Effects on the Phospholipid Patterns of Liver- and Brain-Tissues of Albino Rats Treated with Sodium Nitrite. (아질산염이 백서의 간 및 뇌조직중 연지질조성에 미치는 영향)

  • The amount of total lipids nad phospholipids in the rat liver and brain after sodium nitrite treatment was measured together with the composition ratio of phospholipids. The results obtained were as follows : 1) The amount of total lipids and phospholipids was decreased significantly and this decrease was more outstanding in the rat brain liver. 2) The amount of phosphatidylcholine, phosphatidylethanolamine, and sphingomyelin was decreased, whereas that of phosphatidylinositol and diphosphatidylglycerol nitrite treatment of 120mg/kg/day concentration brings inhibition of lipid metabolism in the rat liver and brain.

Detection of Cytosolic Phosphatidylethanolamine N -Methyltransferase in Rat Brain

  • Phosphatidylethanolamine N-methyltransferase (PEMT) is known to be a membrane-associated protein. However, cytosolic PEMT was detected when sufficient amounts of exogenous phospholipids were added in the incubation media. The methylation of phospholipids was measured by the incorporation of the $[^3H]-methyl$ group from S-adenosylmethionine and the methylated phospholipids were analyzed by thinlayer chromatography. The essence of the assay condition for the cytosolic enzyme was the inclusion of 200 ${\mu}g$ of each substrate, phosphatidylethanolamine (PE), phosphatidyl N-monomethylethanolamine (PME) and phosphatidyl N,N-dimethylethanolamine (PDE), in the reaction mixture of 100 ${\mu}l$. The subcellular fractionation of brain PEMT activities revealed that approximately 38.1 % for PME, 39.5% for PDE, and 22.4% for PC formation was present in the cytosolic fraction. The general properties of cytosolic PEMT were characterized and compared with those of neuronal nuclei PEMT.

Comparison of Lipid Profiles in Head and Brain Samples of Drosophila Melanogaster Using Electrospray Ionization Mass Spectrometry (ESI-MS)

  • Drosophila melanogaster (fruits fly) is a representative model system widely used in biological studies because its brain function and basic cellular processes are similar to human beings. The whole head of the fly is often used to obtain the key function in brain-related diseases like degenerative brain diseases; however the biomolecular distribution of the head may be slightly different from that of a brain. Herein, lipid profiles of the head and dissected brain samples of Drosophila were studied using electrospray ionization-mass spectrometry (ESI-MS). According to the sample types, the detection of phospholipid ions was suppressed by triacylglycerol (TAG), or the specific phospholipid signals that are absent in the mass spectrum were measured. The lipid distribution was found to be different in the wild-type and the microRNA-14 deficiency model ($miR-14{\Delta}^1$) with abnormal lipid metabolism. A few phospholipids were also profiled by comparison of the head and the brain in two fly model systems. The mass spectra showed that the phospholipid distributions in the $miR-14{\Delta}^1$ model and the wild-type were different, and principal component analysis revealed a correlation between some phospholipids (phosphatidylethanolamine (PE), phosphatidylinositol (PI), and phosphatidylserine (PS)) in $miR-14{\Delta}^1$. The overall results suggested that brain-related lipids should be profiled using fly samples after dissection for more accurate analysis.

Effects of Dietary Docosahexaenoic Acid Levels on the Brain Phospholipids and Serum and Liver Lipid Compositions in Rats (Docosahexaenoic Acid의 수준별 섭취가 흰쥐의 뇌인지질 및 혈청, 간의 지질조성에 미치는 영향)

  • The effects of various dietary docosahexaenoix acid(DHA) levels on the brain phospholipids and serum and liver lipid compositions were studied in rats using DHA concentrated oil and corn oil as a control for 4 weeks. Serum total cholesterol and HDL cholesterol levels tended to be the lowest by adding 20% DHA to corn oil. Serum triglyceride levels significantly decreased by adding 30% DHA. Liver cholesterol and triglyceride levels were apparently decreased in the groups added above 20% DHA, especially, the lowest at adding 30% DHA. Brain weight and phospholipid content were not different among groups. The ratios of arachidonic to linoleic acids in serum and liver phosphatidylcholine(PC) were significantly decreased by adding dietary DHA and showed a flat form above 20% of dietary DHA. DHA levels of serum PC were gradually increased according to dietary DHA level. The fatty acid compositions of the brain PC and phosphatidylethanolamine(PE) did not appear any changes with accordance of the dietary DHA levels. However, compared with those of serum and liver in general, linoleic and arachidonic acid levels were very low. Oleic acids were apparently higher than those in the other tissues. DHA were higher than those in the other tissues rigardless of the dietary DHA, especially in brain PE. The ratios of arachidonic to linoleic acid were not apparent tendency in brain PC and PE. However, the ratios of brain PE were above 2 times higher than those of brain PC. As the results, the hypolipidemic effects of dietary DNA were remarkable in liver. Especially in regard to tendency of liver lipid levels and desaturation indices in serum and liver PC, the effects indicated significantly higher by adding 20-30% DHA to diet(n-6/n-3 ratio, about 4-7). Thus, in this study, these dietary DHA levels seemed to be appropriate, at least in these lipid paramenters.(Korean J Nutrition 34(2) : 132∼140, 2001)

Effects of Dietary Fatty Acids on fatty Acid Pattern in Development Rat Brain Phospholipids - Effects on P/M/S and $\omega$3/$\omega$6 Fatty Acid Ratios -

  • Docosahexaenoic acid(DHA), a $\omega$3 series fatty acid and arachidonic acid(AA). a $\omega$6 series fatty acid were found in relatively high concentrations in the phospholipids(PLs) of cell membranes of nerve tissues, and they can be affected by various factors. The present study examined the effects of dietary $\omega$6 and $\omega$3 fatty acid composition on P/M/S and on $\omega$3/$\omega$6 fatty acid ratios in brain PLs of 2nd generation rats. The expeimental diets consisted of 10% fat(by wt), which were computer- searched mixed oil('M') with P/M/S ratio, 1 : 1.4 : 1 and $\omega$6/$\omega$3 ratio, 6 : 1 and safflower oil('S') poor in $\omega$3 fatty acids. The experimental diets were started 3-4 wks prior to conception. During the lactation period, the feeding mothers were switched 1 wk after birth and provided the pups for 2 wks with milk which had compositions different from that of their natural mother. The same diet as their mothers was provided from weaning to 9 wks of age. The 'M'and 'S' rats were again subdivided into MM, MS, SS, SM rats according to diet which their lactating mothers were fed from the begining of the experiment. The relative percentage of P/M/S fatty acids in brain PLs in all experimental groups converged to a very similar value at 9 wks of age, indicating the existence of a control mechanism for the degree of fatty acids, unsaturation. The $\omega$3/$\omega$6 fatty acid ratios of brain PLs converged to about 1.0 in MM & SM groups and to 0.7 in SS & MS groups, suggesting also the existence of some balance between $\omega$3 and $\omega$6 fatty acids in developing rat brain. The concentrations of $\omega$3 fatty acids, especially DHA, in the SM group were increased and became similar to those in MM group at 9 wks of age. The increase in DHA of brain PLs was counterbalanced b)r a decrease in 22 5$\omega$6. Therefore, the ratios of 22 : 6$\omega$6/22 : 5$\omega$6 were higher in both MM & SM groups than those of SS & MS groups at 9 wak of age. Although dietary $\omega$3 and $\omega$6 fatty acids affected 22 : 6$\omega$S and 22 : 5$\omega$6 contained in rat brain PLs reciprocally, the relative percentage of AA did not appear to be significantly influenced by the diet in all groups at 9 wks of age, suggesting that a mechanism for the maintenance of a certain level of AA in brain PLs exists. In conclusion, the $\omega$3/$\omega$6 fatty acid and 22 : 6$\omega$3/22 : 5$\omega$6 ratios, but not P/M/S ratio, of rat brain PLs were affected by the postnatal dietary changes. Futher studies are required to clarify the mechanism(S) of ensuring a certain level of DHA and of maintaining a similar level of AA in rat brain PLs after. weaning(9 wk) regardless of prenatal and postnatal dietary changes.   • This experiment is carried out to study alteration of phospholipid compoent in liver, kidney, testis, and brain tissues of male rats which are fed selenium added high fat diets for 7 weeks. The phospholipids of these tissues are analyzed by TLC method. The results are as follows. 1. The growth rate and food intake levels are not affected by selenium-added high fat diets. 2. The levels of total cholesterol, total lipid and TBA are lower in selenium-added diets than selenium free diets 3. The total phospholipid levels of each tissue are kidney > brain > testis > liver and the levels of phospholipid component are Le>Sph>Ce>LLe for kidney and liver, Le>Sph>LLe>Ce for testis and brain.

Effect of Ginsenosides from Red Ginseng on the Enzymes of Cellular Signal Transduction System (홍삼 사포닌류(Ginsenosides)의 세포 신호 전달계 효소에 미치는 영향)

  • The present study was conducted to assess the effect of total saponins from Korean red ginseng on the biosynthesis of inositol phospholipids in vivo and also effects on the metabolic enzymes, such as phosphatidylinositol-specific phospholipase C(Pl-PLC) and PI-kinases. The administration of 0.1% saponin solution, 0.1 ml 3 times a day intraperitoneally to 5 mice for 30 days has increased a 23% of the body weight when it compared with a control group. The amounts of 32P-phoschorus radioactivity incorporated into the phosphoinositides from the liver and brain tissues have increased a 310% and 260%, respectively, in the saponin treated mice. The activities of PI-PLC from liver and brain were stimulated in the various amounts by the conditions treated with saponins. The PI-kinases from liver and brain were also activated by saponins, but its effect was lower than that of PI-PLC. From these results, it was confirmed that red ginseng saponins have affected positively not only on the biosynthesis of phosphoinositides but also on the PI-PLC and PI-kinases related to the cellular signal transduction.

Changes of Fatty Acid Compositions in Brain Phospholipids of Developing Chicken Embryos (발생중인 닭의 배자에서 뇌 조직내 인지질의 지방산 조성 변화)

  • This study was to investigate the effects of dietary linoleic acid(18:2\omega6, LA) and aipha-linolenic acid(18:3\omega3. \alpha-LNA) levels on brain development and fatty acid compositions of various lipid classes in the chicken embryo brain tissues. Thirty two ISA Brown layers, 52 weeks-old, were divided into four groups. Birds of each group were given corn-soybean meal based diets added with 1) safflower oil 8%, 2) safflower oil 6% + perilla oil 2%, 3) safflower oil 2% + perilla oil 6%, or 4) perilla oil 8%. Mter 15 days fed the diets. the layers were artificially inseminated to obtain fertile eggs. During the incubation. embryonic brains were sampled at 15th and 21st days. Fatty acid contents were quantitated by using heptadecanoic acid (17:0) as an internal standard. No significant differences in brain weight and in contents of various lipids such as phospholipid. triglyceride, cholesterol. cholesterol ester and free fatty acid in the tissues were found among the dietary groups (P<0.05). The ratios of AA/LA in the brain lipid classes were lowered as the dietary levels of perilla oil were increased. Higher LA was found in phosphatidylcholine(PC) than arachidonic acid (20:4\omega6. AA), meanwhile the level of LA was less than AA in phosphatidylethanolamine(PE). Docosahexaenoic acid(22:6\omega3, DHA) was the* major fatty acid in the tissue and its content in PE was 2.5~3 times higher than in PC. DHA level in the phospholipid reached at a peak (1.7~1.8 mg/brain) in dietary groups added with 6% or 8% perilla oil. suggesting that no more increase in that fatty acid level in the brain tissue could be obtained by consuming more \alpha-LNA, the major precursor of DHA.

  • Fresh ginseng is prone to spoilage due to its high moisture content. For long-term storage, most fresh ginsengs are dried to white ginseng (WG) or steamed for hours at high temperature/pressure and dried to form Korean Red ginseng (KRG). They are further processed for ginseng products when subjected to hot water extraction/concentration under pressure. These WG or KRG preparation processes affect ginsenoside compositions and also other ginseng components, probably during treatments like steaming and drying, to form diverse bioactive phospholipids. It is known that ginseng contains high amounts of gintonin lysophosphatidic acids (LPAs). LPAs are simple lipid-derived growth factors in animals and humans and act as exogenous ligands of six GTP-binding-protein coupled LPA receptor subtypes. LPAs play diverse roles ranging from brain development to hair growth in animals and humans. LPA-mediated signaling pathways involve various GTP-binding proteins to regulate downstream pathways like [Ca2+]i transient induction. Recent studies have shown that gintonin exhibits anti-Alzheimer's disease and antiarthritis effects in vitro and in vivo mediated by gintonin LPAs, the active ingredients of gintonin, a ginseng-derived neurotrophin. However, little is known about how gintonin LPAs are formed in high amounts in ginseng compared to other herbs. This review introduces atypical or non-enzymatic pathways under the conversion of ginseng phospholipids into gintonin LPAs during steaming and extraction/concentration processes, which exert beneficial effects against degenerative diseases, including Alzheimer's disease and arthritis in animals and humans via LPA receptors.

Effect of Dietary Fat on Structure and Function of Mammalian Cell Membrane (식이지방이 생체막 구조와 기능에 미치는 영향)

  • The currently accepted model of membrane structure proposes a dynamic, asymmetric lipid matrix of phospholipids and cholesterol with globular proteins embedded across the membrane to various degrees. Most phospholipids are in the bilayer arrangement and also closely associated with integral membrane proteins or loosely associated with peripheral proteins. Biological functions of membrane, such as membrane-bound enzyme functions and transport systems, are influenced by the membrane physical properties, which are determined by fatty acid composition of phospholipids, polar head group composition and membrane cholesterol content. Polar and non-polar region of the phospholipid molecule can interact, with changes in the conformation of a membrane-associated protein altering either its catalytic activity or the protein's interaction with other membrane proteins. Mammalian dietary studies attempted to change the lipid composition of a few cell membranes have shown comparisons, using essential fatty acid-deficient diets. In recent years, Clandinin and a few other workers have pioneered the study proving the influence of dietary fat fed in a nutritionally complete diet on composition of phospholipid classes of cell membrane. Modulation caused by diet fat was rapid and reversible in phospholipid fatty acyl composition of membranes of cardiac mitochondria, liver cell, brain synaptosome and lymphocytes. These changes were at the same time, accompanied by variety of membrane associated functions controlled by membrane-bound enzymes, tranporter and receptor proteins. The findings suggest the basic concept of the necessity of dietary fatty acid balance if consistency of optimal membrane structural lipid composition is to be maintained, as well as the overall inadequacy of describing the nutritional-biochemical quality of a dietary fat solely by its content of linoleic acid. Furthermore, they give light on the possible application to clinical and preventive medicine.
