In this study, we investigated the anticancer effect of rice bran extract in HL-60 human promyelocytic leukemia cells. The extract of rice bran inhibited the proliferation of HL-60 cells. When treated with the rice bran extract, we could observe the apoptotic characteristics such as apoptotic bodies and the increase of sub-G1 hypodiploid cell population, increase of Bax level, decrease of Bcl-2 expression, cleavage of procaspase-3, cleavage of procaspase-9 and cleavage of poly(ADP-ribose) polymerase(PARP) in HL-60 cells. Furthermore, the apoptosis induction of HL-60 cells treated with the rice bran extract was also accompanied by the inactivation of mitogen-activated protein kinases (MAPK) such as ERK1/2 MAPK and p38 MAPK. In addition, the rice bran extract induced the down-regulation of c-myc. These data suggested that the rice bran extract could induce the apoptosis via the inactivation of ERK1/2 MAPK and p38 MAPK, and the down-regulation of c-myc in HL-60 acute pomyelocytic leukemia cells. The results support that the rice bran extract might have potential for the treatment of acute promyelocytic leukemia.
In this study, nutritional analysis was done on regular rice bran and fermented rice bran toward increasing their availability and use. Regular and fermented rice bran were extracted 10 times at $98^{\circ}C$ for 4 hours each with water, extracted with 60% ethanol at $60^{\circ}C$ for 4 hours, then concentrated and extracted twice by freeze-drying. When rice bran was fermented, moisture, protein, and ash contents increased, while fats and carbohydrates decreased. Out of fatty acids, the saturated fatty acid content of regular rice ran was found to be 17.7%, and 20.5% when fermented while the unsaturated fatty acid components of rice ran and fermented rice bran were found to be 82.3 and 79.5%, respectively. In both kinds of bran, palmitic acid, oleic acid and linoleic acid represented over 90% of the fatty acid content. In rice bran the fatty acid composition was 15.1% palmitic acid, 40.6% oleic acid and 39.5% linoleic acid, while that of fermented rice bran was 13.2% palmitic acid, 43.2% oleic acid and 31.3% linoleic acid. Out of free sugars fermented rice bran contained 0% fructose, 0.0099% glucose, 0.0039% maltose and 0.3233% sucrose. These results with which those of regular rice bran were silmilar were according to the normal sugar composition of rice in general. The vitamin C content of rice bran was 53 mg/100 g and that of fermented rice bran 7 mg/100 g. In neither kind of rice bran was vitamin A detected. Out of 18 minerals analyzed, Ca, K, Mg, and Mn were the most abundant minerals in both kinds of rice bran. Fermented rice bran had a higher K content with 3,163 mg/100 g, than normal rice bran, Mg content was 1,178 mg/100g. Fermented rice bran had a higher total mineral content.
This study was conducted to determine the interaction of dietary wheat bran and dietary calcium levels n)n calcium utilization in post-breeding female rats. It was designed to compare the effects of four different levels (2.5, 5, 10 and 20%) of wheat bran and two different levels (0.5 and 1%) of calcium on bone and calcium balance in post-breeding female rats over a ten-week period. The effects of diet on animal weight gain, serum calcium, femur weight, femur calcium concentration, bone mass and calcium balance were determined and statistically analyzed. The addition of 20% wheat bran significantly (p$\leq$0.05) decreased the weight gain of rats. Serum calcium and bone calcium contents were more affected by dietary calcium level than by dietary wheat bran level. There was no significant difference in fat-free solid, ash, percentage of ash to fat-free solid and percentage of calcium to ash among groups. Groups fed the 1% calcium diet had a higher percentage of calcium to fat-free solid. All rats were in positive calcium balance during the three-4ay experimental period. The average calcium balance of the rats fed 1% calcium diet ranged from 25.34 to 53.90mg and the average calcium balance of the rats fed the 0.5% calcium diet ranged from 26.71 to 32.90mg. In rats receiving 2.5% wheat bran, the difference in calcium balance between the group fed the 1% calcium diet and the group fed the 0.5% calcium diet was only 1.37mg, which was not significantly (p$\leq$0.05) different. In rats receiving 20% wheat bran, the difference in calcium balance between the group fed the 1% calcium diet and the group fed the 0.5% calcium diet was 19.S7mg, which was significantly (p$\leq$0.05) different. The addition of wheat bran caused an increase in the calcium balance of the rats adminslesed the 1% calcium diet. On the other hand, the addition of wheat bran had no effect on the calcium balance of the rats adminislesed the 0.5% calcium diet. In conclusion, calcium utilization of rats wire more positively affected by the interaction of both dietary wheat bran levels and dietary calcium levels than either dietary wheat bran levels or dietary calcium levels alone. Moderate wheat bran consumption did not interfere with the calcium metabolism of rats when calcium intake was high enough.
Objective: This experiment was to evaluate the effect of different inclusion levels and adaptation periods on digestible energy (DE) and the apparent total tract digestibility (ATTD) of chemical constituents in diets supplemented with wheat bran. Methods: Thirty-six crossbred barrows with an initial body weight of $85.0{\pm}2.1kg$ were allotted to 6 diets in a completely randomized block design with 6 pigs per diet. Diets included a cornsoybean basal diet and 5 additional diets which were formulated by replacing corn and soybean meal in control diet with 15%, 25%, 35%, 45%, or 55% wheat bran. The experiment lasted for 34 d, and feces were collected from d 8 to 13, 15 to 20, 22 to 27, and 29 to 34 respectively. Results: The results showed no interaction effects between inclusion level and adaptation period on the concentration of DE and the ATTD of gross energy (GE) and crude protein (CP) in wheat bran. The DE value and ATTD of GE in wheat bran decreased (p<0.05) significantly as the inclusion level of wheat bran increased, but no difference in the ATTD of CP was observed. The ATTD of CP in wheat bran increased (p<0.10) significantly as the adaptation period for pigs was prolonged. In addition, the concentration of DE and the ATTD of GE in wheat bran decreased linearly (p<0.05) when pigs were fed either an increased level of wheat bran or given an increased adaptation period to the diets. Conclusion: Wheat bran showed a negative effect on the concentration of DE and ATTD of GE and CP as the inclusion level increased. A longer adaptation period can gradually increase the DE value and ATTD of GE and CP in wheat bran, and at least 14 to 21 d of adaptation might be recommended for growing-finishing pigs fed the high-fiber diets with wheat bran.
The effect of supplementation of Jackfruit (Artocarpus heterrophyllus) tree leaves and mashkalai (Vigna mungo) barn on the intake and digestibility of freshly cut common grass by Black Bengal goats was evaluated. Twelve castrated goats of approximately 5-6 months age were randomly allocated to three dietary treatments: freshly cut common grass ad libitum, common grass+100g Jackfruit tree leaves and common grass+100g mashkalai bran. Although digestibilities of dry matter, organic matter, nitrogen and neutral detergent fibre were very similar in the grass alone and mashkalai bran supplemented goats but the values were significantly (P < 0.01) higher than those observed in the Jackfruit leaves supplemented goats. Supplementation of mashkalai bran promoted higher (p < 0.05) digestible nutrients intake with the consequent higher daily live weight gain (75 g) than those fed grass alone (31 g). It could be concluded that mashkalai bran could be a suitable supplement for goats consuming common grass in the tropics and subtropics.
A bacterium producing the extracellular cellulases was isolated from soil and has been identified as Bacillus sp. The isolate, named Bacillus sp. 79-23, was shown to be very similar to B. subtilis on the basis of its biochemical properties. The carboxymethyl cellulase (CMCase) of culture supernatant was most active at 60$\circ$C and pH 6.0, and retained 90% of its maximum activity at pH 7.0. The additional carbon sources affected the CMCase productivity than nitrogen sources in the culture medium. The carbon sources including wheat bran, rice straw, maltose and glucose increased the enzyme productivty. Especially, the maximum CMCase production was 5.2 units/ml in LB medium supplemented with 3% (w/v) wheat bran, which was 13-folds more than that in LB medium. It was found that the enzyme production was in association with the growth of Bacillius sp. 79-23. But, whean bran did not affect the growth of isolate, suggesting that increasement of CMCase production was owing to the induction of CMCase biosynthesis by wheat bran. In addition, both water-soluble and insoluble components of wheat bran was involved in induction of CMCase biosynthesis.
We had investigated combustion properties of rice bran dusts. Decomposition of rice bran dusts with temperature were investigated using DSC and the weight loss according to temperature using TGA in order to find the thermal hazard of rice bran dusts, and the properties of dust explosion in variation of their dust with the same particle size. Using Hartman's dust explosion apparatus which estimate dust explosion by electric ignition after making dust disperse by compressed air, dust explosion experiments have been conducted by varying concentration and size of rice bran dust.According to the results for thermodynamic stability of rice bran dust, there are little change of initiation temperature of heat generation and heating value for used particle size. But initiation temperature of heat generation decreased with high heating rate whereas decomposition heat increased with particle size. Average maximum explosion pressure was $10kgf/cm^2$ for 60/70 mesh and $1.5mg/cm^2$ dust concentration.
The effect of the addition of wheat bran to the growth medium on the production of cellulolytic enzymes of Trichoderma reesei Rut C30 was studied in batch culture using shake flasks. The activity of cellulase was enhanced by the addition of wheat bran to the cellulase production medium. $KH_2PO_4$-$K_2HPO_4$ buffer was used for pH control during cellulase production. As a result, high cellulase activities were obtained in shake flask culture; a CMC (carboxymethyl cellulose) activity of 125.78 U/ml was obtained from 2% Avicel- and 3% wheat bran-containing medium and an FP (filter paper) activity of 12.85U/ml was obtained from 1% Avicel- and 5% wheat bran-containing medium after 6 days of cultivation.
Acetic acid and butyric acid were produced by the anaerobic fermentation of soil mixed with wheat or rice bran. The concentration of acetic acid produced in the wheat and rice bran-treated soil was 31.2mM and 8mM, respectively, whereas the concentration of butyric acid in the wheat and rice bran-treated soil was 25.0mM and 8mM, respectively. The minimal fungicidal concentration (MFC) for all the fungal strains was 40-60mM acetic acid, 20-40mM butyric acid, and 40-60mM mixture of acetic acid: butyric acid (1:1, v/v). Consequently, the efficacy of mixing wheat-bran with soil to control soil diseases was demonstrated.
It seems possible, by use of vegetable oils, to solve the pollution problem caused by the exhaust gas from diesel-engine vehicles. Recently vegetable oils has received considerable attention as an alternative and clean energy source to the foreseeable depletion of world oil supplies. The objective of this study is to experimentally investigate the characteristics of exhaust emissions of a small diesel engine using light oil, rice-bran oil, heated rice-bran oil, rice-bran oil treated with ultrasonic energy. SO$_2$ emission from the pure and the treated rice-bran oils was not detected at speeds hgher than 1,800 rpm while that from the light oil was detected at all the speeds at 4/4 load. NOx emission form these vegetable oils was generally higher compared to that from the light oil for most of the test conditions. tendency opposite to that of NOx emission. The data obtained in this experiment may be applicable for the desist of small diesel engine using the alternative fuels.
