A Study on the Measurement of Dissociation Constant of Organic Acid with FIA (FIA에 의한 유기산의 해리상수 측정에 관한 연구)

  • The point of present study was focused on developing the method for the determination of dissociation constant by means of FIA conjunction with Diode Array Spectrphotometry or spectrophotometry along with small amount of Bromocresol Green(mg order) within short time. On the calculation with computer, the indicator with pK=7 has been shown the most sensitive reaction when little amount of base or acid has been added. The pKa of Bromothymol Blue and Bromocresol Green were measured as 7.31 and 4.82 respectively with spectrophotometry after activity correction by Kielland method (Reported value by Bishop, 7.30 and 4.79). The pKa of Bromocresol Green by FIA was obtained as 4.78 and was comparable with other values determined by other methods.

  • TLC(Thin Layer Chromatography) 는 제약산업이나 생화학 연구 그리고 여러 산업 현장에서 널리 이용되는 화학 분석 방법이다 . 본 연구에서는 적은 비용으로도 많은 양의 시료를 신속하게 분리할 수 있는 TLC를 이용하여 유기산인 젖산 (Lactic acid) 을 분리는 전개방법을 개발하였다 . 전개용매는 2 가지 용매 (1) nitroethane nitromethane : ethanol: water: I-propanol = 1 : 2 : 3 : 4 : 5 (v/v/v/v/v), (2) diisopropyl ether. formic acid. water = 90 : 7 : 3 (v/v/v) 를 사용하였다 . 발색시약은 A : bromocresol purple reagent I, B . bromocresol purple reagent II, C : bromocresol green-bromophenol blue-potassium permanganate reagent 를 사용하여 분석하는 방법을 개발하였다 $^6)$. 젖산의 분리는 silica gel TLC plate를 이용하는 경우 , 용매 (1) 에 발색시약 B를 사용했을 때 , 분리 확인이 가장 좋았다.

In Situ Detection and Differential Counts of Bifidobacterium spp. Using Bromocresol Green, a pH-dependent Indicator

  • The purpose of this study was to develop a simple detection method, possibly at the species-level, that allows for large-scale screening of bifidobacteria. Human fecal samples were plated on MRS-raffinose agar containing cysteine and neomycin sulfate, serving as selective pressure for bifidobacteria, and 0.003%(w/v) bromocresol green. All of the test strains grew well on this medium at $37{\pm}1^{\circ}C$, forming white colonies surrounded by yellow halos, which presented a sharp contrast against the green background. In this disc assay, the required incubation time to develop a yellowish zone varied with the species of Bifidobacterium that was tested, allowing for differential counts and easy identification at the species-level: 10-14 hr for B. bifidum, 20-22 hr for B. catenulatum and B. infantis. and 24-25 hr for B. longum and B. breve. No apparent color was observed for B. angulatum and B. adolescentis 28 hr after inoculation. To evaluate the results of pH indicator-based identification, individual isolates were subjected to a colony-PCR experiment with genus-specific primers. The amplified products from the isolates were in good accordance with those from the reference strains at a level of 95% agreement. These results suggest that the present method could be conveniently applied to cell counts, as well as to the preliminary identification of bifidobacteria from a variety of sample types including human feces, dairy products, and commercial probiotic supplements.

  • 본 연구에서는 cellulose생산균인 .Acetobacter sp. A9를 UV와 NTG mutagenesis를 통하여 선별된 변이주들을 HS 배지에 bromocresol green이 들어있는 HSB plate에 도말하여 gluconic acid를 생산하지 않는 것을 확인하였다. 그리고 calcofluor WT로 cellulose 생성능을 확인하였다.

Determination of Buprenorphine in Raw Material and Pharmaceutical Products Using Ion-pair Formation

  • A simple and sensitive extractive spectrophotometric method has been described for the determination of buprenorphine either in raw material or in pharmaceutical formulations. The developed method is based on the formation of a colored ion-pair complex (1 : 1 drug/dye) of buprenorphine and bromocresol green (BCG) in buffer pH 3 and extracting in chloroform. The extracted complex shows absorbance maxima at 415 nm. Beer's law is obeyed in the concentration range of 1.32-100.81 μ g mL-1. The proposed method has been applied successfully for the determination of drug in commercial sublingual tablets and injectable dosage form. No significant interference was observed from the excipients commonly used as pharmaceutical aids with the assay procedure.

  • Ionic palladium(II) complex, [($Me_4en)Pd(L)]_2(ClO_4)_4\;(Me_4$en = N,N,N',N'-tetramethylethylenediamine; L = bis-(4-(4-pyridylcarboxyl)phenyl)methane), allows to form a monodisperse submicrosphere without any template or additive. Surface-perchlorates on the submicrosphere have been exchanged by anionic pH indicators such as thymol blue, bromothymol blue, and bromocresol green. The ionic and amphiphilic properties of the palladium(II) complex appear to be primarily associated with the formation and easy surface-anion exchange of submicrosphere. The surface-anion exchange through the electrostatic interaction is a very good tool for the surface-modification. The color of the pH indicator-exchanged submicrospheres is very sensitive to pH and Hg$^{2+}$ cation, and thus they are promising to submicrospherical indicators.

Photocurrent and Its Stability Enhancement of Dye-sensitized Nanoparticle $TiO_2$ Solar Cells (염료감응 나노입자 $TiO_2$ 태양전지의 광전류와 그 안정성 향상)

  • A solar cell based on dye-sensitized photoelectric conversion was studied by electrochemical and spec-trofluorometric methods for the purposes of enhancing its efficiency and stability of $TiO_2$ solar cells. Nanocrystalline $TiO_2$ was used to prepare photoelectrodes, and photosensitizing dyes such as malachite green oxalate, basic blue3, rhodamine B, and bromocresol purple were chosen as sensitizers. Electrochemical oxidation potentials and absorption and emission wavelengths of dyes were used to determine energy levels of the dyes. By comparing excited energy levels of the dyes with the conduction band edge potential $(E_{c,s})\;of\;TiO_2$ calculated by using the flat-band potential $(E_{fb})\;of\;TiO_2$, properties of a dye required to fabricate a high efficient photosensitizing solar cell with high short-circuit current $(J_{sc})$ were suggested. Enhanced stability of photocurrent was obtained by coating a $TiO_2|ITO$ electrode with Polypyrrole that Possibly Prevented the recombination between the conduction band electrons and oxidized dyes and suppressed the direct electrode redox reactions of dyes on ITO.

Spectrophotometric Determination of Some Fluoroquinolone Antibacterials through Charge-transfer and Ion-pair Complexation Reactions

  • Two simple, rapid and sensitive spectrophotometric methods for the determination of three fluoroquinolones, namely levofloxacin, norfloxacin and ciprofloxacin have been performed either in pure form or in their tablets. In the first method, levofloxacin and norfloxacin are directly treated with bromocresol green (BCG) in dichloromethane while ciprofloxacin is allowed to react with the same dye in aqueous acidic buffer. Highly yellow colored complex species were formed instantaneously in case of levofloxacin and norfloxacin or after extraction into dichloromethane for ciprofloxacin. The formed complexes are quantified spectrophotometrically at their absorption maxima at 411 nm for levofloxacin and 412 nm for norfloxacin and ciprofloxacin. The second method involves the reaction of levofloxacin with ${\rho}$-chloranilic acid ( ${\rho}$-CA) and norfloxacin with tetracyanoethylene (TCNE) in acetonitrile to give complexes with maximum absorbance at 521 and 333 nm for the two drugs, respectively. Adopting the first procedure, calibration graphs were linear over the range 1- 20 ${\mu}g\;mL^{-1}$ with mean percentage recoveries of 100.41 ${\pm}$ 0.72, 99.99 ${\pm}$ 0.54 and 100.23 ${\pm}$ 0.91 for the theree drugs, respectively. For the second procedure, the concentration ranges were 15-250 ${\mu}g\;mL^{-1}$ for levofloxacin using ${\rho}$-CA and 0.8-16 ${\mu}g\;mL^{-1}$ for norfloxacin using TCNE with mean percentage recoveries of 99.88 ${\pm}$ 0.45 and 100.26 ${\pm}$ 0.68 for the two drugs, respectively. The proposed methods were successfully applied to determine these drugs in their tablet formulations and the results compared favorably to that of reference methods. The proposed methods are recommended for quality control and routine analysis.

Optimization of Citric Acid Production by Immobilized Cells of Novel Yeast Isolates

  • Citric acid is a commercially valuable organic acid widely used in food, pharmaceutical, and beverage industries. In this study, 260 yeast strains were isolated from soil, bread, juices, and fruits wastes and preliminarily screened using bromocresol green agar plates for their ability to produce organic acids. Overall, 251 yeast isolates showed positive results, with yellow halos surrounding the colonies. Citric acid production by 20 promising isolates was evaluated using both free and immobilized cell techniques. Results showed that citric acid production by immobilized cells (30-40 g/L) was greater than that of freely suspended cells (8-19 g/L). Of the 20 isolates, two (KKU-L42 and KKU-L53) were selected for further analysis based on their citric acid production levels. Immobilized KKU-L42 cells had a higher citric acid production rate (62.5%), while immobilized KKU-L53 cells showed an ~52.2% increase in citric acid production compared with free cells. The two isolates were accurately identified by amplification and sequence analysis of the 26S rRNA gene D1/D2 domain, with GenBank-based sequence comparison confirming that isolates KKU-L42 and KKU-L53 were Candida tropicalis and Pichia kluyveri, respectively. Several factors, including fermentation period, pH, temperature, and carbon and nitrogen source, were optimized for enhanced production of citric acid by both isolates. Maximum production was achieved at fermentation period of 5 days at pH 5.0 with glucose as a carbon source by both isolates. The optimum incubation temperature for citric acid production by C. tropicalis was 32 ℃, with NH4Cl the best nitrogen source, while maximum citric acid by P. kluyveri was observed at 27 ℃ with (NH4)2 SO4 as the nitrogen source. Citric acid production was maintained for about four repeated batches over a period of 20 days. Our results suggest that apple and banana wastes are potential sources of novel yeast strains; C. tropicalis and P. kluyveri which could be used for commercial citric acid production.

Freshness Monitoring of Raw Salmon Filet Using a Colorimetric Sensor that is Sensitive to Volatile Nitrogen Compounds (휘발성 질소화합물 감응형 색변환 센서를 활용한 연어 신선도 모니터링)

  • A colorimetric paper sensor was used to detect volatile nitrogen-containing compounds emitted from spoiled salmon filets to determine their freshness. The sensing mechanism was based on acid-base reactions between acidic pH-indicating dyes and basic volatile ammonia and amines. A sensing layer was simply fabricated by drop-casting a dye solution of bromocresol green (BCG) on a polyvinylidene fluoride substrate, and its color-change response was enhanced by optimizing the amounts of additive chemicals, such as polyethylene glycol, p-toluene sulfonic acid, and graphene oxide in the dye solution. To avoid the adverse effects of water vapor, both faces of the sensing layer were enclosed by using a polyethylene terephthalate film and a gas-permeable microporous polytetrafluoroethylene sheet, respectively. When exposed to basic gas analytes, the paper-like sensor distinctly exhibited a color change from initially yellow, then to green, and finally to blue due to the deprotonation of BCG via the Brønsted acid-base reaction. The use of ammonia analyte as a test gas confirmed that the sensing performance of the optimized sensor was reversible and excellent (detection time of < 15 min, sensitive naked-eye detection at 0.25 ppm, good selectivity to common volatile organic gases, and good stability against thermal stress). Finally, the coloration intensity of the sensor was quantified as a function of the storage time of the salmon filet at 28℃ to evaluate its usefulness in monitoring of the food freshness with the measurement of the total viable count (TVC) of microorganisms in the food. The TVC value increased from 3.2 × 105 to 3.1 × 109 cfu/g in 28 h and then became stable, whereas the sensor response abruptly changed in the first 8 h and slightly increased thereafter. This result suggests that the colorimetric response could be used as an indicator for evaluating the degree of decay of salmon induced by microorganisms.
