Radical Scavenging and Antioxidant Activities of Fermented Laminaria japonica Extracts

  • Radical scavenging and antioxidant activities of Laminaria japonica and fermented its extracts were evaluated. Freeze-dried L. japonica was fermented by Aspergillus oryzae and extracted with distilled water. The extract solution was mixed with ethanol and centrifuged. The supernatant was ethanol soluble fraction, non-polysaccharide fraction (ESF), and residue was ethanol insoluble precipitation, polysaccharide fraction (EIP). ESF was subjected to sequential fractionation with dichloromethane, ethyl acetate, butanol and water. To determine the radical scavenging and antioxidant activities of these, DPPH radical, hydroxyl radical, superoxide anion radical scavenging activities and linoleic acid oxidation were tested. Among the extracts, ESF of fermented L. japonica showed the highest radical scavenging activity. The ESF showed DPPH radical scavenging activity of 64.33% at concentration of $50{\mu}g/mL$. It was higher than 57.70% of vit. C. Ethyl acetate and butanol fraction had high value of radical scavenging and antioxidant activities, especially butanol fraction of fermented L. japonica was 79.48 % of hydroxyl radical scavenging activity at concentration of $50{\mu}g/mL$. The fermented L japonica had radical scavenging and antioxidant activities higher than L. japonica. These results suggest that fermented L japonica is healthy food having radical scavenging and antioxidant activities.

유기노의 항산화 활성 및 멜라닌 생성 억제 효과 (Effect of Artemisia anomala S. Moore on Antioxidant Activity and Melanogenesis)

  • In mammalian melanocytes, melanin synthesis is controlled by tyrosinase, the key enzyme in the pigment synthesis. In this study, to develop a new whitening agent, we have investigated the antioxidant and the inhibitory effect of Artemisia anomala extract on tyrosinase activity and melanigenesis in the B16/F1 melanoma cells. The inhibition ratio of tyrosinase activity of butanol fraction from A. anomala was higher than that of arbutin ($97.5{\pm}0.5%$ at the concentration of 2 mg/ml). The butanol fraction was shown scavenging activities of 1,1-diphenyl-2-picrylhydrazyl (DPPH) radicals and superoxide anion radicals in a dose dependent manner. The highest inhibitory activity of melanogenesis was also butanol fraction ($25.0{\pm}3%$ at the concentration of $200\;{\mu}g/ml$). From these results, we suggest that the A. anomala extract might be used to be a potential agent for skin whitening.

만성(慢性) Acohol 섭취(攝取) mouse에서 alcohol 대사(代謝) 효소활성(酵素活性)에 미치는 인삼(人蔘)의 영향(影響) (Effect of Ginseng on the Hepatic Alcohol Metabolizing Enzyme System Activity in Chronic Alcohol-Treated Mouse)

  • The present study was undertaken to investigate an effect of ginseng butanol fraction(total saponin) on the hepatic ethanol metabolism, we used experimental animals for the subject of study. When, in case of ADH and MEOS, ginseng butanol fraction was added, enzyme activity was increased in a small dose, and on the contrary, in a large dose, showed inhibitory effect. in catalase, the activity showed no significant effect by adding ginseng butanol fraction. In the light of kinetic aspect, when, in reaction mixture, ethanol and ginseng butanol fraction were concurrently added and reacted, Km value of ADH and MEOS was decreased. After pretreated with ginseng butanol fraction and inducement of acute toxic state by ethanol, the activities of ADH and MEOS were increased to an extent of about 25% compared to controls. But catalase activity was not significantly affected. In case that ginseng butanol fraction was given to mice fed with 5% ethanol instead of water for 60 days, the activities of ADH and MEOS were increased about 20% to 50% compared to ethanol-treated group. On the contrary, catalase activity was not affected. But blood concentrations of ethanol were decreased due to ginseng butanol fraction treatment. All these observations suggested that reduction of ethanol blood concentration should be dependent upon increased activities of ADH and MEOS. Thereby it suggests the recovery from alcohol intoxication can be prompted by treatment with ginseng.

아이스플랜트의 항산화 및 HCT116 인체 유래 대장암세포 성장억제 활성 (Antioxidant and growth inhibitory activities of Mesembryanthemum crystallinum L. in HCT116 human colon cancer cells)

  • 본 연구에서는 ice plant의 ethanol 추출물과 분획물의 항산화 및 대장암세포 성장억제 활성을 in vitro 수준에서 평가하는 것을 목적으로 하였다. Ethanol 추출물의 총 폴리페놀 함량 (3.7 mg GAE/g), 총 카로티노이드 함량 ($13.2{\mu}g/g$), DPPH 라디칼 소거활성 (21.0%), 철 환원력 (21.0%)보다 butanol 분획물의 총 폴리페놀 함량 (5.4 mg GAE/g), 총 카로티노이드 함량 ($86.6{\mu}g/g$), DPPH 라디칼 소거활성 (34.9%), 철 환원력 (80.8%)이 더 높았다. 또한 HCT116 대장암세포에서 세포 내 활성산소종 수준을 감소시키거나 세포 성장을 억제하는데 있어서 ethanol 추출물보다 butanol 분획물의 활성이 더 컸다. 대장암세포의 성장을 억제하는데 있어서 butanol 분획물이 ethanol 추출물보다 더 효과적이었던 것은 butanol 분획물의 apoptosis 유도활성이 ethanol 추출물의 활성보다 더 컸고 butanol 분획물만이 G2/M기억류활성을 나타냈기 때문인 것으로 생각된다. 앞으로 이와 같은 결과를 초래하는 주요 활성성분을 분리 동정하고 ice plant의 항산화 활성 및 대장암세포 성장억제 효과가 in vivo 수준에서 재현되는지 검증하며 이와 관련된 세부기전을 탐색하는 심도 있는 연구가 필요할 것으로 생각된다.

첨가제 사용에 의한 Epoxy Resin 용액의 인화점 측정 (Measurement of Flash Points of Epoxy Resin Solutions by Using Additives)

  • The knowledge of the flash point of the various liquid substances is required because of process safety and control in industrial fire protection. The epoxy resin is one of versatile resins that has wide selection of using curing agents and additives to achieve various applications such as coatings, adhesives, interior materials, reinforced plastics and electrical insulation. In this study, the lower flash points for p-xylene+epoxy resin, o-xylene+epoxy resin and n-butanol+epoxy resin systems were measured by using Pensky-Martens closed cup tester. The lower flash points for p-xylene+epoxy resin, o-xylene+epoxy resin and n-butanol+epoxy resin systems rapidly increased 80wt%, 90wt% and 95wt% of epoxy resin concentration, respectively. This results serve as a guide to estimate flash point of any epoxy resin solution.

SPME를 이용한 참외와인의 휘발성 향기성분의 모니터링 (Monitoring of the Changes in Volatile Flavor Components in Oriental Melon Wine Using SPME)

  • 본 연구에서는 SPME를 이용하여 참외 알코올 발효과정 중 주요 휘발성 향기성분의 변화를 조사하였다. 참외 원과의 향기성분은 주로 melon 및 풋내향이 주로 나타났으며 풋내성분은 nonanal, 1-butanol, 1-octen-3-ol 및 benzene으로 동정되었고 상대적 농도는 16.66%로 나타났다. 머스트에서는 풋내성분 중 nonanal은 농도는 감소하였으나 나머지 성분들은 큰 변화가 없었으며, 주로 sweet향이 증가하는 경향으로 나타났으며 특히 benzene의 상대적 함량이 25.58% 증가하여 가장 높은 함량을 차지하였다. 알코올 발효 후 1-butanol을 제외한 풋내유발 성분들의 함량이 매우 감소하였으며 숙성 중 모두 검출되지 않은 것으로 나타났다. 이취의 원인이 되는 풋내유발 물질들은 발효 및 숙성과정을 통해 모두 소멸되었으나 acid류의 휘발성 향기성분들은 알코올 발효과정에서 생성되었으며 특히 불쾌취를 유발하는 octanoic acid는 상대적 농도가 숙성과정 중 60.99%로 매우 높게 나타났으며 숙성 중에 자극적인 신냄새의 acetic acid가 생성되는 경향으로 나타나 향후 참외 와인 제조에 있어 향기 개선을 위한 연구가 요구된다.

Aspergillus niger SFN-416으로부터 생산한 $\beta$-Glucosidase의 정제 및 특성

  • $\beta $-Glucosidase (EC 3.2.1.21) was purified from Aspergillus niger SFN-416 by a sequential process of ammonium sulfate precipitation, Sepadex G-100 and DEAE-Sephacel column chromatography. Molecular weight of the enzyme was 46, 000 daltons. The K$_{m}$ and V$_{max}$ values for PNPG were 0.67 mM and 25 moles/ml $\cdot $min., respectively. The optimum pH and temperature of the enzyme activity were 3.5 and 58$\circ $C, respectively. The enzyme activity was decreased by addition of metal ions, and increased by addition of metanol, ethanol, isopropanol and 1-butanol at a concentration of 10% (v/v). Stability of the enzyme was increased by addition of isopropanol and 1-butanol at a concentration of 10% (v/v).

Inhibitory Effects of Rehmannia glutinosa Liboschitz on Endothelial Cell Proliferation

  • Vascular endothelial growth factor (VEGF) and basic fibroblast growth factor (bFGF) are the most important angiogenic molecules associated with tumor-induced neovascularization. This study was carried out to investigate inhibitory effect of extracts from root of Rehmannia glutinosa LIBOSCHITZ (Rehmannia Radix and Rehmannia Radix Preparata) on endothelial cell proliferation. The methanol extracts from the medicinal herb were fractionated into n-hexane, ethyl acetate, n-butanol and aqueous fractions. Among the four fractions, the n-butanol fraction from R. Radix on exhibited highly effective inhibition (${\approx}79%$ inhibition) on the binding of KDR/Flk-1-Fc to immobilized $VEGF_{165}$ and then ethyl acetate fraction from R. Radix (${\approx}45%$ inhibition) at the concentration of $100\;{\mu}g/ml$. The n-butanol fraction efficiently blocked the VEGF- and bFGF-induced HUVEC proliferation in a dose-dependent manner, but did not affect the growth of HT1080 human fibrosarcoma cells. The n-butanol fraction more efficiently blocked the binding of KDR/Flk-1-Fc to immobilized $VEGF_{165}$ and VEGF- and bFGF-induced human umbilical vein endothelial cell proliferation than the fraction from R. Radix Preparata. Our results suggest that Rehmannia Radix may be used as a candidate for developing anti-angiogenic agent.

결명자 분획물이 당뇨 유발 흰쥐의 혈당에 미치는 영향 (Hypoglycemic Effect of Fractions of Cassia tora Extract in Streptozotocin-Induced Diabetic Rats)

  • The hypoglycemic effect of fractions of methanol extract of Cassia tora was investigated in diabetic rats. Sprague-Dawley male rats weighing 180∼230 g were divided into 6 groups. Diabetes was induced in the male rats by intravenous injection of streptozotocin at a dose of 45 mg/kg dissolved in citrate buffer (pH 4.5). The diabetic animals then had plasma glucose concentration of above 300 mg/㎗. Fractions of methanol extract of Cassia tora were administered orally into the diabetic rats for 14 days after streptozotocin injection. The food intake and body weight gain were monitored and plasma levels of glucose, triglyceride, free fatty acid, HDL-cholesterol, cholesterol and ALT activity were determined. Levels of glycogen, cholesterol and triglyceride in liver were analysed. Weights of liver and kidney were lighter in all the fraction-administered groups than streptozotocin-control group. Plasma glucose level was significantly decreased by the administration of butanol fraction at 14 days. Plasma triglyceride levels were lower in chloroform, butanol and H$_2$O fraction-administered groups compared to streptozotocin-control group. Plasma cholesterol and HDL-cholesterol levels were not significantly different in all groups. Administrations of each of the four fractions have decreased plasma free fatty acid level, ALT activity and liver triglyceride levels in the diabetic rats. Liver glycogen levels in chloroform, butanol and H$_2$O fraction-administered groups were higher than streptozotocin-control group. It is suggested from the results that butanol fraction of methanol extract of Cassia tora may contain the antihyperglycemic compounds.

TTAB/Brij 35 혼합계면활성제의 미셀화에 미치는 부탄올 이성질체들의 효과 (Effects of Butanol Isomers on the Mixed Micellization of TTAB/Brij 35 Mixed Surfactant Systems)

  • 순수 물 및 부탄올 이성질체의 수용액에서 양이온 계면활성제인 trimethyltetradecylammonium bromide(TTAB)와 비이온 계면활성제인 polyoxyethylene(23) lauryl ether(Brij 35)로 이루어진 혼합계면활성제의 임계미셀농도(CMC)와 반대이온의 결합상수값(B)을 TTAB의 겉보기 몰분율(1)의 함수로서 전도도법과 표면장력계법으로 측정하였다. 이와 같이 측정한 CMC 값에 비이상적 혼합미셀화 모델을 적용함으로써 여러 가지 열역학적 함수값(Xi, i, Ci, aiM, 및 △Hmix)들을 계산하고 분석하였다. TTAB/Brij 35 혼합계면활성제의 미셀화에 미치는 부탄올 이성질체의 효과는 n-부탄올>iso-부탄올>tert-부탄올>물 크기순으로 나타났다.
