0.54% and chemical compositions of their extracts were very different according to the kinds of solvents. The main chemical groups of ethylacetate, methanol, chloroform and butanol extracts were 45.99% phenolics, 36.10% acids, 29.13% phenolics and 18.96% phenolics, respectively.

Effect of Butanol Extracts from Prunus mume on the Growth of Salmonella typhimurium (오매의 Butanol추출물이 Salmonella typhimurium의 생육에 미치는 영향)

  • The antimicrobial properties of extracts of Prunus mume were tested on Salmonella paratyphimurium and S. typhimurium. First, the Prumus mume was extracted with methanol at several temperatures, and then fractionation of the methanol extracts from Prunus mume was carried out by using petroleum ether, chloroform, ethyl acetate or butanol. Secondly, absorption column chromatoraphy (using a Diaion HP 20) was conducted to eliminate some water soluble materials that might inhibit the antimicrobial activity of some extracts. The antimicrobial activitry of each of the Prunus mume extracts was determined using a paper disc method against several food-borne pathogens, The growth inhibition curve was determine using butanol extracts of Prunus mume against Salmonella typhimurium. The extraction temperature did not have any significant effect on the yield of the extract or on the level of antimicrobial activity. The butanol extract of Prunus mume showed strong antimicrobial activity against Salmonella paratyphimurium and S. typhimurium; a 1,000 ppm of butanol extract of Prunus mume retared the growth of S. typhimurium up to 36 hours. Effects of Physiological Active Substance Extracted from Silkworm Fece

  • Silkworm (Bombyx mori ) feces have long been used in the pharmaceutical and food industries as a natural colorant. However, there is limited data on the bioactive compounds that constitute silkworm feces. This research emphasizes the antioxidant activity of different solvent and flavonoid extracts of silkworm feces. The solvents were ethanol, butanol, and water, while the methods utilized included ultrasonification, stirrer, reflux, and reflux after ultrasonification extraction. Results showed that butanol ultrasonification extraction (BUE) yield the lowest extraction (1.75%), while the other methods yielded 7 to 14%. The total polyphenol content utilizing BUE was 3.3 mg TAE/g, while water ultrasonification extraction (WUE) yielded the highest extraction rate with 51.6 mg TAE/g. The total flavonoid content was significantly higher using ethanol reflux extraction (EUE) at 266.8 mg QRE/g BUE, which was 158.3 and 151.3 mg QRE/g. Both DPPH radical scavenging activity and SOD-like (superoxide dismutase) activity, showed significant antioxidant effects. Finally, all other extracts except for BUE had a-glucosidase inhibition at 60%. Therefore, an effective extraction method for physiologically active substances must be selected.

  • To use onion(Allium cepa L.) residue as raw materials of food product, yields of onion-juice according to various extracting methods and optimal extracting condition of quercetin and its related glycosides were carried out. Onion juices gained by the methods of pressing, rotary crushing, freeze pressing and enzyme treatment. The yield by the method of enzyme treatment was higher than others. The yields of juice from fresh onion and heat-treated onion(8$0^{\circ}C$/10min) by pressing were 66% and 83%, respectively. Ethanol extraction of onion was efficient at 75$^{\circ}C$ and acetic acid extraction was proper at 3% concentration for 3 hours at 11$0^{\circ}C$. The onion extract was fractionated in the order of hexane, chloroform, ethylacetate and butanol to test flavonoids. The highest abundant flavonoids were found in ethylacetate and butanol fraction.

High Performance Liquid Chromatography (HPLC) Detection of Malonaldehydethiobarbituric Acid (MA-TBA) Complex in Ground Pork

  • For monitoring lipid oxidation development in cooked ground pork during refrigerationm, malonaldehydethiobarbituric acid(MA-TBA) contents were measured using high performance liquid chromatography(HPLC). As the oxidation proceeded during refergeration, TBA-reaction substances(TBARS) absorbances increased and the corresponding HPLC peak areas also increased proportationately. The correlation coefficient between the HPLC peak areas and MA-TBA absorbance were 0.9979. The treatemtn of cetrimide, an ion pairing agent, gave a complete resolution of the MA-TBA complex and the butanol extraction of the complex increased its recovery by 37.8%. Both cetrimide treatment and butanol extraction are essential steps for analyzing MA-TBA complex in ground pork wiht HPLC. A reliable and specific measurement of NA-TBA in ground pork was successfully performed using HPLC.
