Anti-Cancer Effect of 3-(4-dimethylamino phenyl)-N-hydroxy-2-propenamide in MCF-7 Human Breast Cancer

  • Objectives: In recent years, a number of structurally diverse Histone deacetylase (HDAC) inhibitors have been identified and these HDAC inhibitors induce growth arrest, differentiation and/or apoptosis of cancer cells in vitro and in vivo. This study aimed at investigating the antitumor activity of newly synthesized HDAC inhibitor, 3-(4-dimethylamino phenyl)-N-hydroxy-2-propenamide (IN-2001) using human breast cancer cells. Methods: We have synthesized a new HDAC inhibitor, IN-2001, and cell proliferation inhibition assay with this chemical in estrogen receptor-positive human breast cancer MCF-7 cells. Cell cycle analysis on MCF-7 cells treated with IN-2001 was carried out by flow cytometry and gene expression was measured by RT-PCR. Results: In MCF-7 cells IN-2001 showed remarkable anti-proliferative effects in a dose- and time-dependent manner. In MCF-7 cells, IN-2001 showed a more potent growth inhibitory effect than that of suberoylanilide hydroxamic acid. These growth inhibitory effects were related to the cell cycle arrest and induction of apoptosis. IN-2001 showed accumulation of cells at $G_2$/M phase and of the sub-$G_1$ population in a time-dependent manner, representing apoptotic cells. IN-2001-mediated cell cycle arrest was associated with HDAC inhibitor-mediated induction of CDK inhibitor expression. In MCF-7 cells, IN-2001 significantly increased $p21^{WAF1}$ expression. Conclusions: In summary, cyclin-dependent kinase (CDK) induced growth inhibition, possibly through modulation of cell cycle and apoptosis regulatory proteins, such as CDK inhibitors, and cyclins. Taken together, these results provide an insight into the utility of HDAC inhibitors as a novel chemotherapeutic regime for hormone-sensitive and insensitive breast cancer.

Peanut sprout tea extract inhibits lung metastasis of 4T1 murine mammary carcinoma cells by suppressing the crosstalk between cancer cells and macrophages in BALB/c mice

  • BACKGROUND/OBJECTIVES: As peanuts germinate, the content of the components beneficial to health, such as resveratrol, increases within the peanut sprout. This study examined whether the ethanol extract of peanut sprout tea (PSTE) inhibits breast cancer growth and metastasis. MATERIALS/METHODS: After orthotopically injecting 4T1 cells into BALB/c mice to induce breast cancer, 0, 30, or 60 mg/kg body weight/day of PSTE was administered orally. Angiogenesis-related protein expression in the tumors and the degree of metastasis were analyzed. 4T1 and RAW 264.7 cells were co-cultured, and reverse transcription polymerase chain reaction was performed to measure the crosstalk between breast cancer cells and macrophages. RESULTS: PSTE reduced tumor growth and lung metastasis. In particular, PSTE decreased matrix metalloproteinase-9, platelet endothelial cell adhesion molecule-1, vascular endothelial growth factor-A, F4/80, CD11c, macrophage mannose receptor, macrophage colony-stimulating factor, and monocyte chemoattractant protein 1 expression in the tumors. Moreover, PSTE prevented 4T1 cell migration, invasion, and macrophage activity in RAW 264.7 cells. PSTE inhibited the crosstalk between 4T1 cells and RAW 264.7 cells and promoted the macrophage M1 subtype while inhibiting the M2 subtype. CONCLUSIONS: These results suggest that PSTE blocks breast cancer growth and metastasis to the lungs. This may be because the PSTE treatment inhibits the crosstalk between mammary cancer cells and macrophages and inhibits the differentiation of macrophages into the M2 subtype.

생후 6개월 동안 모유영양아의 단백질 섭취량과 성장과의 관계 (Protein Intakes and Growth of Breastfed and Breastfed Supplemented Infants During the First Six Months of Age)

  • Protein concentration in human from 39 well-norished American women and its adequacy for growth of exclusively breastfed infants(BF) and breastfed infants fed supplementary foods(BFS)from 1-6 months postpartum were studied. Mean protein concentration of breast milk measured by Lowry et al., using human serum albumin as a standard, over the first 6 months lactation was 1.31$\pm$0.13g/dl. Concentration of protein was singnificantly higher at the first month of lactation (1.55$\pm$0.23g/dl)(P<0.05) than any other month studied. Mean volume of breast milk ranged from 662-848ml/day in the BE group and from 415-661ml/day in the BFS group during the first 6 months of lactation. Mean protein intake of infants ranged from 1.3-2.2g/kg in the BF group and from 1.4-2.1g/kg in the BFS group. Mean protein intake (g/kg body weight) of both BF and BFS groups was less than Recmmended Dietary Allowance(1989, USA) of 2.2g/kg except at 1 month of age. However, mean growth of the infants was normal according to NCHS reference, suggesting that the RDA for protein was unrealistically high for infants during 2-6 months of age. Protein provided by breast milk alone appeared adequate for normal growth during this time.

Effects of Indole Oligomers Induced from Indole-3-carbinol on the Growth of MCF-7 Breast Cancer Cells

  • Inhibitory effect of indole oligomers induced from indole-3-carbinol(I3C) on the growth of breast cancer cells was studied. We gernerated the reaction mixtures (RXM) at ambient temperature by treating a stirred aqueous solution of I3C (typeically 0.25ml) at a concentration of 12 $\mu$mol/ml) with hydrochloric acid (typically 28$\mu$l of a 1 mmol/ml solution). RXM was fractionated by the column chromatography. The fractions with similar UV-pattern were further fractionated by HPLC and 3.3'-diindoylmethane (DIM) and other indole oligomers were identified. I3C, RXM, and it derived indole compounds were added to MCF-7 cells and cultured in the presence of 10-7M estradiol for 7 days. the growth-inhibitory effect of I3C and DIM on the growth of MCF-7 cell was very strong. The synthetic DIM also revealed antiproliferative effect on MCF-7 cel. The fractions containing high DIM content (77%), were most effective in inhibiting MCF-7 cell growth induced by estradiol. With these results, we suggest that I3C and DIM might have anticarcinogenic effect on the breast cancer.

  • This study is to identify the characteristics of the growth and deveolpment, and to assess Obesity Index, BMI and Kaup Index of infants paticipated in a healthy breast-feeding contest. The subjects of this study were 94 breast- feeding infants, ages from 6 to 8 months. Their weight and height were measured and compared with Korean Standards. Their developmental state was tested by DDST(Denver Develop- mental Screening Test) standardized in Korea. The data were analyzed by using the SPSS-WIN 10.0. The result is as follows : 1. The weight of infants was more than Korean Standards, but the height was less. 2. There were statistically significant differences in Obesity Index, BMI and Kaup Index. Obese infants estimated by Obesity Index were about 10%, but by BMI and Kaup Index were 35.1% and 27.7%. 3. Spearman's rhos of Obesity Index and Kaup Index, Obesity Index and BMI, and Kaup Index and BMI were individually 0.526, 0.528 and 0.753. In conclusion, BMI should be added to the criteria for assessing healthy breast-feeding infants.

Epigenetic regulation of long noncoding RNA UCA1 by SATB1 in breast cancer

  • Special AT-rich sequence binding protein 1 (SATB1) is a nuclear matrix-associated DNA-binding protein that functions as a chromatin organizer. SATB1 is highly expressed in aggressive breast cancer cells and promotes growth and metastasis by reprograming gene expression. Through genome-wide cross-examination of gene expression and histone methylation, we identified SATB1 target genes for which expression is associated with altered epigenetic marks. Among the identified genes, long noncoding RNA urothelial carcinoma-associated 1 (UCA1) was upregulated by SATB1 depletion. Upregulation of UCA1 coincided with increased H3K4 trimethylation (H3K4me3) levels and decreased H3K27 trimethylation (H3K27me3) levels. Our study showed that SATB1 binds to the upstream region of UCA1 in vivo, and that its promoter activity increases with SATB1 depletion. Furthermore, simultaneous depletion of SATB1 and UCA1 potentiated suppression of tumor growth and cell survival. Thus, SATB1 repressed the expression of oncogenic UCA1, suppressing growth and survival of breast cancer cells.

전이성 유방암 세포주에 대한 당목향의 항암효과탐색 (Anti-cancer Effects by Saussurea lappa Clarke in Highly-metastatic MDA-MB-231 Breast Cancer Cells)

  • Saussurea lappa Clarke is a well-known transitional medicine in Asia including Korea, China and Japan. It has been reported that Clarke has diverse effects such as anti-viral, anti-inflammatory, anti-cancer in human gastric cells and human prostate cancer cells. However, the anti-cancer effects and the mechanism of actions of Saussurea lappa Clarke are still unknown in breast cancer. In this study, we observed that Saussurea lappa Clarke inhibits the cell growth in a dose- and time-dependent manner in highly-metastatic MDA-MB-231 breast cancer cells. In order to examine whether Saussurea lappa Clarke suppresses cell growth inducing apoptosis cell death or cell cycle arrest, we analyzed DNA contents and cell cycle distribution using a flow cytometer and western blotting in MDA-MB-231 cells. We suggest that Saussurea lappa Clarke dose not induced apoptosis and induced G2/M phase cell cycle arrest. Moreover, Saussurea lappa Clarke also decreased the expression level of metastasis-angiogenesis releated protein such as VEGF. However, dose not changed the expression level of metastasis related protease MMP-1 in highly-metastatic MDA-MB-231 breast cancer cells. Therefore, Saussurea lappa Clarke might be good and useful chemotherapy agent highly-metastatic breast cancer patients.

Synergistic Effects of Tamoxifen and Tranilast on VEGF and MMP-9 Regulation in Cultured Human Breast Cancer Cells

  • Background: Vascular endothelial growth factor and matrix metalloproteinases are two important factors for angiogenesis associated with breast cancer growth and progression. The present study was aimed to examine the effects of tamoxifen and tranilast drugs singly or in combination on proliferation of breast cancer cells and also to evaluate VEGF and MMP-9 expression and VEGF secretion levels. Materials and Methods: Human breast cancer cell lines, MCF-7 and MDA-MB-231, were treated with tamoxifen and/or tranilast alone or in combination and percentage cell survival and proliferative activity were evaluated using LDH leakage and MTT assays. mRNA expression and protein levels were examined by real-time RT-PCR and ELISA assay, respectively. Results: LDH and MTT assays showed that the combined treatment of tamoxifen and tranilast resulted in a significant decrease in cell viability and cell proliferation compared with tamoxifen or tranilast treatment alone, with significant decrease in VEGF mRNA and protein levels. We also found that tamoxifen as a single agent rarely increased MMP-9 expression. A decrease in MMP-9 expression was seen after treatment with tranilast alone and in the combined treatment MMP-9 mRNA level was decreased. Conclusions: This combination treatment can able to inhibit growth, proliferation and angiogenesis of breast cancer cells.

유방암생존자의 외상 후 성장에 영향을 미치는 요인 (Factors Influencing Posttraumatic Growth in Survivors of Breast Cancer)

  • Purpose: Posttraumatic growth (PTG) is defined as 'positive psychological change experienced as a result of a struggle with highly challenging life circumstances'. The purpose of this study was to identify the level of PTG and its correlates in Korean patients with breast cancer. Methods: A sample of 120 participants was recruited from outpatients, who had successfully completed primary treatment of breast cancer at a university hospital., Data were collected from June to December, 2014 using Posttraumatic Growth Inventory, Illness Intrusiveness Rating Scale, Cancer Coping Questionnaire, Revised Life Orientation Test and The Multidimensional Scale of Perceived Social Support. Results: Total score for the PTG was $79.18{\pm}17.54$ in patients surviving breast cancer. Bivariate analyses indicated that PTG was positively associated with having a religion, perceived social support, greater optimism, cancer coping, and illness intrusiveness. Results of the regression analysis showed that cancer coping (${\beta}=.29$, p=.001), optimism (${\beta}=0.28$, p=.001) and illness intrusiveness (${\beta}=0.17$, p=.037) were statistically significant in patients' PTG. Conclusion: The research findings show that the variables of cancer coping, optimism and illness intrusiveness significantly explain PTG and these psychological variables can be used to provide improvement in PTG for patients with breast cancer.

모유영양아와 인공영양아의 철분과 아연의 섭취상태에 관한 종단적 연구 (A Longitudinal Study of the Iron and Zinc Intakes of Korean Infants from 1 to 3 Months-Breast-Fed vs Formula-Fed Infants)

  • This study was carried out to longitudinally investigate the iron and zinc intakes and correlation with growth performance of 25 male breast-and formula-fed infants from 1 to 3 months postpartum, longitudinally. There were four groups breast fed(BF) and three formula fed groups((FFM, FFN and FFP). Milk intakes and the concentration of iron and zinc from human milk and the formulas were measured. The iron content of human milk was $2.07\pm{1.05(0.63~5.65)}\mu{g}$/ml. The zinc content was $2.43\pm{1.14(0.70~5.30)}\mu{/ml.}$ Both were not significantly different among postpartum months. The average iron intake of the breast-fed and formula-fed infants was $1.6\pm{0.7mg/}$day and $8.4\pm{2.3mg}$/day, respectively. The iron intake of breast-fed infants was higher than previous reports. And that of formula-fed infants was higher than the RDA. The average zinc intake of the breast-fed group was $1.9\pm{0.9mg/}$day and formula-fed infants' was $2.7\pm{0.7mg/}$day, which was higher than the RDA. There was no correlation between these mineral intakes and the growth performance during 1 to 3 postpartum months. So, extensive studies of the iron content of human milk and wide cross-sectional studies for establishing iron and zinc recommended dietary allowances for infants are needed.
