키토산의 분자량에 따른 랫드에서의 적용 (Application of Rats According to Molecular Weight of Chitosan)

  • Biodegradable films were prepared by solution blend method in the weight ratio of chitosan for the purpose of useful biomaterials. The possibility of biomaterials prepared from natural polymer as a skin substitute was evaluated by measuring biocompatibility. These films were inserts in the back of rat and their biodegradability was investigated by the film weight and hematology as a function of time for the biotransformation. The result of rat test showed that medium, high viscosity chitosan induced some suspects of inbiocompatibility in the tissue by goreign body reaction 48 and 72 hours after implantation. Also, we prepared the official burn ointment which is made by low viscosity chitosan. This burn ointment was covered on the skin wound of artificial burn and their effect of healing was investigated by the evaluation of the naked eye and hematological change as a function of time. The result of rats test showed that burn ointments made from chitosan was effective reductio of inflammation than negative group.

Algin을 이용한 화상연고제의 개발 (Development of Burn Ointment using Algin)

  • Burns can be caused by fire, chemicals, heated object and fluids. Distinguishing a minor burn from a more serious burn involves determining the degree of damage to the tissues of the body. Algin is known as natural polymer marine plants, we prepared the official burn ointment which is made by Algin. This burn ointment was covered on the skin wound of artificial burned and their effect of healing was investigated by the evaluation of histological and hematological change as a function of time. The result of rats test showed that burn ointments made from Algin was effective in formation of the new tissue and reduction of inflammation.

키토산의 백서에서의 생체적합성 (Biocompatibility for the Rat of Chitosan)

  • 점도(평균 분자량)의 차이가 있는 카토산을 펼름형태로 제조하여 실험통불언 백서의 펴하에 삽입하여 백서에서의 적합성을 관찰하였으며 또한 우수한 적합성올 나타낸 키토산을 화상연고제로 제조하여 인와적 2도 화상을 입힌 백서의 피부에 대한 치료전후의 육안적 관찰과 혈액학적 분석올 실시l하여 천연고분자인 치토산의 생체적용 가능성을 연구하였다 제초한 키토산펼룹의 중량 변화와 혈액학적 결과에서 확인하 였듯이 재료의 생처l척합성은 여러 생체내 인자들에 의한 복 합적인 결과임을 확인할 수 있었으며 실험 결과 저점도 키토 산이 비교적 우수한 분해속도와 혈액학적 결과를 보여주는 생체 적합성을 나타내었다. 화상설험으로 기존 수용성 연고 베이스확 유사한 키토산 연고베이스물 제조하고 대조군으로 논 자연치유군(negative군)과 기존 수용성연고베이스(positive군) 웅의 두 군올 설정하여 백서의 화상 피부에서 6일과 12 열 후익 육안적 관찰 및 혈액학쩍 검사블 실시하였다 육안 적 관찰로서 실험가간 중에 사망한 백서는 없였으며 대조군 을 포함한 모든 실험군에서 화상실험 힐주일 후부터 백서의 화상부위에 가피(crust)가 형성되기 시작하였다. 백서의 혈액학적 결파 중 백혈구늠 실험 6일 후에는 백혈구가 감소하였으나 12일 후에는 대부분의 설험군에서 백혈구가 증가하여 염증발현의 가능성을 나타내었다. 특히, 자연치유군으로 설정 한 n$\xi$gative군에서는 높은 백혈구 수치를 나타내었다.

수용액 및 연고기제중의 상피세포 성장 인자의 안정화 (Stabilization of Epidermal Growth Factor in Aqueous Solution and Ointment Base)

  • Epidermal growth factor (EGF) is a mitogen which activate the proliferation of basal cells in skin, which implicate the wound healing in severe skin damage such as burn. To carry out the preclinical test for the pharmacological action of EGF, EGF in transdermal delivery system must be stable. Since EGF is a protein susceptible to proteolysis and unstable in aqueous solution, in vitro stabilization of EGF is prerequisite for the formulation. In this study, effect of additives on the stability of EGF is investigated in vitro. The stability of EGF in aqueous solution was enhanced with the various water-soluble polysaccharides such as HPMC, sorbitol, mannitol and dextrin. EGF was successfully extracted from the ointment with 5% HPMC solution, and EGF in aqueous solution and ointment was also successfully stabilized with 5% HPMC. The ointments prepared with different amount of EGF were applied on the damaged dorsal skin of rats for the determination of optimal concentration of EGF. The ointment with EGF $(10\;{\mu}g/g)$ showed good wound healing action on the damaged skin of rats.
