  • Genetic variation at 25 microsatellite loci, population structure, and genetic bottleneck hypothesis were examined for Ankleshwar poultry population found in Gujrat, India. The estimates of genetic variability such as effective number of alleles and gene diversities revealed substantial genetic variation frequently displayed by microsatellite markers. The average polymorphism across the studied loci and the expected gene diversity in the population were 6.44 and 0.670${\pm}$0.144, respectively. The population was observed to be significantly differentiated into different groups, and showed fairly high level of inbreeding (f = 0.240${\pm}$0.052) and global heterozygote deficit. The bottleneck analysis indicated the absence of genetic bottleneck in the past. The study revealed that the Ankleshwar poultry breed needs appropriate genetic management for its conservation and improvement. The information generated in this study may further be utilized for studying differentiation and relationships among different Indian poultry breeds.

  • In recent years, Zn-Al-Mg alloy galvanized steel sheets have been widely used as coated steel sheets to support social capital in the infrastructure field. A feature of Zn-Al-Mg alloy-coated steel sheets is that they provide a better corrosion protection period than Zn-coated steel sheets. In this study, the corrosion resistance of a new Zn-Al-Mg alloy-coated steel sheet was investigated and compared to that of conventional commercially available coated steel sheets. The investigation confirmed that increasing the Mg concentration in the Zn-Al-Mg-coated steel sheet improved corrosion resistance, which was more than 10 times that of the galvanized steel sheet specified in JIS G 3302. The study findings also confirmed that the corrosion resistance reached more than twice that of the coated steel sheet specified in JIS G 3323. If such galvanized steel sheets are applied to social infrastructures that are exposed to severely corrosive environments, the service life of the infrastructure might be extended.

  • To enable effective notification and promotion of the food poisoning prevention, Ministry of Food and Drug Safety (MFDS) developed a mobile augmented reality (AR) game called 'Sik-Jung-Dok-Jop GO', and demonstrated the game at 11 events including Food Safety Day. At the events, we have conducted a field survey of people who have played the game, and evaluated the awareness change of the food poisoning before and after the game. The level of the respondents' awareness about the 3 methods of the food poisoning prevention has been increased by 53 percent after playing the game, the level of awareness about individual food poisoning micro-organism has been improved by 2.1 times. According to the survey, 95% at the respondents answered that the game helped them understand the food poisoning more, and 90% of the respondents said that they enjoyed the game. In conclusion, we believe that 'Sik-Jung-Dok-Jop GO' will be able to create various educative effects on prevention of food poisoning including causative foods, main symptoms, and other related knowledge when the game is widely applied to students in kindergartens, elementary, middle, and high schools.

  • Gene flow, genetic structure and differentiation of Kutchi, Mehsana and Sirohi breeds of goat from North-Western India were evaluated based on 25 microsatellite markers so as to support breed conservation and improvement decisions. The microsatellite genotyping was carried out using an automated DNA sequencer. The gene diversity across the studied loci for the Kutchi breed varied from 0.57 (ILST 065) to 0.93 (OarFCB 304, OMHC 1, ILSTS 058) with an overall mean of 0.79${\pm}$0.02. The corresponding values for Mehsana and Sirohi breeds were 0.16 (ILST 008) to 0.93 (OMHC 1, ILSTS 058) with an average of 0.76${\pm}$0.04, and 0.50 (ILSTS 029) to 0.94 (ILSTS 058) with an average of 0.78${\pm}$0.02, respectively. The Mehsana breed had lowest gene diversity among the 3 breeds studied. All the populations showed an overall significant heterozygote deficit ($F_{is}$). The Fis values were 0.26, 0.14 and 0.36 for Kutchi, Mehsana and Sirohi goat breeds, respectively. Kutchi and Mehsana were more differentiated (16%) followed by Mehsana and Sirohi (13%).The measures of standard genetic distance between pairs of breeds indicated that the lowest genetic distance was between Kutchi and Sirohi breeds (0.73) and the largest genetic distance was between Mehsana and Kutchi (1.0) followed by Sirohi and Mehsana (0.75) breeds. Mehsana and Kutchi are distinct breeds and this was revealed by the estimated genetic distance between them. All measures of genetic variation revealed substantial genetic variation in each of the populations studied, thereby showing good scope for their further improvement.

  • Canola is a crop globally used for production of oil and biofuel. Cultivation area and import volume of living modified (LM) canola have been increasing every year. As canola import dependence has reached 100% in Korea, efforts have been made for safety management of LM canola and ecological risk assessment. We developed a set of multiplex PCR method for simultaneous detection of 5 LM canola events (Topas 19/2, Rf3, Ms8, RT73 and T45) approved in Korea. The multiplex PCR assay developed allows amplification of estimated products of 5 LM canolas from event specific primer sets. Primer extension time was skipped for a time-consuming process and two annealing steps (20 cycles at $55^{\circ}C$ and 20 cycles at $60^{\circ}C$) were performed for yielding the best result which was sufficient to distinguish five LM canolas. Our results suggest that multiplex PCR method provides a cost and time-effective approach for LM canola detection.

  • AbstractLiving modified organisms (LMO) are one of the most widespread products of modern biotechnology after DNA discovery. Due to the decline of grain self-sufficiency rate and the increase of reliance on LMO imports in Korea, a series of concerns with regard to safety of living modified(LM) crops has been raised. The aim of this study is to establish the detection methods for unintentional release or growing of LMO plants in environmental conditions. To detect LM crop events, general concepts of specific primer design and PCR conditions were provided by the Joint Research Centre (JRC). The certified reference materials of seven LM events (4 soybean, 2 cotton and 1 corn) were obtained from the Institute for Reference Materials and Measurements (IRMM) and the American Oil Chemists' Society (AOCS). Genomic DNA from seven LM events were purified and PCR amplifications were carried out by using individual event-specific primer sets. LM-specific PCR products of all seven events were efficiently amplified by our methods. The results indicate that the established detection method for LMOs is suitable as a scientific tool to monitor whether the crops found in natural environments are LMOs.

  • The capability of microsatellite markers for individual identification and their potential for breed assignment of individuals was evaluated in two Indian horse breeds. The strength of these individual assignment methods was also evaluated by increasing the number of loci in increments of five. The probability of identity of two random horses from the two breeds at all twenty five studied loci was as low as $1.08{\times}10^{-32}$ showing their suitability to distinguish between individual horses and their products. In the phylogenetic approach for individual assignment using Nei's genetic distances, 10.81% of horses associated with breed other than the major cluster of the source breed horses when all twenty five microsatellite loci were implemented. Similar results were obtained when the maximum likelihood approach for individual assignment was used. Based on these results it is proposed that, although microsatellite markers may prove very useful for individual identification, their utility for breed assignment of horses needs further evaluation.

  • The breeding of domestic rabbits (Oryctolagus cuniculus) for human consumption has a long tradition in China. Infections that can affect the production of meat or even be transmitted from animals to humans are important to monitor, especially for public health reasons as well as for their impact on animal health. Thus, a total of 1,132 domestic rabbit sera from 4 regions in China were collected for serological screening for Encephalitozoon cuniculi and for Toxoplasma gondii by ELISA and modified agglutination test (MAT), respectively. Antibodies to E. cuniculi were detected in 248/1,132 (21.9%) sera tested while antibodies against T. gondii revealed a seroprevalence of 51/1,132 (4.5%). We believe that the present results are of epidemiological implications and public health importance due to the acknowledged susceptibility of humans to E. cuniculi and T. gondii infections. Therefore, routine screening tests of domestic rabbits are proposed considering the zoonotic potential of these parasites.

  • The microsatellites are the short tandem repeats of 1 to 6 bp long monomer sequences that are repeated several times. These short tandem repeats are considered to be generated by the slipped strand mispairing. Based on the unique capability of alternating purine-pyrimidine residues to form Z-DNA, the possible role of the microsatellites in gene regulation has been proposed. The microsatellites are highly polymorphic, follow Mendelian inheritance and are evenly distributed throughout the genomes of eukaryotes. They are easy to isolate and the polymerase chain reaction based typing of the alleles can be readily automated. These properties make them the preferred markers for comparison of the genetic structure of the closely related breeds/populations; very high-resolution genetic mapping and parentage testing etc. The microsatellites have rapidly replaced the restriction fragment length polymorphism (RFLP) and the random amplified polymorphic DNA (RAPD) in most applications in the population genetics studies in most species, including the various farm animals viz. cattle, buffalo, goat, sheep and pigs etc. More and more reports are now available describing the use of microsatellites in pigs ranging from measurement of genetic variation between breeds/populations, developing high resolution genetic maps to identifying and mapping genes of biological and economic importance.
