Glucosinolates are one of the important plant secondary metabolites that are produced mainly in Brassicaceae plants. The compounds are primarily involved in defense responses to biotic and abiotic resistance in plants and play important biological roles during plant growth and development. In this study, the glucosinolate profiles in leaves of two different Brassica oleracea populations were compared using high-performance liquid chromatography (HPLC). The nine major glucosinolates compounds in cabbage leaves were identified as belonging to the aliphatic and indolic groups. Among them, sinigrin, which belongs to the aliphatic group, was recorded to be 41% whereas glucobrassicin and 4-methoxyglucobrassicin, which belong to the indolic group, were recorded to be 53.8%. In addition, we performed a genetic analysis to identify regions of the genome regulating glucosinolates biosynthesis in the $F_3$ population of Brassica oleracea. A total of 9 glucosinolates were used for the quantitative trait locus (QTL) analysis. Out of 9, a total of 3 QTLs were identified and they were associated with sinigrin, glucobrassicin, and 4-methoxyglucobrassicin synthesis located in Chromosome 1 and Chromosome 8, respectively. The results of this study will provide valuable information for the breeding of cabbage containing high glucosinolate content, and our next target is to develop component-specific and tightly linked markers for various glucosinolates.
Fluorescence in situ hybridization (FISH) is a powerful tool for the detection of DNA sequences in the specific region of the chromosomes. As well as for the integrated physical mapping, FISH karyotype analysis has to be preceded. Karyotype of Raphanus sativus 'Wonkyo 10039' was analyzed by a dual-color FISH technique; using various repetitive DNA probes, including 5S rDNA, 45S rDNA, and centromere retrotransposon. The length of the somatic metaphase chromosome ranged from 1.35 to $2.06{\mu}m$ with a total length of $15.29{\mu}m$. The chromosome complements comprised of eight pairs of metacentrics and one pair of submetacentric. Bleached DAPI Band analysis revealed a heterochromatin region, covering 28.6% to 50.4% each chromosomes. 5S and 45S rDNA sequences were located on two and three pairs of chromosomes, respectively. The centromere retrotransposon of Brassica (CRB) is a major component in Brassica related species that has been maintained as a common centromere component. CRB signals were detected on the centromere and pericentromeric region of R. sativus 'Wonkyo 10039' and three basic Brassica species (B. rapa, B. nigra, and B. oleracea). These results will provide a valuable background for physical mapping and elucidation of the evolutionary relationship among the Brassica related species.
The distribution of S-haplotypes and genetic relationships were evaluated for 47 accessions of 'Danji' radish (Raphanus sativus L. var. hortensis Baker f. gigantissimus Makino) originating from Jeju Island in South Korea. A total of 22 S-haplotype-specific SCAR markers for the S locus glycoprotein (SLG) and S receptor kinase (SRK) loci were tested, and six primer sets amplified locus-specific PCR fragments from at least one 'Danji' radish accession. S5 and S21 alleles atthe SLG locus were the most frequently distributed, and detected from 87.5% and 64.6% of the accessions, respectively. The frequency of the class-II haplotype at the SLG locus was 75%, more frequent than the class-I haplotype. The S23 allele at the SRK locus was detected from 7 accessions. Grouping of the accessions based on S-allele composition revealed three major groups, while 8 accessions showed a unique allelic composition. The genetic diversity of 47 'Danji' radishes and 1 'Gwandong' radish were also evaluated with 38 RAPD primers. A total of 312 bands were scored, and showed that 138 bands (44.2%) were monomorphic among the accessions, whereas 174 (55.8%) bands were polymorphic. Polymorphism rates ranged from 0.2 to 1.0, indicating significant variations in detecting polymorphism across RAPD primers. The genetic similarity coefficients among all pairs of the 48accessions varied from 0.62 to 0.93, and 42% of the comparisons exhibited values higher than 0.85. All the cultivars could be distinguished based on the DNA fingerprints revealed by RAPD. The comparisons between the dendrograms based on S-haplotypes and RAPDs indicate an unrelated and sporadic distribution for several accessions; however, there was a tendency for accessions with the same S-allelic composition to group into the same cluster.
Alternaria brassicicola (Schwein.) invades Brassicaceae and causes black spot disease, significantly lowering productivity. Mitogen-activated protein kinases (MAPKs) and their upstream kinases, including MAPK kinases (MAPKKs) and MAPKK kinases (MAPKKK), comprise one of the most important signaling pathways determining the pathogenicity of diverse plant pathogens. The AbSte7 gene in the genome of A. brassicicola was predicted to be a homolog of yeast Ste7, a MAPKK; therefore, the function was characterized by generating null mutant strains with a gene replacement method. AbSte7 replacement mutants (RMs) had a slower growth rate and altered colony morphology compared with the wild-type strain. Disruption of the AbSte7 gene resulted in defects in conidiation and melanin accumulation. AbSte7 was also involved in the resistance pathways in salt and oxidative stress, working to negatively regulate salt tolerance and positively regulate oxidative stress. Pathogenicity assays revealed that AbSte7 RMs could not infect intact cabbage leaves, but only formed very small lesions in wounded leaves, whereas typical lesions appeared on both intact and wounded leaves inoculated with the wild-type strain. As the first studied MAPKK in A. brassicicola, these data strongly suggest that the AbSte7 gene is an essential element for the growth, development, and pathogenicity of A. brassicicola.
Glucosinolates (GSLs), beneficial secondary metabolites for human health are abundantly present in radish vegetable. Radish is a member of Brassicaceae family and its seed, leaf and root contain very important GSLs. The objective of this study was to determine the variation of individual and total GSL contents in leaves and roots of 44 radish (Raphanus spp.) germplasm (26 R. sativus L., 3 R. raphanistrum, and 15 R. sativus L. var. raphanistroides Makino), and compare the GSL contents between leaves and roots among three Raphanus species. Thirteen GSLs were identified, being the glucoraphasatin (GRS) and glucobrassicin (GBS) the most abundant aliphatic and indolyl GSLs in both the leaves and roots. Variation in individual and total GSL contents was found among the germplasm of three Raphanus species. The GRS content was higher in roots than that of leaves in all three Raphanus species but the GBS content was higher in leaves than roots. GRS was represented 87.0%, 92.7% and 94.7% of the total GSL in roots of R. sativus L., R. raphanistrum and R. sativus L. var. raphanistroides (Makino) germplasm, respectively. Germplasm of R. raphanistrum exhibited the highest (average, $79.5{\mu}mol/g\;dw$) total GSL with a ranged from 62.7 to $92.9{\mu}mol/g\;dw$. The germplasm IT119288, Joseonmu and IT119262 from R. sativus L., RA 504 and K046542 from R. raphanistrum, and Gyeongju-2003-32 (G2003-32) and IT302373 from R. sativus L. var. raphanistroides (Makino) had high total GSL contents and these could be good candidates for developing the functional compounds-rich varieties in radish breeding program.
Non-heading Chinese cabbage (Brassica rapa L. ssp. chinensis) is a popular vegetable in Asian countries. The diamondback moth (DBM), Plutella xylostella (L.), an insect with worldwide distribution, is a main pest of Brassicaceae crops and causes enormous crop losses. Transfer of the anti-insect gene into the plant genome by transgenic technology and subsequent breeding of insect-resistant varieties will be an effective approach to reducing the damage caused by this pest. We have produced transgenic non-heading Chinese cabbage plants expressing the potato proteinase inhibitor II gene (pinII) and tested the pest resistance of these transgenic plants. Non-heading Chinese cabbages grown for 45 days on which buds had formed were used as experimental materials for Agrobacterium-mediated vacuum infiltration transformation. Forty-one resistant plants were selected from 1166 g of seed harvested from the infiltrated plants based on the resistance of the young seedlings to the herbicide Basta. The transgenic traits were further confirmed by the Chlorophenol red test, PCR, and genomic Southern blotting. The results showed that the bar and pinII genes were co-integrated into the resistant plant genome. A bioassay of insect resistance in the second generation of individual lines of the transgenic plants showed that DBM larvae fed on transgenic leaves were severely stunted and had a higher mortality than those fed on the wild-type leaves.
제초제(除草劑) paraquat는 대표적(代表的)인 비선택성(非選擇性) 제초제(除草劑)로서 식물체(植物體)내에서 광합성(光合成)의 전자(電子)흐름을 방해 하여 독성(毒性)을 나타낸다. 본(本) 실험(實驗)은 paraquat를 뿌리를 통해 흡수시킨 후 잡초종(雜草種)들의 반응(反應)을 통하여 조사하였다. 1. Paraquat에 대한 저항성(抵抗性)은 식물종의 생태적(生態的) 특성(特性)과 깊은 관계가 있었다. 일반적으로 일년생(一年生)보다는 다년생(多年生)이나 월년생식물(越年生植物)의 저항성(抵抗性)이 크게 나타났다. 2. 꿀풀과와 석죽과 십자화과의 식물들이 비교적 paraquat에 저항성(抵抗性)을 보였으며, 반면에 화본과의 식물들이 비교적 감수성(感受性)을 나타내었다. 3 꿀풀과의 쥐깨풀은 강한 저항성을 나타냈고, 대부분의 종(種)이 감수성(感受性)을 보인 화본과와 국화과에 있어서도 참새피와 지칭게 등은 강한 저항성(抵抗性)을 나타냈다. 4. Paraquat는 발아(發芽)에도 영향을 주였으나, 발아(發芽)시기의 반응(反應)과 성식물(成植物)의 반응(反應)과는 일치하지 않는 경향이었다.
순활물기생체인 Plasmodiophora brassicae는 병원성 검정을 위해서 반드시 뿌리혹을 장기적으로 보관하는 것이 매우 중요하기 때문에 그동안 병원성을 유지하는 것이 관건이다. 특히 기존의 방법은 100년 이상 사용되어온 저장법으로 개선이 필요하여 그 효율적인 방법을 밝히고자 하였다. 이 결과 장기적으로 병원성의 저하를 최소화하며 장기간 뿌리혹을 저장할 수 있는 방법은 $-70^{\circ}C$ 냉동고에 보관하는 것으로 확인되었고, 저장 조건은 뿌리혹을 그대로 보관하거나 뿌리혹을 갈아 균질화한 후 여러 겹의 거즈에 거른 것이 6가지 저장조건 중에 가장 효과적인 저장법으로 밝혀졌다.
본 연구에서 루꼴라(Eruca sativa)에 대한 다양한 생리활성을 조사하여 기능성소재 응용가능성을 검토하였다. 루꼴라 추출물은 B. subtilis, E. coli, C. albicans에 대한 항균활성이 매우 우수하게 나타났으며 특히 그람양성세균 B. subtilis에 대한 항균활성이 높았다. 또한, 인볼루크린 발현에 대한 면역조직화학 분석을 인공피부를 이용하여 분석한 결과 루꼴라 추출물을 처리(50 mg/L)한 경우에 0.1% DMSO로 처리한 대조군에 비하여 인볼루크린 발현이 뚜렷하게 증가하였으며 동일농도로 처리된 합성물질 WY14643과 비교하였을 때에도 비슷한 발현양상을 보여 주었다. 이와 같은 결과로 미루어 볼 때 루꼴라 추출물은 피부염 개선 및 피부장벽 기능을 향상할 수 있는 천연물 기반의 소재로 활용하기 위한 매우 효과적인 재료가 될 수 있다고 사료된다.
본 연구에서 루꼴라(E. sativa)에 대한 다양한 생리활성을 조사하여 기능성소재 응용가능성을 검토하였다. 루꼴라 추출물은 멜라노마 세포에 대하여 낮은 세포독성을 나타냈다. 세포독성이 거의 없는 농도에서 루꼴라 추출물 처리 시 항산화 활성($ID_{50}$, 17.60 mg/L)이 매우 우수하게 나타났으며 루꼴라 추출물 처리시 높은 tyrosinase 활성저해($ID_{50}$, 132.54 mg/L) 및 멜라닌 함량 저하($ID_{50}$, 158.90 mg/L)를 보여주었다. 또한, 루꼴라 추출물 농도 의존적으로 tyrosinase 발현이 억제되었으며, 이는 루꼴라 추출물이 직접적인 tyrosinase 활성저해 및 세포 내 tyrosinase 발현을 억제시킴으로서 멜라닌 합성을 저해하는 것으로 판단된다. 이와 같은 결과로 미루어 볼 때 루꼴라 추출물은 피부미백 소재 등 피부개선 효과를 지닌 기능성 화장품에 활용하기 위한 매우 효과적인 재료가 될 수 있다고 판단되며 향후 주름개선 효과 등 추가적인 실험을 통하여 화장품 소재 개발 응용 가능성을 타진할 것이다.
