Enhanced Resistance to Botrytis cinerea Mediated by Transgenic Expression of the Spider Chitinase Gene AvChit in Arabidopsis

  • The AvChit gene encodes for a chitinase from the spider, Araneus ventricosus. This spider, A. ventricosus, is an abundant species in Korea. Arabidopsis thaliana plants were transformed with the AvChit gene using Agrobacterium tumefaciens. Thirteen transgenic lines expressing the AvChit gene were obtained. Functional expression of the AvChit gene in transgenic Arabidopsis was confirmed by Southern, northern and western blot analysis. The AvChit cDNA was expressed as a 61 kDa polypeptide in baculovirus-infected insect Sf9 cells. AvChit protein extracted from transgenic Arabidopsis exhibited high levels of chitinase activity. Phytopathological tests showed that two transgenic Arabidopsis lines expressing the AvChit gene displayed high levels of resistance to gray mold disease (Botrytis cinerea).

  • Chitinase (EC 3.2.1.14) was isolated from the culture filtrate of Streptomyces sp. M-20 and purified by ammonium sulfate precipitation, DEAE-cellulose ion-exchange chromatography, and Sephadex G-100 gel filtration. No exochitinase activity was found in the culture filtrate. The molecular mass of the purified chitinase was 20 kDa, estimated by a sodium dodecyl sulfate-polyacrylamide gel electrophoresis, and was confirmed by activity staining with Calcofluor White M2R. Chitinase was optimally active at pH of 5.0 and at $30^{\circ}C$. The enzyme was stable from pH 4 to 8, and up to $40^{\circ}C$. Among the metals and inhibitors that were tested, the $Hg^+$, $Hg^{2+}$, and p-chloromercuribenzoic acid completely inhibited the enzyme activity. The chitinase activity was high on colloidal chitin, chitotriose, and chitooligosaccharide. The purified chitinase showed antifungal activity against Botrytis cinerea, and lysozyme activity against the cell wall of Botrytis cinerea.

Occurrence of Multiple Resistant Isolates of Botrytis cinerea to Benzimidazole and N-phenylcarbamate Fungicides (Benzimidazole계 및 N-phenylcarbamate계 살균제에 다중 저항성인 잿빛곰팡이병균의 발생)

  • 1994년, 1995년에 주요 채소 작물에서 분리한 잿빛곰팡이병균(Botrytis cinerea)의 방제 살균제에 대한 저항성을 조사한 결과, 1994년 분리된 713균주 중 610균주(85.6%)가 benzimidazole계에 저항성(Ben\ulcorner)이었고, 249균주(34.9%)는 dicarboximide계 살균제에 저항성(Pro\ulcorner)이었으며, benzimidazole계 및 N-phenyl carbamate계 두약제 모두에 저항성인 균주는 분리되지 않았다. 1995년에는 현재까지 520균주를 분리하였는데, 그중 Ben\ulcorner이 317균주(61%)이며 Pro\ulcorner은 214균주(41.2%)이었고, Ben\ulcorner이며 NPC\ulcorner인 균주(Ben\ulcorner+NPC\ulcorner)는 15균주(2.9%)로 나타났다. 이러한 저항성 균주의 최소 억제 농도는 carbendazim 1,000 $\mu\textrm{g}$/ml 이상, diethofencarb 1,000 $\mu\textrm{g}$/ml 이상이었다. 또 Ben\ulcorner+NPC\ulcorner인 15균주 중 3균주는 dicarboximide계 살균제인 procymidone에도 저항성이었으며, 한 균주를 제외한 모든 Pro\ulcorner 균주는 Ben\ulcorner로 나타났다.

Induction of Prochloraz-Resistant Isolates of Botrytis cinerea in Vitro and Their Biological Properties (Prochloraz에 저항성인 잿빛곰팡이균(Botrytis cinerea)의 유기 및 이들 균의 생물학적 특성)

  • 1994~1995년 사이에 대전, 공주, 논산, 부여와 김해 등지에서 채집한 병든 딸기, 토마토, 오이에서 총 1,504 균주의 잿빛곰팡이병균을 분리하여 10 $\mu\textrm{g}$/ml의 약제를 넣은 PDA 배지에서 배양하면서 약제반응을 조사한 결과, benomyl에 대하여는 1,147(76.3%)균주, procymidone에 대하여는 614(40.8%)균주, prochloraz에 대하여는 189(12.6%)균주가 저항성으로 나타났다. UV 처리 및 약제 배지에서의 누대배양을 통하여 prochloraz저항성 균주를 선발하였는데, 이들의 생육 억제 최소 농도(MIC)는 8 $\mu\textrm{g}$/ml 이상이었고, 감수성균의 MIC는 2 $\mu\textrm{g}$/ml 이하였다. 병원력, 포자 발아율, 균핵형성과 같은 적응력과 관련된 생물학적 특성 비교에서 저항성 균주들은 감수성 균주보다 열등하거나 비슷한 정도로 나타났다.

Instability of Dichlofluanid Resistance in Dichlofluanid-Resistant Botrytis cinerea Isolates and Comparison of Their Competitive Ability with Dichlofluanid-Sensitive Isolates (Dichlofluanid 저항성 잿빛곰팡이병균(Botrytis cinerea)의 저항성 불안정 및 감수성균과의 경합 능력 비교)

  • 대전, 공주, 논산, 부여와 김해 등에서 병든 딸기, 토마토, 오이에서 총 992균주의 잿빛곰팡이병균을 분리하여 이들 균주로부터 dichlofluanid 저항성 5균주, 감수성 5균주를 임의로 선발하였다. 분리된 균의 dichlofluanid 저항성을 조사한 결과 105균주(10.6%)가 저항성이었다. 저항성균 균사의 최소 억제 농도(minimum inhibitory concentration)는 100~500$\mu\textrm{g}$/ml 이상이었으나 감수성균은 10 $\mu\textrm{g}$/ml 미만이었다. Dichlofluanid 저항성의 안정성 조사를 위하여 35$^{\circ}C$고온 처리 및 계대 배양할 때 조사된 저항성 5균주 중 3균주는 저항성이 감소하거나 상실되었다. 저온 저장 및 온실에 장기 보관할 때는 조사된 저항성 균주 모두가 저항성이 감소하거나 완전히 상실되는 것으로 나타났다. 이러한 결과는 이 약제에 대한 저항성이 불안정하다는 것을 나타낸다. 오이에 감수성균 및 저항성균을 혼합 접종한 후 재분리할 때 시간이 지날수록 감수성균이 더 많이 분리되어 저항성균의 경합 능력이 감수성 균주에 비하여 열등하였다.

  • The rose is one of the most important commercial cut flowers in the world. Recently stem blight symptoms have often found in hydroponics and soil cultured roses. These symptoms are appeared by four diseases; gray mold caused by Botrytis cinerea, common canker by Coniothyrium fuckelii, anthracnose by Colletotrichum gloeosporioides, and Pythium root rot by Pythium sp. Each symptom and its causal pathogen is described.

  • In order to confirm the possibility of UV utilization as a means for disease protection in grapevine the stilbene contents and the disease incidence in UV irradiated leaves were investigated. UV irradiation significantly increased the contents of resveratrol, piceatannol and piceid, in them, resveratrol and piceid showed in vitro inhibition of spore germination and mycelium growth of Botrytis cinerea. The accumulation of both the stilbenes in UV irradiated leaves seems to be enough to inhibit the B. cinerea growth, since the formation of necrotic spot on the leaves was considerably inhibited when they were inoculated with the conidial spores of B. cinerea. However the stilbenes were accumulated only in the leaves exposed directly to UV showing a limited translocation ability of the compounds. Thus it would be necessary to develop a method to evenly irradiate the entire crown of plant with UV in order to expect to protect them from disease by UV irradiation.

Effect of Gamma Irradiation on Botrytis cinerea Causing Gray Mold and Cut Chrysanthemum Flowers

  • Gray mold caused by Botrytis cinerea is one of the most important postharvest fungal pathogens of cut flowers. Here, gamma irradiation, an alternative for phytosanitary purposes, and sodium dichloroisocyanurate (NaDCC) were used to control B. cinerea in a cut chrysanthemum (Chrysanthemum morifolium Ramat.) cultivar, 'Baekma', one of the cultivars susceptible to B. cinerea. Spore germination and mycelium growth of B. cinerea were inhibited by gamma irradiation in an inversely dose-dependent manner. A dose of 4 kGy completely inhibited the mycelium growth of B. cinerea. A significant change in flower quality (physical properties) on chrysanthemum was shown from gamma irradiation at over 0.2 kGy (p<0.05). Therefore, in this study, the integration of gamma ray (below 0.2 kGy) and NaDCC, an eco-friendly form of chlorine, was investigated to control the disease with low dose of gamma irradiation dose. Interestingly, the gamma irradiated flowers showed more disease severity than the non-irradiated flowers. The combined treatment of gamma irradiation and NaDCC does not affect the severity of the fungal disease, whereas only 70 ppm of NaDCC treatment showed a significantly reduced severity. These results suggest that only chlorination treatment can be applied to control B. cinerea in cut chrysanthemum flowers.

Chemical Responses and Fitness-Related Characteristics of Dichlofluanid-Resistant and -Sensitive Botrytis cinerea Isolates (Dichlofluanid 저항성 및 감수성 잿빛곰팡이병균(Botryits cinerea)의 약제 반응과 생태 적응력과 관련된 특성)

  • Nine hundred and ninety-two isolates of Botrytis cinerea were obtained from infected strawberries, tomatoes and cucumbers in Taejon, Gongju, Puyo, Nonsan and Kimhae in Korea. Six hundred forty-two (64.7%) isolates were benomyl resistant (BR), 245 (24.7%) were procymidone resistant (PR), and 105 (10.6%) were dichlofluanid resistant (DR). In the minimum inhibitory concentration (MIC) test, DR isolates showed mycelial growth on the PDA incorporated with 100 or 500 $\mu\textrm{g}$/ml of dichlofluanid, while dichlofluanid sensitive (DS) isolates did not grow on the PDA incorporated even with 10 $\mu\textrm{g}$/ml of dichlofluanid. Chemical concentrations for inhibition of spore germination were much lower than those for inhibition of mycelial growth. IC50 values, the effective concentrations for 50% inhibition of spore germination, for DR were 0.11~0.29 $\mu\textrm{g}$/ml, whereas they were 0.04~0.09 $\mu\textrm{g}$/ml for DS isolates. In comparison of fitness-related characteristics such as virulence, sclerotial formation, and sporulation, DR isolates were inferior to DS isolates. However, mycelial growth was little different between DR isolates and DS isolates.

Bacillus subtilis S1-0210 as a Biocontrol Agent against Botrytis cinerea in Strawberries

  • Bacillus subtilis S1-0210 was selected as a biological agent against Botrytis cinerea in strawberry. The isolate inhibited mycelial growth of B. cinerea in vitro tests. A wettable powder formulation of B. subtilis S1-0210 significantly reduced infection rates with lower than 5%, compared with higher than 70% of infection rates in untreated control. The formulation showed 85 to 89% control efficacies of gray mold incidences on fruits of strawberry in pots. Pre-treatment of the agent was more effective in controlling gray mold on fruits and leaves than post-treatment at the early stage of disease development. The formulation also showed 70% control efficacy of gray mold incidence on fruits of strawberry in a field trial. The results indicate that B. subtilis S1-0210 in the wettable powder formulation may be a potential biocontrol agent to control gray mold on strawberry.
